Abstract
Die vorliegende Erfindung betrifft ein neues Verfahren zur Herstellung von beta-cyclosubstituierten Ethylaminen der Formel I Ar - CH tief 2 - CH tief 2 - NH tief 2, in der Ar ein aromatisches, heterocyclisches oder nichtheterocyclisches Radikal wie Thienyl, Furfuryl, Pyridyl, Phenyl oder Naphthyl, gegebenenfalls durch Gruppen wie Halogen, Nitro, Amino, Cyano, Carboxy, Alkyl, Halogenalkyl, Alkoxy, Halogenalkoxy oder Phenyl mono- oder polysubstituiert, darstellt, gekennzeichnet dadurch, dass man ein Derivat der Formel II, in der X und Y, gleich oder verschieden sind und ein Alkyl-, Aryl-, Alkoxy-, Aryloxy-, Dialkylamino- oder Diarylaminoradikal darstellen, mit einer Carbonylverbindung der Formel III Ar - CHO, in der Ar wie in der Formel I definiert ist, kondensiert, dann mit einer Base der Formel B hoch - M hoch + umsetzt, um zu einem Carbanion der Formel V zu gelangen, das sich in ein Derivat der Formel VI umwandelt und nach weiterer Behandlung in eine Verbindung der Formel VII uebergeht, die ihrerseits mit Hilfe eines Reduktionsmittels eine Verbindung der Formel VIII ergibt, die dann unter Einwirkung von Saeure zum Derivat der Formel I fuehrt.

Verfahren zur Herstellung von ß-cycloaubstituierten Sthylaminen
Die vorliegende Erfindung betrifft ein neues Verfahren zur Herstellung von ß-cyclosubstituierten Sthylaminen der allgemeinen Formel I
Ar - CH2 - GH2 - MH2
in der Ar ein aromatisches, heterocyclisches oder niohtheterocycliaches Radikal wie Thienyl, Furfuryl, Pyridyl, Phenyl oder ITaphthyl, gegebenenfalls durch Gruppen wie Halogen, Itfitro, Amino, Cyano, Carboxy, Alkyl, Halogen-Alkyl, Alkoxy, Halogen-Alkoay oder Phenyl mono- oder polysubstituiert, darstellt.
3s ist eine große Anzahl von entsprechenden Derivaten der Formel I bekannt, die als Zwischenprodukte bei der Herstellung von nützlichen Verbindungen in der chemischen wie pharmazeutischen Industrie verwendet werden·
Daher kann man beispielsweise unter den nach dem neuen Herstellungsverfahren erhaltenen Derivaten die (Ihienyl-2)- und (Thienyl-3)-2-ethylamine erwähnen, die nach bekannten und einfach durchzuführenden Methoden (vgl. 3· GROM)WITZ et Ξ. SAHDBERG, Arkev-Kemi, ^2, 217 (1970) zu den entsprechenden Tetrahydro-4,5,6,7-thieno-^~3,2-c 7- und ^2,3-c 7-pyridinen führen, von denen bestimmte Derivate den Gegenstand mehrerer Patente bilden, die der Anmelderin für die therapeutische

Verwendung und/oder die Herstellung der Derivate erteilt wurden (3S-PS 73 03 503, S1R-PS 75 03 968, ift-PS 75 20 241, 2R-PS 75 23 786, ER-PS 75 24 486, SR-PS 76 00 003, PS-PS 77 21 517).
Ziel der Erfindung ist die Bereitstellung eines im Hinblick auf den früheren Stand der Technik einfacheren und weniger kostspieligen Verfahrens zur Herstellung von Verbindungen der lOrmel I.
Der Erfindung liegt die Aufgabe zugrunde, leicht zugängige Ausgangs- und Reaktionskomponenten für die Herstellung von ß-cyclosubstituierten JEthylaminen aufzufinden·
In Übereinstimmung mit .em erfindungsgemäßen Verfahren zur Hera teilung der Derivate der Formel I:
a) kondensiert man ein Derivat der Pormel II

Diese Reaktion wird im allgemeinen bei einer Temperatur zwischen -78 0C und +150 0C durchgeführt, wobei sie spezifisch in Abhängigkeit von der verwendeten Base gewählt wird und eher im höheren Bereich liegt, insbesondere bei der Realisierung der Phase c)·
d) Die Verbindung der Formel VII wird unter Einwirkung eines Reduktionsmittels in die Verbindung der Formel VIII

Das Verfahren kann vorteilhafterweise wie folgt durchgeführt werden:
a) Die Organo-Phosphorverbindungen der Formel II, einfach erhältlich nach bekannten Herstellungsverfahren, wie beschrieben von C-POPOPP et Coil· (J.Org* Chenu .28, 2898 (1963)» können in Abwesenheit von Lösungsmittel .und Katalysator mit Carbonylderivat en der Pormel III zur Reaktion gebracht werden} das im Verlauf der Reaktion gebildete Wasser wird gegen Ende der Operation durch geeignete Mittel entfernt· Die Kondensation kann vorteilhaft erweise in einem Lösungsmittel wie einem aromatischen Kohlenwasserstoff, z· B« Toluol, oder einem Alkohol, wie Ethanol, durchgeführt v/erden, mittels derer es möglich ist, das Wasser durch azeotrope Destillation zu entfernen« Die Reaktion kann ebenfalls vorteilhaft realisiert werden (hinsichtlich der Geschwindigkeit), in dem man in Anwesenheit einer katalytischen Menge an

Mineralsäure oder organischer Säure, ζ. Β. para-Toluolsulfonsäure, arbeitet. Die Temperatur bei der diese Umwandlung stattfindet, iat variabel, aber sie liegt im allgemeinen zwischen 20 0O und 120 0C
b) Die in dieser Stufe eingesetzte Baae ΒθΒβ kann ein Alkalimetallhydrid sein, insbesondere die Hydride von Natrium, Lithium oder Potassium ein Amid oder Alkylamid, insbesondere Dialkylamid, eines Alkalimetalls, trie Lithiumdiisopropylamid, eine Organometallverbindung, insbesondere die Organolithiumverbindungen wie n-Butyllithium oder die Organonatrium- oder die Organomagnesiumverbindungen« Man kann auch auf die Alkali- oder Erdalkali-, metallalkoholate, wie die Slethylate von Natrium, Lithium, Kalium, und Magnesium, auf Kalium-tert-Butylat oder auf Hatrium-tert-Amylat zurückgreifen· Man kann auch die Alkali- oder Erdalkalimetallhydroxide, wie die Hydroxide von Natrium, Lithium, Kalium und Magnesium, verwenden* Im allgemeinen verwendet man ein stöchiometrisches Äquivalent der Base BM, sogar einen leichten Überschuß, z. B. 10 % im Verhältnis zur Äquivalenz« Aber es ist ebenfalls möglich,- geringere" mengen an Base einzusetzen, sogar deutlich geringere, als es das stSchiometrische Verhältnis aussagt· Man arbeitet im allgemeinen bei einer Temperatur zwischen -78 0C und +150 0C, wobei diese Temperatur spezifisch in Abhängigkeit von der Base B% gewählt wird und eher im höheren Bereich liegt, insbesondere bei der Realisierung der Phase c).
Die bevorzugten Lösungsmittel sind die linearen oder cyclischen Ether" wie Tetrahydrofuran, die Kohlenwasserstoffe, insbesondere die aromatischen, wie Benzol, Toluol, Xylol, die Alkohole, die Amide, insbesondere Dimethylformamid und die Sulfoxide, insbesondere Dimethylaulfoxid·

Es kann ebenfalls vorteilhaft sein, vor allem, wenn can Metallhydroxide verwendet, in einem Zweiphasensystem 2u arbeiten (Wasser + Lösungsmittel, wie halogenierte Lösungsmittel, z· B« Dichlormethan, oder aromatische Kohlenwasserstoffe, wie Benzol, Toluol, Xylol), in Anwesenheit eines Phasen-Übertragungskatalysators, insbesondere eines quaternären Ammoniumsalaes, wie tetra-n-Butylammoniumjodid oder eines Phosphoniumsalzes. Die üblichen Methoden gestalten die Isolierung der Verbindung der Formel V·
c) Die Reduktion des Derivats der Formel V wird vorteilhafterweise mit Hilfe eines gemischten Alkalimetallhydrids durchgeführt, insbesondere eines Borhydrids, wie Hatrium- oder Kalium-Borhydrid· Die Reduktion findet in einem inerten Lösungsmittel statt, z· B« einem Ether, wie Tetrahydrofuran oder Dioxan oder auch in einem Alkohol, wie Methanol oder Ethanol· Man kann diese Reduktion auch über den Umweg der katalytischen Hydrierung in homogener oder heterogener Phase unter den für diese Umsetzung bekannten Bedingungen durchführen·
d) Die säurekatälysierte Spaltung der Phosphor-Stickstoff-Bindung des Derivats VI kann mittels einer Mineralsäure, wie einer Halogenwasserstoffsäure, z· B9 Salzsäure oder Bromwasserstoffsäure erfolgen, jedoch ebenfalls durch eine organische Säure, insbesondere eine starke Säure, wie eine Sulfonsäure, z« B. Benzolsulfonsäure oder para-Toluol-3ulfonsäure bewirkt werden* Die bevorzugten Lösungsmittel sind Ether, insbesondere cyclische Ether, wie Tetrahydrofuran oder Dioxan, Alkohole wie Methanol oder Ethanol, Amide, insbesondere Dimethylformamid oder Sulfoxide, insbesondere Dimethylsulfoxid. Es ist möglich, in diesen Lösungsmitteln unter Abwesenheit von Wasser zu arbeiten,

aber auch in Kischungen mit Wasser, wobei verschiedene Mengenverhältnisse eingesetzt werden können· Schließlich ist es auch möglich, nur in Wasser allein zu arbeiten·
Im allgemeinen verwendet man zwei stöchiometrische Äquivalente an Säure· Man arbeitet generell bei Temperaturen zwischen 0 0C und 100 0C, insbesondere zwischen 30 0C und 70 °C· Die auf diese Weise erhaltenen Verbindungen der Formel I können dann nach üblichen Methoden isoliert und gereinigt werden» um diese Verfahrensschritte durchzuführen, kann es vorteilhaft sein, die freien Verbindungen der Formel I in ihre Salze zu überführen, z. B· in ihre Säureadditionsaalze durch Reaktion mit organischen oder Mineral säuren· Ausgehend von den Salzen, kann man die Verbindungen der Formel I nach bekannten Methoden freisetzen.
Die Erfindung umfaßt ebenfalls die in den verschiedenen Verfahrensstufen erhaltenen Zwischenprodukte·

Beispiel 1 - Herstellung von (thienyl-2)-2-ethylamin- * hYdrochlorid
Stufe a - Diethyl-li(thenyliden~2)-aminoraethylpho3phonat
- P - CH2 - H = CH -I^s
Zu 16,7 g (0,1 Hol) CDiethyl)-aminomethylphosphonat in 200 ml Ethanol (absolut) fügt man 11,2 g (0,1 Hol) Thenaldehyd-2 und hält die Mischung 30 Minuten lang unter Rückfluß. Das im Verlauf der Reaktion gebildete Wasser wird dann durch azeotrope Destillation entfernt. ITach vollständiger Verdampfung des Lösungsmittels erhält man 28 g (ca· 100 %) eines gelben Öls, rein durch (CPL, CCM und CPG-)· IR (PiIm) C = Ii 1645 cm"1

Zu einer Suspension von 11,2 g (0,1 MoI) Kalium-tert.-Butylat in 180 ml Tetrahydrofuran (THP),' fügt man tropfenweise eine Lösung von 27,9 g (0,1 Mol) (Diethyl)-U(thenyliden-2)_ aminomethylphosphonat in 40 ml THF· Im Verlauf der Addition erhöht sich die Temperatur von 20 0G auf 35 0C· Am Ende der Addition hält man die Temperatur noch für 30 Minuten bei 40 - 45 °C und gießt die Reaktionsmischung dann in 400 ml gesättigte Ammoniumchlorid-Lösung. Die wäßrige Phase wird mit Isopropylether extrahiert, die vereinigten etherischen Phasen werden mit gesättigter Kochsalzlösung gewaschen, über Natriumsulfat getrocknet und eingedampft, um 20,9 g (75 %) des Referenz-Produktes in Form eines gelben Öls zu liefern·

Wie wie oben erhaltenen 20,9 g (0,075 Mol) Diethyl-fl-(thienyl-2)-II-vinylphosphoramidat werden zn einer Lösung Ton 5,1 g (0,075 Mol) Natriumborhydrid in 200 ml Ethanol gegeben·
Während der Addition erhöht sich die Temperatur und stabilisiert sich dann bei 30 0C. Nach 2 Stunden zusätzlichem Rühren wird die Temperatur der Reaktionsmiachung noch 1 Stunde bei 45 - 50 0C gehalten, dann wird das Ethanol verdampft und der Rückstand in einer Mischung von Isopropylether und v/aeser aufgenommen·
Die wäßrige Phase wird mehrmals mit Isopropylether extrahiert und die vereinigten organischen Phasen werden über natriumsulfat getrocknet and eingedampft, um 21 g (ca· 75 % in bezug auf das Aminomethylphoaphonat als Ausgangsstoff) des Referenzproduktes in Form eines gelben Öls zu liefern«

Stufe e - (Thienyl-2)-2-ethylaiaid-Hydrochlorid
Bine gut gerührte Mischung von wie oben erhaltenen 21 g Phosphoramidat und 100 ml einer wäßrigen lösung von 6 Bf Salzsäure wird 11/2 Std· bei 80 - 85 0C gehalten. Uach Abkühlung und Extraktion der Reaktionamis chung mit 30 ml Methylenchlorid wird die wäßrige Phase isoliert und mit wäßriger Soda-Löaung basisch gestellt, anschließend wird sie mit Isopropylether extrahiert.
Die nach Tircknung über natriumsulfat isolierte et her is ehe Phase wird, gegebenenfalls mittels Waschflasche, mit gasförmigem Chlorwasserstoff versetzt, wodurch sich ein.Kristallnieder schlag bildet, den man abfiltriert und reinigt, indem man ihn in Ethanol löst und durch Zusatz von Isopropylether wieder ausfällt. Man erhält auf diese Weise 8,9 g (Gesamtausbeutei 54 %) (Thienyl-2)-2-ethylamin-Hydrochlorid in Form weißer Kristallei

Za einer lösung von 25,5 g (0,1 Mol) (Diethyl)-li-benzylidenaminomethylphosphonat in 200 ml Tetrahydrofuran, fügt man bei 20 0C tropfenweise 35,7 ml (0,1 Mol) einer 2,8 M Lösung von n-Butyllithium in Cyclohexane Gegen Ende der Addition hält man die Reaktionsmischung noch während 30 Minuten bei 35 0C, gießt sie dann in 1 1 gesättigte, wäßrige Ammoniumchloridlösung und extrahiert schließlich mit Isopropylether»
Die mit Wasser gewaschene, über Natriumsulfat getrocknete und eingedampfte etherische Phase liefert ein gelbee öl, das nach Verreihen mit Hexan fest wird.
Nach Filtration und Umlcriat all isation aus Hexan werden die erhaltenen Kristalle mit einer Mischung aus Hexan/Isopropylether (90/10) gewaschen und anschließend im Vakuum bei umgebungstemperatur getrocknet. Man erhält auf diese Weise 17,9 g (70 %) (Diethyl)-ß-phenyl-K-vinyl-pho3phoramidat<

Man erhält auf diese Weise nach Filtration und Trocknung im Vakuum 6,7 g 2-Furyl-2-ethylaniin-» Hydrochlorid (Ausbeute 45 % in bezug auf (Diethyl)-aminomethylphosphonat) in Form weißer Kristalle.

Zu 16,7 g (0,1 Mol) (Diethyl)-aminomethylphosphonat in einer Lösung von 200 ml Toluol fügt man bei umgebungstemperatur tropfenweise 14 g (0,1 Mol) 2-Chlorbenzaldehyd· Am Ende der Zugabe rührt man noch weitere 30 Minuten» Das im Laufe der Reaktion gebildete Wasser wird durch Dekantieren entfernt« Die Toluol-Phase wird mit 50 ml wäßriger gesättigter Hatriumchloridlösung gewaschen, über Natriumsulfat getrocknet und eingedampft· Man erhält 25 g (Ausbeute 100 %) (Diethyl)-H-ortho-chlorbenzyliden-aminomethylphosphonat in Form eines gelben Öls, einen Fleck bildend in CCM (Kieselerde-Platte, Elutionsmittel AcOBt, rf 0,45)·

6. Verfahren nach Punkt 5f gekennzeichnet dadurch, daß die Reaktion bei einer in Abhängigkeit von der Base ge-7/ählten Temperatur durchgeführt wird, die eher im höheren Bereich liegt«
7» Verfahren nach Punkt 2 und 3, gekennzeichnet dadurch, daß die Reaktion in einem organischen Lösungsmittel wie linearen oder cyclischen Ethern, aromatischen Kohlenwasserstoffen, Alkoholen, Amiden oder Sulfoxiden durchgeführt wird.
8. Verfahren nach Punkt 7, gekennzeichnet dadurch, dalB die Reaktion in einem organischen Lösungsmittel durchgeführt wird, ausgewählt unter ietrahydrofuran, Benzol, Toluol, Xylol, Ethanol, Dimethylformamid und Dimethylsulfoxid·
9. Verfahren nach Punkt 1, gekennzeichnet dadurch, daß die Reduktion des Derivats der Formel VII mittels eines Alkalimetall-Borhydride, insbesondere Uatriumborhydrid oder Ksliiunborhydrid durchgeführt wird.
