Abstract
Περιγράφονται πλασμίδια έκφρασης και μέθοδοι που χρησιμοποιούν σύστημα έκφρασης E. coli για την υψηλής κλίμακας παραγωγή FGF18. Τα πλασμίδια χρησιμοποιούν την κωδικοποιητική αλληλουχία της FGF18 με συγκεκριμένες αλλαγές στα νουκλεοτίδια ώστε να βελτιστοποιηθούν τα κωδικόνια και η δευτεροταγής δομή του mRNA για μετάφραση στο E. coli. Χρησιμοποιώντας τα πλασμίδια έκφρασης, το γονίδιο FGF18 παρήχθη στο E. coli σε επίπεδο μεγαλύτερο του 1 g/L σε αντιδραστήρα διαλείποντος έργου. Περιλαμβάνονται επίσης στελέχη E. coli με ανεπάρκεια στην OmpT, καθώς και στελέχη αρνητικά σε OmpT και fhuA μετασχηματισμένα με πλασμίδια έκφρασης FGF18.
