Abstract

Η παρούσα εφεύρεση αναφέρεται σε μία απομονωμένη πολυνουκλεοτιδική αλληλουχία η οποία κωδικοποιεί ένα χιμαιρικό TNFα, περιλαμβάνουσα μία πρώτη αλληλουχία νουκλεοτιδίων η οποία κωδικοποιεί έναν τομέα ή υποτομέα ενός προσδέματος παράγοντα νεκρώσεως όγκων εκτός του TNFα, όπου ο κωδικοποιούμενος τομέας ή υποτομέας αντικαθιστά μία θέση αποκοπής του αυτοφυούς TNFα και μία δεύτερη αλληλουχία νουκλεοτιδίων η οποία κωδικοποιεί έναν τομέα ή υποτομέα του αυτοφυούς TNFα ο οποίος συνδέεται με έναν υποδοχέα TNFα. Ο κωδικοποιούμενος χιμαιρικός TNFα είναι σημαντικά λιγότερο ευαίσθητος σε αποκοπή από την κυτταρική επιφάνεια και, ως αποτέλεσμα, μπορεί να αυξήσει τη συγκέντρωση ενός προσδέματος ικανού να συνδέεται με έναν υποδοχέα TNFα επί της επιφανείας ενός κυττάρου. Ο χιμαιρικός TNFα είναι συνεπώς χρήσιμος σε μεθόδους επαγωγής αποπτώσεως ενός κυττάρου εκφράζοντος έναν υποδοχέα TNFα, επαγωγής της ενεργοποιήσεως ενός κυττάρου του ανοσοποιητικού συστήματος και αγωγής κυττάρων νεοπλασίας, δι' εισαγωγής εντός του ενδιαφέροντος κυττάρου μίας απομονωμένης πολυνουκλεοτιδικής αλληλουχίας η οποία κωδικοποιεί ένα χιμαιρικό TNFα ο οποίος εκφράζεται επί της επιφανείας του κυττάρου.
