Abstract

Η παρούσα εφεύρεση απευθύνεται γενικώς σε ανασυνδυασμένες μεθόδους για παρασκευή ενός επιθυμητού πεπτιδίου. Αυτή(ες) η(οι) μέθοδος(οι) παράγει(ουν) ένα πολυπεπτιδικό προϊόν που περιέχει μειωμένα επίπεδα ισομορφικών προσμείξεων αυτού. Συγκεκριμένα, η παρούσα εφεύρεση απευθύνεται σε (1) μία ανασυνδυασμένη μέθοδο για παρασκευή αυξητικής ορμόνης με μειωμένες ισομορφικές προσμείξεις αυτής και (2) μία ανασυνδυασμένη μέθοδο για παρασκευή ανταγωνιστή αυξητικής ορμόνης, όπως pegvisomant, και της ενδιάμεσης του πρωτεΐνης, που επίσης έχει μειωμένες προσμείξεις αυτού. Πιο συγκεκριμένα οι ισομορφικές προσμείξεις που μειώνονται με την μέθοδο της παρούσας εφεύρεσης είναι οι τρισουλφιδικές και des-phe ισομορφικές προσμείξεις της αυξητικής ορμόνης και του ανταγωνιστή αυξητικής ορμόνης (ή του ενδιάμεσου του) αντιστοίχως.
