Abstract

Παρουσιάζονται μέθοδοι για την απομόνωση και την κρυοσυντήρηση προγόνων από ανθρώπινο ήπαρ, οι οποίες περιλαμβάνουν την κατεργασία ιστού ανθρώπινου ήπατος ώστε να παρασχεθεί ένα ουσιαστικά μοναδικό κυτταρικό εναιώρημα, το οποίο περιλαμβάνει προγόνους και μη προγόνους μιας ή περισσότερων κυτταρικών γραμμών που υπάρχουν στο ανθρώπινο ήπαρ· έκθεση του εναιωρήματος σε ένα βήμα λέπτυνσης, που μειώνει ουσιαστικά τον αριθμό των μη προγόνων στο εναιώρημα και παρέχει ένα λεπτυσμένο εναιώρημα, εμπλουτισμένο με προγόνους που παρουσιάζουν έναν ή περισσότερους δείκτες που σχετίζονται με τουλάχιστον μια από τις μια ή περισσότερες κυτταρικές γραμμές∙ και επιλογή από το αναφερθέν λεπτυσμένο εναιώρημα εκείνων των κυττάρων, τα οποία τα ίδια, οι απόγονοι τους ή πιο ώριμες μορφές αυτών εκφράζουν έναν ή περισσότερους δείκτες που σχετίζονται με τουλάχιστον μια από τις μια ή περισσότερες κυτταρικές γραμμές. Μεταξύ αυτών των δεικτών είναι οι CD14, CD34, CD38, CD45 και ICAM. Οι ηπατικοί πρόγονοι χαρακτηρίζονται από το ότι έχουν διάμετρο 6-15 μ, είναι διπλοειδείς, με γλυκοφορίνη A-, CD45-, AFP+++, ALB+, ICAM+ και με υποπληθυσμούς που διαφοροποιούνται ως προς την έκφραση των CD 14+, CD34++, CD117+. Αυτοί οι προγονικοί υποπληθυσμοί έχουν χαρακτηριστικά τα οποία είναι αναμενόμενα για τα κύτταρα που είναι ιδιαίτερα χρήσιμα στις ηπατικές κυτταρικές και γονιδιακές θεραπείες και για την εδραίωση βιοτεχνητών οργάνων.
