Abstract

Description
本发明涉及新颖的、稳定的粉末状制剂,其中包含脂溶性活性成分,本发明还涉及制备这些制剂的方法。本发明的新颖的组合物可被用作为食物、饮料、动物饲料、化妆品或药物的添加剂,以将所述的脂溶性成分包括进此类应用形式中。
更具体地,本发明涉及稳定的粉末状制剂,其中包含:处于奶蛋白组合物基质(matrix)中的脂溶性活性成分。
本文中使用的术语“奶蛋白”指奶(特别是牛奶)中发现的任何天然蛋白质,以及从中获得的部分水解的蛋白质,例如酪蛋白酸盐/酯,以及水解的或部分水解的酪蛋白酸盐/酯,乳球蛋白(lactoglobuline),乳清蛋白或奶粉或其水解产物,其可通过已知方法获得(例如,通过对蛋白质进行酸处理或碱处理,或通过用蛋白酶进行酶处理),具有例如,多达25%,多达15%或多达10%的水解度(DH)。特别优选的是酪蛋白酸盐/酯(例如酪蛋白酸钠)以及其水解产物或部分水解的酪蛋白酸盐/酯,乳清蛋白分离物或经水解的乳清蛋白,其中蛋白质含量超过80%(按重量计),水解度多达25%、多达15%或多达10%。
在本发明的一个方面,新颖的制剂可以额外含有碳水化合物或碳水化合物衍生物,其作为保护性包裹材料发挥作用,例如糊精、糖浆、果胶、卡拉胶、淀粉和淀粉衍生物、纤维素或纤维素衍生物(例如羧甲基纤维素)、植物蛋白或部分水解的植物蛋白(其作为保护性胶体发挥作用,例如,从马铃薯蛋白、大豆蛋白、小麦蛋白、豌豆蛋白、水稻蛋白或羽扁豆蛋白中获得的)。在本发明的一个特定方面,使用下述植物蛋白水解产物,其中,至少80%都具有小于2500道尔顿的分子量分布。此类额外的碳水化合物或碳水化合物衍生物或蛋白质可以以2-20wt.%的量(基于干制剂中碳水化合物或蛋白质的总量)存在于本发明的制剂中。使用具有碳水化合物或碳水化合物衍生物或植物蛋白或植物蛋白水解产物的混合物可以降低对制剂进行热处理过程中活性成分的损失,以及提高活性成分的稳定性。术语“奶蛋白组合物”由此包含:奶蛋白和/或部分水解的奶蛋白,以及它们与碳水化合物或碳水化合物衍生物和/或植物蛋白或水解植物蛋白的混合物。
本文中使用的术语“脂溶性活性成分”指可溶于脂类并且不溶于水或很少溶于水的任何具有生理活性的成分。此类脂溶性活性成分的例子是:脂溶性维生素,即,维生素A、D、E和K及其衍生物,例如维生素A的酯,例如乙酸维生素A酯和棕榈酸维生素A酯,以及维生素E的酯,例如乙酸生育酚酯;类胡萝卜素和类胡萝卜素衍生物,例如,α-或β-胡萝卜素、8’-阿朴-β-胡萝卜素醛、8’-阿朴-β-胡萝卜素酸酯(例如,乙酯)、角黄素、虾青素、虾青素酯、番茄红素、叶黄素、玉米黄质或藏花酸及其衍生物;多不饱和脂肪酸,例如,二十碳五烯酸、二十二碳六烯酸、花生四烯酸和γ-亚麻酸和/或乙酯。此类脂溶性成分可以以多达55%(按重量计)的量存在于根据本发明的制剂中。
在本发明的另一个方面,新颖的制剂额外含有还原糖,例如,葡萄糖、果糖、木糖,或还原糖的衍生物,例如脱氧糖,例如2-脱氧-D-核糖或鼠李糖,或氨基糖,例如氨基葡萄糖,例如2-氨基-2-脱氧-D-葡萄糖。所述的糖可以以多达15%(按重量计)的量存在,优选为5-15%(按重量计),特别是5-9%(按重量计),上述百分比基于制剂的干物质。可对此类制剂进行热处理,以在Maillard类型的反应中使糖与蛋白质交联。交联还可通过用酶(例如,转谷氨酰胺酶)进行处理来获得。经过交联的制剂被发现能展示出增加的稳定性。
本发明的特别的方面涉及如下制剂,其中,奶蛋白组合物是:
酪蛋白酸盐/酯,特别是酪蛋白酸钠;
酪蛋白酸盐/酯(特别是酪蛋白酸钠)和还原糖(例如果糖)的混合物;
酪蛋白酸盐/酯(特别是酪蛋白酸钠)、水解植物蛋白(特别是水稻或大豆或马铃薯蛋白)和还原糖(例如果糖)的混合物;
酪蛋白酸盐/酯(特别是酪蛋白酸钠)、碳水化合物或碳水化合物衍生物和/或水解植物蛋白(特别是水稻或大豆或马铃薯水解蛋白)和还原糖(例如果糖)的混合物;
水解酪蛋白酸盐/酯(特别是水解酪蛋白酸钠)和还原糖(例如果糖)的混合物;
水解酪蛋白酸盐/酯(特别是水解酪蛋白酸钠)、水解植物蛋白(特别是水稻或大豆或马铃薯水解蛋白)和还原糖(例如果糖)的混合物;
水解酪蛋白酸盐/酯(特别是水解酪蛋白酸钠)、碳水化合物或碳水化合物衍生物和/或水解植物蛋白(特别是水稻或大豆或马铃薯水解蛋白)和还原糖(例如蔗糖)的混合物。
根据本发明,新颖的制剂可以通过如下方法获得,所述方法包括:制备脂溶性活性成分和奶蛋白组合物的水性乳液,如果需要的话,加入还原糖,将乳液转化为干粉,以及,如果需要的话,对干粉进行处理,以通过热处理使糖与蛋白质交联,或通过交联酶的处理使蛋白质交联。
合适地,在本发明的方法的第一步,奶蛋白组合物被分散于水中。此后,将脂溶性活性成分乳化进蛋白质的水性分散液,合适地,进行于液体状态,即通过适当加热和/或作为合适溶剂中的溶液来进行。或者,可以通过合适的手段(例如碾磨),来制备固体活性成分的悬浮液。然后喷雾上乳液,可选地,在移去过量溶剂之后进行喷雾。可用传统的喷雾-干燥技术、喷雾干燥与流化床颗粒化(后者通常被称为流化喷雾干燥或FSD)技术的组合来进行喷雾,或者通过粉末捕捉技术,其中,喷雾上的乳液颗粒被吸收剂床捕获,随后被干燥,所述吸收剂例如是淀粉或硅酸钙或硅酸或碳酸钙或其混合物。
最后,在又一个方面,本发明涉及包含本发明的新颖制剂的食物、饮料、动物饲料、化妆品和药物。
本发明的新颖的制剂还可以含有助剂(adjuvant)和/或赋形剂,例如,单糖、二糖、寡糖或多糖、甘油三酯、水溶性抗氧化剂、脂溶性抗氧化剂、润湿剂(例如,甘油、山梨糖醇、聚乙二醇、丙二醇)、补充剂(extender)和增溶剂、硅酸、硅酸钙、碳酸钙和水中的一种或多种。
还可存在于本发明制剂中的单糖和二糖的例子是:蔗糖、转化糖(invert sugar)、葡萄糖、果糖、木糖、乳糖和麦芽糖。还可存在于本发明组合物中的寡糖或多糖的例子是,黄原胶、阿拉伯胶、果胶、瓜尔豆胶、角豆(caroub)胶、藻酸盐、纤维素、纤维素衍生物(例如羧甲基纤维素)、淀粉、改性淀粉和淀粉水解产物(例如糊精和麦芽糊精,尤其是,例如,在5-65个糊精当量(下文中:DE)范围内的,以及葡萄糖浆,尤其是,例如,在20-95DE范围内的)。术语“糊精当量(DE)”指水解程度,其是对被计算为D-葡萄糖的还原糖的量(基于干重)的量度。天然淀粉具有接近于0的DE,而葡萄糖具有的DE=100。
甘油三酯适于为植物油或脂肪,例如玉米油、向日葵油、大豆油、红花油、菜籽油、花生油、落花生(arachis)油、棕榈油、棕榈仁油、棉籽油或椰子油。
水溶性抗氧化剂可以是抗坏血酸和其盐,例如抗坏血酸钠等。脂溶性抗氧化剂可以是生育酚,例如,dl-α-生育酚(即,合成生育酚)、d-α-生育酚(即,天然生育酚)、β-和γ-生育酚和其混合物;脂肪酸的抗坏血酸酯,例如棕榈酸抗坏血酸酯或硬脂酸抗坏血酸酯;丁基羟基甲苯(BHT)、丁基羟基茴香醚(BHA)、丙基没食子酸酯;或叔丁基羟基喹啉或6-乙氧基-1,2-二羟基-2,2,4-三甲基喹啉(EMQ)。
下述实施例更进一步地阐述了本发明。
实施例1
将75g酪蛋白酸钠加入到300ml水和13.2g甘油中。将混合物加热到60℃,直至发生溶解。向该溶液中加入15.1g果糖,将溶液的pH调为6.5±0.2。此后,将52.3g乙酸维生素A酯(2.1×106IE维生素A/g,用乙氧基喹啉稳定过)乳化进基质溶液,之后在60℃对混合物进行45分钟的搅拌。然后乳液的内相(inner phase)展示出大约350nm的平均颗粒大小。然后用约100ml水对乳液进行稀释,在大约5℃,通过旋转式喷雾嘴设备,将大约300g乳液喷雾到硅酸钙床中的喷雾盘上。通过过筛和干燥,将由此获得的珠粒与过量硅酸钙分离。获得了约120g的干粉,其中具有的维生素A含量为约750000IEA/g。
通过在旋转式干燥器中,于135℃对50g由此获得的干粉进行30分钟的热处理,使粉末发生交联。由此获得的产品具有约450000IE/g的维生素A含量,并且不溶于热水。
实施例2
将75g水解的酪蛋白酸钠(DH为大约3.5%)加入到300ml水和13.2g甘油中。将混合物加热到60℃,直至发生溶解。向该溶液中加入15.1g果糖,将溶液的pH调为6.5±0.2。此后,将52.3g乙酸维生素A酯(2.1×106IE维生素A/g,用乙氧基喹啉稳定过)乳化进基质溶液,之后在60℃对混合物进行45分钟的搅拌。然后乳液的内相展示出大约350nm的平均颗粒大小。然后用约100ml水对乳液进行稀释,在大约5℃,通过旋转式喷雾嘴设备,将大约300g乳液喷雾到硅酸钙床中的喷雾盘上。通过过筛和干燥,将由此获得的珠粒与过量硅酸钙分离。获得了约100g的干粉,其中具有的维生素A含量为约750000IEA/g。

实施例3
将72.4g酪蛋白酸钠和8g大豆蛋白水解产物(分子量分布<2000D≈90%)加入到300ml水和9.5g甘油中,在60℃进行溶解。向该溶液中加入14.5g果糖,将溶液的pH调为6.5±0.2。此后,将48g乙酸维生素A酯(2.1×106IE维生素A/g,用乙氧基喹啉稳定过)乳化进基质溶液,之后在60℃对混合物进行45分钟的搅拌。然后乳液的内相展示出大约380nm的平均颗粒大小。以类似于实施例1中所述的过程,将大约300g乳液喷雾到喷雾盘上。获得了大约100g干粉,其中具有的维生素A含量为约735000IE/g。
通过在流化床式干燥器中,于125℃对50g由此获得的干粉进行30分钟的热处理,使粉末发生交联。由此获得的产品具有约605000IE/g的维生素A含量,并且不溶于热水。
Claims (20)
1.稳定的粉末状制剂,其中包含:处于奶蛋白组合物基质中的脂溶性活性成分。
2.如权利要求1所述的制剂,其中,所述奶蛋白组合物是:天然奶蛋白,或具有多达25%的水解度的部分水解的奶蛋白,或它们的混合物,其具有超过80wt.%的蛋白质含量。
3.如权利要求1所述的制剂,其中,所述奶蛋白组合物是:天然奶蛋白,或具有多达15%的水解度的部分水解的奶蛋白,或它们的混合物,其具有超过80wt.%的蛋白质含量。
4.如权利要求1所述的制剂,其中,所述奶蛋白组合物是:天然奶蛋白,或具有多达10%的水解度的部分水解的奶蛋白,或它们的混合物,其具有超过80wt.%的蛋白质含量。
5.如权利要求1-4中任意一项所述的制剂,其中所述奶蛋白是酪蛋白酸盐/酯或部分水解的酪蛋白酸盐/酯。
6.如权利要求1-5中任意一项所述的制剂,其中所述奶蛋白组合物额外含有植物蛋白或植物蛋白水解产物或它们的混合物。
7.如权利要求6所述的制剂,其中所述植物蛋白水解产物中至少80%的平均分子量小于2500道尔顿。
8.如权利要求6或7所述的制剂,其中所述植物蛋白或植物蛋白水解产物获得自马铃薯蛋白、大豆蛋白、小麦蛋白、豌豆蛋白、水稻蛋白或羽扁豆蛋白。
9.如权利要求1-8中任意一项所述的制剂,其中所述奶蛋白组合物额外含有碳水化合物或碳水化合物衍生物,例如,蔗糖、转化糖、葡萄糖、果糖、木糖、乳糖、麦芽糖、黄原胶、阿拉伯胶、果胶、瓜尔豆胶、角豆胶、藻酸盐、纤维素、纤维素衍生物、淀粉、改性淀粉和淀粉水解产物,例如糊精和麦芽糊精,尤其是,例如,在5-65个糊精当量(下文中:DE)范围内的,以及葡萄糖浆,尤其是,例如,在20-95DE范围内的。

18.如权利要求17所述的方法,其中加入还原糖,并通过热处理对组合物进行交联。
