Description
本发明涉及新的合成的或者天然肽,特别是用于治疗病毒或者寄生虫感染或治疗肿瘤的肽,所述肽的长度少于30个氨基酸,优选少于20个氨基酸,尤其是优选15至20个氨基酸,且特征在于它们在体外特异结合2A型蛋白磷酸酶全酶或其亚基之一。本发明也涉及鉴定该肽的方法以及它们的用途。
由于本发明的肽的作用是调节细胞蛋白磷酸酶2A的活性,因此给出蛋白磷酸酶2A相关的现有知识,它们的生理功能及其和特定的细胞,病毒或者寄生虫蛋白的相互作用的介绍是很重要的。
细胞生理学部分是通过调节蛋白磷酸化来控制。细胞蛋白磷酸化状态依赖于使其发生磷酸化的蛋白激酶和使其发生去磷酸化的蛋白磷酸酶的拮抗作用。
将蛋白磷酸化酶分为主要两组:酪氨酸磷酸酶和丝氨酸/苏氨酸磷酸酶。丝氨酸/丝氨酸磷酸酶依赖于它们的底物和对特定抑制物的敏感性分为两种类型,即1型磷酸酶(PP1)和2型磷酸酶(PP2)。2型磷酸酶本身分成不同类型,包括磷酸酶2A(PP2A),磷酸酶2B或者通过钙来调节其活性的钙调磷酸酶,以及通过镁来调节其活性的磷酸酶2C(PP2C)。
目前已知2A型磷酸酶在进化过程中高度保守,并涉及调节多种生物过程。已经清楚PP2A酶涉及转录调节,细胞周期调控或者病毒转化。更进一步,PP2A为不同的病毒或者寄生虫蛋白的靶蛋白,这暗示PP2A在宿主病原体相互作用中起的作用。

近来,由病原体合成的不同的蛋白,尤其是病毒和寄生虫蛋白,牵涉到调节蛋白磷酸酶2A某些特定的活性。
病毒采用涉及PP2A不同的策略来促进它们在宿主细胞内的复制和存活。例如,副流感病毒将一种受PP2A控制的细胞起源的蛋白PKCζ整合到其病毒微粒中。这可以扰乱宿主蛋白磷酸化并促进其自身复制。(De BP,Gupta S,Barnejee AK.Cellular protein kinaseCζregulates human parainfluenza virus type3 replication.Proc.NatlAcad Sci USA1995;92:5204-8)。

相对于蛋白激酶在寄生虫感染中已经非常明确的作用,在最近3年才开始认识到丝氨酸/苏氨酸在寄生虫学领域作为重要的潜在的调节子。
最初,在恶性疟原虫(Plasmodium falciparum)中确定了两种丝氨酸/苏氨酸磷酸酶,Ppβ和PfPP。通过酶学研究证明了寄生虫中1型和2A型的磷酸酶的活性。最后,纯化拉寄生虫酶PP2A和PP2B。

由以上总结的蛋白磷酸酶2A在病毒宿主或者寄生虫宿主中相互作用的重要作用,可以了解鉴定病毒或者寄生虫蛋白和PP2A全酶或其亚基之一的结合位点的重要性。这样就可以确定这些病毒或者寄生虫病原体的新型的治疗目标。
具体来说,和PP2A相互作用的肽的鉴定可以开展新型的通过竞争性抑制来阻止由病毒或寄生虫蛋白通过和PP2A相互作用诱导的细胞机制,尤其是感染,病原体复制和细胞转化机制的药物。
本发明是关于鉴定长度减少的肽的方法,该肽结合PP2A全酶或其亚基之一。相对于天然蛋白或者大尺寸的多肽结构域,减少太小的肽的尺寸具有利于化学或者细胞体系合成,高产和廉价的优点。对于治疗用途,本发明的肽也更加稳定并易于用合适的载体转化到细胞质或者细胞核。
本发明基于可能鉴定长度少于30个氨基酸的肽,尤其是长度少于20个氨基酸的和PP2A全酶或其亚基之一结合的肽的证实。
具体来说,发明者已经表明使用Frank和Overwing描述的“班点合成”技术(Molecular Biology,1996,66卷:149-169中的方法,EpitopeMapping Protocols edited by:G E Morris Humana Press Inc,TotowaNJ)可以鉴定和PP2A全酶或其亚基之一相互作用的蛋白的结合位点。
举例来说,发明者已经鉴定了长度少于20个氨基酸的体外与纯化的PP2A全酶或其亚基之一结合的肽,该肽来自HIV-1的Vpr蛋白或者小泰勒虫寄生虫的CK2α蛋白。由于其抑制病毒或者寄生虫蛋白和特定PP2A全酶的相互作用,源自这些肽并被选择的拮抗物可以组成新型的抗癌,抗病毒或者抗寄生虫药剂。
发明涉及到鉴定其序列来自病毒,寄生虫或者细胞蛋白的肽的方法,所述的肽特异性地结合2A型蛋白磷酸酶全酶或其亚基之一,方法包括以下几个步骤:
a)以斑点的形式将序列源于病毒,寄生虫或者细胞蛋白的肽沉淀到支持物上,每个斑点对应于一个确定序列的肽的沉淀;
b)将固体支持物和含有蛋白磷酸酶2A全酶或其亚基之一的溶液接触,使用的条件允许支持物上的肽结合该全酶或其亚基之一;并且
c)鉴定固体支持物上和蛋白磷酸酶2A或其亚基之一结合的肽。

通常,可以使用的任何方法,只要能够造成肽阵列沉淀到固体支持物上,可以用来检测沉淀的肽和特殊化合物专一的相互作用即可.
高度优选,一系列沉淀的肽序列包括这些序列来源的病毒,寄生虫或者细胞蛋白的所有序列。这样,该过程可以用单一步骤测试给定蛋白的完整的序列,后者被“分成”有限数目的通常有重叠序列的肽。

A材料和方法
A.1纯化的PP2A蛋白
从猪脑中提取三聚体PP2A1蛋白纯化为同质蛋白。
PP2A重组的结构亚基在E coli中表达并用Cohen等人描述的方法(Cohen P,Alemany S,Hemmings B A,Resink T J,Stralfors P,TungH Y.Protein phosphatase-1 and protein phosphastase2A from rabbitskeletal muscle.Methods Enzymol 1988 159,390-408),或者Bosch等人描述的方法(Bosch M,Cayla X,Van Hoof C,Hemmings B A,OzonR,Merlevede W,Goris J.the PR55 and PR65 subunits of proteinphosphastase 2A from Xenopus laevis.Molecular cloning anddevelopmental regulation of expression.Eur J Biochem1995,230,1037-45)进行纯化。
A.2.鉴定HIV Vpr的PP2A结合位点和小泰勒虫(T parva)CK2α的PP2A结合位点的方法
通过以上描述的“点肽”技术(Frank and Overwing,1996,MethMol Biol 66,149-169)鉴定来自CK2α蛋白(由T parva原虫编码)或者Vpr蛋白(由HIV-1病毒编码)的PP2A的结合蛋白。
该方法包括在纤维素膜上固定的位置原位合成十二肽,其中序列系列覆盖目的蛋白(Vpr或者CK2α)的全序列。膜上两个连续点的肽通过两个氨基酸交叠。
合成覆盖HIV-1的Vpr蛋白的全序列的68个十二肽和覆盖泰勒虫的CK2α蛋白序列的205个十二肽,并将其共价连接到纤维素膜上。
在环境温度下每张制备的膜先用含有5%脱脂的Régilait(牛奶)和3%BSA的TBS饱和1小时,然后在含有4ug/ml纯化的蛋白(PP2A亚基A或者全酶PP2A1)的相同缓冲液中孵育过夜。和蛋白质印迹一样,在将膜和抗结构蛋白A(图1A和2A)的抗体以及识别PP2A蛋白A,B和C(图1B和2B)的抗体混合物一起孵育之后,每种纯化的蛋白(结构亚基A或者三聚体全酶PP2A1)和肽序列的特异的相互作用就显示出来。
用传统的用于孵育的TBST缓冲液(TBS+TWEEN)15分钟冲洗膜5次,在环境温度下用二抗(偶联有过氧化物酶)再孵育1小时。最后用TBST缓冲液15分钟冲洗膜5次并且显现。

将Hela细胞(100l含有104)接种到96孔板(平底)中,使用完全DMEM培养基并加入2.5%青霉素/氨卞青霉素和10%胎牛血清。在37℃下CO2箱(5%)中培养过夜,加入复合体(生物素-抗生素过氧化物酶肽)不同的稀释液。在培养4小时后,吸出上清,用PBS将细胞洗3遍,用胰酶处理并在PBS中计数。在计数之后,细胞置于300ul的裂解液中。
-测量过氧化物酶的活性
50ul的OPD缓冲液加入96孔的ELISA培养板中,并加入50ul的裂解缓冲液或者50ul的细胞裂解液(通常,使用不同的系列稀释(至1/2))。为了显现,加入50ul的OPD溶液(在暗处)。用100ul的1N的HCl停止反应(大约10分钟)

Hela细胞(100ul含有104)接种到96孔板(平底)中,使用含有2.5%青霉素/氨卞青霉素和10%肽牛血清的完全的DMEM培养基。在不同的肽浓度中,细胞在37℃下CO2箱(5%)中培养过夜。在培养72小时后,吸出含有肽的培养基,加入100ul的0.5毫克/毫升的MTT(只在DMEM中稀释)至每个孔。37℃下暗处培养30分钟,然后吸出MTT,并将50ul的DMSO加入到所有孔中。需要等待十分钟完成细胞裂解,并搅动裂解液使得孔中的反应产品均匀分散。然后,用690nm参考滤光器读出平板在570nm处的读数。
B.结果和讨论
B.1.鉴定由两种病原体(HIV-1和小泰勒虫)编码的蛋白的含
有和PP2A(PP2A1和亚基A)结合的位点的肽序列
在用纯化的三聚体PP2A全酶孵育含有覆盖HIV-1的Vpr和Tparva的CK2α序列的肽的膜之后得到的结果允许确定Vpr和CK2α肽的5个可以特定结合PP2A的序列并在下表显示:

更精确地,鉴定含有HIV-1Vpr的同PP2A1蛋白(图1B,“位点1”)以及同亚基A(图1A,“位点1”和“位点2”)的结合位点的两条肽序列。也鉴定含有小泰勒虫CK2α的同PP2A1蛋白(图2B,“位点3”)以及和结构亚基A(图2A,“位点1”,“位点2”和“位点3”)的结合位点的三条肽序列。
B.2使用结合PP2A的HIV-1Vpr肽的重要性
外源表达或者由于HIV-1的Vpr前病毒感染引起的表达诱导Hela细胞,T淋巴细胞系和初级淋巴细胞调亡(Stewart等人,1997 J Viroi71:5579-9)。最初使用Vpr突变体使得该影响与细胞停止在细胞周期G2期相关。最近已经表明,Vpr也能够诱导同G2期,停止不相关的调亡(Nishizawa等人,2000,Virology 27,16-26)。
据报道在和E4或f4腺病毒蛋白相互作用后PP2A的活化诱导转化细胞的调亡(Shtrichman R等人,2000,Oncogene19,3757-3765)。类似的,Vpr的表达也诱导转化细胞的调亡。(Stewart等人,1999,PNAS,96,12039-12043)。
进一步地,本领域已知的Vpr突变体的分析表明由本发明方法鉴定的并特异结合PP2A蛋白的肽包含了与Vpr调亡前作用(pro-apototic)相关的序列。
这样,和PP2A相互作用的并由本发明方法鉴定的病毒蛋白Vpr和E4orf4的片段在诱导肿瘤细胞的调亡中非常有用。
鉴定的肽也自然用于抑制HIV或者其它相关的病毒和逆转录酶病毒引起的感染。
B.3.使用结合PP2A的小泰勒虫CK2α序列的重要性
Okadaic酸和SV40小T抗原的使用证明了PP2A通过新的涉及PI3-激酶,PKCζ(鉴定为MAP-激酶-激酶-激酶或者MEKK),MEK蛋白和两种MAP激酶ERk-1和ERk-2,以及转录因子NF-kB和Sp1的磷酸化级联反应途径控制细胞增殖。(Sontag E,Sontag J M,GarciaA(1997),EMBO J,16,5662-5671;Ayllon V,Martinez A,C,Garcia A,Cayla X和Rebollo A(2000),EMBO J 19,1-10,A Garcia,S Cereghini,E Sontag(2000),J Biol Chem275,9385-9389)。进一步地,PP2A在调节MAP激酶级联反应中的功能已经由Chambaz研究组提出(Hériché等人,(1997),Science,276,952-955),研究显示过表达细胞的CK2α亚基活化去磷酸化MEK蛋白的PP2A。
Ole-Moi等人的研究(Embo J.1993,12,1621-1631)表明泰勒虫转化诱导宿主蛋白高度磷酸化。该效应部分是由于组成性活化细胞的CK2α造成的,细胞CK2α本身依赖于由寄生虫编码并分泌到转化细胞的细胞液中的CK2α型亚基的作用。
正如以下所示,由本发明方法鉴定的对应3个同PP2A的结合位点的序列的比较,可以鉴定一类基序的存在:K-I-G/L-R/K,其在位点2部分重复。
位点1:
有趣的是,CK2α的ATP结合位点部分覆盖位点1和位点2,这表明在CK2α和亚基A相互作用后抑制激酶活性。进一步地,可从下表2看出,含有小泰勒虫的CK2α的与PP2A的结合位点的3个序列在包括恶性疟原虫和利什曼虫寄生虫的多个物种中保守。

仔细分析相互作用表明来自不同种的CK2α应该和PP2A相互作用;例如图2中描述的来自小泰勒虫CK2α的肽131可以结合PP2A,其中位点3的最初4个氨基酸被缺失。这表明最初3个氨基酸不同的的利什曼虫,恶性疟原虫的CK2α可以结合PP2A。这和KILRLI基序具有重复的可以作为PP2A结合位点的K/R-II/L-I/L序列这一事实相一致。
细胞内存在的这些肽,对应于蛋白同PP2A的体内结合位点,可以干扰这些寄生虫的发育。
B.4.本发明的肽化合物对细胞的生物学影响
表3列出的不同的肽以生物素化的形式合成,用HPLC纯化(Neosystem)并在Hela细胞中分析它们对胞内渗透和细胞存活的影响。表1中显示的一系列肽的研究允许确定6条有可能穿透进入Hela细胞的肽(图3)。
■FD6:12个AA的肽,来自PP2A亚基A和小泰勒虫CK2α蛋白相互作用的一个位点。
■FD7:18个AA的肽,对应于3个6AA六基序的重复体;该序列来自恶性疟原虫CK2α,和与PP2A结合的小泰勒虫序列同源;
■FD8:来自鱼精蛋白的肽(一种已知的PP2A的激活剂);
■FD11:18个AA的肽,对应于3个6AA六基序的重复体,来自FD14序列;
■FD14:FD14,其复制已鉴定的HIV-1Vpr的同PP2A的结合位点;
■FD13:该肽对应于HIV-1Vpr序列,它和FD14肽序列同源,代表了另外一种HIV-1Vpr的PP2A结合位点。
进一步地,在表1中显示的系列肽中进行的存活研究可以鉴定3种抑制Hela细胞存活的肽:
■FD8:影响Hela细胞的存活(图4A);
■FD14:清除地影响Hela细胞的存活(图4B);
■F12:18个AA的肽,其序列来自FD11肽(R突变为A)。该肽和Chlamydia muridarum的葡糖胺转移酶蛋白同源,影响Hela细胞的存活(图4C)。这种生物影响归因于和质膜的相互作用。

a)以斑点的形式将序列源于病毒,寄生虫或者细胞蛋白的肽沉淀到支持物上,每个斑点对应于一个具有确定序列的肽的沉淀;
b)将固体支持物和含有蛋白磷酸酶2A全酶或其亚基之一的溶液接触,使用的条件允许支持物上的肽结合该全酶或其亚基之一;并且
c)鉴定固体支持物上和蛋白磷酸酶2A或其亚基之一结合的肽。
18.根据权利要求17的方法,其特征在于以斑点形式沉淀的肽长度小于20个氨基酸。
19.根据权利要求17的方法,其特征在于以斑点形式沉淀的肽长度小于15个氨基酸。
20.根据权利要求17-19中任一项的方法,其特征在于肽沉淀在纤维素膜上。
21.根据权利要求17的方法,其特征在于一系列沉淀的肽序列覆盖这些序列所来源的病毒,寄生虫或者细胞蛋白的完整序列。
22.一种制备肽或者多肽的方法,包括用权利要求7中定义的细胞表达载体来转化宿主细胞,随后在培养基中培养转化的宿主细胞并回收肽。
