Description

人的皮肤经过某些老化过程,其中一些老化过程是由于固有过程(年龄老化),和一些老化过程是由外源性因素引起的(环境,例如光老化)。另外,皮肤的外观会出现暂时性和永久性的变化,例如粉刺、油性或干性皮肤,角质层、红斑痤疮、对光敏感、发炎、红斑反应、过敏反应,或自免疫反应,例如皮肤病、光照性皮肤病和其它,仅仅部分地了解影响它们的确切原因。
外源性因素例如特别包括日光或具有相应范围的人造辐射源,以及可通过辐射产生的化合物,例如未确定的反应性光产物,它们还可以是自由基或离子性的。但是,这些因素还包括有害或有活性的化合物,例如臭氧,自由基例如羟基自由基,单价氧和其它活泼的氧或氮化合物,香烟的烟雾,天然和合成毒物,以及其它能干扰皮肤的自然生理学或形态学的因素。这些因素的影响会特别导致对皮肤细胞的DNA、对细胞外基质的骨胶原、弹性蛋白或葡糖胺多糖分子(它们使得皮肤紧致)产生直接损害。此外,信息转导链会受到影响,导致有害因素例如基质降解酶的活化。这些酶的重要代表例子是基质金属蛋白酶(MMP,例如骨胶原酶,明胶酶,溶基质素),它们的活性另外通过TIMP调节(基质金属蛋白酶的组织抑制剂)。

另一个结果是发炎反应,和特别是释放出免疫调节化合物,例如白介素、前列腺素和组胺。结果,抗免疫细胞被抽取,发炎反应加重。
老化的结果是皮肤变薄,表皮和真皮的较弱成网,细胞数目的降低和供应血液的组织减少。老化过程导致形成细纹和皱纹,皮肤变粗糙、发黄和开始松弛,还发生色素紊乱。
具有抗氧化效果的化合物通常在用于抗衰老的皮肤病学或化妆品制剂中使用。此外,它们还可以用于减少在人或动物皮肤中出现的有害的或不利的氧化过程。公知的是,这些过程在皮肤老化中起到重要作用。由于形成过氧化物和氢过氧化物,皮肤暴露在永久性氧化应力下,这些物质中的一些是来源于皮肤的外部环境,但是一些也是内源性形成的。为了抵抗这种应力,皮肤具有许多自身的保护机制。但是,这些保护机制不足以完全防止皮肤中的氧化过程。相比之下,通常认为这些氧化过程是皮肤老化的主要原因,而且也是皮肤中的一般性或病理性变化的主要原因。
特别是,通常认识到类脂的过氧化作用对于老化的重要性。W.A.Prior描述了类脂氢过氧化物以及其分解产物的有害作用(ACS Sysup.Ser.(1985),277,77-96)。对于过氧化物、氢过氧化物或过氧化氢的分解,已经在化妆品方面描述了各种体系,例如使用金属卟啉(JP 3273082),肌醇六磷酸锌盐(JP 08104635),过氧化氢酶(JP 08175035)和其它酶(JP67165553)。另外,JP 06345797公开了含有半胱氨酸的二肽用于美白皮肤、用于防止类脂过氧化和用于防止类脂过氧化物分解的用途。为了帮助内生保护机制,所以将具有抗氧化效果的组分,即O-或C-自由基清除剂,加入化妆品制剂和皮肤病学制剂中(例如DE19739349)。但是,实际达到的效果迄今还远不能满足期望。特别是,抗氧化剂的添加量的增加并不能总是获得相对较高的抗氧化效果。
本发明的目的是提供用于化妆品或皮肤病学制剂的活性成分,它能显著提高抗氧化效果。
本发明的另一个目的是提供一种用于化妆品或皮肤病学制剂的活性成分,它能保护皮肤免受氧化性损害。

虽然常规的抗氧化剂基本上是O-或C-自由基清除剂,但是本发明的目的是通过进一步的措施更有效地防止皮肤受损,即在上述路线中的另一个位置干扰该机理。为此,按照以下路线的离子性和还原性攻击是适宜的。
现在,已经发现还原性过氧化物分解剂的使用具有优异的效果。另外,已经发现使用作为自由基清除剂的抗氧化剂与还原性过氧化物分解剂的组合,具有优异的协同效果。在这种情况下,必须选择过氧化物分解剂,使得它在体外比相应有效的皮肤固有的含硫化合物例如胱氨酸或半胱氨酸具有显著更高的活性。
具体而言,我们发现该目的由具有有效含量的以下组分的化妆品或皮肤病学制剂来实现:
a)至少一种用作O-或C-自由基清除剂的抗氧化剂,和
b)至少一种含硼的有机化合物,它将过氧化物或氢过氧化物还原成相应的醇,且不会形成活性自由基后续阶段。
根据本发明的制剂特别适用于避免或减少皮肤受到内源性或外源性形成的过氧化物或氢过氧化物的损害。
所述化妆品或皮肤病学制剂通常含有基于最终制剂计的0.001-30重量%、优选0.01-10重量%、特别是1-5重量%的抗氧化剂(a)和0.001-30重量%、优选0.01-10重量%、特别是1-5重量%的至少一种过氧化物或氢过氧化物分解剂(b)。
过氧化物或氢过氧化物分解剂(b)具有比皮肤固有的化合物例如胱氨酸或半胱氨酸显著更大的分解(还原)作用。特定化合物是否适用于本发明的用途可以在例如在体外从以下事实观察到:在室温下,在于70℃储存30分钟后以0.055m/L的摩尔浓度溶解在极性或非极性溶剂中,它们使得过氧化物或氢过氧化物的浓度降低至少10%,特别是20%,优选50%,特别是90%。过氧化物或氢过氧化物的浓度通常是0.5m/L。
本发明进一步提供含硼的有机化合物b)在化妆品或皮肤病学制剂中的用途,所述含硼的有机化合物b)将过氧化物或氢过氧化物还原成相应的醇,且不会形成活性自由基后续阶段。
本发明还涉及包含下列组分的组合物在化妆品或皮肤病学制剂中的用途:

另外,根据本发明的活性成分可以在用于以下用途的化妆品组合物中使用:用于护发的组合物,例如卷发剂、美发乳、头发冲洗剂、发用乳液、分叉液,用于烫发的中和剂,热油处理制剂、调理剂、定型液、香波、染发剂和着色剂、喷发胶、烫发液、烫发定型液、闪光喷雾剂、亮发剂、发用定型用品、发用营养剂、脱发护理组合物等。
化妆品或皮肤病学制剂可以根据应用领域是喷雾剂(泵式喷雾或气雾剂)、泡沫、凝胶、凝胶喷雾剂、液、霜、摩丝、软膏、悬浮剂或粉剂的形式。

实施例1(过氧化物分解的检测)
根据本发明使用的化合物列在表1和2中,根据以下的实验研究它们的过氧化物分解作用,并与胱氨酸和半胱氨酸比较。
实验的描述
制备以下溶液:
1、氢过氧化叔丁基在适宜溶剂中的0.05mol/l的溶液
2、潜在氢过氧化物分解剂在适宜溶剂中的0.055mol/l的溶液
合适的溶剂可以是甲苯-d8或CD3COOD。
从上述溶液,在每种情况下通过将350μl的溶液1和350μl的溶液2混合来制备检测溶液;将检测溶液立即加入NMR管中并转移到NMR仪器中。溶液的制备和检测总是在22℃下进行。在检测之前,将溶液于70℃储存在恒温浴中30分钟。所有检测使用来自Varian公司的INOVA 500型500MHz的NMR光谱仪进行。对于每次溶液的检测,记录1H-NMR谱图和2D-HSQC(1H/13C)谱图。氢过氧化叔丁基和叔丁醇各自的CH3-质子信号非常接近;所以参考2D-HSQC谱来确定信号的氢过氧化叔丁基或叔丁醇归属。两种组分的相对比例通过将相应组分在1H-NMR谱图中的信号或HSQC谱图的十字峰的信号积分来进行(Lit:W.Wilker等,Magn.Reson.Chem.31,287-292(1993))。

Claims (17)
1、一种含有以下组分的化妆品或皮肤病学制剂:
a)至少一种用作O-或C-自由基清除剂的抗氧化剂,和
b)至少一种含硼的有机化合物,它将过氧化物或氢过氧化物还原成相应的醇,且不会形成活性自由基后续阶段。
2、根据权利要求1的化妆品或皮肤病学制剂,它含有基于最终制剂计的0.001-30重量%的抗氧化剂(a)和0.001-30重量%的至少一种含硼化合物(b)。
3、根据权利要求1的化妆品或皮肤病学制剂,它含有作为过氧化物或氢过氧化物分解剂(b)的化合物,该化合物在体外在室温下,在于70℃储存30分钟后以0.055m/l的摩尔浓度溶解在极性或非极性溶剂中,使得过氧化物或氢过氧化物的浓度降低至少10%。
4、含硼的有机化合物在化妆品或皮肤病学制剂中的用途,所述含硼的有机化合物将过氧化物或氢过氧化物还原成相应的醇,且不会形成活性自由基后续阶段。
5、包含下列组分的组合物在化妆品或皮肤病学制剂中的用途:
a)至少一种用作O-或C-自由基清除剂的抗氧化剂,和
b)至少一种含硼的有机化合物,它将过氧化物或氢过氧化物还原成相应的醇,且不会形成活性自由基后续阶段。
6、根据权利要求4和/或5的用途,用于避免皮肤受到由内源性或外源性因素形成的过氧化物或氢过氧化物的损害。
7、根据权利要求4和/或5的用途,用于随后清除和/或缓解皮肤受到过氧化物或氢过氧化物的损害。
8、根据权利要求4和/或5的用途,其中使用0.001-30重量%的含硼化合物b)。
11、根据前述权利要求任一项的含硼化合物,其中使用式(III)的化合物,
其中R1、R2和R3具有上述含义。
12、根据前述权利要求任一项的含硼化合物,其中使用式(IV)的化合物,
其中R1和R2具有上述含义,且R1和R2可以通过闭环来桥接。
15、根据前述权利要求任一项的含硼化合物,其中使用下式(VII)的化合物:
其中R1、R2和R3具有上述含义。
