Abstract

在许多领域中,尤其在食品和口腔护理领域中,意图给予人们一种化合物,比如活性成分或芳香产品。根据应用:粉末,颗粒,胶囊,微胶囊,脂质体,微球体等,已开发了各种给药方法。这些不同的方法一般呈现与所施用的化合物的保护和/或释放有关的特性,适合于最终的产品。尤其,它们可以是物理的(机械的,温度等),化学的,(pH,水,离子强度等)或耐酶的,或不溶性,或渐溶性(必须吞服的化合物,耐胃酸(gastro-resistant compounds)化合物等)。

美国专利6238690提到了这个问题.在这专利的以前工艺的描述中,具体说到:用明胶型成膜材料制造封皮,常规地形成胶囊,这种成膜材料的缺点是,除了需要使用复杂的生产方法外,溶解缓慢,同时在口腔中留下不舒服残留.在这个专利中,提供的解决办法,是不用成膜材料,而是用玻璃态的碳水化合物作为封皮材料.
然而,对于包括成膜材料封皮的胶囊,问题仍然存在。
大量研究后,本申请人设计和开发了新的胶囊,它的封皮包含成膜聚合物,用于内含物的快速加溶和释放。本发明的胶囊加溶非常快,另外,不呈现皮层效应;所以,封皮的加溶是最适佳的。本发明因此涉及包括中心部分和至少一个封皮的胶囊,封皮包括至少一种成膜聚合物,其特征在于:其封皮的总加溶时间,根据下面定义的试验A,小于或等于85秒。
在该试验A中,使用在法国或美国药典(USP XXIII,724)中所述的具有连续流动池的溶解装置。这种装置使模拟口腔中胶囊溶解性成为可能。对胶囊优选使用连续流动池,而不是常规的溶解装置,这是因为这样一个事实:胶囊内含物可能是完全亲脂性的,所以有时浮在表面,随着时间,不可能总是得到均质的样品。

根据试验A,本发明胶囊封皮的总加溶时间小于或等于85秒,优选小于或等于80秒,更优选小于或等于70秒。本发明胶囊的破裂时间小于或等于30秒,优选小于或等于20秒,更优选小于或等于15秒。
这样一种胶囊,在摄取时,在口中产生它所含活性化合物的立即感觉。
本发明的胶囊可就此使用,作为最终产品或作为新的药用形式使用,或作为另一药用形式或另一产品的成分使用。它可在使用者摄取后加溶,或在使用者将要服用的介质中加溶。
在工艺的意义上,本发明的胶囊不是软胶囊;它是感觉硬的胶囊,在手指间用力压时会破裂。它的硬度大小为1-5公斤/平方厘米。
在一个实施例中,本发明的胶囊是球形的或基本上球形的,优选球形的。它的直径可变化,优选1-7毫米。该直径取决于使用,可由本领域的技术人员容易地选择。本发明胶囊的重量是可变的;从0.5-170毫克。在优选的实施例中,本发明胶囊的直径为4.5-6.5毫米,重量为45-140毫克;例如,直径为4.5-5.5毫米,重量为45-80毫克。
