发明详述
目前,受高死亡率困扰的AIDS患者的性二态性疾病卡波西肉瘤(KS)仍无法得到有效的治疗。最近的报道(Lunardi-Iskandar Y等,1995,自然375:64-68)表明胎盘糖蛋白激素,即人绒毛膜促性腺激素(HCG)及其β亚单位在小鼠异种移植体中可抑制卡波西肉瘤细胞的致瘤性和转移。随后,在临床试验中阐明了商购HCG的抗KS效力(Gill PS等,1996,新英格兰医学杂志335:1261-1310;Gill PS等,1997,J.Natl.Cancer Inst.89:1797)。另外,Bourinbaiar(Bourinbaiar AS等,1992,FEBS.Lett.309:82-84;Bourinbaiar AS等,1995,Immunol Lett 44:13-18)和Gallo小组(Lunardi-Iskandar Y等,1998,天然药物4:428-434)的早期研究表明HCG的β亚单位(或其衍生的肽)具有抗HIV作用。
本申请人花了几年的时间研究激素调控KS的细胞和分子方面。申请人实验室的最新研究证实商购的HCG制品(已知约为25%纯)以依赖于剂量的方式表现出显著的抑制作用。然而,纯的和生物活性的HCG对培养中的卡波西肉瘤的生长不起作用,这表明杂质(或降解产物)可能是活性剂。
申请人根据分子大小对商购的HCG制品进行了再次分级分离,检测每个级分抑制KS细胞生长,HCG放射性受体结合和类固醇生成生物活性的情况。申请人的结果表明抗KS活性是由低分子量的组分而不是大分子的HCG引起的。有趣的是,申请人已鉴定出转录因子,该因子可能是抗KS组分调控的靶。申请人得出结论:商购HCG制品中存在的尚未鉴定出的分子决定着以前被错误地归功于HCG的生长抑制作用。
令人惊奇地是,本发明鉴定出具有抗KS和抗HIV药物活性的纯化的HIP蛋白质。此蛋白质是HCG样抑制蛋白,并被吸附至聚丙烯塑料支持物上,其具有选自下列的氨基酸序列:

HCG的来源
检测了两个商购的HCG样品,第一个是由Wyeth-Ayerst,Montreal提供的,其商品名为APL(Lot#C84662A是受人惠赠的,cat#DIN-02168936是购买的),应强调的是早期研究中使用了APL(Lunardi-Iskandar Y等,1995,自然375:64-68;Gill PS等,1996,新英格兰医学杂志335:1261-1310)。两个样品也购自Sigma,St-Louis,Mo(lot#26H 1040)。纯的HCG二聚体以及α-HCG和β-HCG得自NIDDK(Bethesda,MD)。重组的HCG得自Organon,Oss,荷兰。将经冻干储存的所有HCG样品溶解于PBS中,并冷冻成等分试样。
细胞增殖的评估
KS-Y1(Lunardi-Iskandar Y等,1995,自然375:64-68)分离自HIV患者,而原始KS-SLK细胞系的被称为N-1506的亚系(Lunardi-Iskandar Y等,1995,自然375:64-68)来源于免疫抑制的受试者(Herndier BG等,1994,AIDS 8:575-581)。这些细胞系是由Lunardi-Iskandar博士(N.I.H.,Bethesda)提供的。在上述任何HCG样品的存在或缺乏下将KS细胞传代24-96小时,每隔1天更换1次培养基。按文献所述(Guo WX等,1996,Am J Pathol148:1999-2008;Guo WX等,1995,Am J Pathol 146:727-734)测定3H-胸苷的掺入。在大多数实验中,数据被表示为4次测定结果的平均值±SEM。由斯氏t-检验测定统计学分析结果。
在SEPHADEXTM G-100上分级分离APL
通过溶解于1.5ml 0.05M NH4HCO3收集3管APL(10000 IU/管)以用于分级分离。将透明的溶液上样于已用相同溶剂平衡过的SEPHADEXTM G-100柱(Pharmacia,Baie d’Urfe,Qc)(1.5×90cm)。收集1.7ml级分并集中成7个级分(见图3)。从各级分中各取出一小部分以评估HCG当量活性,将各级分的其它部分冻干。
HCG受体结合活性
与LH受体有效结合的激素HCG的方便试验是按文献详述(Sairam MR,1983,激素蛋白质和肽,Li C.H编,p1-79;Manjunath P等,1982,生物化学杂志,257:7109-7115)使用成年猪睾丸的膜制品进行放射性受体试验。按文献所述(Sairam MR,1983,激素蛋白质和肽,Li C.H编,p1-79)检测标准(NICHD,Bethesda的CR-125 HCG)或受试样品的125I-HCG结合。计算7个级分中每一个的总结合活性,表示为μg HCG当量/级分。
类固醇生成活性
HCG是很有效的类固醇生成激素,因此,一个可靠的生物测定法是将小鼠间质(Leydig)肿瘤细胞(MA-10,最初得自M.Ascoli博士,Iowa)与上述受试样品一起温育(Sairam MR,1983,激素蛋白质和肽,Li C.H编,p1-79)。通过放射性免疫测定法估计培养基中的孕酮(Sairam MR,1983,激素蛋白质和肽,Li C.H编,p1-79)。
电泳迁移率凝胶移位分析(EMSA)
根据Smeal的原始方法(Smeal T等,1989,基因进展,3:2091-2100)由KSY-1细胞培养物制备核提取物。按文献所述(Smeal T等,1989,基因进展,3:2091-2100,Saatcioglu F等,1994,Semin.Cancer Biol,5:347-359)进行AP-1位点(TRE,TPA反应元件)的结合反应。用32P-ATP末端标记序列5’-GGATCCGATGAGTCAGCCA-3’的合成胶原酶TRE寡核苷酸探针,并按文献所述(Smeal T等,1989,基因进展,3:2091-2100)进行EMSA。通过使用100摩尔过量的未经标记的TRE以确定特异性。通过使用MolecularDynamics(Sunnyvale,CA)的软件进行磷图象仪分析以定量信号。
纯的HCG对KS细胞没有抑制活性(图1)
设计最初的实验是为了证实HCG的抑制作用。比较了对两个不同KS细胞系的作用。在经商购的HCG制品(Sigma或APL)预先处理过的细胞中,在所有的KS细胞系中都引发了抑制作用(p<0.05)。在初步实验中,注意到对细胞生长的剂量依赖型抑制作用。
检测了两个商购的HCG产品(APL和Sigma),得到几乎相同的抑制作用(图1,右边的两个方块)。然而,一些HCG货品的效力比其它的更强。
所用样品的当量浓度为50U/ml(图1)。相对于经载体处理的细胞(C)而言,用Sigma-HCG(S)或Ayerst-HCG(APL)处理之后的KS细胞的生长显著受阻。与之形成对照的是,使用高度纯化的HCG制品没有产生抑制作用。图中符号:1=二聚体HCG;2=αHCG;3=βHCG;4=不相关的人尿蛋白质收集物;*P<0.05。
接着,证实了被清楚表征的纯的二聚体HCG,纯的α或β亚单位和重组HCG的抗KS活性。这些纯的HCG中无一能抑制KS生长(图1,#1-3)。通过在经培养的间质细胞中诱导类固醇生成以检查这些化合物的生物活性。如所预期的,重组或纯的HCG引发了典型的生物反应,而α或βHCG都未显示出任何的类固醇生成作用。
粗HCG的分子筛分(图2)
一般而言,经生物活性和生化分析评价,临床所用的装入安瓿瓶中的孕激素中HCG的纯度仅约为25%(Manjunath P等,1982,生物化学杂志257:7109-7115)。使用SEPHADEXTM层析将商购的HCG(APL)分成7个不同的级分(图2)。
将3管临床级APL(各为10,000 IU)中的内容物溶解于0.05M NH4HCO3中,在SEPHADEXTM G-100柱(1.5×90cm)上进行分子筛分。在A230nm处监测(E组)的经洗脱的蛋白质/肽级分被分成7个收集物,即X轴上的级分收集物#1-7。总共收集120管(1.75ml/管),在KS培养基(不含血清)中重构每个收集物的冻干物,评价其对细胞增殖的影响(A组实验)。测定猪睾丸膜中的HCG受体结合情况(B组实验)和MA-10细胞中的类固醇生成活性(C组实验)。D组实验:条形图显示出AP-1结合的定量光密度扫描,插入部分表示级分(fr)2,4和7的实际EMSA蛋白质-DNA复合物;nd=未测定。用当量浓度为100U/ml的指定试剂将KSY-1细胞处理4天,观察到对性腺细胞的HCG激素活性(收集物2)和对KS细胞的抑制作用(收集物7)有明显的差异,*p<0.05。
从最初2个集中的级分中回收到85%以上的HCG受体结合活性(图2B),其中会出现大小与纯的HCG相同的高分子量蛋白质。早期的主要级分(收集物#2)的Ve/Vo比率对应于大分子的HCG。除了粗提取物中存在的其它未被鉴定的物质外,这些级分还含有激素亚单位(α/β)或其降解产物。以前的研究(Sairam MR,1983,激素蛋白质和肽,Li C.H编,p1-79)表明级分#7由相对较小的肽以及APL制剂中存在的其它试剂组成。在第2个级分中(图2B和C),HCG(而不是其亚单位)的特征,即类固醇生成或结合活性是最高的。这些结果与受体结合检测的结果一致,在该检测中,仅有二聚体的(α-β结合的)HCG而不是各个亚单位或其裂解产物是具有生物活性的(Sairam MR,1983,激素蛋白质和肽,Li C.H编,p1-79;Manjunath P等,1982,生物化学杂志,257:7109-7115)。与此相反,只有级分#7含有KS抑制活性。
通过HCG组分下调AP-1结合(图3)
激活蛋白-1(AP-1)是由12-O-十四烷酰佛波醇-13-乙酸酯(TPA)肿瘤启动子、几个生长因子和多个细胞外刺激物所诱导的转录激活物(参见SaatciogluF等,1994,Semin.Cancer Biol,5:347-359)。AP-1由jun和fos家族的蛋白质组成,它们连接形成同-(jun/jun)或异二聚体(jun/fos),并识别AP-1调控基因上存在的已知为TPA反应元件(TRE)的共有序列5’-TGAG/C TCA-3’。据认为AP-1复合物在如生长刺激,分化,神经元兴奋和转化的几个信号传导途径中起着重要作用(Saatcioglu F等,1994,Semin.Cancer Biol,5:347-359)。APL-HCG和级分7中的组分显著抑制KSY-1细胞中AP-1与TRE的结合(图2D)。处理3,6和12小时之后,APL-HCG分别抑制AP-1结合达1.5,3和2倍(图3)。
将细胞与50IU/ml APL-HCG(+)或载体(-)温育指定的时间段(图3)。制备核提取物并进行EMSA。所示结果是3个实验的代表。箭头指向游离的探针。100ex=100倍过量的未标记探针。图3的上部显示出实际的凝胶移位,而图3的下部提供了主要带(箭头所指)的定量磷图象仪测定结果;*表示相对于经载体处理的细胞而言,p<0.05。
也观察了剂量-反应,在约100IU/ml时观察到接近最大的效应。因此,抑制AP-1可能是抑制KSY-1细胞的重要途径。
使用反相-HPLC纯化HIP(图4)
APL购自Wyeth-Ayerst Cat.#DIN 02168936,其被装入配有冷冻剂的绝热的盒子中。收到货品之后,将APL储存于4℃。室温下,用随APL安瓿一起出售的1ml试剂重构1管APL(其含有干产物)lot#JA(L)3YYF-AB并在1小时之内进行HPLC处理。粉末易于溶解产生均匀的“溶液”。将该“溶液”注入装配有7.8×300mm C-18TM柱的WaterTM HPLC装置。使用渐增线性等梯度的乙腈(溶于含0.1%三氟乙酸的水中)进行柱洗脱。梯度从5%增加至75%乙腈。在洗脱的过程中以220波长监测吸光度,并将级分手工收集至硅烷化的聚丙烯管中。当使用标准的(即未经硅烷化的)试管时,会出现生物活性丢失的现象。收集之后,立即将级分置于SavantTM Speed-vac装置中以干燥样品。图4中显示出梯度;右侧或Y轴显示出%乙腈(%B;B:溶于含0.1%三氟乙酸的水中的80%乙腈),X轴表示时间(分钟)。在Y轴上记录220nm的吸光度。随后发现箭头表示的两个峰(D和E)含有KS抑制活性(见下面的图5)。
HPLC分离之后对收集的级分进行生物检测(图5)
冻干图4中箭头所示的级分(峰),用1ml RPMI培养基(不含血清)重构每个级分并使用KS-Y1细胞检测其生物活性。由于最初的原料是以10,000IU HCG提供的,依此类推,任意假定一个级分应任意含有10,000IU的抗KS活性。根据这种假设,贯穿本申请的始末一直对剂量进行评价。在缺乏(0)或存在不同剂量(10,100和200IU/ml)的情况下检测生物活性。以“mix”表示的级分代表通过将等量的级分A-E混合而制得的1个收集物。从图5可以看出级分D,E和“mix”显示出抑制活性。
通过MALDI-TOF质谱分析活性级分(HIP)(图6和7)
简单地说,将各个样品的等分试样包埋于低分子量的UV-吸附基质(α-氰基-4-羟基肉桂酸)以增强样品的离子化,然后在Voyager-DelayedExtraction系统(Perseptive Biosystem,Framingham,MA)中进行MALDI-TOF(Matrix Assisted Laser Desorption Ionization Time of Flight)质谱分析。
观察到一个主要的峰含有约为13000道尔顿的组成成分(图6)。为了进行比较,也显示出级分A-C和E的质谱分析(图7),注意到在级分D和E中都存在13000道尔顿的组成成分(图6)。
HIP对HIV表达的影响(图9)
较早显示出可抑制KS细胞增殖的低分子量的级分#7(Kachra等,1997,Endocrinology,138:4038-4041),检测该级分的抗HIV活性。按文献所述(Tremblay等,1994,EMBO J,13:774)用病毒HIV-IIIB感染原代培养的人淋巴细胞。感染后立即每天用1-250IU当量的指定剂量的受试物(HCG级分或重组的HCG)处理细胞达10天。随后,裂解细胞,按文献所述(Tremblay等,1994,EMBO.J,13:774)检测HIV病毒蛋白质p24的表达。注意到用高剂量的含HIP的级分处理之后p24表达显著降低,而重组的HCG未显示出显著的影响(图9)。
对HIP级分中所含的蛋白质进行测序(图10A和10B)
HPLC分离之后,按上述检测级分的生物活性(图4和5)。使用自动测序装置(Applied Biosystem生产的比475型先进的470型气相测序仪)对含有最高生物活性的2个级分进行蛋白质测序。使用的内标由pTH-Nor-Leu组成。最初的生产效力约为50±20pmol。15轮循环之后,使用常规的蛋白质分析检查所产生的数据。将推定的氨基酸序列与公开的GenBankTM数据库相比较。发现两个序列与HCG的α和β亚单位具有显著的同源性。
结论
本发明的结果提供证据表明:存在一种潜在重要的化合物,该化合物可能通过AP-1途径的信号传导抑制KS生长。尽管目前正在对纯化的活性分子进行测序,但很明显该活性分子既不是HCG也不是其典型未知的亚单位中的任一种。
由其凝胶渗透层析洗脱结果判断,其大小相对较小,可能低于10,000-14,000。为了获得进一步的分离,使用了HPLC技术,将蛋白质分成独立的和不同的峰。发现特定的单峰具有抗KS活性。为了得到其它数据,通过聚丙烯酰胺凝胶电泳,接着进行银染色以分析单峰。当能肉眼看到蛋白质时,观察到“模糊的”带,这表明蛋白质包括密切相关的种类。
此时,人们仅可推测是否它作为一裂解肽衍生自HCG。实际上,已知糖蛋白激素被代谢成更小的多肽(Sairam MR,1983,激素蛋白质和肽,LiC.H编,p1-79)。推定的裂解物(或相关产物)会经由修饰的HCG受体产生作用。实际上,已知HCG受体基因可以受发育调节的方式被表达成选择剪接的变体转录物(Segaloff DL等,1993,Endocr Rev 14:324-347),产生了这样一种可能性,即推定的产物可介导激素作用的不同方面。平行实验进一步强化了这一假说,该实验的结果表明:KS组织和细胞系表达显著水平的HCG受体,其大小和细胞内的分布不同于典型的靶细胞(Cao H.,Sairam M.R和Antakly T.,美国癌症研究协会1996年会,摘要#1543)。或者,活性物质可能是β-HCG亚单位的降解产物(例如但不限于β-核),其在三维结构上与几个生长因子同源(Lapthorn AJ等,1994,自然369:455-461)。由于KS细胞的启动和增殖在很大程度上依赖于生长因子,β-核片断可能用作生长因子受体的拮抗剂(参见Guo WX等,1996,美国病理学杂志148:1999-2008)。
本发明所含的部分序列支持这样一个观点,即HIP蛋白可能衍生自hCG,作为:1)α或β亚单位的选择表达;或2)hCG亚单位的酶解加工。
尽管结合具体的实施方案描述了本发明,但应理解也能对本发明进行其它修饰,本申请欲覆盖根据本发明的原理的任何变动、用途或修改,并包括本发明所属领域技术人员根据已知或常规实践所得的相对于本申请公开内容的偏离,这一点也适用于上文所述的必要技术特征,它们都包括在所附权利要求书的范围之内。

a)使APL-HCG的生物活性级分或含有所述化合物或蛋白质的尿提取物与聚丙烯塑料支持物接触一段足以吸附所述化合物或蛋白质的时间;和
b)洗涤支持物并释放其上吸附的化合物或蛋白质。
12.评价抗KS和抗HIV化合物的抑制活性的方法,所述方法包括测定AP1基因活性。
13.权利要求12的方法,其中通过测定与DNA反应元件的结合来测定所述AP1基因的活性。
