Description
Technical Field
The application relates to the field of skin care products, in particular to a peptide-containing composition for repairing multiple skin barriers and a preparation method thereof.
Background
With the rapid development of economy and the improvement of the living standard of people, the demands of consumers for anti-aging products are increasing. Especially skin, which is the largest organ of the human body, tends to give priority to aging symptoms.
The aging symptoms of skin often show symptoms such as thinning of skin, abnormal stratum corneum, increased wrinkles, deepening of wrinkles, loose elasticity, increased dead skin, dryness, lack of water and the like. Among them, the most common is that skin elasticity is reduced, sagging and wrinkles are increased and deepened. This is mainly due to collagen loss by elastase.
Although the prior art has greatly advanced research on the anti-aging effect of skin, the prior art has a plurality of defects for developing anti-aging raw materials and skin care products. At present, anti-aging raw materials and skin care products have one-sided performance, and many of the raw materials and skin care products use a single substance with an anti-oxidation effect or use low-content plant extracts, so that the problem of skin aging cannot be solved from root sources. For solving the skin aging problem, the key point is to repair the skin barrier, so that a perfect skin barrier structure can be established and skin dysfunction phenomenon can be avoided, and further skin aging problems caused by the problems of loose skin, skin itch, skin inflammation, increased fine wrinkles, coarse pores, increased color spots, atrophy, scars, red swelling and the like can be avoided.
Therefore, in order to better solve the problems, the application provides a peptide-containing composition for repairing multiple skin barriers and a preparation method thereof, which can solve the problems of dry skin, skin itch, red swelling, inflammation and the like while repairing and perfecting skin microecological barriers, solve the skin aging problem from the root, and can effectively promote skin elasticity, lighten fine lines and further delay skin aging so as to ensure skin health and beauty.

Model control group: deionized water, dosage of 0.2 mL/piece; sample administration group: 5wt% of the composition obtained in examples and comparative examples was administered in an amount of 0.2 mL/dose.
After the adaptive feeding, shearing off the hair at the neck and the back of the mouse by scissors one day before the experiment, unhairing by using unhairing paste to expose the skin, wherein the area of a unhairing area is about 2cm x 2cm, and smearing and administering the mice with intact skin; each dose is administered 1 time a day for 2 days continuously; itching: after the last administration for 1 hour, each group of mice was subcutaneously injected with 0.2mL (10 mL/kg) of 0.05% histamine phosphate physiological saline, and the front paws of the mice were observed to scratch the head, the back paws were sowed to scratch the trunk, and the mouth-biting parts of the whole body were used as the itching reaction indexes. And observing and recording the itching times of each group of mice within 30min, judging the itching relieving effect intensity of the test group according to the itching relieving inhibition rate, and representing the effect of the sample administration group by comparing the itching relieving inhibition rate with that of the model group. Wherein, the calculation formula of the antipruritic inhibition rate is as follows:
antipruritic inhibition ratio (%) = (number of scratches of model control group-number of scratches of administration group)/number of scratches of model control group×100%.
2. Anti-inflammatory efficacy test: the skin care compositions obtained in the examples and the comparative examples were subjected to antipruritic test, and the test subjects were Kunming mice, and the weight of the mice was 20 to 25g.
The testing method comprises the following steps: the mice are grouped randomly, 10 mice in each group are respectively half male and female; (1): blank control group; (2): model control group; (3): a positive control group; (4): the sample administration groups, i.e., examples and comparative examples, each of the experimental mice amounts of the examples and comparative examples were 10.
Model control group: deionized water, dosage of 0.2 mL/piece; positive control group: the dosage of the compound dexamethasone acetate is 0.2 mL/dose; sample group: 5wt% of the composition obtained in examples and comparative examples was administered in an amount of 0.2 mL/dose.
Each group was dosed continuously for 15 days, and each group, except the blank group, induced delayed allergic inflammation in mice using DNCB, in a specific method: the mice were sensitized once on the abdomen on day 1, day 2, and the backs were challenged with 1% DNCB 0uL on day 6, day 9, day 12, and day 15, respectively, using 5% DNCB 25 uL. After the last administration for 1 day, the mice were sacrificed by cervical removal, the skin thickness and weight were measured by punching the dorsal skin, and the inflammatory cytokines were measured by embedding slice pathology observation, immunohistochemistry.
