磷(P)是植物生长和代谢的必要元素之一,直接参与核酸、蛋白质、ATP以及磷脂等生物大分子的合成,涉及调控光合作用、呼吸代谢、能量转化和信号转导等过程,在植物整个生命周期中起到十分重要的作用。据估计,耕地中累积的磷可维持全球最大作物产量约100年。然而,土壤中可被植物吸收利用的P即正磷酸盐(HPO4 2-和H2PO4 -)仅占土壤总P的0.1%~0.5%,难以满足植物正常生长的需要,而其他大部分P主要存在于土壤矿物复合物中,如磷酸三钙、磷酸铁和磷酸铝以及构成土壤有机磷的植酸盐中。在农业系统中通常使用化学磷肥提高土壤肥力和作物产量,对人参适量施磷肥能增强光合作用,增加人参干物质积累。然而,肥料中超过80%的P通过固定、吸附或沉淀等反应,导致植物无法利用,不仅造成资源浪费,还会造成环境污染。
微生物在土壤养分循环中起着关键作用,其中溶磷细菌(Phosphatesolubilizing bacteria,PSB)是土壤磷循环的一个重要组成部分,占土壤可培养细菌的40%以上,能将土壤中不溶性的磷酸盐转化为植物可吸收利用的形式。研究表明,PSB通过分泌有机酸、磷酸酶和植酸酶等代谢产物活化土壤难溶性无机磷和有机磷,释放磷酸根离子供其自身和植物根系吸收。
有鉴于此,特提出本发明。
发明内容
本发明的目的在于提供链霉菌、微生物菌剂及其应用,本发明的链霉菌(Streptomyces pratensis)FF148,是从吉林省敦化紫鑫药业林下基地(128°8'25"E,43°7'45"N,743.68m)分离得到的,向人参施用该菌株或包含该菌株的微生物菌剂,能够提高人参对磷的吸收,进而促进人参的生长。
为了实现本发明的上述目的,特采用以下技术方案:
本发明的一个方面,涉及链霉菌(Streptomyces pratensis)FF148,保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCC No.27219,保藏日期:2023年4月23日;保藏单位地址: 中国 北京。
链霉菌属(Streptomycetaceae)是放线菌中的一属,主要分布于土壤中,其基内菌丝多分枝,一般横隔稀疏,常产生各种水溶性或脂溶性的色素,具有溶磷、产生长素、铁载体和抗生素等功能。
人参等药用植物根际土壤中常见的有益微生物包括芽孢杆菌、假单胞菌和链霉菌等。而链霉菌(Streptomyces pratensis)是林下山参等药用植物根际的优势类群,是有益微生物。本发明从吉林省敦化紫鑫药业林下基地(128°8'25"E,43°7'45"N,743.68m)分离、培养与鉴定了一株链霉菌(Streptomyces pratensis)FF148,该菌株能够溶解土壤中的难溶性磷,促进人参对磷的吸收,促进人参生长,对人参等药用植物栽培产业绿色可持续发展尤为重要。
通过盆栽试验,采用浸泡和灌根方式进行接种,能够确保该菌株稳定定殖。
本发明的另一个方面,还涉及微生物菌剂,包括所述的链霉菌(Streptomycespratensis)FF148及其代谢产物。
含有菌株FF148及其代谢产物的微生物菌剂,具有提高人参吸收磷的能力,能够促进人参的生长。
优选地,所述微生物菌剂包括固体菌剂或液体菌剂。
优选地,所述微生物菌剂为固体菌剂时,所述微生物菌剂含有所述链霉菌(Streptomyces pratensis)FF148的活菌数量为4.2×108~5.6×108cfu·g-1;
优选地,所述微生物菌剂为液体菌剂时,所述微生物菌剂含有所述链霉菌(Streptomyces pratensis)FF148的活菌数量为6.1×108~7.3×108cfu·mL-1。
优选地,每1kg土壤施用100~200mL该微生物菌剂与土壤混合均匀,也可以选择该菌株悬浮液对人参苗进行蘸根和/或灌根处理。
优选地,所述微生物菌剂为固体菌剂时,所述微生物菌剂还包括:载体和粘接剂。
优选地,所述载体包括:蛭石、草炭或纤维素基土壤保水剂中的至少一种。
优选地,所述粘接剂包括:果胶、海藻糖或蚕丝蛋白中的至少一种。
优选地,所述微生物菌剂还包括农业上有效量的肥料和/或农药。
本发明的另一个方面,还涉及一种促进人参生长的栽培方法,向人参施用所述的链霉菌(Streptomyces pratensis)FF148或所述的微生物菌剂。
本发明的另一个方面,还涉及所述的链霉菌(Streptomyces pratensis)FF148或所述的微生物菌剂在促进人参生长或促进人参对磷的吸收中的应用。
与现有技术相比,本发明的有益效果为:
(1)本发明的链霉菌(Streptomyces pratensis)FF148是从林下山参根际土壤分离获得,属于有益微生物。
(2)本发明的链霉菌(Streptomyces pratensis)FF148,能促进人参生长、提升人参的品质、增强对磷素吸收,有效解决人参栽培时,磷的利用效率低的问题。

具体实施方式
下面将结合附图和具体实施方式对本发明的技术方案进行清楚、完整地描述,但是本领域技术人员将会理解,下列所描述的实施例是本发明一部分实施例,而不是全部的实施例,仅用于说明本发明,而不应视为限制本发明的范围。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。实施例中未注明具体条件者,按照常规条件或制造商建议的条件进行。所用试剂或仪器未注明生产厂商者,均为可以通过市售购买获得的常规产品。
实施例1链霉菌(Streptomyces pratensis)FF148的分离与培养
从林下山参根际土壤分离获得。称取根际土壤10g,添加90ml无菌水于500mL锥形瓶中,于28℃,180rpm的摇床充分震荡30min,静置10min后制得稀释度为10-1土壤悬浊液。用无菌水将悬浊液按10倍的浓度梯度依次进行稀释,取稀释倍数为10-3、10-4、10-5的土壤悬液待用。将配好的TSA培养基倒入培养皿中,使其铺满培养皿底部,等其凝固;吸取100μl不同浓度梯度的土壤悬浮液分别加入到TSA培养基中,用涂布棒将土壤稀释液均匀涂布到整个培养基上,使用封口膜封住培养皿,放置28℃恒温培养中培养7d。待TSA培养基长出菌落,选择性状、颜色、大小有差异的单菌落,将单菌挑选至背面划有方格线的TSA培养基中,28℃培养箱中继续培养7天,将单菌挑至含有1mL TSB液体培养基的1.5mL的离心管中,28℃摇床上180r·min-1摇7天至菌液浑浊待用。
实施例2链霉菌(Streptomyces pratensis)FF148的纯化与保存
在鉴定前用平板划线法对分离得到菌株进行纯化。取提前制备好的FF148浑浊菌液,在无菌操作内,使用灭菌的接种环蘸取菌液,在TSA平板上做“Z”字形划线,剩余菌液4℃冰箱保存备用;将接种后的TSA平板于28℃恒温培养箱中倒置培养7d,期间观察菌落生长情况,是否有杂菌污染。
实施例3链霉菌(Streptomyces pratensis)FF148的形态学鉴定
将菌种接种于TSA的培养基上,倒置于28℃培养箱中培养2~3d,出现单菌落后,观察菌落质地、形状和大小、边缘是否整齐、表面是否光滑、有无隆起形状、透明度、菌落及培养基的颜色等。由图1可见FF148菌落较小,微黄色,边缘不整齐,表面粗糙隆起,不透明。
实施例4链霉菌(Streptomyces pratensis)FF148的分子生物学鉴定
取4μL菌液作为PCR扩增的模板,利用细菌16S rDNA的通用引物799F(5′-AACMGGATTAGATACCCKG-3′)和1193R(5′-ACGTCATCCCCACCTTCC-3′)对菌株序列进行扩增,50μL PCR反应体系为菌液模板4μL,2×Taq MasterMix(GenStar)25μL,799F/1193R各1.7μL,dd H2O 17.6μL。PCR反应条件94℃预变性2min,94℃变性30s,56℃退火30s,72℃延伸1min,共30个循环,最后72℃延伸5min。所得PCR产物通过1%的琼脂糖电泳,将能检测到目的条带的PCR原菌液送往生工生物工程(上海)股份有限公司测序,测得的序列输入到NCBI中,应用Blast和DNAMAN软件进行比对分析,在GenBank中找到同源性非常高的相似菌株序列,通过16S rDNA序列长度为392bp,与FF148菌株相似性最高的是Streptomyces pratensis,同源性达到99.74%,根据MEGA 11软件以UPGMA法构建系统发育树(如图2)发现,FF148与Streptomyces pratensis同属一个遗传分支,亲缘关系十分接近。结合形态学分类和分子生物学鉴定结果,可以确认本发明的菌株FF148为Streptomyces pratensis,并将其保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCCNo.27219。菌株FF148的16S rDNA序列如SEQ ID NO.1所示。

实施例5链霉菌(Streptomyces pratensis)FF148的溶磷能力的测定
将链霉菌(Streptomyces pratensis)FF148菌株转移至TSB液体培养基,28℃、180r/min摇床培养3d,OD值调整为0.8。吸取细菌悬浮液0.1mL于无机磷固体培养基中央。每个菌株重复3次,28℃倒置培养7d,测量菌落直径(d)和透明圈直径(D),计算溶磷指数(D/d)。同时,吸取细菌悬浮液1mL接种到装有100mL无机磷液体培养基的250mL三角瓶中,以未接菌株为对照,重复3次,28℃、180r/min摇床培养7d,培养液以10000r/min离心后,取上清液用钼锑抗比色法测定培养液中的可溶性磷含量。结果见表1,菌株FF148具有较好的溶磷能力。

实施例6链霉菌(Streptomyces pratensis)FF148的活化
将在4℃保存的FF148菌种进行活化,吸取10μL冻存的菌液接种于1mL新的TSB液体培养基,28℃下180rpm·min-1摇床培养7d,至菌液浑浊;吸取2μl活化后的菌液于分光广度计上测量600nm的OD值,使用无菌水将菌液OD值调整为0.2,备用。
实施例7链霉菌(Streptomyces pratensis)FF148的人参根系接种定植
选取大小一致,芽孢未损坏且表面消毒的一年生人参苗,用蒸馏水冲洗干净,使用OD值为0.2的菌液将参苗提前浸泡30min待用。将土壤消毒后,装入直径为18cm的花盆中,每盆装1.5kg土。将提前浸泡过菌液的人参移栽到土壤中,移栽后取10mL菌悬液进行灌根处理。对照接种无菌水,土壤用等量无菌水处理,待人参生长90d后,进行观察。
实施例8
(1)链霉菌(Streptomyces pratensis)FF148对人参生长的作用:
分别将对照和接菌处理的人参幼苗取出,分为地上部分和地下部分,测定人参地上部分鲜重、干重、茎长、叶面积、叶绿素、PSⅡ最大光和效率、P含量和P吸收效率;测定人参地下部分鲜重、干重、根长、根表面积、根投影面积、根体积、根尖数量和P含量。

由表2可知,处理组的鲜重、干重、茎长、PSⅡ最大光和效率、P含量和P吸收效率均显著高于对照组。说明菌株FF148能够促进人参地上部的生长,促进对磷的吸收。

由表3可知,处理组的鲜重、根长、根投影面积、根体积和P含量均显著高于对照组,说明菌株FF148能够促进人参地下部的生长,促进对磷的吸收。
(2)链霉菌(Streptomyces pratensis)FF148对人参根际土壤的作用:
分别将对照和接菌处理的人参幼根际土取出,晾干过100目筛,测定人参土壤pH、土壤有机质、土壤C含量、土壤速效磷含量、酸性磷酸酶活性和植酸酶活性。

以上结果表明,与对照处理相比,FF148菌悬液处理的人参磷含量含量增加,根系分支增多,证明链霉菌(Streptomyces pratensis)FF148对人参生长和品质具有显著地促进作用。
尽管已用具体实施例来说明和描述了本发明,然而应意识到,以上各实施例仅用以说明本发明的技术方案,而非对其限制;本领域的普通技术人员应当理解:在不背离本发明的精神和范围的情况下,可以对前述各实施例所记载的技术方案进行修改,或者对其中部分或者全部技术特征进行等同替换;而这些修改或者替换,并不使相应技术方案的本质脱离本发明各实施例技术方案的范围;因此,这意味着在所附权利要求中包括属于本发明范围内的所有这些替换和修改。

1.链霉菌(Streptomyces pratensis)FF148,保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCC No.27219。
2.微生物菌剂,其特征在于,包括权利要求1所述的链霉菌(Streptomyces pratensis)FF148及其代谢产物。
3.根据权利要求2所述的微生物菌剂,其特征在于,所述微生物菌剂包括固体菌剂或液体菌剂。
4.根据权利要求3所述的微生物菌剂,其特征在于,所述微生物菌剂为固体菌剂时,所述微生物菌剂含有所述链霉菌(Streptomyces pratensis)FF148的活菌数量为4.2×108~5.6×108cfu·g-1;
优选地,所述微生物菌剂为液体菌剂时,所述微生物菌剂含有所述链霉菌(Streptomyces pratensis)FF148的活菌数量为6.1×108~7.3×108cfu·mL-1。
5.根据权利要求3所述的微生物菌剂,其特征在于,所述微生物菌剂为固体菌剂时,所述微生物菌剂还包括:载体和粘接剂。
6.根据权利要求5所述的微生物菌剂,其特征在于,所述载体包括:蛭石、草炭或纤维素基土壤保水剂中的至少一种。
7.根据权利要求5所述的微生物菌剂,其特征在于,所述粘接剂包括:果胶、海藻糖或蚕丝蛋白中的至少一种。
8.根据权利要求2所述的微生物菌剂,其特征在于,所述微生物菌剂还包括农业上有效量的肥料和/或农药。
9.一种促进人参生长的栽培方法,其特征在于,向人参施用权利要求1所述的链霉菌(Streptomyces pratensis)FF148或权利要求2~8任一项所述的微生物菌剂。
10.如权利要求1所述的链霉菌(Streptomyces pratensis)FF148或权利要求2~8任一项所述的微生物菌剂在促进人参生长或促进人参对磷的吸收中的应用。
