背景技术
随着时代的变化,对美容的关注日益提高的过程中,毛发占据重要的一部分。毛发是在皮肤表面生产的细而角质化的结构。这不仅具有对外部冲击的缓冲作用,还起到保护人体免受直射光线、寒冷、摩擦、危险等外界刺激,并且使对身体有害的砷、汞、锌等的重金属排出到体外的功能,到了现代还被强调装饰的美容方面。然而,因包括饮食生活变化和内外压力增加在内的各种原因而抱怨脱发的人越来越多。众所周知,对于脱发,遗传因素是最主要的原因,但如上所述,社会压力的增加、环境污染、频繁的烫发和染色以及错误的头皮管理也正成为脱发的原因。
人体头皮的毛发为约10万-15万根左右,每根毛发都有彼此独立的生长周期。在发生初期毛囊中,通过β-连环蛋白的作用由基底层表皮细胞形成基板(placode)。从形成的基板接收信号的真皮成纤维细胞形成皮肤凝聚体(dermal condensate)。同时,基板向真皮方向生长、陷入,与皮肤凝聚体结合,形成一次毛发胚种(primary hair germ)。形成毛发胚种的表皮细胞向发生的真皮方向延伸生长,并形成作为由多层组成的长柱形结构物的毛栓(hair peg),同时皮肤凝聚体形成为球形的毛乳头(dermal papilla)。毛栓的终端逐渐变厚,形成毛球(hair bulb),毛球变成包裹毛乳头的形态,完成毛囊,毛囊内部的表皮细胞分化而生成毛发。
毛发生长周期由三个阶段构成,分类为毛发生长最活跃的生长期(anagenstage)、毛发开始退化的退化期(catagen stage)以及毛发的生长停止或进入休息的休止期 (telogen stage)。具体而言,生长期为3~6年,退行期为2~3周,休止期为3~4个月左右,整个毛发中各自的比重约为88%、1%、11%,大体上呈现与维持期间成正比的倾向。生长期是毛乳头的毛母细胞迅速进行有丝分裂的时期,生成角蛋白,使毛发生长。之后,如果到了退行期,则生长速度变慢,细胞分裂停止。另外,在退行期,毛囊逐渐收缩,毛乳头萎缩,毛根被推上去,导致毛发脱落。当在退行期长出新毛发时,新长出的毛发会推出现有的毛发并与休止期重叠,开始新的生长期(new anagen)。
脱发有多种情况,但如果按生物体周期区分,则区分为生长期脱发症和休止期脱发症。生长期脱发症(Anagen Effluvium)可能是在生长期快速代谢并分裂的毛乳头的细胞受到细胞毒性制剂(例如抗癌剂)的影响而发生的。由于抗癌剂起到杀死癌细胞等快速代谢的细胞的作用,因此诸如生长期的毛母细胞等快速分裂的细胞也会同时受到影响。因此,可能处于以下状态:代谢变慢的休止期的毛发会原样保留,因此仅保留整个毛发中约10%的毛发。这种生长期脱发症一旦去除外部毒性刺激源,就会重新回到原来的状态。
休止期脱发症是,生长期的毛发因外部刺激转化为休止期,从而原本约11%左右的休止期的毛发的比例增加。这种休止期脱发症可因分娩、疾病、严重的头发梳理等而诱发。特别是,生长期的毛发因诸如在日常生活中梳理头发等外力转化为休止期从而诱发的脱发是多数感觉到发量减少的中老年成人正在经历的问题。
日常脱落的毛发每天为50~150根左右,但这些毛发不是在一天内慢慢脱落的,而大部分是因洗头并梳理的过程中施加的外力而脱落的。这样,减少因日常中施加的外力引起的毛发脱落也可帮助防止脱发。
迄今为止已知的脱发防止产品中包含以促进血液循环、强化毛根功能、头皮保湿效果、头屑防止效果、抗氧化效果、毛发生长期延长效果或抑制雄性荷尔蒙作用等为目的的成分等作为有效成分。即,用于改善脱发的现有的方式是向头皮供给营养或者调节毛发生物体周期等的方式,聚焦于使毛发重新生长。然而,聚焦于生发的现有的脱发防止产品无法解决因外力引起的毛发脱落问题。因此,实情是需要开发一种能够有效地减少毛发脱落的头发护理组合物。
发明内容
要解决的课题
本公开要解决的课题是提供一种头发护理组合物,其通过收缩立毛肌来减少毛发脱落,由此可以防止和/或改善脱发。
课题的解决手段
本发明人为了解决传统脱发治疗剂无法解决因外部环境或压力等引起的暂时性毛发脱落问题的问题而进行了深入研究。其结果,本发明人发现,如果将包含烟酸(niacin)和丹醇(deanol)的头发护理组合物施用于头皮,则令人惊讶的是,立毛肌(arrector pili muscle)收缩,从而产生优异的毛发脱落减少效果,由此表现出卓越的脱发防止和/或改善效果。
本公开提供一种用于防止和/或改善脱发的头发护理组合物,其包含烟酸和丹醇作为有效成分。烟酸和丹醇的组合在减少毛发脱落方面产生显著的协同效果。此外,毛发脱落减少效果通过长期使用表现出显著优异的脱发防止和/或改善效果。
在本公开的一个方面,烟酸可以通过激活诸如NIACR1(烟酸受体1,niacinreceptor 1)、NIACR2等G蛋白受体来诱导立毛肌的收缩。另外,丹醇作为乙酰胆碱的前体,可以通过增加末梢组织的血液中乙酰胆碱浓度来收缩立毛肌。由于上述烟酸和丹醇的受体不同,因此可以通过相互不同的机制来收缩立毛肌,从而产生协同效果。由于上述烟酸的收缩效果是直接的和瞬时的,并且上述丹醇的收缩效果是间接的和长期的,因此协同效果可以优异。
在本公开的一个方面,相对于组合物的总重量,本公开的用于防止和/或改善脱发的头发护理组合物中包含的包含烟酸和丹醇的有效成分的总含量可以为0.001至 12重量%,优选0.01至7重量%,更优选0.1至5重量%。当相对于组合物的总重量,上述有效成分的总含量小于0.001重量%时,脱发防止和/或改善效果不显著,并且当超过12重量%时,剂型的稳定性降低,因此不优选。
在本公开的一个方面,相对于组合物的总重量,上述烟酸的含量可以为0.001至10重量%,优选0.01至5重量%,更优选0.1至3重量%。相对于组合物的总重量,上述丹醇的含量可以为0.001至10重量%,优选0.01至5重量%,更优选0.1至3重量%。在本公开中,通过实验确认了当烟酸或丹醇分别在上述含量范围内时,毛发脱落减少功效显著优异。
在本公开的一个方面,上述烟酸和丹醇的重量比可以为1:0.1~10,优选1:0.5~8,更优选1:0.8~5(烟酸:丹醇)。当上述烟酸和丹醇的混合物以上述特定的重量比组合时,毛发脱落减少效果显著优异。
在本公开的一个方面,上述用于防止和/或改善脱发的头发护理组合物可以表现出比与不包含烟酸和丹醇的对照群优异20%以上、优选23%以上、更优选25%以上的毛发脱落减少效果。
在本公开的一个方面,上述用于防止和/或改善脱发的头发护理组合物在与现有的脱发缓解有效成分组合使用时可以进一步提高脱发防止和/或改善效果。除了现有的脱发缓解有效成分的作用机制之外,上述烟酸和丹醇可以通过收缩立毛肌来在防止和/或改善脱发方面表现出提高效果。上述现有的脱发缓解有效成分可以是选自由右泛醇、水杨酸、腺苷、米诺地尔、烟酰胺、七叶素、L-薄荷醇、何首乌提取物、人参提取物、绿茶提取物、咖啡因、生物素、吡啶硫酮锌和生育酚等组成的组的任一种以上,但不限于此。
在本公开的一个方面,上述用于防止和/或改善脱发的头发护理组合物可以用作医药组合物、医药外品(quasi-drug)用组合物、化妆料组合物或健康功能食品组合物。本公开的组合物通常还可以剂型化成可适用于皮肤的任何形式,但优选地可以剂型化成皮肤外用剂的形式。本公开的组合物可以制备成可适用于皮肤的剂型,例如液体、霜状、膏状或固体状等。
当本公开的剂型是液体时,作为载体成分,可以使用溶剂、溶解化剂或乳浊化剂等,例如,可以使用水、乙醇、异丙醇、碳酸乙酯、乙酸乙酯、苯甲醇、苯甲酸苄酯、丙二醇、1,3-丁基乙二醇油、甘油脂肪族酯、聚乙二醇或脱水山梨糖醇的脂肪酸酯等。
当本公开的剂型是膏、霜或凝胶时,作为载体成分,可以使用动物性油、植物性油、蜡、石蜡、淀粉、黄芪胶、纤维素衍生物、聚乙二醇、硅酮、膨润土、氧化硅、滑石或氧化锌等。
当本公开的剂型是粉或喷雾时,作为载体成分,可以利用乳糖、滑石、氧化硅、氢氧化铝、硅酸钙或聚酰胺粉等。特别是,当为喷雾剂型时,还可以包括推进剂,例如氯氟烃、丙烷/丁烷或二甲醚等。
除了上述有效成分之外,本公开的组合物还可以包含通常用作可适用于皮肤的外用剂的成分。例如,可以附加地包含选自由水、表面活性剂、保湿剂、低级醇、螯合剂、杀菌剂、防氧化剂、防腐剂、色素和香料等组成的组的一种以上的添加剂。
另外,本公开提供一种头发或头皮用产品,其包含上述用于防止和/或改善脱发的头发护理组合物。上述头发或头皮用产品可以是选自由生发剂、头皮诊疗(scalp clinic)剂、头皮去垢剂、头皮按摩剂、头皮护理剂、清洁剂、洗发水、滋补剂(tonic)、头发调理剂(hair conditioner)、发露、凝胶、发膜(pack)、霜、精华液、粉、喷雾、油、皂、软膏、头发定型剂(hair styling agent)、染毛剂和烫发剂等组成的组的任一种,但不限于此。在本公开的一个方面,上述用于防止和/或改善脱发的头发护理组合物可以制备成洗发水或处理剂(treatment)剂型(实施例1至4)。
另外,本公开提供一种包含烟酸和丹醇作为有效成分的用于缓解头皮瘙痒症的头发护理组合物、用于改善头皮问题的头发护理组合物、用于减少头皮皮脂的头发护理组合物或用于缓解头皮刺激的头发护理组合物。从本公开的实验例3的官能评价结果来看,对于本公开的头发护理组合物,不仅脱发防止效果,而且头皮瘙痒症缓解、头皮问题改善、头皮皮脂减少以及头皮刺激缓解效果也显著优异。
本公开的用于防止和/或改善脱发的组合物优选地可以以直接涂布或喷洒于皮肤等的经皮给药方法使用。
另外,本公开提供一种脱发防止和/或改善方法,其包括将包含烟酸和丹醇作为有效成分的用于防止和/或改善脱发的头发护理组合物给药于需要预防和/或改善脱发的患者(subject),或者涂布于患者的头皮或毛发的步骤。
本公开中使用的术语“给药”可以是指通过任何适当的方法引入本公开的组合物。对于本公开的组合物的给药途径,可以通过任何一般途径给药,只要能够到达目标组织即可。优选地可以经皮给药,其中,局部涂布是最优选的。本公开的组合物的适用次数可以根据处方、需要或期望而决定。
本公开的用于防止和/或改善脱发的头发护理组合物的使用量可以根据年龄、病变的程度等的个体差异或剂型而适当地调节,通常优选1天1次至数次将适当量涂布于头皮,使用1周至数月。在本公开的实验例1~4中,将用于防止和/或改善脱发的洗发水和处理剂组合物从涂布时间起3分钟后冲洗使用,其结果确认表现出优异的脱发防止和/或改善效果。
本公开中记载的所有成分优选地不超过韩国、中国、美国、日本等的相关法规、规范(例如,化妆品安全标准等相关规定(韩国)、化妆品安全技术规范(中国)、食品公典(韩国)、食品添加物公典(韩国)或健康功能食品公典(韩国))等中规定的最大使用值。即,优选地,本公开的医药组合物、医药外品用组合物、化妆料组合物或健康功能食品组合物以各国的相关法规、规范中允许的含量限度包含本公开的成分。
发明效果
本公开的用于防止和/或改善脱发的头发护理组合物在涂布于头皮时通过收缩立毛肌来减少毛发脱落,由此具有显著优异的防止脱发的效果。

下面,为了帮助理解本发明,将列举实施例等进行详细说明。然而,本发明的实施例可以变形为各种不同形式,并且本发明的范围不应被解释为限于以下实施例。本发明的实施例是为了向本发明所属领域的普通技术人员更完整地说明本发明而提供的。
实施例组合物的制备
以下表1中公开的洗发水处方和表2中公开的处理剂处方为基础,通过常规方法制备洗发水组合物和调理剂组合物。比较例1~4和实施例1~4的有效成分根据下表3 中公开的处方各自不同地包含。另外,将比较例1~4和实施例1~4制备成以相应含量包含各自的有效成分的洗发水组合物和处理剂组合物。具体而言,添加聚合物,然后依次投入表面活性剂,溶解,并添加EDTA·4Na和含水柠檬酸来中和pH。作为其他成分,添加防腐剂、香料、分散剂、粘度调节剂和pH调节剂。

实验例1:实施例组合物的毛发脱落减少效果验证
招募20名苦恼脱发或毛发脱落的受试者,对洗发和造型(styling)中脱落的毛发进行统计并评价。具体而言,在规定的评价室的排水口设置过滤网,并且使受试者使用洗发水和处理剂,以各自平时洗头的方式洗头。但是,为了统一有效成分传递的时间,从涂布时间起3分钟后冲洗干净,这同样适用于洗发水和处理剂。
统计在洗发水和处理剂全部完成后聚集在过滤网上的毛发个数。之后使用毛巾和吹风机弄干湿毛发,进行30次梳理,并将在该过程中脱落的毛发也全部回收,然后比较总统计的毛发数。对于评价,每个实验群各实施3次,对照群使用用上表1和2 的组成制备的洗发水和处理剂,并且实施例的有效成分与下表3中公开的组成相同。在下表3中,对于每个实施例的毛发脱落减少率,将与对照群相比毛发脱落减少的比例以百分比(%)标记。
为了确认烟酸和丹醇组合时的协同效果,采用以下科尔比(Colby)式来比较预测值 (科尔比值)和实测值,并整理于表3中。
科尔比式:E=(A+B)-(A*B/100),E:预测值,A:有效成分A的药效,B:有效成分B的药效,A+B:相加效果,如果实测值大于预测值,则判断具有协同效果。

从上表3中示出的结果来看,从比较例1~4中可以确认,烟酸或丹醇表现出通过收缩立毛肌来减少毛发脱落的效果。此外,从实施例1~4中可以确认,与各自单独使用时相比,烟酸和丹醇的组合对毛发脱落减少效果显著优异,从而产生协同效果。
实验例2:毛发脱落减少效果的比较评价
为了确认与现有的已知具有毛发脱落减少效果的现成产品相比的效果,将实施例4与现成产品进行比较,并比较毛发脱落减少率(图1)。上述现成产品作为脱发症状缓解洗发水,包含咖啡因、水杨酸钠、薄荷醇、生物素、人参前草提取物、生育酚、泛醇、烟酰胺和β-胡萝卜素作为脱发缓解有效成分。
以与上述实验例1相同的方法,招募20名苦恼脱发或毛发脱落的受试者,对洗发和造型中脱落的所有毛发进行统计并比较评价。从图1中公开的评价结果来看,可以确认实施例4的毛发脱落程度显著低于现成产品,因此实施例4的毛发脱落减少效果优异。
实验例3:实施例4的毛发脱落官能评价
招募30名苦恼脱发或毛发脱落的受试者,对当使用实施例4的洗发水和处理剂组合物时能否体感到毛发脱落减少效果进行官能评价,其结果示于表4中。对每个问卷的回答根据下表5中公开的标准进行分数化。作为上述回答,将“根本不是”和“不是”分类为否定回答,将“是”和“非常是”分类为肯定回答,计算各自的比例,并示于表4中。从下表4中示出的结果来看,确认了当使用实施例4的洗发水和处理剂组合物时,脱发缓解、瘙痒症缓解、问题改善、皮脂减少以及头皮刺激缓解效果优异。

实验例4:与现有的脱发缓解成分组合时脱发防止官能评价
为了评价将本发明的头发护理组合物的有效成分与现有的脱发缓解有效成分组合使用时的效果,实施了官能评价。
现有的脱发缓解有效成分使用了食药处认证的脱发缓解用功能性化妆品的有效成分组合,即右泛醇0.2%、水杨酸0.25%、L-薄荷醇0.3%。组合使用例在实施例4 的有效成分中附加地组合上述现有的脱发缓解有效成分而制备。另外,在本实验例中,制备了不包含有效成分的剂型作为对照群。

招募45名苦恼脱发或毛发脱落的男女受试者,每组各15名使用通过上述制备方法制备的洗发水和处理剂3个月。统计了由上述受试者根据下表7中公开的项目的回答,并且统计结果示于表8中。

从上表8中示出的结果来看,当与实施例4相比,在上述组合使用例中毛发脱落减少和脱发防止效果更加优异时,可以确认通过将烟酸和丹醇与现有的脱发防止有效成分组合来收缩立毛肌,从而进一步提高脱发防止效果。
Claims (12)
1.一种用于防止或改善脱发的头发护理组合物,其包含烟酸和丹醇作为有效成分。
2.根据权利要求1所述的用于防止或改善脱发的头发护理组合物,其中,相对于组合物的总重量,所述有效成分的含量为0.001至12重量%。
3.根据权利要求1所述的用于防止或改善脱发的头发护理组合物,其中,所述烟酸和丹醇的重量比为1:0.1~10(烟酸:丹醇)。
4.根据权利要求1所述的用于防止或改善脱发的头发护理组合物,其中,所述组合物通过收缩立毛肌来减少毛发脱落。
5.根据权利要求1所述的用于防止或改善脱发的头发护理组合物,其中,所述组合物是医药组合物、医药外品用组合物、化妆料组合物或健康功能食品组合物。
6.根据权利要求1至5中任一项所述的用于防止或改善脱发的头发护理组合物,其中,所述组合物还包含选自由右泛醇、水杨酸、腺苷、米诺地尔、烟酰胺、七叶素、L-薄荷醇、何首乌提取物、人参提取物、绿茶提取物、咖啡因、生物素、吡啶硫酮锌和生育酚组成的组的任一种以上。
7.一种头发或头皮用产品,其包含权利要求1所述的用于防止或改善脱发的头发护理组合物。
8.根据权利要求7所述的头发或头皮用产品,其中,所述头发或头皮用产品选自由生发剂、头皮诊疗剂、头皮去垢剂、头皮按摩剂、头皮护理剂、清洁剂、洗发水、滋补剂、头发调理剂、发露、凝胶、发膜、霜、精华液、粉、喷雾、油、皂、软膏、头发定型剂、染毛剂和烫发剂组成的组。
9.一种用于缓解头皮瘙痒症的头发护理组合物,其包含烟酸和丹醇作为有效成分。
10.一种用于改善头皮问题的头发护理组合物,其包含烟酸和丹醇作为有效成分。
11.一种用于减少头皮皮脂的头发护理组合物,其包含烟酸和丹醇作为有效成分。
12.一种用于缓解头皮刺激的头发护理组合物,其包含烟酸和丹醇作为有效成分。
