本发明包含小分子肽药物递送平台的设计以及制备技术领域,具体涉及一种基质金属蛋白酶-2响应性小分子肽及应用。
背景技术
化疗是肿瘤治疗的常用方法,但化疗药物缺乏对肿瘤的特异亲和性,最终只有少量药物到达肿瘤部位。同时化疗药物具有严重的毒副反应。纳米药物载体具有独特的增强渗透性和保留性(EPR),在肿瘤治疗领域具有极大的应用前景。自组装多肽具备低毒性、自组装、用途广、可修饰等特点,并且可以在特定条件的诱导下,发生结构变化从而具备新的特性,有望用于肿瘤药物递送。
具有特定响应能力的小分子肽在向肿瘤靶向递送药物的领域中具有极大的探索潜力。

本发明目的在于提供一种基质金属蛋白酶-2响应性小分子肽及应用,该基质金属蛋白酶-2响应性小分子肽,可作为纳米载药载体,可延长包载药物的瘤内滞留时间。

第一:所述基质金属蛋白酶-2响应性小分子肽采用固相合成技术,该技术具有产率高,中间产物少,操作简单等优点;第二:基质金属蛋白酶-2响应性小分子肽载药后可于室温下自组装形成球形纳米颗粒,球形纳米颗粒性质稳定,可长期储存;第三:药物针对性递送平台中含有酶(基质金属蛋白酶-2)敏感基团,利用纳米颗粒所具有的EPR效应,利用RGD肽的选择性靶向作用,增强肿瘤组织的药物摄入;第四:基质金属蛋白酶-2响应性小分子肽纳米载药载体在肿瘤组织表达上调的基质金属蛋白酶-2的诱导下,转化为纳米纤维,可延长包载药物的瘤内滞留时间。

实施例1:载药基质金属蛋白酶-2响应性小分子肽的制备及自组装形貌
本发明的基质金属蛋白酶-2响应性小分子肽是使用微波多肽合成仪,利用固相合成方法合成的。合成纯度在95%以上(见图1、图2),结构式见图15。在室温下,在0.5mmol/L、1mmol/L、4mmol/L的肽溶液中,按质量比加入1/5浓度的抗肿瘤药物DOX后,超声处理3min,放置24h,使用透射电子显微镜(TEM)观测结果。
结果显示:肽溶液在加入DOX后,包载药物的基质金属蛋白酶-2响应性小分子肽自组装为球形纳米颗粒,结果如图3、图4、图5所示。
实施例2:载药纳米颗粒在加入基质金属蛋白酶-2后自组装形貌的改变
取3份肽浓度为1mmol/L的1mL载药肽溶液,分别加入10μL浓度为0.01mg/mL、0.5mg/mL、1mg/mL的基质金属蛋白酶-2溶液,同时分别加入10μL浓度为1mol/L的CaCl2和10μL浓度为15mol/L的NaCl溶液,充分混匀,在37℃下孵育24h后,使用透射电子显微镜观测。
结果显示,包载DOX的肽纳米粒,在基质金属蛋白酶-2的诱导下,载药纳米颗粒发生形态学上的变化,由球形纳米粒转变成纳米纤维结构,如图6、图7、图8所示。
实施例3:基质金属蛋白酶-2响应性小分子肽在包载DOX前后Zeta电位变化
在室温下,配置浓度为1mmol/L的肽溶液1.5mL,为样品1,放置24h后待检;配置浓度为1mmol/L的肽溶液1.5mL,加入肽质量五分之一的DOX,超声处理30min,为样品2,放置24h后待检。配置浓度为1mmol/L的载药(DOX)肽溶液1.5mL,加入15μL浓度为0.5mg/mL的基质金属蛋白酶-2,同时分别加入15μL浓度为1mol/L的CaCl2和15μL浓度为15mol/L的NaCl溶液,为样品3,37℃放置48h;处理完毕后分别测量Zeta电位。
结果显示:样品1的Zeta电位为负,如图9所示;样品2包载疏水性模型药物DOX后,载药肽纳米颗粒的Zeta电位为正,易于吸附到肿瘤细胞的细胞膜,如图10所示;样品3加入基质金属蛋白酶-2后,载药肽的电位减小,但仍为正,此时部分药物被释放,部分药物仍被包载,如图11所示。
实施例4:使用反向高效液相(RP-HPLC)观察基质金属蛋白酶-2酶诱导药物递送载体形变的机理
配置浓度为1mmol/L的肽溶液1.5mL,放置24h后,加入15μL浓度为0.5mg/mL的基质金属蛋白酶-2,同时分别加入15μL浓度为1mol/L的CaCl2和15μL浓度为15mol/L的NaCl溶液,放置于37℃的环境中。样品进样前使用0.22μm的滤头过滤。
结果显示:载药颗粒在加入基质金属蛋白酶-2后,随着洗脱时间的增长,在底物峰的两侧出现新的产物峰。说明此时在基质金属蛋白酶-2诱导下,肽的一级结构发生改变,进一步导致诱导肽载体形貌发生改变,结果如图12所示。
实施例5:包载DOX的纳米药物递送载体的药物释放曲线
采用透析法测量纳米药物递送载体包载DOX的体外药物释放。在室温下,配制浓度为1mmol/L的肽溶液3mL,加入肽质量五分之一的DOX,超声处理30min后,放置24h。将处理好的溶液分为两组,向其中一组加入10μL浓度为0.5mg/mL的基质金属蛋白酶-2溶液,同时分别加入10μL浓度为1mol/L的CaCl2和10μL浓度为15mol/L的NaCl溶液,分别转移到透析袋(MWCO 1000Da)中。通过紫外-可见分光光度计监测载药肽分别在有基质金属蛋白酶-2和无基质金属蛋白酶-2的环境中,载药肽的药物释放行为。将透析袋浸入25mL Hepes溶液中。在设定好的时间,更换透析液,并在每次取样后加入相同体积的新的Hepes溶液。通过紫外-可见分光光度计测量样品在480nm处的吸光度,并计算累积释放的DOX的量。结果如图13所示。
实施例6:使用激光共聚焦扫描显微镜检测小分子肽在肿瘤细胞中的滞留时间
称取4.32mg肽和0.86mg DOX,加入4mL Hepes缓冲液,摇匀,获得浓度为1mmol/L的载药肽(DOX/多肽)溶液;称量0.86mg DOX,加入4mL Hepes缓冲液,摇匀,获得游离DOX溶液;分别超声处理30min,摇床60rpm放置24h待用。将HepG2细胞接种于两块6孔板中,细胞贴壁后,一板加入孔内液体同体积的载药肽(DOX/多肽)溶液;另一板加入孔内液体同体积的游离DOX溶液。分别与HepG2细胞共培养72h后,用DAPI标记HepG2细胞核,用Calcein-AM染液对细胞质染色,DOX自发红色荧光。使用激光共聚焦扫描显微镜观察DOX在细胞中的分布情况,结果如图14所示。
结论:培养72h后,游离DOX组的红色荧光主要集中在细胞质,细胞核几乎观测不到红色荧光强度,表明游离DOX很难滞留在细胞核内;DOX/多肽组的红色荧光在细胞质及细胞核内均可被观测到,且荧光强度均高于DOX组,表明DOX/多肽能够在细胞质及细胞核中长时间滞留。提示本发明所设计的小分子肽所形成的纳米药物递送载体可以延长药物在肿瘤细胞内的滞留时间。

1.一种基质金属蛋白酶-2响应性小分子肽,其特征在于,该肽可作为纳米载药载体,结构式为:

2.一种基质金属蛋白酶-2响应性小分子肽作为纳米载药载体的应用,其特征在于,所述基质金属蛋白酶-2响应性小分子肽为权利要求1所述的可作为纳米载药载体的基质金属蛋白酶-2响应性小分子肽,所述基质金属蛋白酶-2响应性小分子肽载药后在Hepes缓冲溶液中自组装形成球形纳米粒,用以包载抗肿瘤药物。
3.一种纳米载药载体,其特征在于,所述纳米载药载体包含权利要求1所述的可作为纳米载药载体的形成球状纳米粒的基质金属蛋白酶-2响应性小分子肽。
4.根据权利要求3所述的纳米载药载体,其特征在于,所述纳米载药载体响应于肿瘤微环境中的基质金属蛋白酶-2,自组装形貌转变为纳米纤维释药,并延长肿瘤细胞内药物滞留时间;所述基质金属蛋白酶-2的浓度为0.1-10μg/mL。
5.一种纳米载药载体的制备方法,其特征在于,用于制备权利要求3所述的纳米载药载体,该方法包括如下步骤:将基质金属蛋白酶-2响应性小分子肽与抗肿瘤药物共同加入到Hepes溶液中,基质金属蛋白酶-2响应性小分子肽自组装形成载药球状纳米粒。
6.根据权利要求5所述的纳米载药载体的制备方法,其特征在于,Hepes缓冲液的pH为7.4,基质金属蛋白酶-2响应性小分子肽的浓度为0.5-4mmol/L。
7.根据权利要求6所述的纳米载药载体的制备方法,其特征在于,所述基质金属蛋白酶-2响应性小分子肽的浓度为0.5mmol/L。
8.根据权利要求5所述的纳米载药载体的制备方法,其特征在于,包括以下步骤:向浓度为0.5~4mmol/L的肽溶液中,按质量比加入1/5浓度的抗肿瘤药物DOX后,超声3min,室温静置24h后,得纳米载药载体。
