溶菌酶是第一个被报道的人类抗菌蛋白,于1922年由亚历山大·弗莱明从鼻粘液中鉴定报道出来。弗莱明在1928年发现了青霉素,并在20世纪40年代发现了青霉素的治疗用途,并因此获得了1945年的诺贝尔医学奖。这也是“抗生素黄金时代”的开始。在20世纪60年代,耐药微生物病原体的兴起唤醒了人们对AMP作为宿主防御分子的兴趣。AMP的发现可能始于植物,随后是20世纪60年代青蛙中的溴宾蛋白和牛奶中的乳铁蛋白。1981年,汉斯·博曼的一项里程碑式的研究报告了如何将细菌注射到天蚕丝蛾中来诱导一种有效的AMP。另一个重要的进展发生在1987年,扎斯洛夫和他的同事从非洲爪蛙和非洲爪蟾中分离并鉴定了阳离子AMP,命名为“磁蛋白”。在20世纪90年代中期,Hoffmann的研究小组证明了AMP在昆虫宿主防御中的关键作用。1994年,在哺乳动物皮肤中发现了AMP,证明了AMP与哺乳动物宿主防御的相关性。AMP通过静电相互作用与细菌细胞膜结合,以破坏细胞膜或进入细菌以抑制细胞内功能。一些AMP还通过激活免疫细胞或通过引入Toll样受体(TLR)来调节宿主免疫经过40几年的研究,目前已从动物、植物、细菌和病毒中发现了超过2500种抗菌肽。
虽然天然抗菌肽具有普遍的优点,但也存在着某些明显的不足。例如,相当一部分天然的抗菌肽抑菌活性较低、稳定性较差、毒性较高,或者导致真核细胞发生溶血等;另外,部分抗菌肽对耐药菌的抑制效果差,不能满足实际应用的要求;而通过对天然抗菌肽进行改造或全新合成得到的人工抗菌肽可以在很大程度上改善以上某些甚至所有缺点,以适应不同应用需求。
发明内容
本发明针对上述存在的技术问题,提供一种抑菌活性强、可有效抑制耐药菌的抗菌肽AV5。以及提供该抗菌肽在抗菌药中的应用。
为了解决上述技术问题,本发明提供如下技术方案:一种抗菌肽AV5,所述抗菌肽的氨基酸序列为:Alanine-Valine-Glycine-Alanine-Valine。
本发明的抗菌肽AV5中,A和V是短肽前两个氨基酸的缩写,5是氨基酸数,为本领域的常规命名方法。该抗菌肽AV5不但对大肠杆菌和金黄色葡萄球菌有明显的抑制作用,而且具有很低的溶血活性,稳定性好,抗菌活性强,具有高效广谱抑菌作用,可作为抗生素替代品使用。
进一步,所述抗菌肽AV5为α螺旋直链多肽,含有5个氨基酸残基,分子量大小为415.27Da,净电荷数为+1。
所述的抗菌短肽在制备针对开放性伤口的抗菌、抑菌制品中的应用。
所述的应用,所述抗菌肽对大肠杆菌的最小抑菌浓度为150μg/mL;
所述抗菌肽对金黄色葡萄球菌的最小抑菌浓度为250μg/mL;
所述抗菌肽对表皮葡萄球菌的最小抑菌浓度为800μg/mL;
所述抗菌肽对铜绿假单胞菌的最小抑菌浓度为700μg/mL;
所述抗菌肽对副伤寒沙门氏菌B的最小抑菌浓度为500μg/mL;
所述抗菌肽对白色念珠菌的最小抑菌浓度为250μg/mL。
所述的应用,所述的开放性伤口为慢性难愈合伤口、手术缝合伤口、运动损伤、烧烫伤、运动擦伤一类的清洁或污染伤口。
所述的应用,所述抗菌肽应用于制备治疗革兰氏阳性菌或革兰氏阴性菌感染的广谱抗菌药物。
抗生素作为一类重要的抗感染类药物,广泛被应用与临床、畜牧业和水产养殖业等领域,但近年来,由于药物滥用药物残留和细菌耐药性等问题日益严重,越来越多的国家开始寻求抗生素替代品,而AMPs因其独特的生物活性以及不同于传统抗生素的特殊的作用机理,已成为最具潜力的抗生素替代品之一,同时在新型食品、药品、护肤品以及化妆品防腐剂和饲料添加剂等方面的应用也越来越广泛,具有良好的发展前景。
虽然天然AMPs具有普遍的优点,但也存在着某些明显的不足。相当一部分天然的抗菌肽抑菌活性较低、稳定性较差、毒性较高,或者导致真核细胞发生溶血等;另外,部分AMPs对耐药菌的抑制效果差,不能满足实际应用的要求;而通过对天然AMPs进行改造或通过理性设计,合成全新的人工AMPs,可以在很大程度上改善以上某些甚至所有缺点,以适应不同应用需求。

具体实施方式
以下结合具体实施例,对本发明进行详细说明。
本发明抗菌肽AV5产品的氨基酸序列,其序列为:Alanine-Valine-Glycine-Alanine-Valine。
序列特征:序列类型为氨基酸序列,含有5个氨基酸残基(该抗菌肽也可称为抗菌肽AV5),分子量大小为415.27Da,净电荷数为+1。
本实施例抗菌肽AV5产品是采用按照常规多肽固相合成,最终得到的抗菌肽AV5经高效液相色谱分析,其纯度≥97%,其具体的合成步骤如下:
1)树脂溶涨:称取取代度为0.4mmol/g的0.6g 2-Chlorotrityl Chloride Resin树脂,将树脂放入反应管中,加DCM(15ml/g)溶剂,振荡40min。
2)接第一个氨基酸:通过沙芯抽滤掉DCM溶剂,加入3倍树脂物质的量的Fmoc-L-Leu-OH氨基酸,再加入3倍树脂物质的量的DIEA,最后加入少量DMF溶解,振荡1.5h后,用DMF和DCM交替各清洗3遍。
3)脱保护:加10ml 20%哌啶DMF溶液(15ml/g),振荡6min后抽掉哌啶DMF溶液,再加10ml 20%哌啶DMF溶液(15ml/g),振荡20min。
4)检测:抽掉哌啶DMF溶液,取十几粒树脂,用乙醇洗三次,加入茚三酮,苯酚溶液各一滴,105℃~110℃加热3min,变深蓝色为阳性反应。
5)第一次清洗:依次用DMF(10ml/g)清洗两次,
6)封闭:加入甲醇(10ml/g)和3倍树脂物质的量的DIEA,振荡20min后,用DMF和DCM交替清洗3遍。
7)缩合(产生序列中的第二个氨基酸Valine):分别加入3倍树脂物质的量的Fmoc-Lys(boc)-OH氨基酸、PyBoP、HoBT和DIEA到反应管,用尽量少的DMF溶解,反应1.5h。
8)第二次清洗:依次用DMF(10ml/g)清洗一次,DMF(10ml/g)清洗两次。
9)重复二至六步操作,从左到右依次连接序列中的氨基酸,区别在于依次替换步骤2)中的氨基酸为Glycine-Alanine-Valine。
10)最后一个氨基酸连接后,脱保护,按照下列方法洗树脂:DMF(10ml/g)两次,DCM(10ml/g)两次,抽干8min。
11)从树脂上切割多肽:树脂和切割液比例按照10ml/g,恒温振荡150min。(切割液按体积百分数配制,可以为TFA95%、水2.5%、TIS 2.5%)。
12)吹干洗涤:将裂解液用氮气尽量吹干,用乙醚层析出来,再用乙醚洗六次,然后常温挥干。即得粗品肽序。
13)用HPLC-MS分析鉴定多肽:
(1)取粗品肽配制浓度为1mg/ml的溶液。
(2)用0.45μm滤膜过滤溶解液。
(3)分析:取10μl用HPLC-MS分析。流动相是水和乙腈,时间25min,等度洗脱,先将HPLC用等度梯度平衡5min然后进样,梯度水40%,乙腈60%;得到纯度和MS的鉴定。
13)将纯化后的溶液冻干,得到成品。
14)将白色粉末状的多肽,密封包装,-20度保存。
验证试验一:
抗菌肽AV5最小抑菌浓度(MIC)的测定:
采用改良的琼脂平板法评价了该抗菌肽的抗菌活性。以大肠杆菌和金黄色葡萄球菌为模型细菌进行检测和评价。使用前,所有细菌首先在37℃的LB板上培养24h。选择单个菌落,接种到LB肉汤中,在37℃下400rpm振荡孵育过夜。然后,将100μL菌悬液加入5mL新鲜LB肉液中,在37℃孵育至达到对数中期。作为抗菌药物的一个重要指标,本文评价了抗菌肽的最低抑菌浓度(MIC)。简而言之,在离心管中加入4mL浓度为0~400μg/mL的肽灭菌液,然后加入200μL预制备的细菌悬液(1×107CFU/mL)。将其在37℃,200rpm的条件下轻摇孵育2h后,将200μL的混合物均匀涂于LB培养基上,在37℃下孵育24h后计数菌落。仅以微生物悬液作为阴性对照,以含抗生素(青霉素-链霉素)的微生物悬液作为阳性对照。实验重复进行了三次。
验证试验二:
本发明产品抗菌肽AV5的酸碱稳定性试验:
配制1mg/ml的抗菌肽AV5,用各种pH缓冲液(pH=2~12)处理。使用进行高效液相色谱仪进行检测,最后通过观察峰面积大小,来测试抗菌肽AV5 pH的稳定性。最后,以空白对照为基础进行归一化作图,来表现测试抗菌肽AV5 pH的稳定性。结果如图7所示。
结果显示,抗菌肽AV5在pH为2到10时稳定性在80%以上,基于以上结果可知,本发明的抗菌肽AV5具有更广泛的pH适应性,更加方便其用于饲料加工和药物的生产。
验证试验三:
本发明产品抗菌肽AV5的热稳定性检测:
首先,配制1mg/ml的抗菌肽AV5。将其煮沸0、20、40、60、80、100和120分钟,再使用进行高效液相色谱仪进行检测,最后通过观察峰面积大小,来测试抗菌肽AV5的热稳定性。最后,以空白对照为基础进行归一化作图,来表现测试抗菌肽AV5的热稳定性。结果如图8所示。
结果显示,AV5经100℃水浴处理120min,含量仍在50%以上,热稳定性极好。
验证试验四:
首先,配制1mg/ml的抗菌肽AV5。在人工胃液环境中,在不同的时间下,使用进行高效液相色谱仪进行检测,最后通过观察峰面积大小,来测试抗菌肽AV5在人工胃液中的稳定性。结果如图9所示.该肽可以在60min内含量稳定在70%以上,说明该肽能够稳定存在人工胃液中。
验证试验五:
本发明产品抗菌肽AV5的酸碱稳定性试验的抗氧化性测试:
配制1mg/ml的抗菌肽AV5,用各种不同浓度的H2O2处理。使用进行高效液相色谱仪进行检测,最后通过观察峰面积大小,来测试抗菌肽AV5抗氧化性的稳定性。最后,以空白对照为基础进行归一化作图,来表现测试抗菌肽AV5抗氧化性的稳定性。结果如图10所示。
结果显示,在200mM过氧化氢的存在下,该肽仍有80%的存在。当过氧化氢的含量增加后,多肽的含量开始下降。当过氧化氢的量达到600mM时,肽段的量下降得较少,在1000mM前仍保持在20%左右。在200mM过氧化氢的存在下,该肽仍有80%的存在。当过氧化氢的含量增加后,多肽的含量开始下降。当过氧化氢的量达到600mM时,肽段的量下降得较少,在1000mM前仍保持在20%左右。基于以上结果可知,本发明的抗菌肽AV5具有一定的抗氧化性。
验证试验六:
本发明产品抗菌肽AV5的溶血活性检测:
对于抗菌肽急性溶血性能的测定,我们设计实验如下:
(1)取新鲜血液与PBS混合,在4000rpm/min下离心5min,弃去上清液,用无菌的PBS缓冲液洗涤3次,直到离心之后的上清液为无色澄清状态。加入50mL的无菌PBS缓冲液,吹散血细胞,制得血细胞悬浮液。
(2)将抗菌肽用PBS缓冲液溶解,梯度稀释。将每个浓度的溶液与血细胞悬浮液等体积混合,轻微震荡4min,然后在37℃下静置100min,其中Tritonx-100为阳性对照,PBS为阴性对照。
(3)100min后,将混合液在4000rpm/min下离心5min,并将上层液体取出滴加到96孔板中,用酶标仪测定该液体在540nm处的吸光度值。
用SEM来表征小鼠溶血试验后红细胞的形态。将收集的小鼠红细胞浸入2.5%磷酸戊二醛磷酸缓冲液(0.2M,pH7.2)中,4℃过夜,用梯度乙醇(30%、50%、70%、80%、90%、95%、100%)在每个梯度下脱水5分钟。最后,将其进行SEM表征。结果如图11和图12所示。
结果表明,在600μg/ml的浓度下,抗菌肽AV5对红细胞的溶血率仍可忽落不计,说明本发明抗菌肽AV5对红细胞的参透脆性可以忽列不计。
验证试验七:
本发明产品抗菌肽AV5基于MTT的细胞毒性实验:
以L929细胞为模型细胞,采用MTT法和活/死染色法评价抗菌肽的细胞毒性。经过之前的研究,首先制备了一系列不同浓度的样品。简而言之,将L929细胞(每孔10000个细胞)置于96孔板中,在添加10%FBS和1%青霉素-链霉素的RPMI-1640培养液中培养。首先,将制备的抗菌肽在RPMI-1640中浸泡24h,然后向每孔中加入一系列浓度的溶液,并进一步培养24h。每孔加入100μL的MTT(0.5mg/mL),孵育4h。去除培养基后,将细胞完全溶解在100μL的二甲亚砜(DMSO)中,用酶标仪在490nm的测试波长下测定吸光度值。细胞活力以相对于对照组的吸光度表示。
结果如图13和图14所示。通过MTT试验检查抗菌肽AV5的细胞相容性,并通过活/死试验进一步证明。令人惊讶的是,正如培养皿一样,MTT试验显示抗菌肽AV5对L929细胞也几乎没有副作用。
验证试验八:
为了证明该细菌不容易对于抗菌肽产生耐药性,也就是说该抗菌肽有可能抗细菌耐药性。选取大肠杆菌的MIC和对金黄色葡萄球菌的MIC作为实验浓度,同时用抗生素imipenem的MIC作为对比。在细菌培养箱中孵育(37℃,150rpm),每2.5h取一次样(200μL)作为一代,然后取100μL混合液均匀涂抹在LB培养板上孵育(37℃,24h),计数菌落。拍照并将MICn/MIC1数值绘成点线图,观察变化,以大肠杆菌和金黄色葡萄球菌为模型菌,探究抗菌肽修饰物的耐药性。
结果如图15所示。观察变化。抗菌肽在细菌传代14代之内,其对于细菌的最小抑菌浓度没有出现变化,而相对应的抗生素imipenem则在第7代出现了最小抑菌浓度翻倍的情况,标志着细菌针对该药物开始出现耐药性,在第11代该药物的MIC又再次翻倍,细菌耐药性进一步提高。发现的这种新型的抗菌肽具有相当优秀的抗细菌耐药性。
验证试验九:
本发明产品抗菌肽AV5哺乳动物抗感染模型中的体内抗菌试验
小鼠分为四组:分别用等量的消毒PBS、抗菌肽AV5、金黄色葡萄球菌悬液(107CFU/mL)、抗菌肽AV5和金黄色葡萄球菌悬液(107CFU/mL)治疗。每组有4只小鼠。在整个实验过程中,所有组的小鼠都可以自由饮水和进食。在每组中,用消毒剪刀在小鼠的背表面制作2.0mm深度的切除伤口(直径:10mm)。伤口底部是肉膜,没有明显的出血。然后将等分试样(50μL)的对数中期金黄色葡萄球菌(107CFU/ml)接种到小鼠的每个伤口中。在实验组中,接种后创面加入50μL终浓度为20μM(MIC浓度)的抗菌肽AV5。结果如图16所示,在添加抗菌肽AV5的一组,明显的加快了伤口的愈合,单独使用抗菌肽AV5的一组,也并没有对小鼠有影响,所以该抗菌肽AV5具有动物安全性。
验证试验十:
三只健康的豚鼠(300-400g)被用来评估抗菌肽的皮肤刺激性。简言之,将0.3mL抗菌肽溶液直接皮下注射于左背脊骨旁的皮肤上,将0.3mL生理盐水直接皮下注射于右背脊骨旁的皮肤上作为对照。使用表1中报告的标准,在24小时、48小时和72小时评估注射部位周围的皮肤反应,各项积分均为0。结果如图17所示,与皮下注射生理盐水(阴性对照)相比,在给药24、48和72小时的所有豚鼠中,皮下注射抗菌肽AVGAV在豚鼠背部皮肤上没有引起任何皮肤反应。这意味着所开发的水凝胶是一种非皮肤刺激系统,但可以作为全身/局部给药系统的药物载体而不会对皮肤产生任何刺激。

1.一种抗菌短肽在制备针对开放性伤口的抗菌、抑菌制品中的应用,所述的抗菌肽的氨基酸序列为:Ala-Val-Gly-Ala -Val。
2.如权利要求1所述的应用,其特征在于,所述的开放性伤口为慢性难愈合伤口、手术缝合伤口、运动损伤、烧烫伤清洁或污染伤口。
3.如权利要求1所述的应用,其特征在于:所述抗菌肽应用于制备治疗革兰氏阳性菌或革兰氏阴性菌感染的广谱抗菌药物。
