Abstract

The repairing and anti-aging composition provided by the invention has the advantages that various substances are mutually matched, the synergy is realized, the skin barrier is strengthened, the anti-allergy potential of the skin is improved, the elasticity and the tightness of the skin are improved, and the repairing and anti-aging composition has the effects of moisturizing, anti-aging and anti-wrinkle, cutin generation regulation, repairing and regeneration, anti-allergy and relieving, red fading and itching relieving, skin tolerance improvement and the like.

Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to a repairing and anti-aging composition capable of generating hyaluronic acid at double targets and reducing skin collagen loss, and a preparation method and application thereof.
Background
Skin aging refers to the aging damage of skin function, which reduces the protection ability and regulation ability of skin to human body, so that the skin can not adapt to the change of internal and external environment, and the appearance of the whole condition such as color, shape and texture changes. Skin aging is a process by which the human body functions or develops rapidly or slowly naturally under the influence of internal and external factors, mainly due to endogenous aging (time accumulation) and exogenous aging (external exposure to the environment).
At present, common anti-aging skin care products on the market are declared to be anti-aging by anti-oxidation, but the anti-aging mechanism is relatively complex, and although the anti-aging developed according to a certain mechanism can play a certain role, the overall effect is not good.
CN112402280A discloses an efficacy-promoting composition comprising a polymeric hyaluronic acid or a salt thereof, a hydrolyzed hyaluronic acid or a salt thereof, an acetylated hyaluronic acid or a salt thereof, a tetrahydropyrimidine or a derivative thereof, ergothioneine, yeast/rice fermentation product filtrate, and aminobutyric acid. The efficacy-promoting composition disclosed in the document has a certain capability of moisturizing and repairing skin, but has poor antioxidant and anti-aging effects on skin.
Therefore, how to provide a composition with excellent anti-aging effect is a problem to be solved.
Disclosure of Invention
Aiming at the defects of the prior art, the invention aims to provide a repairing and anti-aging composition which can generate hyaluronic acid at double targets and reduce the loss of skin collagen, and a preparation method and application thereof. The components in the repairing and anti-aging composition provided by the invention are matched with each other, and have synergistic effect, so that the generation of sodium hyaluronate can be promoted, collagen elastic fibers are strengthened, the generation of collagen I, III, IV and VI is stimulated, and the tightness, tension and elasticity of skin are improved fundamentally. In addition, the composition has anti-inflammatory and soothing effects, and can rapidly and effectively improve inflammatory response and discomfort of the skin, and maintain the skin's homeostasis and basic health.
In order to achieve the purpose, the invention adopts the following technical scheme:
in a first aspect, the present invention provides a restorative and anti-aging composition for dual-target hyaluronic acid production and reduction of skin collagen loss, the restorative and anti-aging composition comprising myristylaminobutyroylvalinamidobutyric acid urea trifluoroacetate and hydroxypropyl tetrahydropyrane triol.
In the present invention, the repairing and anti-aging composition comprises 0.001-0.01 parts (for example, 0.001 parts, 0.003 parts, 0.005 parts, 0.007 parts, 0.009 parts, 0.01 parts and the like) of tetradecyl aminobutyrylaminobutyramide butyric acid urea trifluoroacetate and 1-2 parts (for example, 1 part, 1.2 parts, 1.4 parts, 1.6 parts, 1.8 parts, 2 parts and the like) of hydroxypropyl tetrahydropyran triol by weight parts.
The repairing anti-aging composition provided by the invention can generate hyaluronic acid at double targets and can supplement the loss of skin collagen. Target 1: the tetradecyl amino butyryl valinamide butyrate urea trifluoroacetate is used for synthesizing CD44 proteoglycan by activating a TGF-BETA receptor, so that the biosynthesis of hyaluronic acid is promoted, the skin barrier is repaired, and the skin is anti-aging and firming. And (3) target point 2: the hydroxypropyl tetrahydropyrane triol integrally improves the expression capacity of glycosaminoglycan GAGS and reduces the degradation of hyaluronic acid in vivo, thereby repairing skin barriers and enabling the skin to achieve anti-aging and anti-tightening effects.

In the invention, the repairing and anti-aging composition also comprises palmitoyl tripeptide-5, acetylated sodium hyaluronate and soluble collagen.
In the present invention, palmitoyl tripeptide-5 may promote the production of collagen, enhance the skin's ability to fight aging, and protect collagen in stimulated keratinocytes. The face is sculpted, resulting in a firmer and firmer skin; reduce large pores and smooth skin.
In the present invention, acetylated sodium hyaluronate increases the penetration into the skin, is up to 13 times more resistant to hyaluronidase (hyaluronidase) than hyaluronic acid without acetylation, and can increase bioavailability.

In the present invention, the repairing and anti-aging composition further comprises 50.001-0.01 parts (for example, 0.001 parts, 0.003 parts, 0.005 parts, 0.007 parts, 0.009 parts, 0.01 parts, etc.) of palmitoyl tripeptide, 0.05-0.15 parts (for example, 0.05 parts, 0.07 parts, 0.09 parts, 0.11 parts, 0.13 parts, 0.15 parts, etc.) of acetylated sodium hyaluronate and 0.001-0.002 parts (for example, 0.001 parts, 0.0013 parts, 0.0016 parts, 0.0019 parts, 0.002 parts, etc.) of soluble collagen by weight.
In the present invention, the rejuvenating anti-aging composition further comprises chlorella/lupin protein fermentation product, kava leaf/root/stem extract and citrus peel extract.
The chlorella/lupin protein fermentation product contains various amino acids, minerals, vitamins and carbohydrates, improves the firmness of the epidermis by promoting the level of adhesion proteins, strengthens the dermis by activating the production of various extracellular matrix components, restores the firmness and elasticity of the skin, and achieves carbon neutralization.
In the invention, the kava leaf/root/stem extract is rich in kavalactones, mainly kavain active substances. Can be used in cosmetic products to achieve the effects of immediately tranquilizing, resisting inflammation, removing acne, and relieving and improving sleep.
In the invention, the citrus peel extract can remarkably relieve the instant stimulation of lactic acid to the skin, remarkably reduce the stimulation score value of the skin to the lactic acid, enhance the skin tolerance and maintain the normal function of the skin; has soothing and repairing effects, and is suitable for sensitive skin.
In the present invention, the repairing and anti-aging composition further comprises 0.0025 to 0.0125 parts (for example, 0.0025 part, 0.0045 part, 0.0065 part, 0.0085 part, 0.01 part, 0.011 part, 0.012 part, 0.0125 part, etc.) of chlorella/lupin protein fermentation product, 0.001 to 0.015 part (for example, 0.001 part, 0.003 part, 0.006 part, 0.009 part, 0.01 part, 0.013 part, 0.015 part, etc.) of kava leaf/root/stem extract, and 0.15 to 0.25 part (for example, 0.15 part, 0.17 part, 0.19 part, 0.21 part, 0.23 part, 0.25 part, etc.) of citrus peel extract, in parts by weight.
The repairing and anti-aging composition provided by the invention can comprehensively repair skin barriers and increase the anti-aging and tightening effects of the skin. By promoting the biosynthesis of hyaluronic acid (activating TGF-BETA receptor and synthesizing CD44 proteoglycan), the expression capacity of glycosaminoglycan GAGS is integrally improved, the degradation of hyaluronic acid in vivo is reduced, collagen elastic fibers are strengthened, the generation of type I, III, IV and VI collagen is stimulated, and the skin tightening effect, the tension and the elasticity of the skin are fundamentally solved. In addition, the composition also has the effects of resisting inflammation and relieving, quickly and effectively soothing the inflammatory reaction and the uncomfortable symptoms of the skin, and maintaining the steady state and the basic health of the skin.
In a second aspect, the present invention provides a method for preparing the repairing anti-aging composition according to the first aspect, the method comprising the steps of: mixing myristylaminobutyroylvalinamide butyrate trifluoroacetate, hydroxypropyl tetrahydropyrane triol, optionally palmitoyl tripeptide-5, optionally acetylated sodium hyaluronate, optionally soluble collagen, optionally chlorella/lupin protein fermentation product, optionally kava leaf/root/stem extract, and optionally citrus peel extract to obtain the anti-aging composition.
In the present invention, the mixing temperature is 50-60 ℃ (for example, 50 ℃, 52 ℃, 54 ℃, 56 ℃, 58 ℃, 60 ℃ etc.).
In the present invention, the sodium acetylated hyaluronate is first soaked in water for 12 to 24 hours (for example, 12 hours, 14 hours, 16 hours, 18 hours, 20 hours, 22 hours, 24 hours, etc.) to form an aqueous solution of sodium acetylated hyaluronate of 0.5% to 1.5% (for example, 0.5%, 0.7%, 0.9%, 1.1%, 1.3%, 1.5%, etc.).
In a third aspect, the present invention provides a use of the repairing anti-aging composition according to the first aspect in the preparation of cosmetics.

This example provides a rejuvenating anti-ageing composition which differs from example 1 only in that it does not contain chlorella/lupin protein fermentation products, the parts by weight of the kava leaf/root/stem extract are increased to 0.015 parts, the other components and the process for their preparation are the same as example 1.

This example provides a rejuvenating anti-aging composition, which is different from example 1 only in that kava leaf/root/stem extract is not contained, the weight part of chlorella/lupin protein fermentation product is increased to 0.015 part, and other components and preparation method thereof are the same as example 1.
Example 9
This example provides a rejuvenating anti-aging composition, which differs from example 1 only in that it does not contain citrus peel extract, the weight fraction of water is increased to 98.3735 parts, and the other components and the method of preparation are the same as in example 1.
Comparative example 1
This comparative example provides a repairing anti-aging composition which differs from example 1 only in that tetradecylaminobutylamidobutanamido urea butyrate trifluoroacetate is not contained, the part by weight of hydroxypropyl tetrahydropyratriol is increased to 1.505 parts, and other components and the preparation method thereof are the same as example 1.
Comparative example 2
This comparative example provides a repairing anti-aging composition, which is different from example 1 only in that hydroxypropyl tetrahydropyrane triol is not contained, the weight part of water is increased to 99.6735 parts, and other components and a preparation method thereof are the same as example 1.
Test example 1
DPPH clearance test.
Testing a sample: examples 1-9 and comparative examples 1-2 provide repair anti-aging compositions.
The test method comprises the following steps: DPPH free radical is a stable free radical, and whether the tested substance has oxidation resistance can be determined by utilizing the characteristic that ethanol solution of DPPH free radical is purple and has a characteristic absorption peak at the wavelength of 517 nm. Transferring 2mL of DPPH-ethanol solution (0.2 mmol/L), adding 2mL of sample to be detected, fully mixing, standing in a dark place for 30min, taking absolute ethyl alcohol as a blank control, measuring the absorbance at 517nm, and calculating the DPPH clearance rate of the sample to be detected according to the following formula:

In the formula, A i Is the absorbance of the sample to be measured and the DPPH reaction system, A j Is the absorbance of the sample to be measured after mixing with absolute ethyl alcohol, A 0 The absorbance of the blank control absolute ethyl alcohol is shown.

Test example 3
Test for soothing efficacy
Testing a sample: examples 1 and 7 to 9 provide the repairing and anti-aging composition.
Test methods experimental methods: RAW264.7 macrophages were used as the study subjects, and cells were stimulated by lipopolysaccharide LPS (bacterial endotoxin) to establish a model of cellular inflammation. The TNF-alpha inhibition rate is calculated by the following formula:
TNF- α inhibition (%) = (inflammatory factor concentration in LPS-test substance-action group inflammatory factor concentration)/(inflammatory factor concentration in LPS-non-stimulation group inflammatory factor concentration) × 100%. The higher the inhibition rate of TNF-alpha, the better the anti-inflammatory effect of the anti-aging composition.

Comparison of examples 1 and 7-9 shows that the citrus peel extract, the chlorella/lupin protein fermentation product, and the kava leaf/root/stem extract of the present invention cooperate with each other to provide a synergistic effect, and have excellent anti-inflammatory effects.
Test example 4
Safety test
Testing a sample: examples 1-9 and comparative examples 1-2 provide repair anti-aging compositions.
The test method comprises the following steps: refer to "cosmetic hygiene Standard" for testing skin patches on human body. 110 healthy, allergy-free, volunteers of 18-60 years old were selected as subjects, which were divided into 11 groups of 10 persons each, and no other skin care products were used during the trial. Selecting a spot tester with an area of 6mm multiplied by 6mm, putting a test object into a small chamber of the spot tester, wherein the dosage is 0.025g, sticking the spot tester with the test object on the back of a subject by using a hypoallergenic adhesive tape, and lightly pressing the spot tester with a palm to uniformly stick the spot tester on the skin for 24 hours. 30min (after the indentation disappears), 24h and 48h after the test object spot tester is removed. Skin reactions were observed according to the criteria shown in table 5 and the observations were recorded.

The test result shows that the anti-aging composition provided by the invention is very mild and has no irritation to the skin.
The applicant states that the present invention is illustrated by the above examples, but the present invention is not limited to the above examples, i.e. it is not intended that the present invention is implemented by relying on the above examples. It should be understood by those skilled in the art that any modification of the present invention, equivalent substitutions of the raw materials of the product of the present invention, addition of auxiliary components, selection of specific modes, etc., are within the scope and disclosure of the present invention.
Claims (10)
1. A repairing and anti-aging composition capable of generating hyaluronic acid at double targets and reducing loss of skin collagen, which is characterized by comprising tetradecyl aminobutyroylvalerian amidobutyric urea trifluoroacetate and hydroxypropyl tetrahydropyrane triol.
2. The repairing and anti-aging composition of claim 1, wherein the repairing and anti-aging composition comprises 0.001-0.01 parts by weight of tetradecylaminobutylamidobutyric acid urea trifluoroacetate and 1-2 parts by weight of hydroxypropyl tetrahydropyrane triol.
3. The remedial anti-aging composition of claim 1 or 2, further comprising palmitoyl tripeptide-5, acetylated sodium hyaluronate and soluble collagen.
4. The repairing and anti-aging composition of any one of claims 1 to 3, further comprising palmitoyl tripeptide-5.001-0.01 parts, acetylated sodium hyaluronate 0.05-0.15 parts and soluble collagen 0.001-0.002 parts by weight.
5. The repair anti-aging composition according to any one of claims 1 to 4, further comprising chlorella/lupin protein fermentation product, piper methysticum leaf/root/stem extract and Citrus pericarp extract.
6. The repairing and anti-aging composition according to any one of claims 1 to 5, further comprising 0.0025 to 0.0125 parts by weight of chlorella/lupin protein fermentation product, 0.001 to 0.015 parts by weight of Piper methysticum leaf/root/stem extract, and 0.15 to 0.25 parts by weight of Citrus reticulata pericarp extract.

A method for preparing the repairing anti-ageing composition according to any one of claims 1 to 6, wherein the preparation method comprises the following steps: mixing myristylaminobutyroylvalinamide butyrate trifluoroacetate, hydroxypropyl tetrahydropyrane triol, optionally palmitoyl tripeptide-5, optionally acetylated sodium hyaluronate, optionally soluble collagen, optionally chlorella/lupin protein fermentation product, optionally kava leaf/root/stem extract, and optionally citrus peel extract to obtain the anti-aging composition.
8. The method of claim 7, wherein the temperature of the mixing is 50-60 ℃.
9. The method according to claim 7 or 8, wherein the mixing is preceded by immersing the sodium hyaluronate in water for 12-24 hours to form a 0.5% -1.5% aqueous solution of the sodium hyaluronate.
10. Use of a repairing anti-ageing composition according to any one of claims 1 to 6 in the preparation of a cosmetic product.
