Technical Field
The invention belongs to the field of skin care products, and particularly relates to a multi-effect anti-aging composition and a preparation method thereof
Background
Skin aging refers to the aging damage of skin function, which reduces the protection ability and regulation ability of skin to human body, so that the skin can not adapt to the change of internal and external environment, and the change of overall appearance conditions such as color, luster, shape, texture, etc. appears. Aging of the skin is classified into intrinsic aging and extrinsic aging. Endogenous aging refers to the natural aging of the skin with aging, a natural process, which has been described systematically by the theory of natural cross-linking of biomolecules. The theory, when demonstrating the molecular mechanisms of aging of organisms, points out: organisms are unstable chemical systems and belong to dissipative structures. Various biomolecules in the system have a large number of active groups, and the active groups must interact with each other to generate chemical reaction, so that the biomolecules are slowly crosslinked to tend to be stable in chemical activity. With the lapse of time, the degree of crosslinking is continuously increased, the active groups of the biomolecules are continuously consumed and reduced, the original molecular structure is gradually changed, and the accumulation of the changes can cause the aging phenomenon of the biological tissues. It is manifested as whitening of the skin, fine wrinkles, decreased elasticity, skin laxity, etc. The most prominent cause of extrinsic aging is photoaging due to sun exposure. Skin discoloration manifested as wrinkles, loose skin, roughness, yellowish or grayish yellow, telangiectasia, formation of pigmented spots, and the like. In the case of the Chinese market, aging resistance has shown a trend towards more and more youthfulness. Survey data from the first financial group show that 59.2% begin to stress aging by age 20-30 years, and 19.64% of younger people aged 20 to 25 feel "anxious" to aging. At the same time, 28% of the time after 00 has begun to use anti-aging products. Therefore, anti-aging cosmetics are the most market-demanding class of cosmetics among functional cosmetic products.
The skin aging inducing factors are numerous, and people develop a plurality of anti-aging skin care products, but the skin care products can only improve the skin aging problem in one aspect, improve the aging by stimulating cell regeneration, and have poor and difficult lasting effect.
The consumer has a higher demand for efficacy due to the positive attitude against aging. At present, many cosmetics cannot meet the requirement of consumers for being very effective, so that part of consumers begin to turn to seeking for the help of medical beauty, the market scale of medical beauty is increased day by day, but in recent years, medical and patient disputes occur in the medical beauty market sometimes, and the effect is greatly reduced due to improper postoperative care, so that consumers very need safe and effective anti-aging products.

Efficacy verification
(I) efficacy test
Firstly, a testing instrument:
1. the invention uses a finland Delfin elastometer which is lightly pressed against the skin with an indenter. When an external force is applied to the skin, the skin resists changes in its shape and the response of the skin under short term loading indicates its immediate elastic properties, resulting in an elasticity value for the skin.
2. Using Delfin antrera 3D photography in finland, antrera 3D used innovative optical methods and complex mathematical algorithms to acquire three-dimensional images. This allows data to be extracted from the image relating to the three-dimensional shape of the skin, which will enable quantification of the effect of the treatment and monitoring of the situation over time. For example, the texture, smoothness of the skin, and depth and width of fine lines and wrinkles are measured. With its multispectral analysis, the ant software can also measure pigmentation and redness, which will provide meaningful information about sun damage, veins, etc.
3. The percutaneous water loss meter, Delfin VapoMeter SWL5201, finland, was used, the VapoMeter having a cylindrical chamber with relative humidity and temperature sensors inside. Shortly after the instrument was brought into contact with the skin, there was a linear increase in relative humidity (RH%) in the measurement cavity. TEWL values were calculated from the increase in RH%. In the measurement gap, the measurement chamber is passively ventilated. The VapoMeter uses a closed-chamber measurement principle, so the measurement is not affected by the gas flow in the chamber. The measurement time is short, ensuring the minimum of the blockage of normal evaporation.
II, experimental principle:
TEWL value, transdermal water loss (TEWL), measures the change in the vapor pressure of moisture adjacent to the skin surface. TEWL values represent the loss of water from the stratum corneum and are an important criterion for evaluating the barrier function of the stratum corneum. A lower TEWL value of the skin indicates a better barrier function of the skin and vice versa. The efficacy of the moisturizing and repairing cosmetic can be evaluated by testing the value of skin moisture loss TEWL, and the moisturizing and repairing cosmetic can also be applied to allergic patch test, contact dermatitis and physical therapyMonitoring of burns and new tissue, finding in time whether the protective function of the skin has been destroyed. The unit of TEWL is: g/hm 3 。
Thirdly, testing the population:
each group comprises 20 persons, 9 groups of volunteers with the age of 30-65 years and meeting the experimental requirements.
Fourthly, testing conditions:
subjects rested for 30 minutes at room temperature 21 ± 1 ℃, relative humidity 50 ± 10%, and were monitored dynamically in real time and were retested for relevant data.
Fifthly, an experimental process:
the experiment is divided into 9 groups, wherein the 1 st group uses the whole face of the invention in the embodiment 1, the second group uses the whole face of the invention in the embodiment 3, the third group uses the whole face of the comparison example 8, the 4-9 groups use the left face of the invention in the embodiment 2, and the right face of each group is respectively coated with the comparison examples 1-6; the face is rubbed 2 times in the morning and evening every day for 4 weeks, the skin elasticity ISE value, the percutaneous water loss rate TEWL value, the average value WRC (mm) of the skin wrinkle length, the average value WRK (mm) of the wrinkle width and the average value WRS (mm) of the wrinkle depth are recorded, then, one part of each volunteer face, the left forehead, the right forehead, the eye corner and the cheek is photographed by an Antera3D, and the whole is stored.
Sixthly, experimental results:
1. skin transdermal water loss Rate (TEWL) test
The test sets the subject's TEWL at week 0 as TEWL0, week 4 as TEWL4, with the TEWL change being Δ TEWL, Δ TEWL being TEWL4-TEWL0, Δ TEWL rate of change being (TEWL4-TEWL0)/TEWL0 + 100%, Δ TEWL rate of change being negative and the lower the number, indicating less moisture loss in the skin after four weeks of use of the product, i.e. better skin barrier function repair, better moisture retention, and healthier skin, table 2 shows the average value of the TEWL change for each group of 20 people after 4 weeks of use compared to the initial value.
TABLE 2
Group of | Delta TEWL rate of change (%) | Group of | Delta TEWL rate of change (%) |
Example 1 | -7.54 | Comparative example 3 | -8.51 |
Example 2 | -14.62 | Comparative example 4 | -10.38 |
Example 3 | -25.43 | Comparative example 5 | -7.79 |
Comparative example 1 | 14.73 | Comparative example 6 | -5.46 |
Comparative example 2 | 2.25 | Comparative example 7 | -6.95 |
The results in table 2 illustrate that:
1) the delta TEWL change rate value, example 1 > example 2 > example 3, therefore, it can be shown that as the addition ratio of the composition of the invention is increased, the delta TEWL change rate value is correspondingly reduced, namely, the less the skin moisture is lost, the more complete the stratum corneum barrier function of the skin is, the better the capability of the skin for resisting the external environment, the less the skin moisture is lost, and the more healthy, moist and full the skin is.
2) The delta TEWL change rate value of the comparative example 1 is 14.73%, which shows that the skin loses moisture and increases after the essence cream of the comparative example 1 is used, namely, the barrier function of the skin cuticle is gradually weakened, the skin becomes more fragile and dry, and the moisture of the skin is more easily lost.
3) The delta TEWL change rate value of comparative example 2 was 2.25%, that is, the increase of the moisture loss rate of the essence cream of comparative example 2 was not obvious, which indicates that the skin barrier function recovery is not well affected by using the essence cream of comparative example 2.
4) It is understood from comparative example 2 and comparative examples 3 to 6 that removal of any one of teprenone, collagen, hydroxypropyl tetrahydropyran triol and diamond micropowder increases the Δ TEWL value, i.e., the skin moisture is more likely to be lost, and the recovery of the stratum corneum barrier function of the skin is not as excellent as that of example 2. The teprenone, the collagen, the hydroxypropyl tetrahydropyrane triol and the diamond micropowder have good synergistic effect.
5) The delta TEWL change values of the compositions in the examples 1-3 are smaller than those of the comparative example 7, so that the composition disclosed by the invention is better than the anti-aging essence cream of the market international brand in the capacity of reducing the skin moisture loss and also better than the anti-aging essence cream of the market international brand in the capacity of keeping the skin moisture and repairing the skin stratum corneum barrier.
2. Skin Elasticity (ISE) test
The test set the subject's measured ISE at week 0 as ISE0 and ISE at week 4 as ISE4 with the ISE change value Δ ISE which is ISE4-ISE0, Δ ISE change rate (ISE4-ISE0)/ISE0 100%, Δ ISE change rate is positive and higher values indicate more increase in skin elasticity value around the product used, higher skin elasticity and more firmer and more elastic skin, and table 3 shows the average Δ ISE change rate values for 20 people per group compared to the initial values after 4 weeks of use.
TABLE 3
Group of | Delta ISE Change Rate (%) | Group of | Delta ISE Change Rate (%) |
Example 1 | 10.31 | Comparative example 3 | 15.44 |
Example 2 | 23.43 | Comparative example 4 | 20.33 |
Example 3 | 34.32 | Comparative example 5 | 12.46 |
Comparative example 1 | 18.44 | Comparative example 6 | 8.83 |
Comparative example 2 | 15.36 | Comparative example 7 | 7.58 |
As can be seen from table 3:
1) the ISE change rate, example 1 < example 2 < example 3, shows that as the addition ratio of the composition of the invention increases, the change rate of the elasticity value of the composition also increases greatly, thus showing that in the addition range of the composition of the invention, the higher the addition amount of each component, the more the elasticity of the skin increases, and the firmer and fuller the skin.
2) The delta ISE change rate value of comparative example 1 < the delta ISE change rate value of example 2, which indicates that the skin elasticity-increasing effect of the composition of the present invention is inferior to that of the composition of the present invention when the composition of the present invention is replaced with retinol, a commercially available anti-aging star ingredient, and the amount of retinol added is the highest amount added.
3) The delta ISE change value of comparative example 2 < delta ISE change value of example 2, indicating that the composition of the present invention was changed to a commercially available peptide in an amount similar to that of example 2, and the skin elasticity-increasing effect was far less than that of the composition of the present invention.
4) Comparative examples 3 to 6 are essence creams prepared by removing any one of the compositions of the present invention in example 2, and the delta ISE change rate is smaller than that of example 2 by comparing with example 2, so that it can be shown that any removal of a certain component of the composition of the present invention results in a smaller delta ISE change rate, and the skin elasticity is not increased as much as that of example 2 of the present invention. The teprenone, the collagen, the hydroxypropyl tetrahydropyrane triol and the diamond micropowder have good synergistic effect.
5) The delta ISE change values of the examples 1-3 are all larger than that of the comparative example 7, so that the composition disclosed by the invention is better than the anti-aging essence cream of the commercial international brand in the capability of increasing the skin elasticity within the adding amount range, and the cosmetics prepared by using the composition disclosed by the invention have more excellent effects of increasing the skin elasticity and compacting the skin.
3. Skin wrinkle Length (WRC value), average wrinkle Width (WRK), average wrinkle depth (WRS) test
The test sets the average value of the wrinkle length, width and depth of the 0 th week measured by a subject as WRC0, WRK0 and WRS0, the average value of the wrinkle length, width and depth of the 4 th week measured by the subject as WRC4, WRK4 and WRS4, the wrinkle change values are Δ WRC, Δ WRK and Δ WRS, Δ WRC is WRC4-WRC0, Δ WRK is WRK4-WRK0, WRS is WRS4-WRS 0; Δ WRC rate of change (WRC4-WRC0)/WRC0 × 100%, (WRK4-WRK0)/WRK0 × 100%, (WRS4-WRS0)/WRS0 × 100%; the change rates of delta WRC, delta WRK and delta WRS are negative numbers, and the lower the numerical value is, the shorter the wrinkle length of the skin is, the narrower the wrinkle width is, the shallower the wrinkle depth is, the better the product has the capability of delaying the wrinkles of the skin, and the smoother and younger the skin is. The results are shown in Table 4 and FIG. 1. Table 4 shows the average wrinkle length, width, depth change rate values for each group of 20 people compared to the initial values after 4 weeks of use.
TABLE 4
Group of | Δ WRC Rate of Change (%) | Δ WRK Change (%) | Δ WRS Change ratio (%) |
Example 1 | -17.10 | -7.46 | -8.49 |
Example 2 | -28.66 | -14.51 | -13.52 |
Example 3 | -45.43 | -20.62 | -21.20 |
Comparative example 1 | -20.55 | -10.53 | -10.14 |
Comparative example 2 | -18.34 | -11.72 | -10.82 |
Comparative example 3 | -15.64 | -9.67 | -9.61 |
Comparative example 4 | -22.38 | -12.81 | -11.94 |
Comparative example 5 | -16.82 | -8.26 | -7.25 |
Comparative example 6 | -13.37 | -7.75 | -5.16 |
Comparative example 7 | -10.25 | -5.48 | -6.01 |
As can be seen from table 4:
1) the change rate values of Δ WRC, Δ WRK and Δ WRS, example 1 > example 2 > example 3, thus it can be shown that as the addition ratio of the composition of the present invention is increased, the change rate values of Δ WRC, Δ WRK and Δ WRS become smaller, namely, the shorter the wrinkle length of the skin, the narrower the width and the shallower the depth, the less obvious the wrinkles of the whole skin become, and the more excellent the anti-aging effect of the product becomes.
2) The values of change Δ WRC,. DELTA.WRK,. DELTA.WRS of comparative example 1 are greater than those of example 2, indicating that the skin is less able to reduce wrinkles overall after the use of the cream of comparative example 1 than the skin is able to reduce wrinkles overall by the composition of the present invention of example 2, i.e., the composition of the present invention is better than 1% retinol at reducing the overall level of wrinkles.
3) The change rate values of Δ WRC, Δ WRK, and Δ WRS of comparative example 2 are greater than those of example 2, indicating that the skin has less overall skin wrinkle reduction capability after the essence cream of comparative example 2, i.e., the composition of the present invention is superior to the same amount of palmitoyl pentapeptide-4 in reducing the overall wrinkle level.
4) Comparative examples 3 to 6 are essence creams prepared by removing any one of the compositions of the present invention in example 2, and compared with example 2, the change rates of Δ WRC, Δ WRK, and Δ WRS are all larger than those of example 2, so it can be shown that any removal of a certain component of the composition of the present invention will increase the corresponding change rates of Δ WRC, Δ WRK, and Δ WRS, i.e., the overall level of wrinkles on the skin, and therefore any removal of a component of the composition of the present invention will reduce the anti-wrinkle effect of the product, i.e., any component of the composition of the present invention will have a positive effect on the anti-aging effect of the final product.
5) The change rate values of Δ WRC, Δ WRK, and Δ WRS of examples 1-3 are all smaller than those of comparative example 7, thus demonstrating that the composition of the present invention has better capability of reducing the whole level of skin wrinkles than the commercial international brand anti-aging essence cream in the addition amount range, i.e. better anti-wrinkle and anti-aging effects than the commercial international brand anti-aging essence cream.
6) Before and after selecting a part of facial wrinkle images of volunteers using the facial wrinkle image obtained in example 2 of the present invention, as shown in fig. 1 in detail, it can be seen visually that the whole level of facial wrinkles is reduced after the facial skin is treated with the essence cream prepared in example of the present invention, indicating that the anti-wrinkle effect of the composition of the present invention is very excellent.
(II) safety test (human body skin patch experiment)
1. The number of tested persons: total 40, 20 men and 20 women, age between 18-60 years.
2. Health condition: the skin of the subject is healthy, the allergic history of the skin disease does not exist, and the voluntary selection standard of the subject is met.
3. The test method comprises the following steps: selecting a qualified spot tester, putting about 0.020-02025g of a test product in the spot tester in a closed spot-sticking experiment mode, externally sticking a special adhesive tape on the back of a test subject, coating 11 coating points on each test subject, removing the test product after 24 hours, observing skin reactions after 0.5, 24 and 48 hours respectively, grading according to skin reaction grading standards in cosmetic hygiene standards, wherein the evaluation standards are shown in table 5, calculating the average value, and recording the result in table 6.
