Further, the mass percentage of the yeast fermentation product extract in the final product is 10.0%, the mass percentage of the lysate of the saccharomyces cerevisiae fermentation product in the final product is 5.0%, the mass percentage of the carnosine in the final product is 0.2%, the mass percentage of the p-hydroxyacetophenone in the final product is 0.15%, and the rest is water.
In another aspect, the invention provides a method for preparing the skin anti-aging composition, comprising the steps of mixing the components of yeast fermentation product extract, yeast fermentation product lysate, carnosine and p-hydroxyacetophenone in solvent water according to a certain proportion, wherein the components are premixed in a container according to a certain proportion in the preparation process of the product, and then the mixture is added into a skin care product. Firstly, weighing the yeast fermentation product extract, the lysate of the yeast fermentation product, the carnosine, the p-hydroxyacetophenone and the water according to the formula ratio in a beaker. P-hydroxyacetophenone was added to solvent water and stirred using an electromagnetic stirrer for 15 minutes until complete dissolution. And then adding the yeast fermentation product extract, the two-split yeast fermentation product lysate and the carnosine into water one by one, and continuously stirring for 5 minutes by using an electromagnetic stirrer until the two components are uniformly mixed. And then pouring the mixed product into a plastic bottle for standby. All operations are performed at room temperature and normal temperature.
In a third aspect of the present invention, there is provided a cosmetic product prepared by the method for preparing the composition for skin aging according to claim 6, and the formulation of the cosmetic product may be one of various formulations including, but not limited to, a solution, an emulsion, a cream, a gel, a solid, a lyophilized powder or a liposome.
In a fourth aspect the present invention provides the use of a composition according to any one of claims 1 to 5 for the preparation of a cosmetic for skin anti-ageing, or alternatively, a cosmetic according to claim 7.
In a fifth aspect the present invention provides the use of the composition of the second aspect for the preparation of a composition for skin anti-ageing or the cosmetic of the third aspect in non-therapeutic cosmetic and/or dermatological applications, acting on the surface of the skin, including skin tissue at the facial, neck or abdominal location.
Yeast is one of the earliest and most widely used microorganisms and is of a wide variety. Yeast extract is a "Baozhen ingredient", and contains skin-friendly ingredients such as monosaccharides, polysaccharides, minerals, vitamins, etc., in addition to amino acids and nucleotides. The yeast extract has the following effects in the skin care field:
1. and (5) moisturizing. Yeast extract contains abundant amino acids, which are essential components of skin itself, and various free amino acids are important components of skin natural moisturizing factors. At the same time, the collagen protein of the skin and the silk fibroin which plays a key role in the skin barrier are also composed of amino acids with different sequences.
2. Whitening. The components such as polypeptide and amino acid contained in the yeast extract have a good effect of inhibiting the formation of melanin, and are therefore effective in improving skin darkness, spots, etc.
3. Anti-inflammatory, antioxidant and antiaging. The yeast extract is rich in vitamin B group, and B5 has antiinflammatory and repairing effects, and B3 (nicotinamide) has antioxidant, antiinflammatory and antiaging effects. Some polysaccharide components (beta glucan) in yeast extract also have anti-inflammatory effects.
4. Mild exfoliating. The organic acid contained in the yeast extract is mostly weak acidic, and can play a role in mildly exfoliating.
5. Other effects. Different brands of yeast extract skin care products also use different skin care efficacy because of the different yeasts used and modes of manufacture. With the increasing awareness of yeast extracts, the market for yeast extracts is also growing, and particularly in the field of skin care, the strong skin care efficacy of yeast extracts has been accepted by more and more people.
The yeast fermentation product extract is added with the pepper in the fermentation substrate, and the fermentation product contains rich polypeptide components and pepper components metabolized by yeast, so that the subcutaneous muscle can be relaxed under the combined action of various active ingredients, and the memory contraction of the subcutaneous muscle is eliminated, thereby achieving the effect of immediately removing wrinkles. The main wrinkle-removing characteristic is quick effect, and can take effect within 15 minutes after use. However, it has a significant disadvantage in that its effect gradually disappears within twenty-four hours after use, and wrinkles rebound quickly once it is deactivated.
The lysate of the fermentation product of the saccharomyces cerevisiae is fermented, cultured, inactivated and decomposed to obtain a metabolite, a cytoplasmic fragment, a cell wall component and a polysaccharide complex.
The lysate of the fermentation product of the saccharomyces cerevisiae has strong anti-immunosuppression activity, can promote DNA repair, can effectively protect skin from being damaged by ultraviolet rays, is used for skin care, sun protection and after-sun care products of emulsion, water-based and hydroalcoholic systems, and helps to prevent photoaging of epidermis and dermis. The saccharomycete fermentation product, namely the sporulation substance, can generate micromolecules comprising vitamin B group, mineral substances, amino acids and the like which are beneficial to skin care, and is high-quality yeast essence only used for skin care. It can strengthen metabolism of horny layer, capture free radical, inhibit lipid peroxidation, and has skin whitening and antiaging effects. The product is rich in nutrients, and has skin nourishing effect.
The lysate of the fermentation product of the saccharomyces cerevisiae has mild property and obvious long-term wrinkle removal effect. But the effect will not appear until after 28 days of use.
Carnosine is widely present in all organ tissues of the body, especially up to 20mmol/L in muscle, brain and eye lenses.
The histidine residue on the carnosine side chain can be used as a hydrogen acceptor and has the functions of capturing hydroxyl radicals, singlet oxygen and peroxidation radicals. These properties have been demonstrated experimentally. For example, beom et al established a system for catalyzing the generation of hydroxyl radicals by iron to degrade deoxyribose, and found that carnosine was effective in inhibiting the degradation of deoxyribose, indicating its ability to capture hydroxyl radicals. Chan et al established an iron ion-hydrogen peroxide system to generate hydroxyl radicals, and EPR was used to monitor the formation of non-hydroxyl radicals after direct interaction of carnosine and related histidine-containing dipeptides with hydroxyl radicals. The results show that dipeptides such as carnosine can quench 49.1% -94.9% of hydroxyl radicals generated by iron ions and hydrogen peroxide.
Carnosine has an antioxidant effect. The antioxidant mechanism of carnosine has now largely reached consensus: (1) Carnosine has the ability to buffer physiological pH, reducing lipid peroxidation due to system pH changes. (2) Carnosine has the ability to sequester metal ions and is able to inhibit fat oxidation caused by metal ions, particularly copper ions. (3) Carnosine has the effects of capturing hydroxyl radicals, quenching singlet oxygen and scavenging peroxidized radicals, and can inhibit fat oxidation caused by non-metals.
Carnosine is a multifunctional physiological dipeptide that has anti-aging properties in addition to some of the functional properties described above. Experiments prove that carnosine can restore the activity of the aged human fibroblasts, and can be regenerated again. The anti-aging mechanism of carnosine is not completely understood. The anti-oxidant, free radical scavenging and metal ion chelating properties of carnosine in vivo can explain in part the anti-aging phenomenon of carnosine but not completely. VE or VC, which also has at least antioxidant properties, does not have the ability to rejuvenate fibroblasts. Carnosine has been shown to react with small carbonyl-containing compounds (e.g., aldehydes, ketones, etc.) to prevent cross-linking between biological macromolecules. Glycosylation (glycation) under protein oxidation and enzyme-free conditions is associated with aging phenomena of cells or tissues. Glycosylation under enzyme-free conditions of proteins will ultimately result in highly complex cross-linked molecules, known as AGES, and often contain reactive carbonyl groups on the AGES molecule. Carbonyl groups on proteins can also be produced by direct reaction of any reactive aldehyde, including the lipid peroxidation product Malondialdehyde (MDA) with reactive oxygen radicals ROS (reactive oxygen species) such as-OH, HCLO-radicals. Whereas aging is associated with excessive accumulation of carbonyl groups on proteins. Carnosine is reported to competitively glycosylate with aldoses or ketoses, thereby protecting proteins and other peptides; carnosine reacts with c=o generated by MDA and HClO-, thereby avoiding cross-linking between proteins or between proteins and DNA, and also protecting cells (because MDA is toxic to cells); carnosine reacts with c=o on oxidized or glycosylated proteins, thereby acting to delay senescence. Such a reaction is called carnosine.
Carnosine can effectively remove wrinkles and improve the health degree of skin cells after long-term use. But the effect alone is not obvious.
Among the three components of the composition of the invention, the yeast fermentation product extract plays a role in instant wrinkle removal; the lysate of the fermentation product of the saccharomyces cerevisiae can strengthen the skin barrier function, nourish subcutaneous tissue and activate subcutaneous cell activity, thereby achieving the effect of reducing wrinkles; carnosine can eliminate free radicals, improve cell firmness and increase skin elasticity. The three components have synergistic effect and can play a better role in removing wrinkles and resisting aging than the single raw material.
Compared with the prior art, the invention has the beneficial effects that:
1. better anti-aging and wrinkle-removing effects, and better and more stable wrinkle-reducing effects than any single component can be obtained by using the combination of the three active ingredients. The effective population is wider than that of single component.
2. The invention combines three components to achieve the effect of the anti-aging skin care product when being used once, can continuously improve the skin condition when being used for a long time, and has no obvious rebound after being used for a period of time.
FIG. 1 is a graph comparing skin anti-aging compositions provided in examples of the present invention with test examples.
Detailed Description
The method used in the present invention to characterize skin aging is a clinical test, with a total of 35 female volunteers selected to participate in the test. Volunteers were between 35-60 years of age. During testing, 0.1 gram of anti-wrinkle toner is uniformly applied to the corners of eyes of volunteers. The volunteer wrinkle volumes were measured at these time points 15 minutes after use, 1 hour after use, 1 day after use, 28 days after continuous use, and 7 days after stopping use, respectively, before use. The equipment used for the test was a PRIMOS CR wrinkle analyzer.

The yeast fermentation product extract, the lysate of the yeast fermentation product, and carnosine are added into deionized water according to the following proportion, and 0.15wt% of p-hydroxyacetophenone is added as a preservative. Stirring until all the materials are dissolved for standby.

Test result analysis method the volume of wrinkles obtained from the equivalent time point test was subtracted from the volume of wrinkles before the test for each volunteer 15 minutes after use, 1 hour after use, 1 day after use, 28 days after continuous use, 7 days after use, and the value of wrinkles reduction at different time points for the volunteer was obtained. This reduction was divided by the pre-test wrinkle volume to obtain the percentage of wrinkle reduction in the volunteer. The percentage reduction in the volume of wrinkles was mathematically averaged for all volunteers to obtain an average reduction in the volume of wrinkles for this test.
Test results: as can be seen from fig. 1, examples 1 and 2 and comparative example 1, comparative example 5, and comparative example 6 all produced good immediate wrinkle removal effect and a significant reduction in wrinkle volume within 15 minutes, 1 hour and 1 day after the use of the examples and comparative examples. However, after 28 days of continuous use, the tendency of the volume of wrinkles of comparative example 1, comparative example 5 and comparative example 6 to decrease was retarded, and the wrinkle-removing effects of comparative examples 2,3 and 4 were gradually developed, and particularly, the long-term wrinkle-removing effect of comparative example 4 was better than that of the carnosine or the lysate of the fermentation product of Saccharomyces cerevisiae alone. However, in combination, the effect of reducing wrinkles was obtained after 28 days of use in examples 1 and 2. Next, after all examples and comparative example 7 were deactivated for days, it was found that examples 1 and 2 were much lighter in wrinkles than comparative example 1, comparative example 5, comparative example 6, and examples 1 and 2 maintained better overall wrinkle removal than comparative examples 2,3, and 4. The combination of the present invention results in an optimal synergistic effect. There is no way to achieve the combined effect of the present invention using a single ingredient or two-by-two combination of ingredients.
The above embodiments are only some of the preferred embodiments of the present invention, and should not be construed as limiting the scope of the present invention, and any insubstantial changes and substitutions made by those skilled in the art on the basis of the present invention are intended to fall within the scope of the present invention as claimed.
Claims (8)
1. A composition for skin anti-aging comprising a yeast fermentation product extract, a yeast fermentation product lysate, carnosine, p-hydroxyacetophenone, and water, wherein
The mass percentage of the yeast fermentation product extract in the composition is 1.0-15.0%, the mass percentage of the two-split yeast fermentation product lysate in the composition is 1.0-10.0%, the mass percentage of carnosine in the composition is 0.05-0.5%, the mass percentage of p-hydroxyacetophenone in the composition is 0.15%, and the rest is water;
the yeast fermentation product extract is added with pepper in a fermentation substrate.
2. The composition for skin aging resistance according to claim 1, wherein the mass percentage of the yeast fermentation product extract in the composition is 5.0 to 15.0%, the mass percentage of the lysate of the yeast fermentation product in the composition is 1.0 to 10.0%, the mass percentage of the carnosine in the composition is 0.05 to 0.5%, the mass percentage of the p-hydroxyacetophenone in the composition is 0.15%, and the balance is water.
3. The composition for skin aging resistance according to claim 2, wherein the mass percentage of the yeast fermentation product extract in the composition is 7.0 to 12.0%, the mass percentage of the lysate of the yeast fermentation product in the composition is 3.0 to 8.0%, the mass percentage of the carnosine in the composition is 0.1 to 0.3%, the mass percentage of the p-hydroxyacetophenone in the composition is 0.15%, and the balance is water.
4. A composition for skin aging according to claim 3, wherein the yeast fermentation product extract is 10.0% by mass of the composition, the lysate of the yeast fermentation product is 5.0% by mass of the composition, the carnosine is 0.2% by mass of the composition, the p-hydroxyacetophenone is 0.15% by mass of the composition, and the balance is water.
5. A process for the preparation of a composition for skin anti-aging according to any one of the claims 1 to 4,
mixing the components of yeast fermentation product extract, a two-split yeast fermentation product lysate, carnosine and p-hydroxyacetophenone in solvent water according to a certain proportion, wherein the components of the yeast fermentation product extract, the carnosine and the p-hydroxyacetophenone are premixed in a container according to a certain proportion in the preparation process of the product, and then the product is added into a skin care product, and the yeast fermentation product extract is added with pepper in a fermentation substrate.
6. A cosmetic product comprising the composition of any one of claims 1 to 4, wherein the cosmetic product is in a form selected from the group consisting of a solution, an emulsion, a cream, a gel, a solid and a liposome.
7. Use of a composition for the preparation of a cosmetic for skin anti-aging according to any one of claims 1 to 4.
8. Use of a composition obtainable by the process according to claim 5 or of a cosmetic according to claim 6 for non-therapeutic cosmetic and/or dermatological treatment, which acts on the surface of the skin, including skin tissue at the facial, neck or abdominal location.
