The content of pyrrolidone carboxylic acid in skin is determined by High Performance Liquid Chromatography (HPLC). Natural Moisturizing Factor (NMF) is mainly composed of free amino acids and their metabolites (such as pyrrolidone carboxylic acid), and thus the production of natural moisturizing factor in skin can be evaluated by measuring the change in the content of pyrrolidone carboxylic acid in skin.
The test personnel: all panelists were sensitive muscles of several dry, pruritic, desquamated, 40-45 year old, disease-free women.
Experimental reagent: sodium pyrrolidone carboxylate (purity is more than or equal to 98 percent), methanol (chromatographic purity), potassium dihydrogen phosphate (analytical purity), sodium alkane sulfonate (chromatographic purity) and phosphoric acid (chromatographic purity).
Experimental equipment: LC310 high performance liquid chromatograph (purchased from Jiangsu Tianrui instruments, inc.), analytical balance (sensory quality is 0.0001G), analytical balance (sensory quality is 0.00001G), and desktop high-speed centrifuge AXTG16G (purchased from Shanghai Zhao laboratory centrifuge, inc.).
Manufacturing a skin patch: the test persons were divided into 18 groups of 5 persons each. Preparation of experimental control solutions: accurately weighing 50mg of sodium pyrrolidone carboxylate, placing the sodium pyrrolidone carboxylate into a 10ml measuring flask, dissolving the sodium pyrrolidone carboxylate with methanol and diluting the sodium pyrrolidone carboxylate to scale, shaking the measuring flask uniformly to obtain a first mixed solution, accurately weighing 1ml of the first mixed solution from the measuring flask, placing the first mixed solution into a 100ml measuring flask, diluting the first mixed solution to scale with 0.02mol/L potassium dihydrogen phosphate solution (containing 5mmol/L sodium heptane sulfonate and regulating the pH value to 2.5 with phosphoric acid) to prepare a second mixed solution containing 50 mu g of sodium pyrrolidone carboxylate per 1ml, and diluting the second mixed solution by 10 times to obtain a control solution.
The experimental method comprises the following steps: the test sample solution and the test control solution were subjected to measurement under chromatographic conditions (column: thermo Betabasic Phenyl (250 mm. Times.4.6 mm,5 μm)), mobile phase: methanol-0.02 mol/L potassium dihydrogen phosphate solution (containing sodium heptanesulfonate 5mmol/L, phosphoric acid adjusted pH to 2.5) (3. Example 4 differs from example 1 in that in example 4, the mass ratio of the yucca schidigera extract, the trimeprazole alisma extract and the opuntia ficus indica extract in the soothing and repairing essence is 5:5:2, the relieving and repairing essence compounded according to the proportion is proved to be capable of further promoting the generation of natural skin moisturizing factors, and better increasing the water content of the horny layer to achieve the effect of moisturizing the skin.
Examples 5 to 7 showed no significant change in pyrrolidone carboxylic acid content compared with example 4, and thus it was confirmed that the simultaneous addition of 5 plant extracts of yucca brevicaulis root extract, trimedia extract, opuntia ficus indica extract, ampelopsis grossedentata extract, and moutan root bark extract enhanced the generation effect of natural moisturizing factors of the soothing and repairing essence to some extent.
Experiment 2
The oxidation resistance of the soothing repair essence is measured by a DPPH free radical clearance experiment.
Experimental reagent: DPPH (purity is more than or equal to 99%), absolute ethyl alcohol (analytically pure)
Experimental equipment: ultraviolet-visible spectrophotometer (available from Shanghai Jing and analytical instruments, inc.).
The experimental method comprises the following steps: the preparation concentration is 2.0 multiplied by 10 -4 A DPPH ethanol solution of mol/L (20 mgDPPH is weighed and added with absolute ethyl alcohol to be dissolved, and then transferred to a 250ml volumetric flask to be constant in volume); respectively weighing 1ml of the soothing repair essence to be tested, 1ml of absolute ethyl alcohol and 1ml of DPPH ethanol solution, uniformly mixing, immediately measuring the absorbance value at 517nm, calculating as A1, reacting for 30min, and thenMeasuring the absorbance value at 517nm, and recording as A2; weighing 1ml of the soothing repair essence to be detected and 1ml of absolute ethyl alcohol again, uniformly mixing, reacting for 30min, and measuring the absorbance value at 517nm, and recording as A3; DPPH clearance was calculated according to this formula: DPPH (%) = [1- (A1-A2)/A3]* The results of the measurements are shown in Table 3, at 100%.

Through comparison between the example 4 and the examples 5-7, it can be shown that the soothing and repairing essence containing 5 plant extracts of yucca brevifolia root extract, trimeresura alata extract, opuntia ficus indica extract, ampelopsis grossedentata extract and peony root bark extract has the effect of obviously improving the removal of free radicals, and the soothing and repairing essence containing 5 plant extracts can effectively improve the oxidation resistance of the skin, so that the repairing effect of a sebaceous membrane is better, and the skin barrier of sensitive muscles can be better repaired.
By comparing the example 7 with the comparative example 4 and the example 7 with the comparative example 5, it can be shown that the effect of obviously improving the clearance rate of free radicals can not be achieved by adding the ampelopsis grossedentata extract or the peony root bark extract alone, and meanwhile, by adding the 5 plant extracts of the yucca brevifolia root extract, the sanzhi largeflower acanthus extract, the ampelopsis grossedentata extract and the peony root bark extract, the clearance rate of free radicals can be obviously improved, and the relieving and repairing essence has better oxidation resistance.
Comparison of examples 8-10 with example 7 shows that the addition of six plant extracts, yucca brevifolia root extract, mirabilis javanica extract, opuntia ficus-indica extract, vitis vinifera extract, paeonia suffruticosa bark extract and purslane extract, can significantly increase the clearance of free radicals.
By comparing the example 10 with the comparative example 6, it can be shown that when the purslane extract is compounded with the yucca schidigera root extract, the trifingerlarea extract, the opuntia ficus-indica extract, the ampelopsis grossedentata extract and the peony root bark extract, the clearance rate of the soothing repair essence to free radicals is obviously improved, the compounding of the six plant extracts can obviously improve the oxidation resistance of the purslane extract, and in the example 10, the improvement of the oxidation resistance of the purslane extract is not only dependent on the function of the trifingerlarea extract for promoting the efficient absorption of other components.
Experiment 3
And judging the continuous moisturizing effect of the soothing and repairing essence by using a skin detector. The moisture-preserving effect of the soothing and repairing essence can be judged by detecting the water content of the stratum corneum, the repairing effect of the soothing and repairing essence on the skin barrier can be judged by detecting the TEWL value, and the smaller the TEWL value is, the better the skin barrier function is shown.
Experimental equipment: ASA-M100 skin stratum corneum moisture content tester (purchased from guining (shanghai) experimental equipment ltd.) and ASCH percutaneous moisture loss tester (purchased from guining (shanghai) experimental equipment ltd.).
The test personnel: all panelists were sensitive muscles of several dry, pruritic, desquamated, 40-45 year old, disease-free women.
The experimental method comprises the following steps: the test persons were divided into 18 groups of 5 persons each. Each group of testers respectively uses the soothing and repairing essence lotion in each embodiment and comparative example in the morning and evening, before each use, the face needs to be cleaned, then the soothing and repairing essence lotion is uniformly smeared on the face, then the face is beaten by gently massaging hands for 2min, and the testers need to continuously use the soothing and repairing essence lotion for 28 days.
Before use, on the 28 th day after use, the water content of the horny layer in the same area of the skin of a tester is detected by an ASA-M100 horny layer water content tester according to the formula: the water content increase rate (%) of the skin stratum corneum = [ (water content of skin stratum corneum after use-water content of skin stratum corneum before use)/water content of skin stratum corneum before use ] × 100%, the water content increase rate of the skin stratum corneum was calculated, and the experimental results are shown in table 4.
The TEWL values of the same area of the skin of the test subjects were measured using the ASCH transdermal Water loss Meter before, on days 2, 7, 14, 21, and 28, respectively, and the results are shown in Table 5.
