Abstract
The invention belongs to the field of scalp care daily chemicals, and particularly relates to a preparation method of a pichia pastoris extract and a scalp care composition containing the same. By extracting the scalp care active ingredients in pichia pastoris, reasonably building a composition system architecture and limiting key parameters of raw materials, the composition can solve the problems of dryness, sensitivity, greasiness, hair follicle blockage, alopecia, excessive grease secretion and the like of some scalps and the defects of the existing hair care products, and provides a refreshing product for hair care.
Description
Technical Field
The invention relates to the field of scalp care daily chemicals, and particularly relates to a preparation method of a pichia pastoris extract and a composition containing the pichia pastoris extract.
Background
The root cause of alopecia is degenerative atrophy of hair follicles, and the common types of alopecia are mainly the following types: (1) healthy people have 50-100 hairs shed every day, which is caused by the normal metabolism of the human body. The growth of hair is periodic, for example, the hair falls off in the catagen and telogen periods due to combing hair or excessive force of shampooing, so as to form dynamic balance. (2) Androgenetic alopecia is one of the most common types of alopecia, also known as "seborrheic alopecia," and is often accompanied by increased hair scalp oil. The human body suffering from such alopecia is not characterized by abnormal androgen levels, but hair follicles in the area of alopecia are sensitive to androgen levels, resulting in a decrease in hair follicle function under the continuous action of androgen, a decrease in newly grown hair, and an increase in hair loss. Furthermore, sebaceous glands are also sensitive to androgens, resulting in the secretion of large amounts of oil and the appearance of seborrheic dermatitis leading to dandruff and the like. (3) Alopecia areata: organ-specific autoimmune diseases mediated by cells affect the hair follicles in the anagen phase, causing premature hair loss. Alopecia areata is also called round alopecia. The alopecia has no inflammation and no subjective symptom on local parts, and is often discovered in an unintentional way. The traditional Chinese medicine considers that the pathogenesis of the disease is related to factors such as emotional internal injury, deficiency of qi and blood, dysfunction of viscera and the like. (4) Postpartum alopecia: postpartum alopecia is mainly related to unstable hormone secretion level in the body during pregnancy, and may be related to mental factors and dietary factors. Levels of hormone secretion are unstable, mainly abnormal estrogen or androgen secretion. Abnormal secretion of hormones in the body after delivery can affect normal metabolism of hair, so that a pregnant woman can cause emotional instability, increased mental stress and possible postpartum depression due to various reasons after delivery, thereby causing endocrine metabolism disorder in the body and causing hair loss due to stimulation of other factors.
In recent years, more and more people pay attention to scalp care products, prevent hair loss and nourish hair, and keep scalp health becomes a new hotspot in the field of cosmetics. Various anti-alopecia products are launched on the market, and the consumer demand of the anti-alopecia and hair-growth market generally focuses on health preservation and prevention, so that the demand has the potential. Young consumers can stay up to lose hair and buy a hair-nourishing order at the same time. The young people with the bald heads are hooked with the working pressure directly, the anti-hair loss consumption of young consumer groups in nearly three years is structurally changed, and the cost proportion of scalp essence and a high-tech hair growth instrument is continuously improved.
The pichia pastoris extract is widely applied to the food industry as a natural and safe active substance, and can also exert unique effects in the field of cosmetics. Yeast extract is rich in amino acids, oligopeptides and trace elements including various vitamins and minerals. Amino acids play an indispensable role in skin care as components contained in the skin itself; the oligopeptide components can be used as signal molecules to participate in various biological reactions such as competition with tyrosinase so as to inhibit the generation of melanin; yeast extract promotes mitochondrial fusion, thereby increasing energy production for hair follicle development; limiting the over-expression of signal molecules involved in androgenetic alopecia; can reduce epigenetic dysregulation in alopecia models; can stimulate synthesis of pluripotent proteoglycan, a specific proteoglycan for long-term hair generation, thereby maintaining activity of dermal papilla and inducing ability of hair follicle development. Can improve the growth of hair follicles treated with DHT1 by maintaining the synthesis of pluripotent proteoglycans and Ki-67. The technical scheme of applying the composition to the scalp care composition can make a contribution to the field of scalp and hair care, and provides a product with alopecia prevention and comprehensive care function.
Disclosure of Invention
In view of the above, the invention aims to provide a preparation method of pichia pastoris extract and a composition containing the same, which can solve the problems of dryness, sensitivity, greasiness, hair follicle blockage, alopecia, excessive grease secretion and the like of some scalps and the defects of the existing hair care products by extracting scalp care effective components in pichia pastoris, reasonably building a composition system architecture and limiting key parameters of raw materials, and provide a fresh product for hair care.
The technical scheme of the invention for solving the technical problems is as follows:
the invention provides a preparation method of a pichia pastoris extract, which comprises the following steps:
A. adding deionized water into a container, adding yeast paste or yeast powder of Pichia pastoris into water, stirring thoroughly for dissolving, and filtering with 200-250 mesh filter cloth to obtain clear water phase; centrifuging, and taking the lower layer liquid with darker color as a first centrifugate; resuspending the first centrifugate with PBS, centrifuging again, and taking the supernatant as the second centrifugate;
B. crushing the second centrifugate, and adding a buffer solution to adjust the pH value to be between 5 and 6; preferably, this step should be broken until more than 50% of the yeast cells are broken as observed under the microscope. This step breaks the yeast cells sufficiently to about 1 micron, releasing the intracellular material. In some embodiments, the disruption step is a 1min high pressure cell disruption instrument disruption, 1min pause followed by another 1min disruption, and 30 repetitions.
C. B, maintaining the temperature of the solution obtained in the step B in a reaction kettle at 58-60 ℃, and carrying out autolysis for 24-48 hours by using yeast self-hydrolase to obtain a self-solution; this step allows the cell endoplasmic to be completely autolyzed, and then allows more effective components to be obtained by controlling the autolysis time. Preferably, the autolysis period is also followed by intermittent stirring, for example every 3 hours for 25min.
D. In a microwave electromagnetic field, quickly heating the autolysis liquid to 100 ℃ to inactivate yeast hydrolase, then cooling to 50-55 ℃ to perform microwave extraction for 1-3 times with water, keeping the microwave power at 550-650W, and extracting for 20-45min to obtain an extract; this step can be used to extract the active ingredients of the yeast cells from the solution after autolysis is completed using microwave extraction.
E. Extracting the extractive solution with microporous membrane, filtering, removing residue, standing, centrifuging for 20-30min, settling, and separating to obtain organic phase to obtain crude extractive solution;
F. concentrating the crude extractive solution under reduced pressure to 10-12% of the original volume, and making into powder to obtain Pichia pastoris extract. In some embodiments, milling uses an ultrasonic gas atomization and rapid cooling milling technique to obtain pichia extract powder. The ultrasonic atomization fast cooling powder preparation process is that the liquid medicine added with maltodextrin forms high-speed pulsating airflow through a Laval nozzle with a valve, so that the temperature of the liquid can be rapidly reduced, meanwhile, the fine pichia pastoris extract powder is prepared by matching with ultralow temperature freeze drying, is convenient to store and transport, and is dissolved into liquid by water when being used for the next time.
The invention also provides a scalp care composition containing the pichia pastoris extract; preferably, the cactus stem extract is also contained.
Further, the scalp care composition comprises the following components in percentage by weight: 0.3 to 0.7 percent of pichia pastoris extract, 0.2 to 0.5 percent of opuntia ficus-indica stem extract, 9.5 to 22.8 percent of auxiliary agent and the balance of water to be up to 100 percent.
Further, the scalp care composition comprises the following components in percentage by weight: 0.3 to 0.7 percent of pichia pastoris extract, 0.2 to 0.5 percent of opuntia ficus-indica stem extract, 0.5 to 1.2 percent of caffeine, 2 to 4 percent of encapsulated salicylic acid with the salicylic acid content of 10 to 99 percent, 0.1 to 0.5 percent of dipotassium glycyrrhizinate, 0.5 to 1 percent of caprylyl glycine, 0.5 to 1 percent of skin conditioner, 5 to 7 percent of ethanol, 0.5 to 5 percent of polyhydric alcohol, 0.05 to 0.2 percent of freshener, 0.2 to 0.5 percent of solubilizing emulsifier, 0.05 to 0.2 percent of flavoring agent, 0.1 to 1 percent of preservative, and also contains a pH regulator which is enough to regulate the pH value to the required value and the balance of water.
Further, the skin conditioner is at least one of betaine, sodium PCA, niacinamide, vitamin B6, and tocopherol acetate. The skin conditioner is used as a moisturizing factor and plays a role in moisturizing and nourishing.
Further, the polyalcohol is at least one of propylene glycol, 1, 3-propylene glycol, butanediol and glycerol.
Further, the cooling agent is at least one of menthol, menthol lactate and encapsulated menthol lactate. Has effects of refreshing, relieving itching, and shrinking follicular orifice.
Further, the solubilizing emulsifier is at least one of PEG-40 hydrogenated castor oil and PPG-26-butanol polyether-26. Has effects of solubilizing essence and refreshing agent
The invention also provides a preparation method of the scalp care composition, which comprises the following steps:
s1, dissolving caprylyl glycine in hot water of 80-85 ℃ accounting for 10-20% of the total amount, and neutralizing with alkali until the pH value is 5.0-6.0;
s2, adding polyalcohol into the aqueous solution prepared in the S1, and then standing and cooling to room temperature;
s3, adding ethanol, and uniformly stirring;
s4, dispersing and dissolving the skin conditioner and dipotassium glycyrrhizinate by using a small amount of water, and adding the mixture into the solution;
s5, dissolving caffeine and coated salicylic acid by using water accounting for 20% of the total amount, and adding the dissolved caffeine and coated salicylic acid into the solution;
s6, dispersing and dissolving the pichia pastoris extract powder completely by using part of water, adding the pichia pastoris extract powder and the opuntia ficus-indica extract into a system, and stirring uniformly;
s7, mixing and fully dissolving the freshener, the aromatizing agent, the preservative and the solubilizing emulsifier, adding the mixture into a system while stirring, and uniformly stirring;
s8, aging at low temperature for 12 hours, and filtering with 200-mesh filter cloth to obtain the scalp care composition.
The invention has the following beneficial effects:
the invention adopts the technologies of high-pressure crushing, enzymolysis and ultrasonic extraction to extract the pichia pastoris, the hydrolysis degree is thorough under the optimal condition, and the yield of effective substances is high. Meanwhile, the active substances are separated in the extraction step, and the enzyme is inactivated, so that the time consumption of the working procedure and the step consumption are saved. And finally, an ultrasonic atomization freezing powder preparation technology is adopted during concentration, so that the activity of effective substances in more yeasts is better preserved by rapid cooling, the problem that the activity of the effective substances is reduced due to the use of other drying modes is avoided, the utilization rate of raw materials and the yield of finished products are improved, and the waste of the raw materials is avoided. The prepared powder finished product is convenient to store and transport, and can be added into a system after being pre-dissolved by water when in use, so that the working efficiency and the economic benefit are improved. The Pichia pastoris extract contains more amino acids, oligopeptides and trace elements including various vitamins and minerals than the extract prepared by the general extraction method, and can promote hair growth and improve the influence of androgen on hair follicle development.
The pichia pastoris extract is matched with other components to be applied to the formula of the scalp care composition, so that the scalp state can be improved, the unhealthy state of the scalp can be reduced, the cell renewal can be promoted, the stimulation caused by scalp injury and external factors can be reduced, and the scalp renewal cycle can be accelerated. Meanwhile, the scalp water-oil balance is regulated, 5 alpha-reductase is inhibited, skin oil secretion is reduced, the expression of signal molecules is limited, and the scalp micro-environment is improved. The cactus stem extract of the prickly pear and the pichia pastoris extract have a synergistic effect, and can enhance the effects of preventing hair loss, nourishing hair, controlling oil and relieving the tension of the pichia pastoris extract.
The pichia pastoris extract is applied to a hair care composition, all components in the extract components are mutually complementary and play a role together, compared with the common commercial care products, the pichia pastoris extract has more excellent quick-drying effect and a certain relieving and oil controlling effect, and after the extract is added, the pichia pastoris extract can keep the hair fresh and repair the scalp barrier, so that the scalp is prevented from being damaged by ultraviolet rays in the environment and chemical factors.
Detailed Description
The present invention will be described in detail with reference to examples, which are only preferred embodiments of the present invention and are not intended to limit the present invention.
The following examples and comparative examples were prepared and carried out using the following methods:
a preparation method of a pichia pastoris extract comprises the following steps:
A. adding deionized water into a container, adding yeast paste or yeast powder of Pichia pastoris purchased from the market into water, fully stirring and dissolving, and fully filtering by using 200-mesh filter cloth to obtain a clear water phase; centrifuging by using a micro centrifuge, and taking the lower layer liquid with darker color as a first centrifugate; resuspending the first centrifugate with PBS, centrifuging again, and taking the supernatant as a second centrifugate;
B. crushing the second centrifugate by using a high-pressure cell crusher for 1min, performing 1min intermission, crushing again for 1min, repeating for 30 times, and adding a buffer solution to adjust the pH to 5.5;
C. keeping the temperature of the solution obtained in the step B at 58-60 ℃ in a reaction kettle, carrying out autolysis for 24 hours by using yeast self-hydrolase, and stirring for 25min every 3 hours during the autolysis period to obtain an autolysis solution;
D. in a microwave electromagnetic field, quickly heating the autolysis liquid to 100 ℃ to inactivate yeast hydrolase, then cooling to 50-55 ℃ to perform microwave extraction, keeping the microwave power at 550 watts, and extracting for 30min to obtain an extract liquid;
E. extracting the extractive solution with microporous membrane, filtering, removing residue, standing, centrifuging for 30min, settling, and separating to obtain a crude extractive solution;
F. and (3) concentrating the crude extract to 10% of the original volume by using a rotary evaporator under reduced pressure, adding maltodextrin in proportion, and then obtaining pichia pastoris extract powder by using an ultrasonic gas atomization quick cooling powder preparation technology.
The yield of the pichia pastoris extract process (weight of extracted target substance/usage of raw material x 100%) is 46.7%.
A method of preparing a scalp care composition comprising the steps of:
s1, dissolving caprylyl glycine in hot water of 85 ℃ accounting for 15% of the total amount, and neutralizing with alkali until the pH value is 5.5;
s2, adding polyhydric alcohol into the aqueous solution prepared in the S1, and then standing and cooling to room temperature;
s3, adding ethanol, and uniformly stirring;
s4, dispersing and dissolving the skin conditioner and dipotassium glycyrrhizinate by using a small amount of water, and adding the mixture into the solution;
s5, dissolving the caffeine and the coated salicylic acid by using water with the total amount of 20%, and adding the dissolved caffeine and the coated salicylic acid into the solution;
s6, completely dispersing and dissolving the pichia pastoris extract powder by using part of water, adding the powder and the opuntia ficus-indica extract into a system, and uniformly stirring;
s7, mixing and fully dissolving the freshener, the aromatizing agent, the preservative and the solubilizing emulsifier, adding the mixture into a system while stirring, and uniformly stirring;
s8, ageing at low temperature for 12 hours, and filtering by using 200-mesh filter cloth to obtain the scalp care composition.
The formulations of three examples and four comparative examples of a scalp care composition containing pichia pastoris extract are shown in the following table.
For the above examples and comparative examples, 20 volunteers arbitrarily selected were subjected to instrument efficacy evaluation and human trial sensory self-evaluation after one month using the above 3 example compositions and 4 comparative examples, respectively, to finally obtain average results as shown in the following table. The used instruments comprise a hair detector, a scalp care physiotherapy instrument and a scalp grease tester.
The results of examples 1 to 3 show that, after the scalp care composition claimed in the present invention is used, the examples show significant effects of promoting hair growth, reducing hair loss and increasing hair density, and further, can reduce the oil content of scalp, increase the freshness of hair, and have a significant effect of scalp soothing. Examples 1-3 and comparative example 1 show that 0.3-0.7% of pichia pastoris extract is a more preferable addition concentration, and the increase of the addition amount for preventing alopecia and other effects is limited. Comparative example 2 shows that pichia pastoris is indispensable to the effects of hair loss prevention, hair care, oil control and relieving, and combined with examples and comparative examples 2-3, the cactus stem extract and the pichia pastoris extract have a synergistic effect, so that the effects of hair loss prevention, hair growth promotion, oil control and relieving of the pichia pastoris extract can be enhanced, and meanwhile, the effect of the yeast extract on scalp oil control is remarkably promoted. The pichia pastoris extract is matched with other components to be applied to the formula of the scalp care composition, so that the scalp state can be improved, the unhealthy state of the scalp can be reduced, the cell renewal can be promoted, the stimulation caused by scalp injury and external factors can be reduced, and the scalp renewal cycle can be accelerated. Meanwhile, the scalp water-oil balance is regulated, 5 alpha-reductase is inhibited, skin oil secretion is reduced, the expression of signal molecules is limited, and the scalp micro-environment is improved. The pichia pastoris extract is applied to a hair care composition, all components in the extract components are mutually complementary and play a role together, and compared with a common nursing product sold in the market, the pichia pastoris extract is a scalp care composition which has more excellent quick-drying effect and certain relieving and oil controlling effects, and after the extract is added, the pichia pastoris extract can keep the hair fresh and repair the scalp barrier, so that the scalp is prevented from being damaged by ultraviolet rays in the environment and chemical factors.
The preferred embodiments of the present invention have been described in detail, however, the present invention is not limited to the specific details of the above embodiments, and various simple modifications may be made to the technical solution of the present invention within the technical idea of the present invention, and these simple modifications are within the protective scope of the present invention.
It should be noted that the various features described in the foregoing embodiments may be combined in any suitable manner without contradiction. The invention is not described in detail in order to avoid unnecessary repetition.
In addition, any combination of the various embodiments of the present invention can be made, and the same should be considered as the disclosure of the present invention as long as the idea of the present invention is not violated.
Claims (9)
1. A preparation method of a pichia pastoris extract is characterized by comprising the following steps:
A. adding deionized water into a container, adding yeast paste or yeast powder of Pichia pastoris into water, stirring thoroughly for dissolving, and filtering with 200-250 mesh filter cloth to obtain clear water phase; centrifuging, and taking the lower layer liquid with darker color as a first centrifugate; resuspending the first centrifugate with PBS, centrifuging again, and taking the supernatant as the second centrifugate;
B. crushing the second centrifugate, and adding a buffer solution to adjust the pH value to be between 5 and 6;
C. keeping the temperature of the solution obtained in the step B at 58-60 ℃ in a reaction kettle, and carrying out autolysis for 24-48 hours by using yeast self-hydrolase to obtain a self-solution;
D. in a microwave electromagnetic field, quickly heating the autolysis liquid to 100 ℃ to inactivate yeast hydrolase, then cooling to 50-55 ℃, performing microwave extraction for 1-3 times by using water, keeping the microwave power at 550-650 watts, and extracting for 20-45min to obtain an extract;
E. extracting the extractive solution with microporous membrane, filtering, removing residue, standing, centrifuging for 20-30min, settling, and separating to obtain clear solution;
F. and (3) concentrating the crude extract under reduced pressure to 10-12% of the original volume, and making powder to obtain the pichia pastoris extract.
2. A scalp care composition comprising the pichia pastoris extract of claim 1.
3. A scalp care composition according to claim 2, characterized by comprising the following components in percentage by weight: 0.3 to 0.7 percent of pichia pastoris extract, 0.2 to 0.5 percent of opuntia ficus-indica stem extract, 9.5 to 22.8 percent of auxiliary agent and the balance of water to be up to 100 percent.

A scalp care composition according to claim 2, characterized by comprising the following components in percentage by weight: 0.3-0.7% of pichia pastoris extract, 0.2-0.5% of opuntia ficus-indica stem extract, 0.5-1.2% of caffeine, 2-4% of encapsulated salicylic acid with the salicylic acid content of 10-99%, 0.1-0.5% of dipotassium glycyrrhizinate, 0.5-1% of caprylyl glycine, 0.5-1% of skin conditioner, 5-7% of ethanol, 0.5-5% of polyhydric alcohol, 0.05-0.2% of freshener, 0.2-0.5% of solubilizing emulsifier, 0.05-0.2% of flavoring agent, 0.1-1% of preservative, pH regulator capable of regulating the pH value to the required value and the balance of water.
5. A scalp care composition according to claim 4 characterized in that said skin conditioning agent is at least one of betaine, sodium PCA, niacinamide, vitamin B6, tocopherol acetate.
6. A scalp care composition according to claim 4, characterized in that the polyhydric alcohol is at least one of propylene glycol, 1, 3-propanediol, butylene glycol, glycerol.
7. A scalp care composition according to claim 4, wherein the cooling agent is at least one of menthol, menthol lactate, and encapsulated menthol lactate having a menthol lactate content of 10-99%.
8. A scalp care composition according to claim 4, wherein the solubilizing emulsifier is at least one of PEG-40 hydrogenated castor oil, PPG-26-Butaneth-26.
9. A method of preparing a scalp care composition according to any one of claims 2 to 8 comprising the steps of:
s1, dissolving caprylyl glycine in hot water of 80-85 ℃ accounting for 10-20% of the total amount, and neutralizing with a pH regulator until the pH value is 5.0-6.0;
s2, adding polyhydric alcohol into the aqueous solution prepared in the S1, and then standing and cooling to room temperature;
s3, adding ethanol, and uniformly stirring;
s4, dispersing and dissolving the skin conditioner and the dipotassium glycyrrhizinate by using a small amount of water, and adding the mixture into the solution;
s5, dissolving the caffeine and the coated salicylic acid by using water with the total amount of 20%, and adding the dissolved caffeine and the coated salicylic acid into the solution;
s6, dispersing and dissolving the pichia pastoris extract powder completely by using part of water, adding the pichia pastoris extract powder and the opuntia ficus-indica extract into a system, and stirring uniformly;
s7, mixing and fully dissolving the freshener, the aromatizing agent, the preservative and the solubilizing emulsifier, adding the mixture into a system while stirring, and uniformly stirring;
s8, aging at low temperature for 12 hours, and filtering with a 200-mesh filter cloth to obtain the scalp care composition as claimed in any one of claims 2 to 8.
