Abstract
本发明公开了一种改善食欲和/或促进消化吸收的组合物及其应用,包括按照重量百分比计的以下组分:益生菌0.01‑2%,富锌酵母3‑15%,益生元5‑65%,矫味剂20‑60%;所述益生菌包括乳双歧杆菌和/或长双歧杆菌。本发明所提供的组合物采用益生菌、益生元和富锌酵母进行组合,三重功效原料共同助力提升食欲和/或促进消化吸收。并且该组合物在原料的选择上,考虑了口感、味道、性状之间的相互配合,无需添加防腐剂或香味剂等人工合成添加剂,口感好且安全健康,无副作用、无依赖性。
Description
技术领域
本发明涉及益生菌技术领域,尤其涉及一种改善食欲和/或促进消化吸收的组合物及其应用。
背景技术
益生菌是一种活的微生物,当给予足够的量时,会给宿主带来健康益处。研究表明益生菌能通过自身代谢维持肠道菌群平衡,抑制有害细菌生长。因此肠内益生菌与人类健康密切相关,在维持肠黏膜屏障功能、调节机体免疫功能和促进营养物质吸收方面具有重要作用。例如,益生菌可分泌水解酶,参与营养物质的分解和吸收,并参与许多维生素的合成;益生菌在代谢过程中可产生乳酸、醋酸及其他有机酸,降低肠道pH值,促进矿物质(钙、铁、镁和锌)和维生素D的吸收。
肠道菌群不仅能够干预宿主的一般生理调节,甚至能对中枢神经系统产生显著地干预作用,调节宿主的行为和情绪。食欲是中枢调节的行为结果之一,这也为肠道菌群改变宿主的食欲状态提供了潜在的可能。近几年来,越来越多的证据表明肠道菌群紊乱和宿主的食欲紊乱存在一定联系。有研究表明神经性厌食症的患者和肥胖症患者均存在肠道菌群紊乱的现象。
虽然体内只有少量锌,但是其在许多活性酶系统中起辅助因子的关键作用,例如影响消化的醛缩酶、肽酶和磷酸酶的活性都依赖于锌,因此,锌在许多酶、蛋白质、核酸、基因表达和细胞免疫发育的产生和运行中起着重要作用。同时,锌在人体发育、认知、味觉调节中也发挥重要作用。锌元素参与儿童生长发育与正常食欲和味觉维持、免疫功能等多种生理功能密切相关。
食欲减退是小儿缺锌的早期症状,由于缺锌妨碍核酸和蛋白质的合成,从而影响味蕾的新陈代谢和发育。唾液中含锌离子的味觉素为味蕾及口腔粘膜提供营养,缺锌时味觉就会减退且易患复发性口腔炎。同时缺锌时唾液中的磷酸酶减少,味蕾功能也会减退。另外缺锌所引起的粘膜增生和角化不全,可使脱落的粘膜阻塞味蕾小孔,使食物不能有效地引起味觉。因此缺锌的儿童常出现挑食、偏食、厌食、异食癖等食欲减退的表现。
目前,补充益生菌是调节肠道健康的常用手段,却鲜有资料表明锌与益生菌的联合使用具有有益作用,未有锌和益生菌联合使用用于改善食欲和促进消化吸收的产品问世。
发明内容
为了克服现有技术的不足,本发明的目的之一在于提供一种改善食欲和/或促进消化吸收的组合物。
本发明的目的之二在于提供所述改善食欲和/或促进消化吸收的组合物的应用。
本发明的目的之一采用如下技术方案实现:
一种改善食欲和/或促进消化吸收的组合物,包括按照重量百分比计的以下组分:益生菌0.01-2%,富锌酵母3-15%,益生元5-65%,矫味剂20-60%;所述益生菌包括乳双歧杆菌和/或长双歧杆菌。
作为本发明的一个优选方案,所述乳双歧杆菌为乳双歧杆菌HN019,所述长双歧杆菌为长双歧杆菌BB536;所述益生元包括低聚果糖、低聚半乳糖和抗性糊精中的一种或任意组合;所述矫味剂包括罗汉果粉、木糖醇和甜橙粉中的一种或任意组合精。
作为本发明的一个优选方案,所述组合物包括按照重量百分比计的以下组分:乳双歧杆菌HN019和/或长双歧杆菌BB5360.01-2%,富锌酵母3-15%,低聚果糖和/或低聚半乳糖5-15%,罗汉果粉0.1-5%,木糖醇15-35%,甜橙粉5-15%,抗性糊精30-50%。
作为本发明的一个优选方案,所述组合物包括按照重量百分比计的以下组分:乳双歧杆菌HN0190.01-2%,富锌酵母3-15%,低聚果糖和/或低聚半乳糖5-15%,罗汉果粉0.1-5%,木糖醇15-35%,甜橙粉5-15%,抗性糊精30-50%。

作为本发明的一个优选方案,所述组合物包括按照重量百分比计的以下组分:乳双歧杆菌HN0190.01-1%,长双歧杆菌BB5360.01-1%,富锌酵母5-15%,低聚果糖2-10%,低聚半乳糖2-10%,罗汉果粉0.1-5%,木糖醇15-35%,甜橙粉5-15%,抗性糊精30-50%。
作为本发明的一个优选方案,所述乳双歧杆菌在所述组合物中的活菌量为1.0×109~1.0×1011CFU/克;所述长双歧杆菌在所述组合物中的活菌量为1.0×109~1.0×1011CFU/克。
作为本发明的一个优选方案,所述组合物用于预防、改善或治疗功能性消化不良,从而改善食欲和/或促进消化吸收。
作为本发明的一个优选方案,所述组合物为口服制剂,所述口服制剂的剂型是口服液、片剂、粉剂、胶囊剂或颗粒剂。
本发明的目的之二采用如下技术方案实现:

相比现有技术,本发明的有益效果在于:
(1)本发明所提供的组合物,采用益生菌、益生元和富锌酵母进行组合,三重功效原料共同助力提升食欲和/或促进消化吸收。
(2)本发明所提供的组合物,在原料的选择上,考虑了口感、味道、性状之间的相互配合,无需添加防腐剂或香味剂等人工合成添加剂,口感好且安全健康,无副作用、无依赖性。
具体实施方式
下面,结合具体实施方式,对本发明做进一步描述,需要说明的是,在不相冲突的前提下,以下描述的各实施例之间或各技术特征之间可以任意组合形成新的实施例。在下述实施例中所采用的原材料、设备等除特殊限定外均可以通过购买方式获得。
实施例1-5
一种改善食欲和/或促进消化吸收的组合物,由益生菌粉、富锌酵母、益生元和矫味剂组成。其中,益生菌粉为乳双歧杆菌HN019菌粉和长双歧杆菌BB536菌粉的其中一种或两种组合;益生元为低聚果糖和/或低聚半乳糖,以及抗性糊精的组合;矫味剂为罗汉果粉、木糖醇和甜橙粉中的一种或任意组合。具体详见表1。

实施例1-5的组合物按照以下方法制备而成:
步骤一 烘干抗性糊精
把抗性糊精放入沸腾式烘干机中,设置进风温度为120℃,加热10min,将其水分活度降低至0.05以下,干燥后的物料转移至暂存间暂存。
步骤二 菌粉解冻
将益生菌粉原料(乳双歧杆菌HN019菌粉和/或长双歧杆菌BB536菌粉)从冷库取出后,放置在温度为18℃-26℃的环境解冻,时间大于等于12小时,小于等于48小时。
步骤三 称量
根据表1分别按照配方量称取各个原辅料。
步骤四 过筛和预混合
4.1将罗汉果粉和低聚半乳糖或/和低聚果糖进行预混。混合时间:10分钟。得预混料1。
4.2益生菌粉与部分抗性糊精按照1:10的比例预混。混合时间:10分钟。得预混料2。
4.3首先将预混料1过筛,投入混料罐中,随后将预混料2过筛,投入预混料中,再将剩余的其他物料(富锌酵母、木糖醇、甜橙粉和余下的抗性糊精)分别过14目筛后,投入混料罐中,备用。
步骤五 总混
将步骤四中的物料进行混合,混合时间:30分钟。
步骤六将步骤五的混合粉进行包装、检验,得到所述粉剂产品。
其中,在制备实施例1-5的组合物时,生产环境要求如下:步骤一至步骤五均在C级洁净区进行,而且环境温度≤25℃,相对湿度≤40%。步骤六在一般区进行,环境温度≤25℃。
本发明实施例在制备过程中,工艺上设计多重混合方法,梯度混合,确保各个原料均匀。
对比例1-7
对比例1-7分别提供了一种组合物,其中,对比例1-4与实施例5的区别在于矫味剂的不同。对比例5-7与实施例5的区别在于益生菌粉、益生元或富锌酵母的不同。具体配方详见表2。

在对比例1-7的制备方法中,根据原料的不同,其制备方法相比实施例1-5的制备方法做适当调整,具体如下。
对比例1的制备方法与实施例相比,区别在于:调整步骤四中的4.1为使用等份额的抗性糊精代替罗汉果粉。
对比例2的制备方法与实施例相比,区别在于:调整步骤四中的4.3为使用等份额的抗性糊精代替木糖醇。
对比例3的制备方法与实施例相比,区别在于:调整步骤四中的4.3为使用等份额的抗性糊精代替甜橙粉。
对比例4的制备方法与实施例相比,区别在于:调整步骤四中的4.1为使用等份额的抗性糊精代替罗汉果粉,同时调整4.3为使用等份额的抗性糊精代替木糖醇和甜橙粉。
对比例5的制备方法与实施例相比,区别在于:省略步骤二,及步骤四中的4.2。
对比例6的制备方法与实施例相比,区别在于:调整步骤四中的4.1为使用等份额的麦芽糊精代替低聚半乳糖和低聚果糖。4.2、4.3中的抗性糊精也全部替换为麦芽糊精。
对比例7的制备方法与实施例相比,区别在于:步骤4.3中省略富锌酵母的加入。
效果测试
一、感官测试
差异程度(DoD)是通过差异性评分来评价样品和参考样品之间是否存在感官差异。本次DoD测试涉及样品的整体口感和酵母味。
本次测试以实施例5为标准样品,实施例1-4和对比例1-4分别为测试样品,10名经过训练的品评员首先品尝标准样品,然后逐一品尝测试样品,并将测试样品与标准样品的差异性进行打分,分数范围为0-4。其中,0代表没有差异,1代表有轻微差异,2代表有一点差异,3代表有明显差异,4代表有非常明显差异。如有出现打分为3或者4的情况,则要求品评员描述具体的差异现象。
样品准备:分别取样品2g,用20ml水溶解,且要求水温控制在40℃以下。
样品的对应信息和品评结果分别如下表3和表4所示,表4中每个样品的得分为10名品评员的平均分。

如表4所示,实施例1-5的样品在整体口感和酵母味道上均十分相似,无差别。但是对比例1-4与实施例相比,差异明显,经过品评员描述,实施例1-5口感好,有甜味和水果味,无酵母粉的刺激味,非常易接受。而对比例1-4口感寡淡,且酵母味十分明显,非常不容易接受。
本发明实施例所提供的改善食欲和/或促进消化吸收的组合物,作为益生菌类功能性食品,目标人群主要为儿童和青少年,产品具有良好的、能被接受的食用口感是非常重要的。综上感官测试结果,可以看出本发明实施例所提供的改善食欲和/或促进消化吸收的组合物口感好,能完全掩盖酵母粉的刺激味道,非常易接受。
二、改善功能性消化不良测试
1、造模。制备功能性消化不良小鼠模型。取160只小鼠,予以不规则喂食和夹尾刺激双因素法造模,即双日正常进食,逢单日禁食不禁水,连续2周后,继续以上喂养方法的同时,用长海绵钳夹大鼠尾巴远端1/3处,以不破皮为度,以激怒小鼠,每次夹尾刺激20min,每隔3h刺激1次,3次/天,连续刺激1周,所有小鼠均出现饮食不佳,大便稀溏,气味臭秽刺鼻,性情暴躁,易被激怒等症状,表示造模成功。
2、测试。将消化不良的小鼠随机分成8组,每组20只,每天分别将2g实施例1-5及对比例5-7的样品用5ml水溶解后,灌胃喂养,每天1次。其余时间小鼠自由进食和进水。另外选取与模型小鼠重量接近的20只小鼠作为对照组。对照组小鼠自由进食和进水,不做其他干预。分别在第1、2、3、4周,记录各组小鼠的日常表现、体重增加量以及饲料的消耗量。为了准确记录每一组小鼠的进食量,所有小鼠均单笼饲养于不锈钢笼内,并配以塑料水瓶和不锈钢吸水管。小鼠饲料配方为:小麦粉20%,玉米粉35%,豆饼粉25%,麸皮10%,鱼粉5%,酵母粉1%,植物油1.5%,骨粉1%,矿物质添加剂1%,维生素添加剂0.5%。
平均体重日增加量=(测试末体重量-测试开始体重量)/测试天数;
平均食物日摄入量=(每周饲料提供量-每周饲料剩余量)/每周天数。
如下表5展示了测试期间小鼠的状态,表6展示了测试期间小鼠的体重情况,表7展示了小鼠每周体重的变化情况,表8展示了测试期间小鼠的摄食量。表5-表8中,所有数据均为该组20只小鼠的平均值。

注:情绪反应指标中,稳定是最佳状态,其次是较稳定,最差的是易怒。
由表5中可得,实验期间,对照组小鼠的外观行为等未见异常。与对照组相比,一周后,实施例1-5组小鼠虽依旧表现出毛发不泽、活跃程度减弱、情绪易怒,但是大便情况已经由稀溏转变成便溏,有所好转;而对比例1-3组小鼠依旧表现出毛发不泽、活跃程度减弱、情绪易怒,稀溏。随着测试时间的延长,实施例1-5组的小鼠状态逐渐恢复,在2周后,实施例1-5组小鼠在皮肤毛发、活跃程度、情绪反应、大便状态等方面均明显的改善,与对照组无明显差异。然而实施例1-3组小鼠在4周后,虽然情况有所好转,但是依旧没有回复至正常状态。
综上所述,本发明实施例采用益生菌(乳双歧杆菌HN019和长双歧杆菌BB536)、益生元和富锌酵母进行组合,能够使得功能性消化不良小鼠得状态得到改善,特别是实施例5,在服用样品第一周后改善效果是最佳的。而对比例5缺了益生菌粉(乳双歧杆菌HN019和长双歧杆菌BB536),对比例7缺了富锌酵母,对比例5和对比例7的效果是最差的,对比例6缺了益生元,虽然效果比对比例5和对比例7好点,但是依然差于对照组,不能使得造模小鼠完全恢复。

表7的数据是在表6的基础上获得的,不考虑表6的标准差,仅以体重平均值为差值进行计算,每一个数据均为与上一周数据的差值。例如,实施例1第2周的数据,是表6中实施例1第2周数据减去第1周数据的差值。
从表6中可得,在测试的第1周,食用实施例1-5样品的小鼠体重虽然略低于对照组,但是明显高于食用实验对比例1-3组的小鼠。并且从第2周结束时开始,食用实施例1-5样品的小鼠体重已经追平并开始超过对照组的小鼠。说明实施例1-5的样品可以有效治愈小鼠的功能性消化不良,并且能够促进消化吸收。
从表7中可得,实验开始后,所有测试组小鼠体重均有所增加。测试的第1周,对照组小鼠增重最多,实验例小鼠由于身体在恢复中,因此实验例小鼠体重增加值低于对照组。但是,测试第一周结束后,食用实施例1-5组的小鼠和食用对比例1-3组的小鼠体重增加平均值分别为34.2g、17g,可以看出食用实施例1-5样品的小鼠比食用对比例5-7样品的小鼠体重明显增加。并且从实验的第2周开始,食用实施例1-5样品的小鼠已经恢复至正常状态,可以看出食用实施例1-5样品的小鼠增重情况均高于对照组和对比例5-7的小鼠。

由表8可知,实验开始后,所有测试组小鼠的每周摄食量随着周龄的增大呈现逐级递增的趋势,食用实施例1-5样品的小鼠自第2周开始,每周摄食量即与对照组接近,并且在第4周时略高于对照组,说明实施例1-5可在1周后改善或治愈小鼠的功能性消化不良。食用对比例5-7样品的小鼠摄食量并没有恢复至对照组的情况,说明对比例5-7对功能性消化不良并无明显改善。
综上所述,本发明实施例采用益生菌(乳双歧杆菌HN019和长双歧杆菌BB536)、益生元和富锌酵母进行组合,能够使得功能性消化不良小鼠得状态得到改善,特别是实施例5,对功能性消化不良小鼠的体重增长情况和摄食量改善情况时最佳的。而对比例5缺了益生菌粉(乳双歧杆菌HN019和长双歧杆菌BB536),对比例7缺了富锌酵母,对比例5和对比例7的效果是最差的,对比例6缺了益生元,虽然效果比对比例5和对比例7好点,但是依然差于对照组,对功能性消化不良并无明显改善。
上述实施方式仅为本发明的优选实施方式,不能以此来限定本发明保护的范围,本领域的技术人员在本发明的基础上所做的任何非实质性的变化及替换均属于本发明所要求保护的范围。
Claims (4)
1.一种改善功能性消化不良的组合物,其特征在于,由按照重量百分比计的以下组分组成:乳双歧杆菌HN019 0.05-0.1%,长双歧杆菌BB536 0.05-0.1%,富锌酵母5-15%,低聚果糖5-10%,低聚半乳糖5-10%,罗汉果粉1-5%,木糖醇15-35%,甜橙粉5-15%,抗性糊精30-50%。
2.如权利要求1所述的改善功能性消化不良的组合物,其特征在于,所述组合物的剂型是口服液、片剂、粉剂、胶囊剂或颗粒剂。
3.权利要求1所述的组合物在制备改善功能性消化不良产品中的应用。
4.如权利要求3所述的应用,其特征在于,所述产品为功能性食品或保健食品。
