Abstract
可用于通过内源基因激活生产人蛋白质的人细胞系的选择方法使得可以经济上可行的量、适于生产药物组合物的形式生产蛋白质。除此之外还公开了在用此方式鉴定出的细胞系中生产人蛋白质的方法。
Description
本发明涉及可用于通过内源基因激活生产人蛋白质之人细胞系的选择方法,以便以经济的产量及适于制备药用制品的形式生产人蛋白质。此外,本发明还涉及在以此方式鉴定出的细胞系中制备人蛋白质的方法。
在人细胞系中通过内源基因激活制备人蛋白质的方法是已知的。例如,WO93/09222、WO94/12650和WO95/31560中描述了通过内源基因激活而在人细胞系中生产人促红细胞生成素及其它人蛋白质。
然而,上述文件中并没有指出在筛选可用于制备人蛋白质的细胞系时必须遵守某些准则。因此不能确保在为其生产所选择的细胞系中能以所希望的产量和形式并且无污染地获得目的人蛋白质。所以在上面所提到的文件中通常只获得低产量的人蛋白质。
本发明的问题是要消除本领域这一状况中的不利因素,尤其是提供在选择适用于预定靶基因内源激活的人起始细胞系时的有关准则。
这一问题通过选择可用于经激活内源性存在于其中的靶基因而制备人蛋白质的人细胞系的方法解决,其特征在于:
(a)检测具有以下特征的人细胞系:
(i)带目的核酸序列的靶基因,
(ii)悬浮培养14天内数量至少倍增5次,及
(iii)在无血清培养基中培养14天内数量至少倍增5次,及
(b)用具特征(i)、(ii)、(iii)的细胞系作为起始细胞系进行靶基因的内源激活。
根据本发明,如果需要通过基因打靶激活人细胞系中的人靶基因和获得能以满意的产量和所期望的形式生产靶蛋白质的细胞,则需检测各细胞系是否有某些特征,这些特征是细胞系作为后面大规模生产靶蛋白质的合适候选者所必须具备的。优选如此检测无限增殖化细胞系,尤其是肿瘤细胞系,因为它们较非无限增殖化细胞在培养能力上具有重要的优势。
按特征(i),检测人细胞系以观察靶基因,即将通过内源基因激活而活化的基因是否确实具有所期望的核酸序列,该序列通常是天然靶基因的核酸序列。这是因为永久培养中的肿瘤细胞系及其它细胞系常常在其基因组中有一系列的突变。因此选择合适细胞系的一个重要方面是该细胞是否具有编码所期望产物的正确基因。序列的测定可通过以普通方式培养细胞并将靶基因测序而完成。如果需要,可通过PCR或其它扩增方法在测序前完成靶基因的扩增。
选择细胞系的另一重要特征是其悬浮培养的能力。悬浮细胞更易发酵,且此发酵更易适用于较大的规模,如在容积约10-50,000升的大发酵罐中发酵。因此所选择的细胞应或者是悬浮细胞或者是易于悬浮培养的细胞。为此,在持续搅拌下培养该细胞14天。如果在此期间细胞显示出数量至少翻了5番,就认为它们适于悬浮培养。细胞倍增数可通过周期性地测定细胞读数而确定,如用细胞计数或检测细胞悬浮液的光密度。
选择人细胞的另一重要特征是在无血清培养基中的培养能力。因为从无血清细胞培养物中纯化蛋白质方便得多,且在无血清培养物中不存在诸如病毒之类动物病原体污染的危险,所以被选择的细胞应能生长于无血清培养基中。为此,将该细胞以1-10×105个细胞/ml的密度在装有无血清培养基(如来自Boehringer Mannheim有ITS的RPMI 1650)的培养容器中培养14天。如果通过细胞计数测定出该细胞在此培养期间其数量至少翻了5番,则认为它们适于无血清培养。
对于本发明来说优选的另一重要特征是世代时间(iv)。所选择的细胞在诸如含小牛全血清10%的DMEM或含小牛全血清10%的RPMI1640之类的培养基中应有较高增殖水平,即在培养一周内它们的数量应倍增16-256次,优选64-128次。为此,以0.1-10×105个细胞/ml的浓度、优选0.5-2×105个细胞/ml的浓度将细胞接种于培养皿中,在经过或未经胰酶消化后通过细胞槽方式完成细胞计数。世代时间足够短的细胞特别适于通过内源基因激活大规模生产人蛋白质。
另一优选特征是缺乏靶基因的任何可检测性内源表达,即转录和翻译(v)。为了进行内源基因的激活,优选基本上无靶基因内源表达的细胞系。为此,可以0.01-2×106个细胞/ml、优选0.5-1×106个细胞/ml培养基的密度将细胞接种培养24小时。在预定时间,如24小时后,取出细胞上清液,弃去细胞,并用已知检测方法如ELISA测定细胞上清液中靶蛋白的含量。以EPO为例,检测限度是例如10pg/EPO/ml。以106个细胞/ml的接种量培养时合成少于10pg蛋白质的细胞被认为是不产生该蛋白质而尤其适合的种类。
还有另一重要且优选的特征是被选择细胞中靶基因的多体性(vi)。细胞中靶基因存在两个以上染色体拷贝能有效提高同源重组的频率。对于基因位于第7号染色体上的EPO的生产来说,具有3个7号染色体的Namalwa细胞(Nadkarni等,癌症23(1969),64-79)或Hela53细胞[Puck等,实验医学杂志(J.Exp.Med.)103(1956),173-284]被证实是尤为适合的。含多拷贝7号染色体的细胞系的其它例子是结肠腺癌细胞系SW-480(ATCC CCL-228;Leibovitz等,癌症研究,36(1976),4562-4567)、恶性黑素瘤细胞系SK-MEL-3(ATCC HTB69;Fogh和Tremp《体外人肿瘤细胞》,第115-159页,J.Fogh(编辑),Plenum出版社,纽约,1975)、结肠腺癌细胞Colo-320(ATCC CCL-220;Quinn等,癌症研究39(1979),4914-4924)、黑素瘤细胞系MEL-HO(DSM ACC62;Holzmann等,国际癌症杂志(Int.J.Cancer)41(1988),542-547)和肾癌细胞系A-498(DSM ACC55;Giard等,J.Natl.Cancer Inst.51(1973),1417-1423)。
可通过使用对特定染色体和/或靶基因位点特异的DNA探针来核查细胞系基因组中染色体的数目。
可用于内源基因激活的起始细胞系的又一优选特征是目的靶蛋白的正确糖基化(vii)。与例如天然靶蛋白相比,以类似糖基化方式、尤其是有关唾液酸数目类似的方式合成靶蛋白的人细胞系是优选使用的细胞系。正确糖基化方式的检测优选通过用含有处于细胞中有活性的启动子控制之下的目的靶基因的染色体外载体瞬间转染该细胞来完成。靶基因短暂表达后,用等电聚焦分析细胞上清液和/或提取物。以EPO为例,正确糖基化的存在是非常明显的。因此,非糖基化EPO,如来自大肠杆菌细胞的重组EPO在体外实验中具有类似于糖基化EPO的活性,然而在体内实验中,非糖基化EPO基本上是低效的。为了确定起始细胞系是否能产生正确糖基化的EPO,可用尿EPO作一比较,也可用来自CHO细胞的重组EPO作比较(已知它有一种在人体中有活性的糖基化形式,且它的糖基化作用与尿EPO大致相同)。优选用等电聚焦完成糖基化作用的比较。
在人细胞系选择中还有另一优选特征是所研究的细胞系无传染性的污染物(vii),如无侵染性病毒颗粒或支原体。可通过细胞培养、体内分析和/或特异病毒蛋白质的检测等方式完成病毒污染物存在情况的检查。
本发明还涉及在人细胞系中通过内源基因激活制备人蛋白质的方法,其特征在于使用的细胞系符合上述特征(i)、(ii)和(iii),并另外符合上述特征(iv)、(v)、(vi)和(vii)中至少一项。
本发明的方法特别适用于制备人因子,诸如EPO,血小板生成素(TPO),诸如G-CSF或GM-CSF之类的集落刺激因子,诸如tPA之类影响凝血的蛋白质,诸如IFN-α、IFN-β或IFN-γ之类的干扰素,诸如IL-1至IL-18之类的白细胞介素,诸如MIP之类的趋化因子,诸如NF或BDNF之类的神经营养因子,诸如IFG-BP之类影响骨生长的蛋白质,刺猬状蛋白质(hedgehog protein),诸如TFG-β之类的肿瘤生长因子,诸如HGH、ACTH、脑啡肽(encephalin)、内啡肽之类的生长激素,诸如可容性或膜结合形式的白细胞介素或胰岛素受体之类的受体,及其它蛋白结合蛋白质。特别优选EPO的制备方法。
可用已知方法完成内源基因激活本身,优选该方法包括以下步骤:
(a)制备人起始细胞系,该细胞系含至少一拷贝带目的核酸序列的内源靶基因,且已通过本发明的选择准则进行了检查而鉴定为适于表达靶基因;
(b)用包含以下部分在内的DNA构建体转染细胞:
(i)与靶基因位点区同源的两段侧翼DNA序列以便发生同源重组,
(ii)正选择标记基因,
(iii)若需要还有负选择标记基因,
(iv)若需要还有扩增基因,和
(v)在人细胞中有活性的异源表达调控序列;
(c)在能够对正选择标记基因的存在和任选负选择标记基因的缺失进行选择的条件下培养转染细胞;
(d)分析按步骤可选择出的细胞;
(e)鉴定产生目的靶蛋白的细胞,和
(f)若需要,还对所选择细胞中的靶基因进行扩增。
用于制备能产生目的人蛋白质之细胞的DNA构建体包含与靶基因位点区域同源的两段侧翼DNA序列。合适侧翼序列的选择可通过如WO90/11354和WO91/09955所述方法进行。优选每段侧翼序列长度至少为150bp。
特别优选同源DNA序列是选自靶基因的5’区域,如5’-非翻译序列、编码信号序列的外显子和位于该区域中的内含子。
正选择标记基因可以是任何适于真核细胞的选择标记基因,它的表达产生可选择的表型,如抗生素抗性、辅源营养等。一个特别优选的正选择标记基因是新霉素磷酸转移酶基因。
可任选存在负选择标记基因,例如HSV胸苷激酶基因,在选择剂存在时由于它的表达细胞会被破坏。负选择标记基因位于两个侧翼同源序列区之外。
若需要扩增人细胞中内源激活的靶基因,则DNA构建体可含扩增基因。合适的扩增基因的例子是二氢叶酸还原酶基因、腺苷脱氨酶基因和鸟氨酸脱羧酶基因等。特别优选的扩增基因是二氢叶酸还原酶基因,尤其是编码二氢叶酸还原酶-精氨酸突变体的基因,它比野生型基因对选择剂(氨甲喋呤)具更高的灵敏性(Simonsen等,美国国家科学院院报80(1983),2495)。
此外,用于内源基因活化的DNA构建体还含有在人细胞中有活性的异源表达调控序列。该表达调控序列包括启动子及优选的附加表达促进序列,如增强子。启动子可以是能调控的或组成型的启动子。优选的启动子是强病毒启动子,如SV40或CMV启动子。特别优选的是CMV启动子/增强子。
本发明还涉及在内源性地存在于细胞中的靶基因被激活后由上述方法鉴定出的人细胞系用于获取由活化靶基因编码的多肽,优选在包括如大发酵罐在内的大规模生产方法中获得该多肽的用途。
本发明还有另一主题是含有与人细胞中具活性之异源表达调控序列可操作连接的一拷贝内源基因、并在无事先基因扩增的情况下每106个细胞24小时内能产生至少200ng由该内源基因编码的多肽的人细胞。根据本发明,优待该人细胞能产生200-3000ng多肽/106个细胞/24小时,最优选能产生1000-3000ng多肽/106个细胞/24小时。
最后,本发明的又一主题是可通过基因扩增而由上述细胞获得、含多拷贝内源基因(每个拷贝的内源基因均与在该人细胞中有活性的异源表达调控序列可操作连接)每106个细胞能在24小时内产生至少1000ng由该内源基因编码的多肽的人细胞。尤其优选通过基因扩增可获得的人细胞系能产生1000-25000ng多肽/106个细胞/24小时,最优选能产生5000-25000ng多肽/106个细胞/24小时的人细胞系。
按照本发明的一个细胞实例是EPO生产性克隆“Aladin”,该克隆在1997年7月15日按布达佩斯协议的条款保藏于Deutsche Sammlung vonMikroorganismen und Zellkulturen(DSMZ),Mascheroder Weg 1b,38124Brunswick,保藏号DSM ACC 2320。

实施例
实施例1选择方法
1.1测定世代时间
将检测过的细胞系以0.1-5×105个细胞/ml的浓度、优选0.5-2×105个细胞/ml的浓度分别接种于含DMEM和10%FCS或RPMI1640和10%FCS的培养皿中,并在如ATCC为特殊细胞推荐的培养基及合适条件下培养,每2-3天,经过或未经胰酶消化后用计数槽、如Neubauer测定细胞数量。培养一周内细胞倍增16-256次、优选倍增64-128次的细胞被评估为阳性(+,++或+++)。
1.2悬浮培养能力
为了测定悬浮培养能力,将细胞在带相应附件的Belico旋转仪中于37℃和7%二氧化碳的条件下,在培养基(见1.1,添加和不添加血清,如胎牛血清)中持续搅拌培养14天。在此期间至少倍增5次的细胞被评估为适于悬浮培养(+)。
1.3在无血清培养基中的培养能力
为了测定细胞在无血清培养基中的培养能力,将细胞以1-10×105个细胞/ml的密度,在如1.1所述条件下,用ATCC为特殊细胞推荐并含ITS(Boehringer Mannheim,Cat.NO.1074547)的基本培养基(即未添加血清)于培养容器里培养14天。被证实在此期间数量至少倍增5次(通过细胞计数测定)的细胞被评估为适于无血清培养。
1.4测定靶基因的内源表达
为了确定靶蛋白是否在所选择的细胞系中内源性产生,将细胞以0.01-2×106个细胞/ml的密度、优选0.5-1×106个细胞/ml培养基的密度接种培养24小时。24小时后取出细胞上清液,弃去细胞,并用已知方法,如用对特定蛋白质特异的免疫测定法确定细胞上清液中细胞蛋白质的含量。
在EPO的情形中,通过ELISA测定含量。为此,用生物素化的抗EPO抗体(Boehringer Mannheim)包被由链霉亲和素包被的微量滴定板,并与含有蛋白质(1%W/V)的溶液温育,以阻断非特异性结合。然后加入0.1ml培养上清液并保温过夜。洗涤后加入过氧化物酶偶联的单克隆抗EPO抗体(Boehringer Mannheim)2小时。用ABTS为底物,在Perkin-Elmer光度计上于405nm测定过氧化物酶反应。
在此试验中EPO检测限度是10pg EPO/ml。以106个细胞/ml接种量接种产生少于10pg EPO/ml的细胞被认为不产生此蛋白质,因而被评估为合适的细胞(+)。
1.5测定靶基因拷贝数
为了确定细胞系中靶基因的拷贝数,从约108个细胞中分离人基因组DNA并定量(Sambrook等,分子克隆,实验室手册(1989),冷泉港实验室出版社)。用如AgeI和AscI或Bam HI、HindIII和SalI等限制酶切割DNA后,通过琼脂糖凝胶电泳按大小分别分离这些DNA片段,最后将它们转移至尼龙膜上并固定。
用对靶基因位点或靶基因所处的染色体特异的地高辛标记DNA探针与该固定化DNA杂交,在严谨条件下漂洗。用辐射敏感胶片通过化学发光法检测特异性杂交信号。
1.6测定靶基因的核酸序列
用DNA分离试剂盒(如QIAGEN Blood & Cell Culture DNA试剂盒)从约107个细胞中分离基因组DNA。
用一对PCR引物扩增靶基因。引物序列与靶基因编码区侧翼序列互补,因而能够扩增靶基因的整个编码区。
PCR产物被直接用于序列分析或克隆到载体中后再测序。所用测序引物与来自靶基因内含子区域的序列互补,以便能够得到完整的靶基因外显子区域序列。按制造商所给信息,用“PrismTM Ready Reaction DyeTerminator Cycle Sequencing”试剂盒(PE/ABJ,Forest City,美国)在PE/ABI自动测序仪上测序。
1.7确定糖基化方式
为了确定EPO的糖基化方式,用含有处于SV40启动子控制之下的人EPO基因序列4Kb HindIII/EcoRI片段的质粒pEPO227转染待检测细胞系(Jacobs等),自然313(1985),806;Lee-Huang等,基因128(1993),272)。在脂转染胺存在下,用可购买到的试剂盒,按制备商说明书转染细胞。2-5天后用ELISA测定所获取的细胞上清液中EPO的含量。
浓缩细胞上清液,并通过等电聚焦与已知EPO产品比较[Righetti,P.G.,于Work,T.S.,Burdon,R.H.(编)《等电聚焦:原理、方法学和应用》,Elsevier Biomedical出版社,Amsterdam(1983)]。具有类似于已知EPO产品,诸如尿EPO之类的糖基化方式的人细胞被评估为合适的(+)。
1.8病毒污染物的测定
1.8.1通过细胞培养分析
为了检测待查人细胞系中可能具有的感染性病毒污染物,将该细胞裂解液与检测细胞系共温育以检测致细胞病变效应。此外,还进行血吸附作用分析。
为了制备裂解液,用快速冻融法裂解悬浮于1ml缓冲液中的106个细胞。离心分离细胞残余物,将上清液与检测细胞系混合。所用的检测细胞系是Hep G2(ATCC HB-8065,自然282(1979),615-616)、MRC-5(ATCC-1587)和Vero(ATCC CCL-171,Jacobs,自然227(1970),168-170)。Polio SV和流感型病毒被用作阳性对照。阴性对照是在无裂解液状态下培养的检测细胞系。为了检测致细胞病变效应,在至少14天的期间内定期研究检测细胞系。
为了进行血吸附作用分析,对已与细胞裂解液及与对照温育过的Vero细胞在7天后用鸡、猪或人的红细胞处理。红细胞附着到单层培养细胞上表明培养物中有病毒污染。
1.8.2体内分析
按1.8.1所述制备待研究之细胞系的裂解液,并以0.1ml的量经腹膜内或脑间注射入新生小鼠体内。14天后观察小鼠的致病率和死亡率。
1.8.3病毒蛋白质的特异性检测
通过将具EBV阳性带的人血清与待检细胞系的固定化细胞混合来检测诸如EB病毒蛋白质(核蛋白或衣壳抗原)之类的特殊病毒蛋白质的存在情况。然后通过添加补体和相应抗人补体C3荧光素偶联物(用于检测核抗原)和通过添加抗人球蛋白荧光素(用于检测衣壳抗原)完成病毒抗原的检测。
实施例2选择结果
用第1点所述方法检验人细胞系HepG2、HT1080、Namalwa、HeLa和HeLaS3。结果列于下表1中。

从表1可见,细胞系HT1080、Namalwa和HeLaS3被认为适用于EPO的内源基因激活。Namalwa和HeLaS3尤其适合。
实施例3克隆EPO同源区
用基因组胎盘DNA(Boehringer Mannheim),通过PCR扩增EPO基因的同源区。从EPO基因5’-非翻译序列、外显子1和内含子1区域6.3Kb长的同源区制备了两种PCR产物(参看图1)。用于制备PCR产物1的引物具以下序列:5’-CGC GGC GGA TCC CAG GGA GCT GGG TTG ACC GG-3’(SEQID NO.1)和5’-GGC CGC GAA TTC TCC GCG CCT GGC CGG GGT CCC TCA GC-3’(SEQ ID NO.2)。用于制备PCR产物2的引物具以下序列:5’-CGC GGCGGA TCC TCT CCT CCC TCC CAA GCT GCA ATC-3’(SEQ ID NO.3)和5’-GGC CGC GAA TTC TAG AAC AGA TAG CCA GGC TGA GAG-3’(SEQID NO.4)。
用限制酶裂解法从PCR产物1和2中切出需要的片段(PCR产物1:HindIII,PCR产物2:HindIII和EcoRV),并克隆入已用HindIII和EcoRV裂解的载体pCRII(Invitrogen)中。以此方式获得的重组载体被称为5epopcr1000(参见图2)。
实施例4 EPO基因打靶载体的构建
4.1将含NEO基因、DHFR基因和CMV启动子/增强子的基因激活序列(参见图3)插入含EPO同源区的质粒5epopcr1000之Agel位点,从而得到质粒p176(参见图4a)。为了使CMV启动子尽可能靠近EPO基因的翻译起始位点,将限制性酶切位点AscI和AgeI(部分裂解)之间963bp长的片段删除掉,从而得到质粒p179(图46)。
4.2为了达到表达的最优化,用合成序列Met-Ser-Ala-His的核苷酸替换编码EPO前导序列Met-Gly-Val-His起始部分的外显子1内核苷酸。通过基因组EPO DNA序列的扩增获得该序列,例如用相应的引物,以含有处于SV40启动子控制之下的人EPO基因序列3.5Kb BstEII/EcoRI片段(包括外显子1-5)的质粒pEPO148(Jacobs等,自然313(1985),806,和Lee-Huang等,基因128(1993),227)为模板扩增该序列。从而获得质粒p187(图4b)。
4.3将来自Psvtk-1的单纯疱疹病毒胸苷激酶基因(HSV-TK)(PvuII/NarI片段)(图4c)插入质粒p187,制备得到质粒p189。HSV-TK基因处于内含子13’-末端与CMV启动子反方向的SV40启动子(EcoRV/ClaI)的控制之下,可用于同源重组的负选择。
4.4为了构建质粒p190,将含Simonsen等(美国国家科学院院报80(1983),2495)所述DHFR精氨酸突变体cDNA的质粒pHEAVY之SfiI/BglII片段亚克隆入经SfiI和BglII酶切的质粒pGenak-1中,该质粒包含处于RSV启动子控制之下并以晚期SV40聚腺苷酸化位点为终止子的NEO基因,处于早期SV40启动子、作为终止子的早期SV40聚腺苷酸化位点控制之下的鼠DHFR基因(Kaufmann等,分子细胞生物学,2(1982),1304;Okayama等,分子细胞牲学3(1983),280和Schimke,生物化学杂志263(1988),5989)和CMV启动子(Boshart等,细胞41(1995),521)。然后将含有DHFR精氨酸突变体编码cDNA的HpaI片断连接到经HpaI酶切的质粒p189中,从而获得质粒p190(图4d)
4.5为了得到无HSV-TK基因的转染载体,将质粒p190的含基因激活序列之AscI/NheI酶切片段连接至含外显子1的质粒p187之AscI/NheI片段中。产生的质粒被称为p192(图4e)。
实施例5转染细胞
为EPO生产选择多种细胞系并用打靶载体转染。
5.1 Namalwa细胞
将细胞培养于T150组织培养瓶中,通过电穿孔法转染(1×107个细胞/800μl电穿孔缓冲液,缓冲液含20mM Hepes、138mM氯化钠、5mM氯化钾、0.7mM磷酸氢二钠、6mM D-葡萄糖-水化物pH7.0、10μg线性化DNA,960μF,260V BioRad Gene Pulser)。电穿孔后,在40个96孔板上将细胞培养于含RPMI1640、10%(V/V)胎牛血清(FCS)、2mM L-谷氨酰胺、1mM丙酮酸钠的培养液中。两天后将细胞置于含G-4181mg/ml的培养基中培养10-20天。通过固相ELISA检验上清液中EPO的产量(见实施例1.4)。在24孔板及T-25组织培养瓶中扩增产生EPO的克隆。冻存等分试样,并用FACS(Ventage,Becton Dickin son)亚克隆诸细胞。重复检验这些亚克隆的EPO产量。
5.2 HT 1080细胞
除了将HT1080细胞培养于含DMEM、10%(V/V)FCS、2mM L-谷氨酰胺、1mM丙酮酸钠的培养基中外,HT1080的其它条件与Namalwa细胞培养中所述相同。为了进行电穿孔转染,用胰酶消化使细胞从培养容器的壁上脱离下来。电穿孔后,在5个96孔板上将1×107个细胞培养于DMEM、10%(V/V)FCS、2mM L-谷氨酰胺、1mM丙酮酸钠中。
5.3 Hela S3细胞
除了将Hela S3细胞培养于RPMI1640、10%(V/V)FCS、2mM L-谷氨酰胺、1%(V/V)NEM非必需氨基酸(Sigma)和1mM丙酮酸钠中之外,其它条件如为Nama lwa细胞所述。为了进行电穿孔转染,用胰酶消化使细胞从培养容器的壁上脱离下来。电穿孔的条件是960μF/250V。电穿孔后,在T75组织培养瓶中将细胞培养于RPMI1640、10%(V/V)FCS、2mM L-谷氨酰胺、1%(V/V)NEM、1mM丙酮酸钠中。电穿孔24小时后,用胰酶消化细胞并于10个96孔板上在含600μg/ml G-418的培养基中培养10-15天。
实施例6 EPO基因扩增
为了提高EPO的表达量,在氨甲喋呤(MTX)浓度逐步提高(100pM-1000nM)的条件下培养产生EPO的克隆。在每一MTX浓度下经ELISA(见实施例1.4)检验克隆的EPO产量。通过有限稀释对高水平生产者进行亚克隆。
实施例7鉴定产生EPO的细胞系
选用三个不同的细胞系(Namalwa、HeLa S3和HT1080)进行EPO基因的激活。通过用质粒p179、p187、p189、p190或p192转染获得产生EPO的克隆(参阅实施例2和3)。
检验约160,000个NEO抗性克隆的EPO产量,其中12-15个克隆可重复性地以明显产量分泌EPO至细胞上清液中。
其中未通过MTX的基因扩增即令人惊奇地鉴定出共7个克隆,它们能以足够用于大规模工业生产的量产生EPO。这些克隆的EPO产量范围为200ng/ml至高于1000ng/ml/106个细胞/24小时。
用500nM MTX进行基因扩增后,所鉴定的EPO克隆的EPO产量提高至超过3000ng/ml/106个细胞/24小时。进一步提高MTX浓度至1000nM,导致EPO产量高于7000ng/ml/106个细胞/24小时。
在无血清培养条件下及MTX缺乏时,所获得的克隆也显示有EPO产生。

Claims (14)
1.可用于通过内源激活内源性地存在于细胞中的靶基因而制备人蛋白质的人细胞系的选择方法,其特征在于:
(a)检验人细胞系中是否存在以下特征:
(i)有所需核酸序列的靶基因,
(ii)悬浮培养14天内数量至少倍增5次,及
(iii)在无血清培养基中培养14天内数量至少倍增5次,和
(b)使用符合特征(i)、(ii)和(iii)的细胞系作为靶基因内源激活的起始细胞系。
2.根据权利要求1的方法,其特征在于:使用的人细胞系(iv)其世代时间在无血清培养基中一周内数量倍增16-256次。
3.根据权利要求2的方法,其特征在于:使用的人细胞系在无血清培养基中一周内数量倍增64-128次。
4.根据权利要求1-3中任一项的方法,其特征在于:使用的人细胞系(v)其中的靶基因没有显著内源性表达。
5.根据权利要求1的方法,其特征在于:使用的人细胞系(vi)含有2个以上染色体拷贝的靶基因。
6.根据权利要求1的方法,其特征在于:使用的人细胞系(vii)以类似于天然靶蛋白的糖基化方式合成细胞蛋白质。
7.根据权利要求1的方法,其特征在于:使用的人细胞系(viii)无可检测到的感染性污染物。
8.在人细胞系中通过内源基因激活制备人蛋白质的方法,其特征在于包括以下步骤:
(A)选择人细胞系,其中:
(a)检验人细胞系中是否存在以下特征:
(i)有所需核酸序列的靶基因,
(ii)悬浮培养14天内数量至少倍增5次,及
(iii)在无血清培养基中培养14天内数量至少倍增5次,和
(b) 使用符合特征(i)、(ii)和(iii)的细胞系作为靶基因内源激活的起始细胞系。
(B)激活按照步骤(A)选择出的细胞系内的内源靶基因;和
(C)获得由在步骤(B)中激活的靶基因所编码的蛋白质。
9.根据权利要求8的方法,其特征在于:步骤(B)中所用细胞系还符合如权利要求2、4、5、6和7中任一项所限定的特征(iv)、(v)、(vi)、(vii)和(viii)中至少一条。
10.根据权利要求8的方法,其中所获得的蛋白质是选自下组的人类因子:EPO,TPO,集落刺激因子,影响凝血的蛋白质,干扰素,白细胞介素,趋化因子,神经营养因子,影响骨生长的蛋白质,刺猬状蛋白质,肿瘤生长因子,生长激素,ACTH,脑啡肽、内啡肽和受体。
11.根据权利要求10的方法,其中所述蛋白质是EPO。
12.根据权利要求8-11中任一项的方法,其中在步骤(C)中,所述蛋白质是在大型发酵罐中获得的。
13.一种EPO生产人细胞系,其特征在于:该细胞系是含有与在该人细胞中有活性的异源启动子可操作连接的一拷贝内源EPO基因的Namalwa、HT1080或HelaS3细胞系,当在补充了10v/v%胎牛血清、2mM L-谷氨酰胺、1mM丙酮酸钠的RPMI1640或DMEM中培养时,该细胞系能够以至少200ng多肽/106个细胞/24小时的量生产该内源基因所编码的EPO多肽,并且可以通过权利要求1-7中任一项的选择方法得到。
14.可通过基因扩增由权利要求13的细胞系获得的EPO生产人细胞系,其特征在于:该细胞系是含有均与在该人细胞中有活性的异源启动子可操作连接的多拷贝内源EPO基因的Namalwa、HT1080或HelaS3细胞系,当在补充了10v/v%胎牛血清、2mM L-谷氨酰胺、1mM丙酮酸钠的RPMI 1640或DMEM中培养时,该细胞系能够以至少1000ng多肽/106个细胞/24小时的量生产该内源基因所编码的EPO多肽。
