本发明提供了荞麦多酚在降脂中的应用,提取工艺,包括如下步骤:(1)荞麦粉末制备:将荞麦样品清洗除杂,然后进行干燥、脱壳、粉碎、过筛后得到荞麦粉末,于‑20‑‑10℃保存备用;(2)游离酚提取:将所述的荞麦粉末溶解于溶剂中,提取2‑4次后,过滤后收集上清液与滤渣。(3)结合酚提取:在所述的滤渣中加入碱性溶液进行消化,调节溶液的pH值,向其中加入溶剂,提取3‑6次,收集上清液即成。本发明所述的荞麦多酚的提取工艺提取荞麦游离酚与结合酚,保证了多酚物质较高的提取率、纯度和质量。本发明所述的荞麦多酚的提取工艺采用特定的提取溶剂、离心分离协同超声波辅助提取的方式从荞麦中提取多酚,方法简单,可工业应用。

技术领域
本发明属于食品领域,尤其是涉及荞麦多酚在降脂中的应用。
背景技术
荞麦营养丰富,含有丰富的多酚、黄酮、芦丁、γ-氨基丁酸等生物活性成分,具有多种抗氧化、抗炎症、缓解肥胖和糖尿病的药理作用,具有独特的食疗保健功能,在保持机体健康方面有重要意义。
研究表明荞麦改善机体健康部分原因是含有丰富的植物化学物质。其中,多酚是化学结构具有苯环并结合多个羟基的物质的总称,主要为黄酮类化合物,是植物体内重要的次生代谢产物。植物多酚在清除自由基方面有较好的成效,其能够阻止活性氧引起的生物大分子的氧化损伤,并且对由自由基诱发的生物大分子损伤起到保护作用,所以多酚具有较强的抗氧化作用。由于荞麦多酚类成分极性较大,受热不稳定,因此在提取荞麦多酚时多用传统的溶剂萃取法,但存在提取率较低、耗时长等问题。
发明内容
有鉴于此,本发明旨在克服现有技术中的缺陷,提出荞麦多酚在降脂中的应用。
为达到上述目的,本发明的技术方案是这样实现的:
一种荞麦多酚的提取工艺,包括如下步骤:
(1)荞麦粉末制备:将荞麦样品清洗除杂,然后进行干燥、脱壳、粉碎、过筛后得到荞麦粉末,于-20--10℃保存备用;
(2)游离酚提取:将所述的荞麦粉末溶解于溶剂中,提取2-4次后,过滤后收集上清液与滤渣。
(3)结合酚提取:在所述的滤渣中加入碱性溶液进行消化,调节溶液的pH值,向其中加入溶剂,提取3-6次,收集上清液即成。
进一步,所述的步骤(1)中的干燥步骤的温度为50-80℃,时间为20-48小时;所述的步骤(1)中的粉碎步骤为超微粉碎;所述的步骤(1)中的过筛步骤的筛网孔径为40-80目。
进一步,所述的步骤(2)中的溶剂为浓度为50-90%的丙酮溶液;所述的步骤(2)中的荞麦粉末与溶剂的固液比为1:1-20。
进一步,所述的步骤(2)中的提取步骤具体为:超声提取5-20min,离心5-20min;所述的超声步骤的转速为3000rpm。
进一步,所述的步骤(3)中的碱性溶液为浓度为2-10mol/L的NaOH溶液;所述的步骤(3)中的滤渣与碱性溶液的固液比为1:1-20;所述的步骤(3)中的消化步骤的时间为30-120分钟。
进一步,所述的步骤(3)中的溶液的pH值为1-3;所述的步骤(3)中的调节溶液的pH值的步骤采用浓盐酸。

下面结合实施例来详细说明本发明。
实施例1
一种荞麦多酚的提取工艺,包括如下步骤:
(1)荞麦粉末制备:将荞麦样品清洗除杂,然后进行于60℃鼓风干燥箱中干燥48h,超微粉碎,荞麦粉末过60目筛,得到荞麦粉末,于-20℃保存备用;
(2)游离酚提取:称取荞麦粉末样品,溶解于80%丙酮溶液(固液比1:10,w/v),在40~50℃温度下,超声提取10min,5000rpm离心10min,再重复上述浸提步骤3次,收集上清液;
(3)结合酚提取:在所述的滤渣中加入10倍体积4mol/L NaOH溶液消化1h,滴加浓盐酸调节溶液pH值至2;向消化液中加入10mL乙酸乙酯,在40~50℃温度下,超声提取5min,3000rpm离心5min,再重复上述浸提步骤3-6次,收集上清液;
一种使用所述的提取工艺制得的荞麦多酚。
一种降脂组合物,所述的组合物包含质量比为50%的所述的荞麦多酚。
对上述提取工艺制备得到的荞麦多酚的含量进行测定:
吸20μL步骤(2)与(3)所得上清液至酶标孔板上,加入250μL质量分数为2%的Na2CO3溶液,混匀后于室温反应4-6min,加入15μL体积分数为50%Folin–Ciocalteu试剂,混匀,静置30min后,将酶标孔板用多功能酶标仪在700nm处测光吸收值,以不添加荞麦粉末作为空白对照;通过标准曲线分别计算荞麦游离酚与结合酚的含量。
以吸光度(A)对浓度(c)绘制标准曲线,经最小二乘法得到标准曲线方程:A=0.0032c+0.0524,R2=0.9959。说明在0-0.160μg/mL浓度区间中,吸光度和荞麦多酚浓度呈良好的线性关系。计算可得荞麦多酚含量,结果见表1。

实施例3
一种荞麦多酚的提取工艺,包括如下步骤:
(1)荞麦粉末制备:将荞麦样品清洗除杂,然后进行于60℃鼓风干燥箱中干燥48h,超微粉碎,荞麦粉末过60目筛,得到荞麦粉末,于-20℃保存备用;
(2)游离酚提取:称取荞麦粉末样品,溶解于80%丙酮溶液(固液比1:10,w/v),在40~50℃温度下,超声提取10min,5000rpm离心10min,再重复上述浸提步骤3次,收集上清液;

一种荞麦多酚的提取工艺,包括如下步骤:
(1)荞麦粉末制备:将荞麦样品清洗除杂,然后进行于60℃鼓风干燥箱中干燥48h,超微粉碎,荞麦粉末过60目筛,得到荞麦粉末,于-20℃保存备用;
(2)游离酚提取:称取荞麦粉末样品,溶解于80%丙酮溶液(固液比1:10,w/v),在40~50℃温度下,超声提取10min,5000rpm离心10min,再重复上述浸提步骤3次,收集上清液;
(3)结合酚提取:在所述的滤渣中加入10倍体积4mol/L NaOH溶液消化1h,滴加浓盐酸调节溶液pH值至2;向消化液中加入10mL乙酸乙酯,在40~50℃温度下,超声提取5min,3000rpm离心5min,再重复上述浸提步骤3-6次,收集上清液;

Claims (10)
1.一种荞麦多酚的提取工艺,其特征在于:包括如下步骤:
(1)荞麦粉末制备:将荞麦样品清洗除杂,然后进行干燥、脱壳、粉碎、过筛后得到荞麦粉末,于-20--10℃保存备用;
(2)游离酚提取:将所述的荞麦粉末溶解于溶剂中,提取2-4次后,过滤后收集上清液与滤渣。
(3)结合酚提取:在所述的滤渣中加入碱性溶液进行消化,调节溶液的pH值,向其中加入溶剂,提取3-6次,收集上清液即成。
2.根据权利要求1所述的荞麦多酚的提取工艺,其特征在于:所述的步骤(1)中的干燥步骤的温度为50-80℃,时间为20-48小时;所述的步骤(1)中的粉碎步骤为超微粉碎;所述的步骤(1)中的过筛步骤的筛网孔径为40-80目。
3.根据权利要求1所述的荞麦多酚的提取工艺,其特征在于:所述的步骤(2)中的溶剂为浓度为50-90%的丙酮溶液;所述的步骤(2)中的荞麦粉末与溶剂的固液比为1:1-20。
4.根据权利要求1所述的荞麦多酚的提取工艺,其特征在于:所述的步骤(2)中的提取步骤具体为:超声提取5-20min,离心5-20min;所述的超声步骤的转速为3000rpm。
5.根据权利要求1所述的荞麦多酚的提取工艺,其特征在于:所述的步骤(3)中的碱性溶液为浓度为2-10mol/L的NaOH溶液;所述的步骤(3)中的滤渣与碱性溶液的固液比为1:1-20;所述的步骤(3)中的消化步骤的时间为30-120分钟。
6.根据权利要求1所述的荞麦多酚的提取工艺,其特征在于:所述的步骤(3)中的溶液的pH值为1-3;所述的步骤(3)中的调节溶液的pH值的步骤采用浓盐酸。
7.根据权利要求1所述的荞麦多酚的提取工艺,其特征在于:所述的步骤(3)中的溶剂为滤渣与溶剂的固液比为1:1-20;所述的步骤(3)中的提取步骤为超声提取1-20min,离心1-20min;所述的超声步骤的转速为3000rpm。
8.一种使用权利要求1-7任一项所述的提取工艺制得的荞麦多酚。
9.一种降脂组合物,其特征在于:所述的组合物包含质量比为1-70%的权利要求8所述的荞麦多酚。
10.权利要求8所述的荞麦多酚的应用,其特征在于:所述的荞麦多酚在制备降脂药物中的应用;所述的荞麦多酚在食品添加剂中的应用。
