Abstract

本发明公开了一株鱼类致病气单胞菌噬菌体及其应用,该噬菌体在2021年11月15日保藏于武汉市的中国典型培养物保藏中心,保藏编号为CCTCCNO:M20211408。所述噬菌体对致病气单胞菌特异性强,裂解能力强,裂解量可达105PFU/cell。体外和体内实验都有效证实了该噬菌体对致病气单胞菌有强烈的裂解作用,对预防和治疗由致病气单胞菌引起的水生动物疾病提供了有效的治疗方法。

Description

一株鱼类致病气单胞菌噬菌体φA008及其应用
技术领域
本发明属于生物技术领域,尤其涉及一株鱼类致病气单胞菌噬菌体及其应用。
背景技术
嗜水气单胞菌是革兰氏阴性细菌,能够引发鳗鲡的败血症、赤鳍病、烂鳃病等,同时也会造成鳗鲡的脱粘、溃疡、肾脏发生病变、鳃部出血等症状。这些年来造成了我国多个地区淡水养殖鱼类的大量死亡,给众多养殖户造成了巨大损失的同时也对我国的淡水养殖业带来了巨大的冲击。
多年来,水产养殖业在细菌性病害的治疗一直依赖于使用抗生素,但是长期和滥用抗生素取得的效果是有限的,并具有使病原菌产生耐药性的危险。因此,亟须寻找新的治疗方法来代替抗生素。噬菌体制剂作为新兴药物,在水产细菌性疾病的防控和治疗上有着显著的效果,并且其特异性强,不会对非靶标生物造成危害以及对环境友好,是当前生物技术研究的热点。
发明内容
针对上述问题,本发明提供了一株嗜水气单胞菌噬菌体及其应用。该噬菌体特异性强,对嗜水气单胞菌有强烈的裂解能力,能够在一定程度上有效的解决水产养殖中出现的嗜水气单胞菌病害感染问题,具有良好的应用前景。
具体的,本发明涉及以下技术方案:
本发明的第一方面,提供了一株嗜水气单胞菌噬菌体该噬菌体自福建省的一个鳗鲡养殖场分离得到的,已于2021年11月15日保藏于中国典型培养物保藏中心,保藏地址为武汉市武昌珞珈山武汉大学校内。保藏编号为CCTCC NO:M20211408。所述噬菌体具有如下特征:
电镜下观测到:该噬菌体为廿面体,头部长为66.7nm,头宽81.8nm,根据国际病毒分类委员会第九次报告的分类标准鉴定,最后结合全基因组比对可确定该噬菌体属于肌尾噬菌体科(Myoviridae)。
在本发明中:所述的噬菌体在30℃~50℃下放置1h其活性稳定,在60℃下处理1h完全失活;在pH为4~10左右时,噬菌体保持较高活性;当pH为2时,噬菌体完全失活;所述噬菌体裂解谱较广,裂解能力强,裂解量可达105PFU/cell;有效效价高,感染复数为0.1时噬菌体效价最高,为4.00×108pfu/mL;噬菌体φA008潜伏期短,裂解量大,适合用于噬菌体制剂的研发。
优选地,本发明的嗜水气单胞菌噬菌体(Aeromonas hydrophila phage)具有SEQ ID No.1所示的核苷酸序列。
本发明的第二方面,提供了嗜水气单胞菌噬菌体(Aeromonas hydrophilaphage/>)在如下(1)~(4)至少一项中的应用:
(1)制备杀灭嗜水气单胞菌的产品;
(2)制备抑制嗜水气单胞菌生长的产品;
(3)制备预防和/或治疗嗜水气单胞菌所致鱼类疾病的产品;
(4)制备预防和/或治疗由嗜水气单胞菌引起的炎症反应的产品。
本发明的第三方面,提供一种噬菌体制剂,其特征在于,所述噬菌体制剂以权利要求1或2所述的嗜水气单胞菌噬菌体(Aeromonas hydrophila phage/>)为活性成分。
优选地,所述噬菌体制剂中,嗜水气单胞菌噬菌体(Aeromonas hydrophilaphage/>)的剂量大于或等于105PFU/cell。
上述噬菌体制剂可以以本发明的嗜水气单胞菌噬菌体(Aeromonas hydrophilaphage/>)作为唯一活性成分;还可以含有除本发明的噬菌体以外的其他噬菌体,其与本发明的噬菌体复配使用可以得到与之相当或者更好的防治嗜水气单胞菌的效果。
上述噬菌体制剂还可以包括载体,所述载体可以为固体载体或液体载体;其中固体载体可以选自矿物材料、植物材料或高分子化合物;液体载体可以选自有机溶剂、植物矿物油和水。
上述噬菌体制剂可以被制备成各种使用形式,如液体制剂、冻干制剂或口服固体制剂等,经喷雾、注射或口服等方式用于防治嗜水气单胞菌。
本发明的第四方面,提供上述噬菌体制剂在如下(1)或(2)中的用途:
(1)制备预防和/或治疗嗜水气单胞菌所致鱼类疾病的产品;
(2)制备预防和/或治疗由嗜水气单胞菌引起炎症反应的产品。
本发明的第五方面,提供包含上述嗜水气单胞菌噬菌体(Aeromonashydrophila phage/>)为有效成分的饲料添加剂、饮水用添加剂或环境杀菌剂。
本发明的第六方面,提供一组扩增嗜水气单胞菌噬菌体的特异性引物,所述特异性引物的序列如SEQ ID No.2、SEQ ID No.3、SEQ ID No.4、SEQ ID No.5所示。
本发明的有益效果:
1.本发明的嗜水气单胞菌噬菌体φA008,具有较强的耐酸能力,可以特异性地部分或完全灭活嗜水气单胞菌,为工业化生产噬菌体杀菌剂提供噬菌体株来源;
2.本发明的嗜水气单胞菌噬菌体φA008是从自然界中分离的烈性噬菌体,不含毒力基因或不良基因,安全、无任何副作用,对预防和治疗由致病气单胞菌引起的水生动物疾病提供了有效的治疗方法;
3.本发明的嗜水气单胞菌噬菌体φA008具有宽宿主谱,对嗜水气单胞菌属(Aeromonas hydrophila)中的多株不同菌株均有裂解效果,在水产养殖中具有广泛的应用前景;
4.本发明的嗜水气单胞菌噬菌体φA008有效效价高,感染复数为0.1时噬菌体效价最高,为4.00×108pfu/mL;噬菌体感染宿主菌的潜伏期为20min左右,爆发时间为100min左右,爆发量为105PFU/cell;其潜伏期短、裂解量高,是治疗嗜水气单胞菌的强效噬菌体。
5.本发明的嗜水气单胞菌噬菌体φA008在30℃~50℃之间能够保持较好的稳定性和活性,可以很好的适应水产养殖环境,在水产养殖中应用具有极大的可能性。
6.本发明的嗜水气单胞菌噬菌体φA008,可用于制备组合物,试剂,或试剂盒;可应用于嗜水气单胞菌的快速检测,其包括但不限于以试纸、试剂盒等形式对目标样本中的嗜水气单胞菌进行检测,或对临床样本中的目标致病菌进行筛选,可有效确保检测的灵敏度;
7.本发明的嗜水气单胞菌噬菌体φA008可以单独或混合使用,用作为鱼类生物杀菌剂,饲料添加剂,或因嗜水气单胞菌引起的细菌感染的治疗药物、保健品及医疗器械;嗜水气单胞菌噬菌体φA008可应用于治疗或预防由嗜水气单胞菌引起的感染性疾病;
8.本发明的嗜水气单胞菌噬菌体φA008可以应用于工业生产,可由宿主菌特异性扩增,可应用标准病毒纯化方法高度纯化,具有极好的推广前景。本领域技术人员可以根据本发明的记载及本领域常识将本发明所述嗜水气单胞菌或其组合物制备成应用于医疗,检测,消毒及食品防护等方面的各种产品加以工业应用。

具体实施方式
为了更好地理解本发明,下面结合实施例和附图对本发明做进一步的详细说明,但本领域技术人员了解,下述实施例不是对本发明保护范围的限制,任何在本发明基础上做出的改变和变化,都在本发明的保护范围之内。
下述实施例中所使用的实验方法如无特殊说明,均为常规方法。
下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
实施例1噬菌体的分离培养与生物学特性
(一)菌种的复苏培养和菌悬液的制备
挑取嗜水气单胞菌的冻存菌液在LB营养琼脂平板上分区划线,分离单菌落,30℃恒温培养箱中培养10~12h。挑取单菌落,接种20ml LB肉汤培养基中,200rpm/min震荡培养8~12h,得到单一菌悬液。
(二)噬菌体的分离与纯化
将取自福建省范围内鳗鲡养殖场的淡水样品与上述嗜水气单胞菌增殖液在30℃震荡培养箱中,200rpm/min震荡培养8~12h,富集可能存在的嗜水气单胞菌噬菌体。取混合液放入离心机,12000rpm离心2min,离心后取上清液用0.22μm滤膜将细菌过滤。
滤液应用涂布点斑法来判断是否存在噬菌体。取宿主菌液100μl涂布于含LB琼脂平板上,再取上述滤液20μl均匀点在平板四角方位上,待滤液干后,将平板倒置于30℃恒温培养箱中培养10~12h,之后观察是否出现空斑。
利用双层平板法将得到的噬菌斑进行纯化。挑取单个噬菌斑置于1ml SM缓冲液中,涡旋震荡30s,置于4℃冰箱过夜浸出,之后再通过离心机12000rpm离心2min,取上清用0.22μm滤膜过滤。滤液梯度稀释至10-2,按照1:1的比例与宿主菌混合,30℃孵育5min,后加入8ml的LB上层培养基中,混匀后迅速倒入LB琼脂营养平板上,待其凝固后倒置30℃恒温培养箱中培养10~12h。纯化3-5次后,得到形态一致的噬菌斑(如图1所示)。
(三)噬菌体的增殖
将上述纯化完毕的噬菌体,采用固体增殖的方式,选取空斑数最为密集的双层板,挖取上层于10ml SM缓冲液中,搅拌均匀,放置于4℃冰箱过夜,之后取出置于离心机,12000rpm离心2min,上清液用0.22μm滤膜过滤,即得到噬菌体增殖液。
(四)噬菌体效价测定
对噬菌体液用SM缓冲液以10倍梯度进行稀释,使用双层平板测定稀释至10-6、10-7、10-8的噬菌体效价,每个梯度三个平行。30℃条件下过夜培养后对噬菌斑进行计数并计算效价。
测定得到该噬菌体增殖液的效价为2.3×1010pfu/mL。
(五)噬菌体电镜检测
取上述噬菌体富集液20μl于铜网上,静置吸附10min,用滤纸吸去多余菌液,再取适量3%磷钨酸染色5min,干燥后电镜观察。
结果如图2所示,该噬菌体为廿面体,头部长为66.7nm,头宽81.8nm。
(六)噬菌体基因组测定
扩增基因组的特异性引物如表1所示

按照常规噬菌体基因组检测方法,测得噬菌体全基因组序列如SEQ IDNo.1所示。
如图3所示,将噬菌体的基因组序列用NCBI网站的blastn(megablast)程序进行比对。在得到的同源序列中,噬菌体与气单胞菌噬菌体60AhydR15PP(MH179477)和气单胞菌噬菌体phiAS4(HM452125)的覆盖率仅为2%,一致性分别为89.04%和89.70%。从基因组来看,噬菌体/>与这二者最为接近。二者在分类上均属于有尾噬菌体目,肌尾噬菌体科。这与噬菌体/>电镜结果不同。使用Mauve软件对3个基因组进行共线性比对分析(如图4所示),发现噬菌体/>与二者并无同源片段。但是噬菌体60AhydR15PP和菌噬菌体phiAS4存在2个同源性片段,可确定该噬菌体属于肌尾噬菌体科(Myoviridae)。
该噬菌体的基因组结构如图5所示。
(六)噬菌体热稳定性检测
分装噬菌体增殖液1ml于2ml离心管中,分别置于30℃、40℃、50℃、60℃、70℃、80℃、90℃恒温培养箱中培养1h,之后用双层平板法对噬菌体效价进行测定。
具体结果如图6所示,噬菌体在30℃~50℃下放置1h其活性稳定,在高于50℃的温度下培养1h其噬菌体滴度开始下降,在60℃的培养1h后则完全失活。
(七)噬菌体的pH稳定性检测
以NaOH溶液和HCl溶液对含LB肉汤培养基调节其pH为2、4、6、7、8、10,分别取调节完毕的肉汤培养基900μl与100μl的噬菌体液混合,置于30℃恒温培养箱中1h,双层平板法计算噬菌体效价,分析噬菌体对pH的耐受性。
结果参见图7,在pH为4~10左右时,噬菌体保持较高活性;当pH≤2时,噬菌体完全失活。
(八)噬菌体裂解谱测定
采用斑点法来测定噬菌体的裂解谱。取20μl噬菌体液分均点在由气单胞菌涂布的琼脂平板上,液滴干燥后倒置于30℃恒温培养箱中培养10~12h。通过观察是否有空斑形成来鉴定是否对菌株有裂解能力。宿主菌株包括15株嗜水气单胞菌和5株维氏气单胞菌。
通过上述方法检测噬菌体对20株不同气单胞菌的裂解能力。如表2所示,其中噬菌体/>能够裂解9株嗜水气单胞菌,裂解谱较广。

(九)噬菌体最佳感染复数的测定
按照感染复数1000、100、10、1、0.1、0.01、0.001、0.0001、0.00001的比例取菌体菌液和宿主菌液各100μl,加入800μl的LB肉汤培养基,置于30℃恒温摇床培养5h,之后于离心机12000rpm离心2min,0.22μm滤膜过滤。取滤液采用双层平板法对其进行效价测定。
结果如表3所示,感染复数为0.1时噬菌体效价最高,为4.00×108pfu/mL。

按照MOI=0.1,噬菌体菌液与宿主菌液1:1体积混合,总体积为10ml,放入30℃恒温摇床培养10min,12000rpm离心2min,弃上清,用培养基洗涤3次,再取1ml培养基重悬沉淀后转移到50ml的培养基中,置于30℃恒温摇床中进行培养,分别在0、10、20、30、40、50、60、90、120、150min时间节点采取双层平板法对噬菌体效价进行实时检测,每个时间点三个平行,以此结果绘制一步生长曲线。
如图8所示,噬菌体的裂解周期时长为150min,噬菌体感染宿主后,在20min噬菌体数量增长较慢,表明噬菌体潜伏期为20min;噬菌体侵染宿主后的120min内,噬菌体数量急剧增加,可见噬菌体的爆发期为100min;在随后的30min内,噬菌体数量基本保持不变,进入稳定生长期。噬菌体裂解量为105PFU/cell。噬菌体/>潜伏期短,裂解量大,适合用于噬菌体制剂的研发。
(十一)噬菌体的毒力基因或不良基因缺失检测试验
对噬菌体进行生物信息学分析,结果如表4所示,噬菌体/>不含有毒力基因或不良基因。

(十二)噬菌体的系统进化分析
以DNA连接酶和相关尾纤蛋白构建的系统进化树(如图9、图10所示)可以看出,噬菌体在系统进化树上与其他噬菌体基因组都不在一个分支上,且全基因组比对的同源性序列均没出现在进化树的分支上。通过比对发现,噬菌体/>的DNA连接酶序列与数据库中其他DNA连接酶序列的相似性均在30%以下(图9),相关尾纤蛋白与数据库中序列的相似性也均在50%以下,这也说明噬菌体/>与其他噬菌体的亲源关系较远(图10)。
(十三)噬菌体预防嗜水气单胞菌感染鳗鲡效果测定
将体重40g左右的健康鳗鲡随机分成9个桶,每组10条,预防组、攻毒组和空白组都设置3个平行。实验用鱼都已经过3~5天的暂养。各组按照质量比3%进行饲料的投喂。预防组将嗜水气单胞菌、噬菌体与鳗鲡饲料均匀混合,攻毒组用嗜水气单胞菌和鳗鲡饲料均匀混合,空白用清水拌料。处理过的饲料中嗜水气单胞菌含量为1×108CFU/g,噬菌体含量为1×106PFU/g,阴干后再进行投喂。记录各组30天内的鳗鲡死亡情况,并计算死亡率及保护率。预防保护率(%)=(攻毒组平均死亡率-预防组死亡率)/攻毒组平均死亡率。具体结果见表5。

实验结果及分析:根据实验数据(表5)显示,攻毒组的鳗鲡在没有投喂带有噬菌体的拌料,经过投喂带有嗜水气单胞菌后死亡率在80%以上;而预防组有在饲料中添加噬菌体,30天内的死亡率仅为10%,保护率在88%以上。说明噬菌体作为饲料添加剂添加在鳗鲡饲料中能够有效预防由此类气单胞菌引起的水产疾病,降低养殖风险。
(十四)噬菌体治疗气单胞菌疾病的效果测定/>
将体重40g左右的健康鳗鲡随机分成9个桶,每组10条,治疗组、攻毒组和空白组都设置3个平行。实验用鱼都已经过3~5天的暂养。试验开始时,将治疗组和攻毒组的所有鳗鲡在背鳍相同位置都做一个小创伤处理,之后将其浸泡于菌剂量为5×105CFU/mL的水中。当其中有一个桶开始出现濒死的鳗鱼时,将治疗组和攻毒组鳗鲡转移到新的养殖桶中,并在治疗组的水中加入噬菌体噬菌体终含量为1×104PFU/mL。其他组不做处理。记录各组10天内的鳗鲡死亡情况,并计算死亡率及保护率。治疗保护率(%)=(攻毒组平均死亡率-治疗组死亡率)/攻毒组平均死亡率。具体结果见表6。

实验结果及分析:根据结果数据显示,经创伤处理并浸泡于嗜水气单胞菌中的攻毒组,在没有加入噬菌体于养殖系统的情况下,10天内的死亡率在90%以上,而治疗组在经过噬菌体的治疗后,死亡率最高为30%,保护率达到67%以上。说明噬菌体在治疗由此类气单胞菌引起的疾病上效果明显,可以用于治疗气单胞菌疾病的噬菌体药剂。
综上,本发明嗜水气单胞菌噬菌体φA008,具有较强的耐酸能力,可以特异性地部分或完全灭活嗜水气单胞菌,为工业化生产噬菌体杀菌剂提供噬菌体株来源;是从自然界中分离的烈性噬菌体,不含毒力基因或不良基因,安全、无任何副作用,对预防和治疗由致病气单胞菌引起的水生动物疾病提供了有效的治疗方法;具有宽宿主谱,对嗜水气单胞菌属中的多株不同菌株均有裂解效果,在水产养殖中具有广泛的应用前景;有效效价高,感染复数为0.1时噬菌体效价最高,为4.00×108pfu/mL;噬菌体感染宿主菌的潜伏期为20min左右,爆发时间为100min左右,爆发量为105PFU/cell;其潜伏期短、裂解量高,是治疗嗜水气单胞菌的强效噬菌体;在30℃~50℃之间能够保持较好的稳定性和活性,可以很好的适应水产养殖环境,在水产养殖中应用具有极大的可能性;可用于制备组合物,试剂,或试剂盒;可应用于嗜水气单胞菌的快速检测,其包括但不限于以试纸、试剂盒等形式对目标样本中的沙门氏菌进行检测,或对临床样本中的目标致病菌进行筛选,可有效确保检测的灵敏度。

Claims (8)

1.一株嗜水气单胞菌噬菌体,其特征在于,所述嗜水气单胞菌噬菌体为嗜水气单胞菌噬菌体φA008 (Aeromonas hyd rophila phage φA008) ,其保藏编号为CCTCC NO:M20211408。
2.根据权利要求1所述的嗜水气单胞菌噬菌体φA008 (Aeromonas hydrophila phageφA008)在如下(1)~(4)至少一项中的应用:
(1)制备杀灭嗜水气单胞菌的产品;
(2)制备抑制嗜水气单胞菌生长的产品;
(3)制备预防和/或治疗嗜水气单胞菌所致鱼类疾病的产品;
(4)制备预防和/或治疗由嗜水气单胞菌引起的炎症反应的产品。
3.一种噬菌体制剂,其特征在于,所述噬菌体制剂以权利要求1所述的嗜水气单胞菌噬菌体φA008 (Aeromonas hydrophila phage φA008)为活性成分。
4.根据权利要求3所述的噬菌体制剂,其特征在于,所述噬菌体制剂中,嗜水气单胞菌噬菌体φA008 (Aeromonas hydrophila phage φA008)的剂量大于或等于5×105 PFU/ml。
5.根据权利要求3所述的噬菌体制剂,其特征在于,所述噬菌体制剂还包括载体,所述载体为固体载体或液体载体。
6.根据权利要求3所述的噬菌体制剂,其特征在于,所述噬菌体制剂的剂型为液体制剂、冻干制剂或口服固体制剂。
7.权利要求3~6任一项所述的噬菌体制剂在如下(1)或(2)中的用途:
(1)制备预防和/或治疗嗜水气单胞菌所致鱼类疾病的产品;
(2)制备预防和/或治疗由嗜水气单胞菌引起炎症反应的产品。
8.包含权利要求1所述的嗜水气单胞菌噬菌体φA008(Aeromonas hydrophila phageφA008)为有效成分的饲料添加剂或环境杀菌剂。
