Abstract
本发明涉及在免疫细胞的培养过程中通过添加到培养液能够活化免疫细胞的物质及利用其的免疫细胞的活化方法,其能显著增强免疫细胞对癌细胞或感染细胞的细胞杀伤能力,从而可以显著增强包含免疫细胞作为有效成分的细胞治疗剂的癌症治疗或预防功效。
Description
技术领域
本发明涉及免疫细胞活化物质及利用其的免疫细胞的活化方法。更具体地,涉及在免疫细胞的培养过程中可通过与上述免疫细胞一起进行培养来增强上述免疫细胞的免疫活性的组合物。
背景技术
胰腺癌(pancreatic cancer)是最常见的原发性癌症之一的恶性肿瘤,在确诊后平均存活率为5年的患者在所有患者中的比例小于5%。与其他癌症相比,已知该癌症是唯一一个几十年来平均存活率没有提高的癌症。目前,已知对胰腺癌有效的治疗方法是手术切除。胰腺癌的癌肿块组织主要由纤维组织组成,由于癌细胞仅仅为其中一部分,因此很难评估抗癌治疗后对癌症的治疗反应,并且众所周知,胰腺癌为一种抗癌治疗不是非常有效的癌症,因此建议外科手术切除而不是采用抗癌化学疗法。临床上使用的抗癌剂有吉西他滨(Gemcitabine)(礼来公司(Eli Lilly and Company)),但一直提出有关其治疗效果的疑问。但是,没有任何可以替代其的物质及治疗方法。因此,迫切需要开发能够有效治疗胰腺癌且副作用少的抗癌剂或能够增强现有抗癌剂效果的辅助剂。
作为这种抗癌剂的辅助剂,细胞治疗剂备受关注。自然杀伤细胞(常用作细胞治疗剂的有效成分之一的细胞)被各种细胞因子活化(activation)而扩增,并分泌溶细胞功能(cytolytic function)物质,活化的自然杀伤细胞分泌趋化因子(chemokine)的同时,还分泌干扰素-γ、肿瘤坏死因子(tumor-necrosis factor,TNF)、自体粒细胞-巨噬细胞集落刺激因子(granulocyte/macrophage colony-stimulation factor;GM-CSF)以引起炎症反应。并且,通过诱导单核细胞(monocyte)、树突状细胞(dendritic cells)、粒细胞(granulocyte)的分化和生长,影响适应性免疫(NK细胞在人类癌症免疫治疗中的应用前景,自然评论,2007年(Prospects for the use of NK cells in immunotherapy ofhuman cancer,NATURE REVIEWS,2007))。
另一方面,在参与免疫调节方面,除了干扰素-γ(IFN-γ)等蛋白质外,来源于植物的多糖类活化各种巨噬细胞功能(对肿瘤细胞和微生物的细胞毒性活性、吞噬活性及特定细胞因子(如α-致癌坏死因子和白细胞介素)的分泌),并促进细胞的分化和增殖。
因此,可以通过使用特定物质来提高如上所述的免疫细胞的免疫活性增强效果。正尝试着各种有效地增强免疫细胞的免疫活性并最终利用这些免疫细胞实现癌症治疗效果的方法。
发明内容
发明要解决的问题
本发明的一目的在于,提供一种可以增强免疫细胞的免疫活性的细胞治疗剂制备方法,其中免疫细胞为细胞治疗剂的有效成分。
本发明的另一目的在于,提供一种增强用于治疗或预防癌症的细胞治疗剂的治疗效果的组合物。
本发明的其他目的和优点将从以下本发明的详细描述、所附权利要求书和附图中变得更加明确。
用于解决问题的手段
本发明的一实现例涉及一种包含免疫细胞作为有效成分的细胞治疗剂的制备方法,包括:将免疫细胞与由下述化学式1或2表示的化合物、褪黑素或其组合进行培养的步骤;以及从上述培养物分离免疫细胞的步骤。

在本发明中,由上述化学式1表示的化合物对应于5-硝基-2-糠醛缩对羟基苯甲酰肼(5-Nitro-2-furaldehyde p-hydroxybenzoylhydrazone),也可用“硝呋齐特(Nifuroxazide)(例如,AMBATROLTM、ANTINALTM、BACIFURANETM、DIAFURYLTM)”表示。
在本发明中,由上述化学式2表示的化合物对应于3-(2,4-二氯-苯氧基甲基)-5-三氯甲基-[1,2,4]恶二唑(3-(2,4-dichloro-phenoxymethyl)-5-trichloromethyl-[1,2,4]oxadiazole)。
在上述免疫细胞培养步骤中,培养液可包含褪黑素。在本发明中,褪黑素(melatonin)是在哺乳动物、植物、细菌、霉菌等中发现的物质之一,已知其影响睡眠时间、血压调节、季节性繁殖等生理功能的活动周期(circadian rhythm)。作为褪黑素可以单独使用褪黑素,或者与选自人参皂甙(ginsenoside)及β-葡聚糖(β-glucan)中的一种以上组合(以下称为,MTAM1)使用。MTAM1可用于培养淋巴细胞,能够提高培养后获得的免疫细胞中自然杀伤细胞的增殖率,由此进一步增强上述自然杀伤细胞对癌细胞的杀伤能力,从而可以更有效地预防或治疗癌症。在MTAM1存在的环境培养淋巴细胞的情况下,上述淋巴细胞可优先与CD3进行培养。上述MTAM1还可包括CD56、白细胞介素-18(IL-18)或其组合。根据需要,可以同时、依次或交替地进行MTAM1与额外的免疫活化因子,如CD3、CD56、IL-18或其组合的培养。
在本发明中,上述“细胞治疗剂(cellular therapeutic agent)”是指使用从个体分离、培养及通过特殊操作制备的细胞和组织,其是用于治疗、诊断和预防目的的药物(美国FDA规定),并且是指为了恢复细胞或组织的功能,通过在体外增殖和筛选活的自体、同种异体或异种细胞,或者以其他方式改变细胞生物学特性等的一系列行为,来用于治疗、诊断和预防目的的药品。在具体例中,上述细胞治疗剂可包含免疫细胞作为有效成分。
在本发明中,“预防”是指通过本发明的组合物的给药来抑制或延迟癌症的进行的所有行为。
在本发明中,“治疗”及“改善”是指通过本发明的组合物的给药来改善或有益地改变癌症症状的所有行为。
在本发明中,上述“免疫细胞”可以指可以存在于体内或者可以分离到体外的所有具有免疫活性的细胞。免疫细胞可以是例如自然杀伤细胞、T细胞、B细胞、树突状细胞、巨噬细胞、或其组合。在一具体例中,上述免疫细胞可以是淋巴细胞。在另一具体例中,上述免疫细胞可以是自然杀伤细胞(natural killer cells,NK cells)。
在一具体例中,上述化学式1或2的化合物可以通过接触分离的自然杀伤细胞来增强其免疫活性。从人体获得的免疫细胞是各种免疫细胞的混合物形态。因此,在培养过程诱导分化成特定免疫细胞,或者仅活化其增殖是很常见的。如下述实施例所示,本发明的一具体例即使应用于分离的自然杀伤细胞也可以成功地增强其免疫活性。该效果表明,即使对于单纯含有自然杀伤细胞作为有效成分的细胞治疗剂的最终产品,也可以获得预期的免疫活性。
在本发明中,上述“培养”可以指维持或保存细胞处于存活状态,包括使免疫细胞暴露或接触于特定环境。
在一具体例中,将由上述化学式1或2表示的化合物、或其组合与免疫细胞混合培养,从而可增强免疫细胞的免疫活性。上述免疫活性例如可以是指对癌细胞或感染细胞的细胞杀伤能力(cytotoxicity)。最终,可以进一步增强包含免疫细胞作为有效成分的细胞治疗剂的治疗或预防癌症的效果。在本发明中,上述癌症例如可选自由胰腺癌、肝癌、肺癌、大肠癌、乳腺癌、前列腺癌、卵巢癌、大肠癌、宫颈癌、甲状腺癌、喉癌、白血病、脑肿瘤、神经母细胞瘤、视网膜母细胞瘤、头颈癌、唾液腺癌、淋巴瘤组成的组,优选地,可以为胰腺癌。
从上述培养物分离免疫细胞的步骤实质上去除除免疫细胞之外的培养液和残留于培养液的成分。分离方法例如可以使用膜过滤、透析、沉淀法、细胞粘附和洗涤等多种方法,但不限于此。为了避免在将免疫细胞应用于个体时可能发生的不必要的生物反应,可以采用分离免疫细胞的步骤,但是,由于由化学式1或2表示的化合物、褪黑素或其组合仅为了增强免疫细胞的免疫活性而少量使用,因此,即使与免疫细胞一起残留,对个体的影响也可能非常微不足道。
在一具体例中,上述细胞治疗剂的制备方法还可包括将分离的免疫细胞剂型化成细胞治疗剂的步骤。上述剂型化包括将细胞治疗剂加工成以对需要其的个体进行给药时合适的形态的所有行为,例如,可以意味着将分离的免疫细胞与药学上可接受的载体进行混合。药学上可接受的载体可以与盐水、灭菌水、林格溶液、缓冲盐水、右旋糖溶液、麦芽糖糊精溶液、甘油、乙醇、脂质体和这些组分中的一种以上混合使用,根据需要可添加其他常规的添加剂,例如抗氧化剂、缓冲剂、抑菌剂等。并且,可通过额外添加稀释剂、分散剂、表面活性剂、粘合剂和润滑剂来制剂化成注射用剂型(如水溶液、悬浮液、乳浊液)、丸剂、胶囊剂、颗粒或片剂,并且可将靶器官特异性抗体或其他配体与上述载体结合使用,以特异性作用于靶器官。进而,可利用本技术领域的适当的方法或雷明顿文献(Remington'sPharmaceutical Science(最新版),Mack Publishing Company,Easton PA)中公开的方法,根据每种疾病或组分来制剂化成优选状态。
上述细胞治疗剂可以是液剂、悬浮剂、分散液、乳剂、凝胶、可注射液剂及活性化合物的缓释制剂等,优选地,可以是注射剂。当上述细胞治疗剂被剂型化成注射剂的情况下,在上述剂型化步骤中,为了确保根据注射剂处方的流通的产品稳定性,使用可用作注射剂的酸水溶液或磷酸盐等缓冲溶液调节pH,从而可以制备物理以及化学上非常稳定的注射剂。
根据本发明的另一实现例,提供包含由上述化学式1或2表示的化合物、褪黑素或其组合的细胞治疗剂的治疗效果增强用组合物。
上述细胞治疗剂的治疗效果增强用组合物与细胞治疗剂接触,以增强作为上述细胞治疗剂的有效成分的免疫细胞的免疫活性,结果可以增强细胞治疗剂的癌症治疗或预防效果。
上述细胞治疗剂的治疗效果增强用组合物可以用作用于培养免疫细胞的培养液的添加剂,以使其与免疫细胞接触。
在一具体例中,为了使细胞治疗剂在个体中表现出快速有效的免疫活性,上述细胞治疗剂的治疗效果增强用组合物可以在用于需要细胞治疗剂的个体之前,优先与上述细胞治疗剂进行混合。可包含1μM至100μM、1μM至80μM、1μM至60μM、或1μM至40μM的由上述化学式1或2表示的化合物。上述细胞治疗剂的治疗效果增强用组合物可以在对个体进行给药之前从与细胞治疗剂的混合物中除去,从而防止个体中诱发不必要的生物反应。然而,由于由化学式1或2表示的化合物仅为了增强免疫细胞的免疫活性而少量使用,因此,即使以混合物状态给药或残留,对个体的影响也是微不足道。
根据本发明的另一实现例,提供试剂盒,包括:第一单元,包含由上述化学式1或2表示的化合物、褪黑素或其组合;以及第二单元,包含免疫细胞作为有效成分。
在本发明中,上述“单元”仅仅是区分试剂盒的构成的手段,其可意味着可以以固体、粉末、溶液、悬浮液、凝胶等形态存在,并包括在可分离的合适的容器中。
上述第一单元和第二单元在上述试剂盒中是分离开的,在用于个体之前彼此混合,可通过增强第二单元的免疫细胞的免疫活性来增强细胞治疗剂的治疗效果。
上述试剂盒还可包括可从第一单元和第二单元的混合物分离免疫细胞的结构。分离上述免疫细胞的结构可用于从上述第一单元和第二单元的混合物中实质上去除化学式1或2、褪黑素或其混合物。然而,由于第一单元的成分仅为了增强免疫细胞的免疫活性而少量使用,因此,即使以混合物状态给药或残留,对个体的影响也是微不足道。
以上,针对细胞治疗剂的制备方法的说明可以被解释为与细胞治疗剂的治疗效果增强用组合物或试剂盒的相同构成的说明具有相同的含义。
发明的效果
利用本发明提供的由上述化学式1或2表示的化合物、或其组合的细胞治疗剂制备方法及其组合物能够显著增强免疫细胞对癌细胞或感染细胞的细胞杀伤能力,从而可以显著增强包含免疫细胞作为有效成分的细胞治疗剂的癌症治疗或预防功效。

具体实施例
以下,通过下述实施例对本发明进行详细说明。但是,下述实施例仅用于示例本发明,本发明的内容并不限于下述实施例。
实施例1:确认化学式1和2的免疫细胞的免疫活性增强能力
1-1.细胞培养
作为胰腺癌细胞系的Panc-1购自韩国细胞系银行(首尔大学)。细胞利用含10%的胎牛血清(FBS)和1%的青霉素链霉素的达尔伯克氏改良伊格尔氏培养基(Dulbecco'smodified Eagle's medium,DMEM,海克隆(Hyclone),洛根(Logan),UT;Panc-1细胞),在37℃、5%的CO2培养箱培养。作为自然杀伤细胞系的NK-92细胞购自美国典型培养物保藏中心(ATCC)(弗吉尼亚州马纳萨斯(Manassas,VA)),利用含20%的胎牛血清和1%的青霉素链霉素、10ng/ml的重组人白细胞介素-2(recombinant human IL-2)的低限量伊格尔-α培养基(Minimum Essential Medium Eagle alpha,MEM-α),在37℃、5%的CO2培养箱中培养。
1-2.确认化学式1和2的细胞毒性
为了评估实验物质对自然杀伤细胞的毒性,使用化学式1和2分别以1μM、5μM、10μM、20μM及40μM的浓度对实施例1-1的NK-92细胞进行处理,并孵育4小时。然后,以D-PlusTM细胞活力检测试剂盒(D-PluTM cell viability assay kit)(CCK-3000,Donginbiotech Co.,韩国)通过众所周知的方式以0.1X处理后,测量450nm处的吸光度。
结果确认,在所试验的所有浓度条件下存活率为约85%,对NK-92细胞没有直接毒性(图1)。
1-3.化学式1和2对免疫细胞的免疫活化评估
将实施例1-1的NK-92细胞分别与40μ的化学式1和2培养4小时后,分离NK-92细胞,并将其与作为癌细胞的Panc-1细胞按各个比例(E:T=2.5、5、10、20)共培养。4小时后,针对于癌细胞存活率,使用检测乳酸脱氢酶细胞毒性检测试剂盒(LDH cytotoxicitydetection kit)(日本宝生物(Takara,Japan))以众所周知的方式进行实验,并测量490nm处的吸光度。
结果确认,与对照组相比,与化学式1和2一起培养的NK-92细胞对Panc-1细胞具有显著优异的杀伤能力(图2)。
实施例2:确认褪黑素对免疫细胞的免疫活性增强能力
2-1.淋巴细胞的获得和培养
从患者的血液中分离淋巴细胞,悬浮于培养液后,将其置于其中固相化有CD3的T-25培养瓶(Flask)。然后,将5μm的D56和10%的自体血浆添加到T-25培养瓶后,在37℃、5%的CO2培养箱(incubator)培养了1~2天。转移到T-75培养瓶后,在37℃、5%的CO2培养箱中培养了3~4天。此时,就试验例1和2的每一个而言,在T-75培养瓶中使用CD56 5μl、IL-18以及作为免疫细胞活化物质的褪黑素、人参皂甙和β-葡聚糖的混合物(以下称为MTAM1))1.5μl或40μl来进行处理。并且,作为比较,就对照组而言,在T-75培养瓶中使用CD56 5μl、IL-18进行处理后,在CO2培养箱培养了3~4天。然后,转移到T-175培养瓶,并在37℃、5%的CO2培养箱培养了48小时。
然后,就试验组1和2而言,在T-175培养瓶中使用10μl或20μl的MTA M1分别进行处理,并加入5%的自体血浆。并且,作为比较,就对照组而言,加入5%的自体血浆后,在CO2培养箱培养了48小时。
然后,就试验组1和2而言,在T-175培养瓶中培养中的细胞悬浮液使用IL-18和MTAM1进行处理,观察细胞的活化状态后,将T-150培养瓶转移到装有1000ml容量的透气性培养袋的培养液后,通过进一步培养7~9天来增殖大量细胞。就对照组而言,将T-150培养瓶转移到装有1000ml容量的透气性培养袋的培养液后,通过进一步培养7~9天来增殖大量细胞。
2-2.免疫细胞的获得
就试验组1和2、对照组中的每一个而言,装在1000ml容量的CO2透气性培养袋的细胞和培养液分别分装并转移到500ml离心管,离心17分钟(min)以获得细胞。以不使细胞脱落的方式小心弃去上清液,然后用250ml的灭菌生理盐水洗涤细胞。通过再次将离心管以调平衡状态放入离心机后,离心15分钟收获细胞。以不使细胞掉落的方式小心弃去上清液,使用注射器(syringe)从100ml的注射用灭菌生理盐水袋中取30ml。使用30ml的注射用灭菌生理盐水对细胞进行悬浮。将悬浮的细胞倒入细胞筛(cell strainer)中并过滤。将过滤后的细胞悬浮液加入注射用灭菌生理盐水袋并进行悬浮。
2-3.免疫细胞的免疫活性评估
通过分析从实施例2-2中获得的试验组1和2、对照组的细胞表面抗原,进行纯度测试,以便以定量方式测量淋巴细胞的种类,结果分别示于图3至图6。其中,图3至5分别为对试验组1、试验组2、对照组按培养条件检查细胞表面抗原的曲线图,图6示出每一个的NK细胞与NKT细胞的比例、T细胞比例。
结果可知,与对照组相比,试验组1和2的NK细胞与NKT细胞的比例显著增加(图6)。
并且,就细胞数量和存活率而言,与对照组相比,试验组1和2的细胞数量和存活率提高(图7及表1)。

最后,细胞毒性(Cytotoxicity)试验结果可知,与对照组的细胞伤害活性能力相比,试验组1和2的细胞伤害活性能力(%)显著提高(图8及表2)。

这些结果是由于添加了MTAM1,其中,MTAM1通过包含植物来源的褪黑素、人参皂甙及β-葡聚糖中的至少一种,优选地,一起包含褪黑素、人参皂甙及β-葡聚糖来制备。NK细胞是通过抑制肿瘤的生长和扩散来控制许多类型的肿瘤的先天免疫系统的调节性淋巴细胞,在没有其他刺激的情况下直接发挥细胞的细胞毒性,并分泌如干扰素-γ(IFN-γ)的免疫刺激性细胞因子,可以控制局部肿瘤(local tumor growth)的生长和转移(metastasis)。
尤其,被白细胞介素-2(IL-2)活化的NK细胞与细胞毒性T淋巴细胞(CTL)相似,通过穿孔素(perforin)和颗粒酶B(granzyme B)连续性杀伤靶(target)细胞后自己仍能存活,此时,IL-2具有积累耗尽的NK细胞的细胞毒性和颗粒的能力。并且,NK细胞具有抗体依赖性细胞毒性(antibody dependentcellular cytotoxicity,ADCC)。在NK细胞中表达的Fc受体中的FcγRIII(CD16)识别如IgG的抗体(结合到被病原体(pathogen)感染的细胞的表面)的Fc区。Fc受体的活性诱导NK细胞的活性,以诱导分泌溶细胞性细胞因子(cytolyticcytokine),这种NK细胞的ADCC作用是只有NK细胞具有的特性,与CTL不同。
褪黑素增加淋巴细胞产生谷胱甘肽,具有提高淋巴细胞的功能的效果。通过单核细胞表面的褪黑素受体促进IFN-γ和白细胞介素-1、白细胞介素-2、白细胞介素-6、白细胞介素-12(Interleukin-1、2、6、12)等的细胞因子的分泌,以诱导NK细胞的活性。本发明的MTAM1增加NK细胞对病毒感染细胞和癌细胞的杀伤能力,并通过抑制培养物中的活性氧来增加细胞存活率。
并且,人参皂甙(Ginsenoside)活化C-SRC激酶(C-Src Kinase),使细胞内钙离子浓度增加2至3倍。作为细胞内信号转导体系的早期步骤的C-Src激酶相关途径的活化及细胞内钙离子的增加会使特定基因的表达增加,并促进细胞增殖。尤其,人参皂甙-Rp1在树突状细胞中以恒定浓度特异性诱导免疫细胞因子的产生,并能够在低浓度条件下增加NK细胞的活性。
另一方面,β-葡聚糖增加肿瘤坏死因子-α(TNF-α)和IFN-γ的表达,并在中性粒细胞中表现出抗凋亡(Anti-apoptosis)效果。尤其,从掌状海带(Laminaria digitata)分离出来的作为葡聚糖的匹卡林(Phycarine)能增加吞噬细胞活性,不仅增加IL-1、IL-6、TNF-α的产生和分泌,而且匹卡林(Phycarine)通过补体第三受体(CR3)增加NK细胞的癌细胞凋亡作用。
图9为示出MTAM1的细胞生死辨别试验(Cell viability test)结果的图表,表明本发明的MTAM1没有细胞内毒性,进而还对增殖有效。
并且,图10及图11为示出活性氧(Reactive Oxygen Species;ROS)测量结果的图表,通过图10确认了对NK细胞人为增加ROS时处理MTAM1后ROS减少。通过图11确认了将NK细胞在具有MTAM1的培养基中培养14天后测量ROS的结果,褪黑素可阻断活性氧。其中,H2O2为增加活性氧的刺激剂,NAC为ROS清除剂(Scavenger)。
在细胞表面抗原分析中可以确认,使用由褪黑素、人参皂甙及β-葡聚糖组成的免疫细胞活化物质处理后进行培养并收获的细胞中,其以具有CD3-CD56+CD16+表型的NK细胞为74.2%、NKT细胞为7.9%、T细胞为12.7%的比例构成(图12)。与图6的未处理对照组的结果相比,通过上述免疫细胞活化物质处理并收获的细胞内NK细胞与NKT细胞的比例显著增加。
以上,对本发明的特定部分进行详细描述,对于本领域普通技术人员而言,这些具体技术只是优选的实现例,显然本发明的范围并不限于此。因此,本发明的实质范围将由所附权利要求及其等同物来定义。
工业实用性
本发明涉及免疫细胞活化物质及利用其的免疫细胞的活化方法。更具体地,涉及在免疫细胞培养过程中可通过与上述免疫细胞一起进行培养来增强上述免疫细胞的免疫活性的组合物。

所述免疫细胞为淋巴细胞。
3.根据权利要求1所述的细胞治疗剂的制备方法,其特征在于,
所述免疫细胞为自然杀伤细胞。
4.根据权利要求1所述的细胞治疗剂的制备方法,其特征在于,
由所述化学式1或2表示的化合物与分离的自然杀伤细胞一起培养以增强所述自然杀伤细胞的免疫活性。
5.根据权利要求1所述的细胞治疗剂的制备方法,其特征在于,
用于增强所述免疫细胞的免疫活性。
6.根据权利要求1所述的细胞治疗剂的制备方法,其特征在于,
所述细胞治疗剂用于治疗或预防癌症。
7.根据权利要求5所述的细胞治疗剂的制备方法,其特征在于,
所述免疫活性是对癌细胞或感染细胞的细胞杀伤能力。
8.根据权利要求6或7所述的细胞治疗剂的制备方法,其特征在于,
所述癌症选自由胰腺癌、肝癌、肺癌、大肠癌、乳腺癌、前列腺癌、卵巢癌、大肠癌、宫颈癌、甲状腺癌、喉癌、白血病、脑肿瘤、神经母细胞瘤、视网膜母细胞瘤、头颈癌、唾液腺癌、淋巴瘤组成的组。
9.根据权利要求1所述的细胞治疗剂的制备方法,其特征在于,
还包括:将所述分离的免疫细胞剂型化成细胞治疗剂的步骤。
11.根据权利要求10所述的细胞治疗剂的治疗效果增强用组合物,其特征在于,
在对需要所述细胞治疗剂的个体使用之前,优先将所述细胞治疗剂的治疗效果增强用组合物与所述细胞治疗剂进行混合。
12.根据权利要求10所述的细胞治疗剂的治疗效果增强用组合物,其特征在于,
包含1μM至100μM的由所述化学式1或2表示的化合物。
13.一种试剂盒,其特征在于,包括:
第一单元,包含由下述化学式1或2表示的化合物、褪黑素或其组合;以及
第二单元,包括细胞治疗剂,所述细胞治疗剂包含免疫细胞作为有效成分。
14.根据权利要求13所述的试剂盒,其特征在于,
所述第一单元和第二单元分离,
并且,通过混合所述第一单元和所述第二单元来增强所述第二单元的免疫细胞的免疫活性。
