Description

Mulberry leaf polyphenol-based mouthwash and preparation method thereof
Technical Field
The invention relates to the technical field of mouthwashes, in particular to a mulberry She Duofen-based mouthwash and a preparation method thereof.
Background
The mouthwashes in the market are various, and most mouthwashes can achieve the effects of cleaning the oral cavity and keeping fresh, but have the defects in preventing dental caries and dental ulcer. With the increasing pressure of modern people, the diet is irregular, and the two diseases are more common. At present, people pay more attention to the oral health problem, and meanwhile, the pursuit of beauty promotes the vigorous development of the mouthwash related industry, and focuses more on the technical improvement on dental caries and dental ulcer.
Caries is commonly known as dental caries and dental caries, and is a bacterial disease, and the main bacteria causing caries are streptococcus mutans, streptococcus light and streptococcus blood, which can secondary pulpitis and periapical periodontitis, and even can cause alveolar bone and jawbone inflammation.
Canker sores are commonly known as "aphtha", and are a common ulcerative injury disease occurring on oral mucosa. The occurrence of canker sores is the result of a combination of factors including localized trauma, stress, food, medications, malnutrition, altered hormone levels, and vitamin or trace element deficiency; bacteria such as Streptococcus sanguis and helicobacter pylori are also closely related to canker sores.
Sang Sheshi dried leaves of Morus alba L.China is the country of the world where the mulberry planting area is the largest and the annual output of mulberry leaves is happy. The mulberry leaf is rich in various active ingredients such as flavonoid, alkaloids, polysaccharides and the like, has various functions of resisting oxidation, reducing blood fat, reducing blood sugar and the like, and can be widely applied. Related researches show that mulberry leaf polyphenol, namely a bioactive phenol secondary metabolite extracted from mulberry leaves, is an important material basis for forming the oxidation capacity of Sang Shekang. The mulberry leaf polyphenol has stronger DPPH free radical, hydroxyl free radical and superoxide anion free radical scavenging capability. Although the mulberry leaves and polyphenol extracts thereof in Chongqing are rich in resources, the mulberry leaves are not high in utilization rate, are mostly used for raising silkworms, a large amount of mulberry leaves are not effectively utilized, the resource waste degree is high, and especially the mulberry leaves polyphenol is not effectively applied as an active ingredient of the mouthwash.
The invention enhances the effective application of the mulberry leaf polyphenol natural product, improves and innovates the components of the mouthwash, not only enhances the antibacterial effect on the basis of cleaning the oral cavity, but also enhances the efficacy of preventing decayed teeth and canker sores, reduces the oral problems of contemporary people, and has better life.

Detailed Description
The invention is further described in detail below in connection with specific embodiments.
Example 1
In order to further prove that the mouthwash has a strong antibacterial effect, the embodiment combines experimental researches to prove that the mulberry leaf polyphenol mouthwash has good efficacy on oral health.
1. Bacteriostasis experiment
A single colony of Staphylococcus aureus was cultured in 20mL of LB at 37℃with shaking for 16h. Mixing 1mL of bacterial solution with 9mL of mulberry leaf polyphenol mouthwash, performing gradient dilution with 0.85% sterile physiological saline, and respectively diluting to 10 -1 、10 -2 、10 -3 、10 -4 、10 -5 And 10 -6 . Under sterile conditions, the sample dilutions were dripped onto nutrient agar plates for colony counting and incubated at 37℃for 16h.
2. Experiment for eliminating biofilm
Taking a single bacterium of staphylococcus aureusFalls into 20mL of LB, and is cultured for 16h at 37 ℃. 1mL of the bacterial liquid was mixed with 9mL of mulberry leaf polyphenol mouthwash, and added to a 50mL sterile centrifuge tube containing stainless steel (2.0X12.0X10.1 cm). After stationary culture at 32℃for 48 hours, the bacterial culture was removed and the stainless steel sheet was washed 2 times with 10mL of 0.85% sodium chloride. The liquid in the centrifuge tube was removed and the stainless steel plate was washed 2 times with 0.85% sodium chloride. After all the washing buffer was removed, 10mL of 0.2% crystal violet was added to the centrifuge tube and treated at 25℃for 30min, and the stainless steel plate was washed with 0.85% sodium chloride 2 times and naturally air-dried. Subsequently, the biofilm on the stainless steel plate was dissolved with 1mL of 95% ethanol, and OD was measured 600 The smaller the value, the smaller the amount of biofilm, the better the biofilm removal effect of the mouthwash.
Example 2
Single factor test: determining the influence of the content of each component on the antibacterial effect by controlling the variable, soaking the black tea, the honeysuckle, the wild chrysanthemum and the phellodendron in proportion with pure water, boiling, and decocting with small fire to obtain a medicinal soup; adding pure water into the pot, and decocting again to obtain decoction. Mixing the two obtained medicinal decoctions, adding folium Mori polyphenol extract and xylitol, mixing with water to 100%, and pasteurizing under the variable setting conditions shown in table 1.
TABLE 1 mouthwash Each component content gradient
The results are shown in figures 4-6, and the contents of the honeysuckle and the phellodendron have remarkable differences and have the content with the optimal antibacterial effect, so that the content is directly determined. The optimal content ratio is determined by performing response surface test because the polyphenol extract of mulberry leaves, the black tea and the wild chrysanthemum have no significant difference.

Mixing the two obtained medicinal soup, adding folium Mori polyphenol extract and xylitol, mixing with water to 100%, and pasteurizing.
The remaining examples are similar to examples 3-5, except for the amounts of polyphenol extract from mulberry leaves, black tea, honeysuckle, wild chrysanthemum and phellodendron, which are shown in Table 2.
TABLE 2 summary of the effects of mouthwash inhibition and biofilm removal for each example formulation
Note that: total index = bacteriostatic effect x 0.5+ biofilm x 0.5.
Response surface analysis and optimal condition determination: the response surface analysis chart (figures 1-3) is drawn by using Design-Expert 8.0, and as can be seen from the response surface chart, interaction exists among the mulberry leaf polyphenol extract (A), the black tea content (B) and the wild chrysanthemum content (C), and the interaction between the black tea content (B) and the wild chrysanthemum content (C) is not obvious. And combining a response surface and a regression model of the influence of interaction of all factors on the comprehensive index of the mouthwash, and when the content (A) of the polyphenol extract of the mulberry leaves, the content (B) of the black tea and the content (C) of the wild chrysanthemum are respectively 3.22mL,2.53g and 1.25g, the effect of the comprehensive index is predicted to be optimal.
The above embodiments are only for illustrating the technical solution of the present invention and not for limiting the same, and although the present invention has been described in detail with reference to the preferred embodiments, it should be understood by those skilled in the art that modifications and equivalents may be made thereto without departing from the spirit and scope of the technical solution of the present invention, which is intended to be covered by the scope of the claims of the present invention.
