Abstract
The invention discloses a polypeptide and plant extraction composition for fading wrinkles and application thereof, aiming at providing a composition for timely supplementing elastin to skin, promoting collagen synthesis, removing free radicals, supplementing moisture required by skin, increasing skin elasticity and fading wrinkles; the technical key points comprise that: 1-5 parts of polypeptide compound, 1-6 parts of peony extract, 0.5-4 parts of centella asiatica extract, 1.2-3.5 parts of secondary fission yeast fermentation product filtrate, 2-7 parts of spirulina maxima extract, 3-7 parts of magnolia sieboldii extract, 0.5-1.5 parts of saccharomycete polypeptide, 0.5-1.5 parts of arginine/lysine polypeptide, 0.5-1 part of alanine, 0.5-1 part of acetylcysteine and 0.5-1 part of acetyl tyrosine; belongs to the technical field of cosmetics.
Description
Technical Field
The invention discloses a composition, in particular to a polypeptide and plant extraction composition for fading wrinkles and application thereof, belonging to the technical field of cosmetics.
Background
The skin covers the body surface and sequentially comprises epidermis, dermis and subcutaneous layer from outside to inside. The dermis is generally 1-2 mm thick and is differentiated from mesoderm and is divided into a papillary layer and a reticular layer, and no obvious boundary exists between the papillary layer and the reticular layer. The dermis protrudes with numerous papillae and is embedded into the deep surface of the epidermis, which is connected with the superficial fascia through connective tissue fiber bundles.
Wrinkles are the "natural enemies" of women, but everyone cannot prevent the lapse of time. From about age 35, the metabolism of a human begins to slow down, sebaceous and sweat gland functions slowly decline, internal fiber reserves supporting the skin surface gradually deplete, the tension in the skin outward continuously decreases, local areas begin to sink, and wrinkles develop and become increasingly apparent. Particularly, in the eye area where the skin is thinner and the fiber reserve is less, problems such as fishtail lines and eyelid laxity are more likely to occur.
The main reason is that the skin is affected by the external environment to form free radicals, which destroy collagen and active substances in normal cell membrane tissues. Resulting in a gradual decrease in collagen content in the dermal layer of the skin. The net-shaped supporting body can also become thick and hard and lose elasticity, and when the elasticity and the water retention of the real cortex are reduced, the skin can lose elasticity and become thin and aged; the epidermis forms loose wrinkles.
The dermis is mainly composed of two proteins, collagen and elastin. Collagen is a biological macromolecule, exists in skin, bones, tendons, blood vessels and other connective tissues, occupies 70 percent of the skin, forms an elastic net of the skin, can moisturize, inhibit wrinkles, reduce pigmentation and make the skin on the face look elastic and glossy, and is tough like glue but is essentially a protein. Almost 20-25% of the proteins in the whole body of the human body are collagen.
However, with age, collagen begins to be lost, like when the sponge begins to be in a dehydration state, the sponge is drier along with the faster water loss, and the drier the sponge is, the more the sponge can not lock water, and the cycle is repeated, so that a vicious circle can be formed, the water and oil of the skin are seriously unbalanced, and the first stage of skin aging is caused; the skin elasticity is weaker and weaker after a large amount of water is lost, the collagen net begins to break along with the lapse of time, and when the collagen net begins to break, fine lines at the canthus and the ordinance lines begin to appear on the face because no way is available for supporting the skin, and finally true wrinkles are formed.
Elastin is an important component of skin composition, elastin only contains 5% in skin, but is a key for stimulating the generation of skin collagen and elastic fibers, collagen accounting for 70% of skin determines the smoothness and plumpness of skin, elastic fibers determine the tightness and elasticity of skin, and 90% of elastic fibers are composed of elastin.
The elasticity of the skin depends on the elastic fibers in the inner layer of the skin, which is equivalent to a plurality of elastic fiber nets supporting the skin tissues. While 90% of the elastic fibers are composed of elastin, the amount of elastin determines the elasticity of the skin and whether wrinkles are present. However, elastin in the skin is not invariable and is reduced, which is an important cause of skin aging.
Therefore, in order to reduce skin wrinkles and delay aging, the skin is tender, plump, smooth, compact and glossy, and moisture, collagen and elastin, which are required by the skin, need to be supplemented in time, and free radicals need to be eliminated.
Disclosure of Invention
In view of the above disadvantages, the present invention aims to provide a polypeptide and plant extract composition for wrinkle reduction, which has synergistic effects of the components, and can supplement elastin, promote collagen synthesis, scavenge free radicals, supplement moisture required by skin, increase skin elasticity, and reduce wrinkles.
Therefore, the first technical scheme provided by the application is as follows:
a polypeptide and plant extraction composition for fading wrinkles comprises the following components in parts by weight: 1-5 parts of polypeptide compound, 1-6 parts of peony extract, 0.5-4 parts of centella asiatica extract, 1.2-3.5 parts of secondary fission yeast fermentation product filtrate, 2-7 parts of spirulina maxima extract, 3-7 parts of magnolia sieboldii extract, 0.5-1.5 parts of saccharomycete polypeptide, 0.5-1.5 parts of arginine/lysine polypeptide, 0.5-1 part of alanine, 0.5-1 part of acetylcysteine and 0.5-1 part of acetyl tyrosine.
Furthermore, the polypeptide and plant extraction composition for fading wrinkles further comprises 1-4 parts of hibiscus syriacus flower extract and 1-3 parts of ginger flower root extract.
Furthermore, the polypeptide and plant extract composition for fading wrinkles further comprises 1-2 parts of a magnolia biondii extract.
Furthermore, the polypeptide and plant extraction composition for fading wrinkles also comprises 0.1-0.3 part of caprylyl hydroximic acid and 0.5-2 parts of ethylhexyl glycerol.
Furthermore, the polypeptide and plant extraction composition for fading wrinkles further comprises 10-15 parts of butanediol and 10-20 parts of water.
Further, the polypeptide and plant extract composition for fading wrinkles comprises acetyl hexapeptide-8, tripeptide-1, tetrapeptide-1 and nonapeptide-1 in equal mass ratio.
Further, the polypeptide and plant extract composition for fading wrinkles is prepared by the following steps of:
1) cleaning Magnolia sieboldii root, drying, pulverizing, and sieving with 50-100 mesh sieve to obtain Magnolia sieboldii root powder;
2) adding the magnolia sieboldii roots obtained in the step 1) into an extraction tank, adding 8-12 times of extraction solvent, placing in an ultrasonic oscillator, treating for 45-60min under 50-55kHZ ultrasonic waves, filtering, repeating for 3 times, combining 3 times of filtrate, and concentrating under reduced pressure to 1/4-1/5 of the original volume;
3) putting the concentrated solution in the step 2) on an HP-20 macroporous adsorption resin column, eluting for 3-4 times by using water, discarding the eluent, eluting by using an ethanol water solution with the mass concentration of 75%, recovering the eluent, and concentrating under reduced pressure until the eluent is dried to obtain the magnolia sieboldii root extract.
Furthermore, the extraction solvent of the polypeptide and plant extraction composition for wrinkle reduction is ultrapure water.
The second technical scheme provided by the invention is as follows:
a cosmetic containing polypeptide and plant extract composition for removing wrinkle is provided, wherein the content of the polypeptide and plant extract composition for removing wrinkle is 3-10% of the total weight of the cosmetic.
Compared with the prior art, the invention has the following beneficial effects:
(1) according to the technical scheme provided by the invention, the polypeptide compound is compounded with the plant extraction composition, and acetyl hexapeptide-8 participates in competition for SNAP-25 at the site of the bubble-melting complex, so that the formation of the complex is influenced, the muscle contraction is weakened, and the formation of wrinkles is prevented; the plant extract composition and tripeptide, acetyl tetrapeptide-1 and pentapeptide-3 further stimulate and promote the generation of collagen, fibronectin, elastin, hyaluronic acid and the like; inhibiting glycosyl, reducing the problem of disorderly and loose collagen fibers, and recovering elasticity; the skin is restored to be young and smooth.
(2) According to the technical scheme provided by the invention, the peptide compound is compounded with a peony extract, a centella asiatica extract, a schizosaccharomyces cerevisiae fermentation product filtrate, a spirulina maxima extract, a magnolia sieboldii extract, saccharomycete polypeptides, an amino acid/lysine polypeptide, alanine, acetylcysteine and acetyltyrosine, so that the inhibition of elastase and glucuronidase is further increased, the inhibition effect on the activity of cathepsin and the activity of fiber cells is further improved, the effects of eliminating free radicals and inhibiting the generation of nitric oxide are achieved, the degradation of collagen fibers can be slowed down, the cell renewal is stimulated, the service life of cells is prolonged, and the skin elasticity can be maintained.
(3) According to the technical scheme provided by the invention, the peptide compound is compounded with a peony extract, a centella extract, a secondary fission yeast fermentation product filtrate, a spirulina maxima extract, a magnolia sieboldii extract, yeast polypeptides, an amino acid/lysine polypeptide, alanine, acetylcysteine and acetyltyrosine, so that the softening of the horny layer can be effectively promoted, the hydration function of cells is increased, the water content of the horny layer is improved, and the peptide compound has good moisture retention, deep water replenishing and skin moistening and elasticity.
(4) According to the technical scheme provided by the invention, the hibiscus extract is further compounded, the ginger root extract and the magnolia biondii extract can improve the oxidation resistance of skin, and meanwhile, the components are synergistic to synthesize collagen and enhance the cell proliferation speed; butanediol and water are used as solvents, and caprylyl hydroximic acid and ethylhexyl glycerol are used as preservatives, so that the preservative is beneficial to maintaining and adding, and the moisturizing performance is further improved.
(5) The technical scheme provided by the invention can effectively supplement iodine, various mineral substances, vitamins, proteins and amino acids, accelerate metabolism, maintain skin vitality, increase skin oxygen resistance, resist inflammation and the like, thereby achieving the effects of firming skin, reducing wrinkles, increasing elasticity and delaying aging.
(6) According to the technical scheme provided by the invention, the Magnolia sieboldii extract is extracted by combining ultrasonic waves with an adsorption resin column, the extraction efficiency is high, phenols and magnoflorine of the Magnolia sieboldii extract are effectively kept, the oxidation resistance and the anti-aging performance of the Magnolia sieboldii extract are improved, the degradation of collagen fibers is slowed down, and the skin elasticity can be maintained.

The technical solutions in the embodiments of the present invention are clearly and completely described below, and it is obvious that the described embodiments are only a part of the embodiments of the present invention, and not all embodiments. For process parameters or conditions not specifically mentioned, it can be carried out with reference to conventional techniques.
The raw materials used in the present invention are all purchased from commercial sources.
Example 1
The invention provides a polypeptide and plant extraction composition for fading wrinkles, which comprises the following components in parts by weight: 3g of polypeptide compound, 1g of peony extract, 4g of centella asiatica extract, 2.5g of secondary fission yeast fermentation product filtrate, 3g of spirulina maxima extract, 4g of magnolia sieboldii extract, 0.5g of yeast polypeptide, 1g of arginine/lysine polypeptide, 0.5g of alanine, 1g of acetylcysteine, 0.6g of acetyltyrosine, 10g of butanediol, 20g of water, 0.1g of caprylyl hydroximic acid and 2g of ethylhexylglycerol.
Example 2
The invention provides a polypeptide and plant extraction composition for fading wrinkles, which comprises the following components in parts by weight: 5g of polypeptide compound, 4g of peony extract, 1g of centella asiatica extract, 2g of secondary fission yeast fermentation product filtrate, 7g of spirulina maxima extract, 5g of magnolia sieboldii extract, 1g of yeast polypeptide, 1.5g of arginine/lysine polypeptide, 1g of alanine, 0.5g of acetylcysteine, 0.5g of acetyltyrosine, 15g of butanediol, 15g of water, 0.15g of caprylyl hydroximic acid and 1.5g of ethylhexylglycerol.

The polypeptide complexes described in examples 1 to 10 consist of acetyl hexapeptide-8, tripeptide-1, tetrapeptide-1, pentapeptide-3, nonapeptide-1 in equal mass ratios.
The magnolia sieboldii extract described in examples 1-10 was prepared by the following method:
1) cleaning Magnolia sieboldii root, drying, pulverizing, and sieving with 50-100 mesh sieve to obtain Magnolia sieboldii root powder;

A composition comprising the following components in parts by weight: 3g of peony extract, 2g of centella asiatica extract, 2g of secondary fission yeast fermentation product filtrate, 4g of spirulina maxima extract, 4g of magnolia sieboldii extract, 3g of saccharomycete polypeptide, 13g of butanediol, 12g of water, 0.2g of caprylyl hydroximic acid and 1g of ethylhexyl glycerol.

A composition comprising the following components in parts by weight: 2g of polypeptide compound, 3g of peony extract, 2g of centella asiatica extract, 2g of secondary fission yeast fermentation product filtrate, 4g of spirulina maxima extract, 4g of magnolia sieboldii extract, 1g of yeast polypeptide, 3.25g of alanine, 13g of butanediol, 12g of water, 0.2g of caprylyl hydroxamic acid and 1g of ethylhexyl glycerol.
In the comparative example, the magnolia sieboldii extract was prepared in the same manner as in the example.
Comparative example 4
A composition comprising the following components in parts by weight: 2g of polypeptide compound, 3g of peony extract, 2g of centella asiatica extract, 2g of secondary fission yeast fermentation product filtrate, 4g of spirulina maxima extract, 4g of magnolia sieboldii extract (sold in the market), 1g of yeast polypeptide, 1g of arginine/lysine polypeptide, 0.75g of alanine, 0.75g of acetylcysteine, 0.75g of acetyltyrosine, 13g of butanediol, 12g of water, 0.2g of caprylyl hydroximic acid and 1g of ethylhexyl glycerol.

Test samples: compositions of examples 1, 5, 8, 10, comparative examples 1-4.
The test method comprises the following steps: determining the collagen contents of I, III and IV of fibroblasts by an enzyme-linked immunosorbent assay, transferring the fibroblasts into a 96-hole culture plate after being digested by trypsin, filling marginal holes with a sterile phosphate buffer solution, transferring the cells into an incubator to continue culturing, using a microsyringe to sequentially add 20 mu L/hole of the composition sample solution of examples 1, 5, 8 and 10 and comparative examples 1-4 when the cells are completely attached to the wall, adding 0.01MPBS with the same volume into a control group, determining the collagen contents of I, III and IV according to the instruction of a kit after 48h of a sample, using 3 multiple holes of each group, using ultrapure water as the control group, and determining the collagen synthesis amount to be 100%. The test results are shown in table 1.

Experiment 2 moisturizing effect experiment
The specific test method is as follows:
reference standard: refer to QB/T4256-.
The testing personnel: 40 healthy volunteers aged 30-40 years were selected, and randomly grouped into 5 groups. The samples were applied twice daily in the morning and evening. During the experiment, the subject was not able to apply any other cosmetic product at the experimental site.
And (3) testing conditions are as follows: the capacitance method skin moisture tester Corneometer CM825 is used for testing the temperature of 20-22 ℃ and the humidity of 40-60%.
The test principle is as follows: the capacitance method is used for measuring the moisture content of the human skin stratum corneum, and is based on the obvious difference of dielectric constants of water and other substances, the capacitance values of the measured skin are different according to the moisture content of the skin stratum corneum, and the parameters can represent the moisture content (MMV) of the skin.
Test samples: the experimental group is that the compositions provided by examples 1, 5, 8 and 10 and the compositions of comparative examples 1 to 4 are added into the same emulsion matrix, the addition amount is 5 wt%, and samples 1, 5, 8 and 10 to be tested and control samples 1 to 4 are obtained correspondingly in sequence.
The test method comprises the following steps: each area is fixedly measured at three points according to a certain sequence, and an average value of the three points is obtained. Experiment 3 skin elasticity test

Experiment 4 DPPH radical scavenging test
0.2mmol/L DPPH solution is prepared by 50% ethanol and stored away from light. The experimental group is that the compositions provided by examples 1, 5, 8 and 10 and the compositions of comparative examples 1 to 4 are prepared into 0.2mg/mL solution to be tested, the absorbance A is measured at 517nm, the absorbance A is measured in parallel for 3 times, the average value is taken, and the experimental result is shown in Table 4.

As is apparent from tables 1 to 4, the composition provided by the invention has synergistic effect, can supplement elastin to skin in time, promote collagen synthesis, remove free radicals, supplement moisture required by skin, increase skin elasticity and fade wrinkle.
Claims (9)
1. The polypeptide and plant extraction composition for fading wrinkles is characterized by comprising the following components in parts by weight: 1-5 parts of polypeptide compound, 1-6 parts of peony extract, 0.5-4 parts of centella asiatica extract, 1.2-3.5 parts of secondary fission yeast fermentation product filtrate, 2-7 parts of spirulina maxima extract, 3-7 parts of magnolia sieboldii extract, 0.5-1.5 parts of saccharomycete polypeptide, 0.5-1.5 parts of arginine/lysine polypeptide, 0.5-1 part of alanine, 0.5-1 part of acetylcysteine and 0.5-1 part of acetyl tyrosine.
2. The composition of claim 1, further comprising 1-4 parts of hibiscus syriacus flower extract and 1-3 parts of zingiber officinale root extract.
3. The composition of claim 1, further comprising 1-2 parts of an extract of gomphrena globosa.
4. The composition of claim 2, further comprising 0.1-0.3 parts of caprylyl hydroxamic acid and 0.5-2 parts of ethylhexylglycerin.
5. The composition of claim 3, further comprising 10-15 parts of butanediol and 10-20 parts of water.
6. The composition of claim 1, wherein the polypeptide complex comprises acetyl hexapeptide-8, tripeptide-1, tetrapeptide-1, pentapeptide-3, nonapeptide-1 in an equal mass ratio.
7. The polypeptide and plant extract composition for lightening wrinkles as claimed in claim 1, wherein the Magnolia sieboldii extract is prepared by the following method:
1) cleaning Magnolia sieboldii root, drying, pulverizing, and sieving with 50-100 mesh sieve to obtain Magnolia sieboldii root powder;
2) adding the magnolia sieboldii roots obtained in the step 1) into an extraction tank, adding 8-12 times of extraction solvent, placing in an ultrasonic oscillator, treating for 45-60min under 50-55kHZ ultrasonic waves, filtering, repeating for 3 times, combining 3 times of filtrate, and concentrating under reduced pressure to 1/4-1/5 of the original volume;
3) putting the concentrated solution in the step 2) on an HP-20 macroporous adsorption resin column, eluting for 3-4 times by using water, discarding the eluent, eluting by using an ethanol water solution with the mass concentration of 75%, recovering the eluent, and concentrating under reduced pressure until the eluent is dried to obtain the magnolia sieboldii root extract.
8. The composition of claim 7, wherein the extraction solvent is ultrapure water.
9. A cosmetic containing the polypeptide and plant extract composition for wrinkle reduction according to claim 1, wherein the polypeptide and plant extract composition for wrinkle reduction accounts for 3-10% of the total mass of the cosmetic.
