Abstract
The invention discloses a polypeptide and plant extract composition for removing wrinkles and application thereof, and aims to provide a composition for supplementing elastin to skin in time, promoting synthesis of collagen, scavenging free radicals, supplementing water required by skin, increasing skin elasticity and removing wrinkles; the technical key points include: 1-5 parts of polypeptide compound, 1-6 parts of paeonia lactiflora extract, 0.5-4 parts of centella asiatica extract, 1.2-3.5 parts of fermentation product filtrate of two-split yeast, 2-7 parts of spirulina maxima extract, 3-7 parts of magnolia sieboldii extract, 0.5-1.5 parts of saccharomycete polypeptide, 0.5-1.5 parts of arginine/lysine polypeptide, 0.5-1 part of alanine, 0.5-1 part of acetylcysteine and 0.5-1 part of acetyltyrosine; belongs to the technical field of cosmetics.
Description
Technical Field
The invention discloses a composition, in particular relates to a polypeptide and plant extraction composition for removing wrinkles and application thereof, and belongs to the technical field of cosmetics.
Background
The skin is covered on the body surface, and is sequentially an epidermis, a dermis and a subcutaneous layer from outside to inside. The dermis is generally 1-2 mm thick and is differentiated from mesoderm, and is divided into a papillary layer and a reticular layer, and no obvious limit exists between the papillary layer and the reticular layer. The dermis is a bulge with numerous papillae embedded in the deep surface of the epidermis, which is connected to the superficial fascia by connective tissue fiber bundles.
Wrinkles are a "natural enemy" of women, but everyone cannot prevent the passage of years. Starting from about 35 years old, the metabolism of the person begins to slow down, the sebaceous and sweat gland functions slowly decline, the internal fibre reserves supporting the skin surface gradually deplete, causing the tension inside the skin to decrease outwards and the local areas to subside, so that wrinkles are produced and become more apparent. Particularly, in the periocular region with thinner skin and less fiber reserve, problems such as fish tail, eyelid relaxation and the like are more likely to occur.
The main reason is that the skin is influenced by the external environment to form free radicals, and collagen and active substances in normal cell membrane tissues are destroyed by the free radicals. Resulting in a gradual decrease in collagen content of the dermis layer of the skin. The reticular supporting body can also become thicker and harder and lose elasticity, and when the elasticity and the water retention degree of the dermis layer are reduced, the skin can lose elasticity and become thinner and aged; the epidermis forms loose wrinkles.
The dermis layer mainly contains two proteins, namely collagen and elastin. Collagen is a biopolymer, which is present in skin, bones, tendons, blood vessels and other connective tissues, and occupies 70% of the skin, forming an "elastic net" of the skin, which can moisturize, inhibit wrinkles, reduce pigmentation, make the skin on the face look elastic and shiny, and is as tough as a gel, but is essentially a protein. The protein of the whole body of the human body has more or less 20-25% of collagen.
However, as the age increases, collagen begins to run off, just as when the sponge begins to be in a dehydrated state, the sponge is drier as the water runs off, the sponge is drier and the water is not locked, and cycle by cycle, the cachexia circulation is formed, so that serious unbalance of water and oil of the skin is caused, and the first stage of skin aging is caused; following a significant loss of moisture, the skin elasticity becomes weaker and, over time, the collagen network begins to break, and when it begins to break, there is no way to support the skin, the face will begin to appear fine lines at the corners of the eyes, and french lines, eventually forming true wrinkles.
Elastin is an important component of skin composition, and only 5% of elastin is in the skin, but is a key for exciting generation of skin collagen and elastic fibers, 70% of collagen in the skin determines softness and plumpness of the skin, and 90% of elastic fibers are composed of elastin.
The elasticity of the skin depends on the elastic fiber of the inner layer of the skin, which is equivalent to innumerable elastic fiber webs supporting skin tissues. Whereas 90% of the spandex is composed of elastin, the amount of elastin determines the elasticity of the skin and whether wrinkles occur. However, elastin in the skin is not constant, and elastin is reduced, which is an important cause of skin aging.
Therefore, in order to reduce wrinkles in skin, delay aging, make skin tender, plump, elastic, compact and glossy, it is necessary to supplement moisture, collagen and elastin required by skin in time, and remove free radicals.
Disclosure of Invention
Aiming at the defects, the invention aims to provide a polypeptide and plant extract composition for removing wrinkles, which has the advantages that all the components are synergistic, elastin can be timely supplemented to skin, collagen synthesis is promoted, free radicals are eliminated, moisture required by the skin is supplemented, skin elasticity is increased, and the wrinkles are removed.
For this purpose, the first technical solution provided in the present application is as follows:
a polypeptide and plant extract composition for reducing wrinkles comprises the following components in parts by weight: 1-5 parts of polypeptide compound, 1-6 parts of paeonia lactiflora extract, 0.5-4 parts of centella asiatica extract, 1.2-3.5 parts of fermentation product filtrate of two-split yeast, 2-7 parts of spirulina maxima extract, 3-7 parts of magnolia sieboldii extract, 0.5-1.5 parts of saccharomycete polypeptide, 0.5-1.5 parts of arginine/lysine polypeptide, 0.5-1 part of alanine, 0.5-1 part of acetylcysteine and 0.5-1 part of acetyltyrosine.
Further, the polypeptide and plant extract composition for reducing wrinkles further comprises 1-4 parts of the extract of the hibiscus syriacus flower and 1-3 parts of the extract of Jiang Huagen.
Further, the polypeptide and plant extract composition for reducing wrinkles further comprises 1-2 parts of a flos mori extract.
Further, the polypeptide and plant extract composition for reducing wrinkles further comprises 0.1-0.3 part of octanoyl hydroxamic acid and 0.5-2 parts of ethylhexyl glycerol.
Further, the polypeptide and plant extract composition for reducing wrinkles further comprises 10-15 parts of butanediol and 10-20 parts of water.
Further, the polypeptide compound for reducing wrinkles and the plant extract composition consists of acetyl hexapeptide-8, tripeptide-1, tetrapeptide-1 and nonapeptide-1 in equal mass ratio.
Further, a polypeptide and plant extract composition for reducing wrinkles as described above, wherein the magnolia sieboldii extract is prepared by the following method:
1) Cleaning radix Magnolia sieboldii, drying, pulverizing, and sieving with 50-100 mesh sieve to obtain radix Magnolia sieboldii powder;
2) Adding the magnolia sieboldii obtained in the step 1) into an extraction tank, adding an extraction solvent with the volume of 8-12 times, putting into an ultrasonic vibration instrument, treating for 45-60min under the ultrasonic wave of 50-55kHZ, filtering, repeating for 3 times, combining the 3 filtrates, and concentrating under reduced pressure to 1/4-1/5 of the original volume;
3) And (3) loading the concentrated solution in the step (2) on an HP-20 macroporous adsorption resin column, eluting with water for 3-4 times, discarding the eluent, eluting with 75% ethanol water solution, recovering the eluent, and concentrating under reduced pressure to dryness to obtain the magnolia sieboldii extract.

Compared with the prior art, the invention has the following beneficial effects:
(1) According to the polypeptide complex compounding and plant extraction composition provided by the technical scheme of the invention, acetyl hexapeptide-8 participates in competition of SNAP-25 at the site of the foam-melting complex, so that the formation of the complex is influenced, and the muscle contraction is weakened, and the formation of wrinkles is prevented; the plant extract composition and tripeptide, acetyl tetrapeptide-1 and pentapeptide-3 can further stimulate and promote the generation of collagen, fibronectin, elastin, hyaluronic acid, etc.; inhibiting glycosyl, reducing the problem of disordered and loose collagen fibers, and recovering elasticity; the skin is restored to be youthful and smooth.
(2) According to the technical scheme provided by the invention, the peptide compound is compounded with the paeonia lactiflora extract, the centella asiatica extract and the fermentation product filtrate of the two-way yeast, the spirulina maxima extract, the magnolia sieboldii extract, the saccharomycete polypeptides, the amino acid/lysine polypeptides, the alanine, the acetylcysteine and the acetyltyrosine, so that the inhibition of elastase and glucuronidase is further increased, the effects of promoting the activity of cathepsin and the activity of fiber cells, eliminating free radicals and inhibiting the generation of nitrogen oxides are achieved, the degradation of collagen fibers can be slowed down, the renewal of cells is stimulated, the service life of the cells is prolonged, and the skin elasticity can be maintained.
(3) According to the technical scheme provided by the invention, the peptide compound is compounded with the paeonia lactiflora extract, the centella asiatica extract and the fermentation product filtrate of the two-split yeast, the spirulina maxima extract, the magnolia sieboldii extract, the saccharomycete polypeptides, the amino acid/lysine polypeptides, the alanine, the acetylcysteine and the acetyltyrosine, so that the softening of the horny layer can be effectively promoted, the hydration function of cells can be increased, the water content of the horny layer can be improved, the moisture retention property is good, the deep water supplementing is realized, and the skin is moist and has high elasticity.

(6) According to the technical scheme provided by the invention, the magnolia sieboldii extract is extracted by combining ultrasonic waves with the adsorption resin column, so that the extraction efficiency is high, phenols and magnaline of the magnolia sieboldii extract are effectively maintained, the oxidation resistance and anti-aging performance of the magnolia sieboldii extract are improved, the degradation of collagen fibers is slowed down, and skin elasticity is maintained.

The polypeptide complexes described in examples 1 to 10 consist of acetyl hexapeptide-8, tripeptide-1, tetrapeptide-1, pentapeptide-3, nonapeptide-1 in equal mass ratios.
The magnolia sieboldii extracts of examples 1-10 were prepared by the following method:

Comparative example 3
A composition consists of the following components in parts by weight: 2g of polypeptide compound, 3g of paeonia lactiflora extract, 2g of centella asiatica extract, 2g of fermentation product filtrate of schizosaccharomyces cerevisiae, 4g of spirulina maxima extract, 4g of magnolia sieboldii extract, 1g of saccharomycetes polypeptide, 3.25g of alanine, 13g of butanediol, 12g of water, 0.2g of octanoyl hydroxamic acid and 1g of ethylhexyl glycerol.
The procedure for the preparation of magnolia sieboldii extract in the comparative example was the same as in the examples.
Comparative example 4
A composition consists of the following components in parts by weight: 2g of polypeptide compound, 3g of paeonia lactiflora extract, 2g of centella asiatica extract, 2g of fermentation product filtrate of schizosaccharomyces cerevisiae, 4g of spirulina maxima extract, 4g of magnolia sieboldii extract (commercially available), 1g of saccharomycetes polypeptide, 1g of arginine/lysine polypeptide, 0.75g of alanine, 0.75g of acetylcysteine, 0.75g of acetyltyrosine, 13g of butanediol, 12g of water, 0.2g of octanoyl hydroxamic acid and 1g of ethylhexyl glycerol.
The preparation method of the composition comprises the following steps: the components are weighed according to any one group of weight numbers in examples 1-10 and comparative examples 1-4, and are stirred and mixed uniformly at normal temperature to obtain the composite material.
In order to better illustrate the advantages of the present invention, the following provides experimental effects of the technical solution provided by the present invention:
experiment 1 Effect on collagen Synthesis
Test sample: examples 1, 5, 8, 10, compositions of comparative examples 1-4.
The testing method comprises the following steps: the contents of collagens I, III and IV of fibroblasts are measured by an ELISA method, the fibroblasts are digested by trypsin and transferred into 96-well culture plates, edge wells are filled with sterile phosphate buffer solution, the culture is continued in an incubator until the cells are completely adherent to the wall, 20 mu L/well of the composition sample solution of examples 1, 5, 8 and 10 and comparative examples 1-4 are sequentially added by using a microscale sample adding device, 0.01MPBS with the same volume is added in a control group, the contents of collagens I, III and IV are measured according to a kit instruction after 48 hours of adding samples, 3 compound wells are used as a control group, and the synthesis amount of the collagens is 100%. The test results are shown in Table 1.

The specific test method is as follows:
reference standard: reference is made to the QB/T4256-2011 guidelines for evaluation of moisturizing efficacy of cosmetics.
The test person: 40 healthy volunteers were selected, 30-40 years old, randomly grouped, 5 per group. The samples were applied twice daily in the morning and evening. During the experiment, the subject was unable to apply any other cosmetics at the experimental site.
Test conditions: the skin moisture meter Corneometer CM825 by capacitance method has the testing temperature of 20-22 ℃ and the humidity of 40-60%.

Experiment 3 skin elasticity test

0.2mmol/L DPPH solution is prepared by 50% ethanol and stored in a dark place. The experimental groups were prepared by preparing 0.2mg/mL of the tested solution from the compositions provided in examples 1, 5, 8 and 10 and the compositions of comparative examples 1-4, measuring the absorbance A at 517nm, measuring 3 times in parallel, taking an average value, and the experimental results are shown in Table 4.

Claims (3)
1. A polypeptide and plant extract composition for reducing wrinkles, which is characterized by comprising the following components in parts by weight: 1-5 parts of polypeptide compound, 1-6 parts of paeonia lactiflora extract, 0.5-4 parts of centella asiatica extract, 1.2-3.5 parts of fermentation product filtrate of two-split yeast, 2-7 parts of spirulina maxima extract, 3-7 parts of magnolia sieboldii extract, 0.5-1.5 parts of saccharomycete polypeptide, 0.5-1.5 parts of arginine/lysine polypeptide, 0.5-1 part of alanine, 0.5-1 part of acetylcysteine, 0.5-1 part of acetyltyrosine, 1-4 parts of harness hibiscus flower extract, 1-3 parts of Jiang Huagen extract and 1-2 parts of telescoping flower extract; 0.1-0.3 part of octanoyl hydroxamic acid and 0.5-2 parts of ethylhexyl glycerol; 10-15 parts of butanediol and 10-20 parts of water;
the polypeptide compound consists of acetyl hexapeptide-8, tripeptide-1, tetrapeptide-1, pentapeptide-3 and nonapeptide-1 in equal mass ratio;
the magnolia sieboldii extract is prepared by the following method:
1) Cleaning radix Magnolia sieboldii, drying, pulverizing, and sieving with 50-100 mesh sieve to obtain radix Magnolia sieboldii powder;
2) Adding the magnolia sieboldii obtained in the step 1) into an extraction tank, adding an extraction solvent with the volume of 8-12 times, putting into an ultrasonic vibration instrument, treating for 45-60min under the ultrasonic wave of 50-55kHZ, filtering, repeating for 3 times, combining the 3 filtrates, and concentrating under reduced pressure to 1/4-1/5 of the original volume;
