Preferably, the veterinary drug composition for treating urinary calculus of dogs and cats comprises the following components in parts by weight: 10-20 parts of plantain herb, 8-15 parts of desmodium styracifolium, 6-25 parts of rhizoma alismatis, 7-15 parts of semen coicis, 5-15 parts of pyrrosia lingua, 15-28 parts of cluster mallow seed, 3-18 parts of corn stigma and 2-12 parts of liquorice.

The invention adopts water extraction method which is a general method in industry to prepare: weighing the medicinal materials according to the required weight parts, adding water which is 8-10 times of the total weight of the medicinal materials, decocting twice for 1.5 hours each time, filtering, combining filtrates of the two times, refining and removing impurities, concentrating the filtrate under reduced pressure until the relative density is 1.15-1.20 (60 ℃), and preparing the corresponding preparation according to the preparation process of the required dosage form.
The application mode of the composition in the pharmaceutical field is as follows: the composition can be made into oral liquid, tablet, capsule, paste, etc.
The invention also provides application of the traditional Chinese medicine composition in preparation of a traditional Chinese veterinary medicine for preventing and treating urinary calculus of pets.

The traditional Chinese medicine composition for preventing and treating the urinary calculus of the pet is obtained according to the theory of Chinese veterinarian and through practice, has the effects of inducing diuresis and excreting dampness, clearing heat and removing toxicity, and inducing diuresis to alleviate edema, has the effects of improving various urinary calculus diseases of the pet, promoting dissolving and discharging of calculus, relieving inflammation, and reducing the recurrence rate of calculus, and is a veterinary medicine composition for the pet which is not reported in the market. Each tablet is 0.5g, and the content of the composition is 1.5 g.
Example 2
A traditional Chinese medicine composition for preventing and treating urinary calculus of pets, wherein the components and the weight of the traditional Chinese medicine composition are randomly selected from efficacy classifications as follows: 25 parts of plantain herb, 10 parts of desmodium styracifolium, 15 parts of rhizoma alismatis, 9 parts of semen coicis, 7 parts of pyrrosia lingua, 15 parts of cluster mallow seed, 8 parts of corn stigma and 3 parts of liquorice, adding water which is 8-10 times of the total weight of the medicinal materials, decocting twice for 1.5 hours each time, filtering, combining filtrates obtained in two times, refining and removing impurities, concentrating the filtrate under reduced pressure until the relative density is 1.15-1.20 (60 ℃), carrying out vacuum spray drying to obtain a dried substance, adding auxiliary materials, uniformly mixing, granulating and tabletting. Each tablet is 0.5g, and the content of the composition is 1.5 g.
Example 3
A traditional Chinese medicine composition for preventing and treating urinary calculus of pets, wherein the components and the weight of the traditional Chinese medicine composition are randomly selected from efficacy classifications as follows: 20 parts of plantain herb, 15 parts of desmodium styracifolium, 8 parts of rhizoma alismatis, 10 parts of semen coicis, 9 parts of pyrrosia lingua, 25 parts of cluster mallow seed, 5 parts of corn stigma and 2 parts of liquorice, adding water which is 8-10 times of the total weight of the medicinal materials, decocting twice for 1.5 hours each time, filtering, combining filtrates obtained in two times, refining and removing impurities, concentrating the filtrate under reduced pressure until the relative density is 1.05-1.10 (60 ℃), and adding water to dilute until 1g of the raw medicinal materials is contained in each 1ml of filtrate.
Example 4
A traditional Chinese medicine composition for preventing and treating urinary calculus of pets comprises the following components in parts by weight: 15 parts of desmodium styracifolium, 10 parts of rhizoma alismatis, 8 parts of semen coicis, 15 parts of cluster mallow seeds, 7 parts of corn stigma and 2 parts of liquorice are weighed according to the required weight parts, water which is 8-10 times of the total weight of the medicinal materials is added, the decoction is carried out twice, each time is carried out for 1.5 hours, the filtration is carried out, the filtrates of the two times are combined, the refining and the impurity removal are carried out, the filtrate is decompressed and concentrated to the relative density of 1.15-1.20 (60 ℃), the vacuum spray drying is carried out to obtain a dried substance, auxiliary materials are added, the mixture is uniformly mixed, No. 1 capsule is prepared, and the composition contained in each capsule is 1.5g of the original medicinal material.
1. Safety test study of the invention
1.1 test methods
Test dogs were selected, and in example 1, mental states, food intake, drinking, exercise, feces, urine color, and the like were recorded daily for 14 consecutive days in tablets at 5 times the clinical recommended dose (recommended dose: 2 tablets/5 kg body weight) per day. Collecting the saphenous vein of hind limb 1 day before and 14 days after the test, wherein one part is used for routine blood detection, and the other part is used for separating serum and carrying out serum biochemical index detection: ALT (alanine aminotransferase), AST (aspartate aminotransferase), BUN (urea), Creatinine (CREA), serum calcium (Ca), and the like.
1.2 test results
1.2.1 clinical observations
During the test period, all test dogs are normal in mental, sports, food intake and drinking conditions, and have no conditions of food refusal, vomiting and the like, and the properties of excrement and urine are not abnormal.
1.2.2 blood test
The blood routine and biochemical index detection results are shown in the following tables 2 and 3 according to statistics, and according to the data, the detection results of various physiological index specifications of the test dog have no abnormal change compared with the detection results before the test, and are all within a normal range, so that the safety of the invention is very high.

1.2.3 conclusion
The test dog is fed with the calculus forming tablets with 5 times of dosage, adverse reactions are not seen in the two-week feeding period, general physical sign observation, blood routine and biochemical indexes are not abnormal, and the clinical use safety of the calculus forming tablets is high.
2. Experimental study of the invention
2.1 test methods
20 test dogs were selected and randomly divided into a blank control group, a model group, and example 1, example 3, and example 4 groups. During the test period, the animals received free access to drinking water in the following manner as shown in table 4.

2.2 results
2.2.1 general signs
Before administration, except for the blank group, dogs in other groups had depressed mental state, decreased food intake, normal stool state and deepened urine color. After the administration of the examples 1, 3 and 4, the state is gradually recovered, and after the administration of the examples 1 and 3, the mental state, the feed intake and the urine color of the dog are recovered to be normal 21 days, the dog becomes active and well-moving, and the urine color is gradually changed into light yellow from reddish brown or dark yellow; example 4 recovered slowly compared to examples 1 and 3, and urine color did not return to normal 21 days after dosing. The aspects of the model group gradually recovered from 14 days after the test, and the urine color improved after 21 days.
2.2.2 urine changes
Table 5 shows that on the 21 st day of the test, the urine character score of the model group is improved relatively slowly, which shows that the effect is relatively poor depending on the self restoring force of the animal body; the urine character scores of the groups of examples 1 and 3 are obviously improved, and the color of urine is basically recovered to be normal; the urine color of the example 4 group is not completely recovered but is better than that of the model group due to the lower urine trait scores of the example 1 and the example 3 groups.

2.2.3 Biochemical index detection
The measurement of urea nitrogen and creatinine in serum showed: on the 14 th day of the test, the urea nitrogen of the groups of examples 1, 3 and 4 starts to decrease, wherein the decrease of the groups of examples 1 and 3 is more obvious, and the level of the model group is increased; creatinine levels began to rise for the examples 1, 3, 4 and model groups; at day 21 of the trial, the increase in creatinine levels was more pronounced in the model group compared to the creatinine levels in the examples 1, 3, and 4 groups.
The results of the blood calcium and urine calcium measurements show that: on the 14 th day of the test, the urinary calcium of the groups of examples 1, 3 and 4 begins to decrease, and the blood calcium does not change obviously; on day 21 of the test, the urine calcium reduction was more significant in the groups of examples 1, 3 and 4 than in the model group, and the results are shown in tables 6 and 7, in which the groups of examples 1 and 3 were superior to the group of example 4.

2.3 conclusion
The animals fed with the feed of the examples 1, 3 and 4 are fed to the urinary calculus dogs every day, and after the feed is used for 21 days, symptoms such as urinary calculus can be effectively improved, the content of serum urea nitrogen and urine calcium can be reduced, the recovery of calculus symptoms is facilitated, and the effect of the groups of the examples 1 and 3 is better than that of the group of the example 4.
3. Clinical case efficacy reports of examples 1 and 3 of the present invention
3.1 Experimental methods
45 sick dogs with urinary calculus symptoms are screened and randomly divided into 3 groups. In case screening, cases with urinary tract diseases are excluded, and urinary tract calculus cases show the following clinical manifestations: the pet has bad appetite, lassitude, frequent urination, dribbling urine, hematuria and strong ammonia smell, difficult urination, tense after long-time squatting or urination, frequent licking of genitals, etc.
The positive control group is a conventional treatment method of the current western medicine, and adopts a laser lithotripsy technology to discharge along with urine or open the bladder to remove stones by an operation according to the blocking condition of urinary calculi when the urinary calculi is completely blocked; when the obstruction is not completely blocked, medicines such as diuretic, potassium acetate, aminophylline, etc. are used for treating.
Example 1 administration of 2 tablets/4 kg body weight daily, 2 times a day for 21 consecutive days; example 3 administration was performed 2 times a day for 21 consecutive days at a dose of 3ml/4kg body weight per day.
3.2 therapeutic efficacy criteria
Rehabilitation: the mental state, the eating and drinking water and the behavioral activities are normal, the pain of the urine disappears, the color is normal, and the calculi are discharged;
the method has the following advantages: the mental state, the eating and drinking of water and the behavior are improved, the pain of urination gradually disappears, the color is dark, and the calculi are reduced;
and (4) invalidation: mental state, food intake, drinking water, no improvement or deterioration of behavior, persistent pain in urination, deepened color or hematuria, and increased calculus amount and volume.

3.3 clinical results
The statistics are shown in Table 8. The results in Table 8 show that the total effective rates of the group of example 1 and the group of example 3 are not significantly different from the control group (P >0.05),
the total effective rate of the two groups is more than 80%; the relapse rates of the example 1 group and the example 3 group were significantly different from those of the control group (P < 0.05),
the recurrence rate of the two groups was significantly lower than the control group; shows that the traditional Chinese medicine composition has better treatment effect on the urinary calculus symptom of pets,
can effectively dilute urine, dissolve calculi, promote the discharge of calculi and reduce the recurrence rate of calculi.

4. Examples 2 and 3 clinical case efficacy reports of the present invention
3.1 Experimental methods
In case screening, cases with complete obstruction of urinary calculus are excluded, and 30 incompletely-obstructed dogs and 15 cats are screened and randomly divided into two groups. The following clinical manifestations are presented in the cases of urinary tract stones: the pet has bad appetite, lassitude, frequent urination, dribbling urine, hematuria and strong ammonia smell, difficult urination, tense after long-time squatting or urination, frequent licking of genitals, etc.
The positive control group is a certain brand of urinary tract prescription food sold in the market at present and is fed according to requirements.
Example 2 was administered 2 tablets per 4kg body weight per day 2 times a day for 21 consecutive days.
Example 3 was used 2 times a day for 21 consecutive days at a dose of 3ml/4kg body weight per day.
In both cases, the treatment was performed in the same manner except for pet food.
3.2 therapeutic efficacy criteria
Rehabilitation: mental state, drinking water and normal behavior, pain of urination disappears, color is normal, and calculi is discharged;
the method has the following advantages: the mental state, the eating and drinking of water and the behavior are improved, the pain of urination gradually disappears, the color is dark, and the calculi are reduced;
and (4) invalidation: mental state, food intake, drinking water, no improvement or deterioration of behavior, persistent pain in urination, deepened color or hematuria, and increased calculus amount and volume.
3.3 clinical results
The statistics are shown in Table 9. The results in table 9 show that the effective rates of the groups in examples 2 and 3 are significantly higher than those of the control group (P < 0.05), the effective rates in pet feed are all more than 80%, the recurrence rate is significantly lower than that of the control group (P < 0.05), and the effect is still ideal. The traditional Chinese medicine composition has a good treatment effect on pet urinary calculus symptoms, can effectively dilute urine, dissolve calculus, promote calculus discharge and reduce calculus recurrence rate.

By combining the experimental researches, the traditional Chinese medicine composition can be added into tablets, oral liquid and capsules to be used, can well play the roles of diluting urine, dissolving calculus, promoting the discharge of calculus and reducing the recurrence of calculus, can be widely used for preventing and treating urinary calculus symptoms of pets, shortens the course of disease and quickly restores normal physiological functions.
The preferred embodiments of the present invention have been described in detail, however, the present invention is not limited to the specific details of the above embodiments, and various changes may be made to the technical solution of the present invention within the technical idea of the present invention, and these simple modifications are included in the protective scope of the present invention.
It should be noted that, in the above embodiments, the various specific technical features and steps described in the above embodiments may be combined in any suitable manner, and in order to avoid unnecessary repetition, the present invention does not separately describe various possible combinations.

The traditional Chinese medicine composition for preventing and treating urinary calculus of pets according to claim 1, which is characterized in that the medicine composition comprises the following main materials in parts by weight: 8-15 parts of plantain herb, 12-15 parts of desmodium styracifolium, 6-15 parts of rhizoma alismatis, 5-15 parts of semen coicis, 5-13 parts of pyrrosia lingua, 10-25 parts of cluster mallow seed, 5-12 parts of corn stigma and 1-5 parts of liquorice.

The traditional Chinese medicine composition for preventing and treating urinary calculus of pets according to claim 2, wherein the weight parts of the main materials in the medicine composition are preferably as follows: 12 parts of plantain herb, 12 parts of desmodium styracifolium, 9 parts of rhizoma alismatis, 6 parts of semen coicis, 6 parts of pyrrosia lingua, 20 parts of cluster mallow seed, 6 parts of corn stigma and 2 parts of liquorice.

The traditional Chinese medicine composition for preventing and treating urinary calculus of pets according to any one of claims 1 and 2, which is characterized in that the preparation method comprises the following steps: weighing the medicinal materials according to the weight parts, adding water which is 8-10 times of the total weight of the medicinal materials, decocting twice for 1.5 hours each time, filtering, combining filtrates obtained in the two times, refining and removing impurities, concentrating the filtrate under reduced pressure until the relative density is 1.15-1.20 (60 ℃), and performing vacuum spray drying to obtain a dried substance.
5. The traditional Chinese medicine composition for preventing and treating urinary calculus of pets according to any one of claims 1 and 2, wherein the traditional Chinese medicine composition can be used in a form of being added into tablets, oral liquid, capsules, nutritional paste and grains.
6. The use of the veterinary drug composition according to any one of claims 1 and 2 in the preparation of a medicament for preventing and treating urinary calculus of pets.
7. The use as claimed in claim 6, wherein the companion animal is a cat or a dog.
