Compared with the prior art, the invention has the following beneficial effects:
1) the plant extract composition with anti-inflammatory and allergy-relieving functions takes the extracts of the three substances of the pseudo-ginseng root, the gardenia fruit and the kava root as main analgesic and anti-inflammatory functional components, and the raw materials are selected from natural plant components, so that the plant extract composition with anti-inflammatory and allergy-relieving functions has no toxicity or irritation to human skin and high safety.
2) The plant extract composition with anti-inflammatory and allergy-relieving functions of the invention has the functions of effectively resisting stabbing pain and relieving redness and burning sensation, and the efficacy and the action of the plant extract composition are obviously better than those of a single preparation material.

Detailed Description
Embodiments of the present invention will be described in detail below with reference to examples, but it will be understood by those skilled in the art that the following examples are only illustrative of the present invention and should not be construed as limiting the scope of the present invention.
Example 1
A method for preparing plant extract composition with anti-inflammatory and anti-allergy effects comprises the following steps:
step 1, adding 30g of pseudo-ginseng root into 300mL of 80% ethanol solution, and performing reflux extraction at 60 ℃ for 2 hours to obtain pseudo-ginseng root extracting solution;
step 2, adding 40g of gardenia fruits into 400mL of 50% ethanol solution, and performing reflux extraction at 50 ℃ for 2 hours to obtain gardenia fruit extracting solution;
step 3, adding 30g of kava root into 300mL of 70% ethanol solution, and carrying out reflux extraction at 70 ℃ for 2h to obtain kava root extracting solution;
step 4, uniformly mixing the pseudo-ginseng root extract, the gardenia fruit extract and the kava root extract according to the proportion of 3:4:3 of pseudo-ginseng root, gardenia fruit and kava root to obtain a mixed solution;
step 5, adding 1g of hydroxypropyl cyclodextrin into the mixed solution obtained in the step 4, uniformly mixing, and then concentrating at 70 ℃ until no alcohol exists to obtain a concentrated solution; heating the concentrated solution at 70 deg.C for 1 hr, filtering at room temperature under-0.1 MPa, and vacuum drying at 70 deg.C for 4 hr to obtain plant extract composition DCR550 with antiinflammatory and allergy relieving effects.
Example 2
A method for preparing plant extract composition with anti-inflammatory and anti-allergy effects comprises the following steps:
step 1, adding 35g of pseudo-ginseng root into 300mL of 75% ethanol solution, and performing reflux extraction at 60 ℃ for 2 hours to obtain pseudo-ginseng root extracting solution;
step 2, adding 35g of gardenia fruits into 400mL of 60% ethanol solution, and performing reflux extraction at 50 ℃ for 2 hours to obtain gardenia fruit extracting solution;
step 3, adding 30g of kava root into 300mL of 70% ethanol solution, and carrying out reflux extraction at 70 ℃ for 2h to obtain kava root extracting solution;
step 4, uniformly mixing the pseudo-ginseng root extract, the gardenia fruit extract and the kava root extract according to the proportion of 3.5:3.5:3 of pseudo-ginseng root, gardenia fruit and kava root to obtain a mixed solution;
step 5, adding 1.5g of hydroxypropyl cyclodextrin into the mixed solution obtained in the step 4, uniformly mixing, and concentrating at 50 ℃ until no alcohol exists to obtain a concentrated solution; heating the concentrated solution at 60 deg.C for 1 hr, filtering at room temperature under-0.1 MPa, and vacuum drying at 50 deg.C for 6 hr to obtain plant extract composition DCR550 with antiinflammatory and allergy relieving effects.
Example 3
A method for preparing plant extract composition with anti-inflammatory and anti-allergy effects comprises the following steps:
step 1, adding 40g of pseudo-ginseng root into 400mL of 75% ethanol solution, and performing reflux extraction at 50 ℃ for 3 hours to obtain pseudo-ginseng root extracting solution;
step 2, adding 20g of gardenia fruits into 200mL of 60% ethanol solution, and performing reflux extraction at 45 ℃ for 2.5 hours to obtain gardenia fruit extracting solution;
step 3, adding 40g of kava root into 400mL of 70% ethanol solution, and performing reflux extraction at 65 ℃ for 1.5h to obtain kava root extracting solution;
step 4, uniformly mixing the pseudo-ginseng root extract, the gardenia fruit extract and the kava root extract according to the proportion of 4:2:4 of pseudo-ginseng root, gardenia fruit and kava root to obtain a mixed solution;
step 5, adding 1.5g of hydroxypropyl cyclodextrin into the mixed solution obtained in the step 4, uniformly mixing, and concentrating at 50 ℃ until no alcohol exists to obtain a concentrated solution; heating the concentrated solution at 60 deg.C for 1 hr, filtering at room temperature under-0.1 MPa, and vacuum drying at 50 deg.C for 6 hr to obtain plant extract composition DCR550 with antiinflammatory and allergy relieving effects.
Example 4
A method for preparing plant extract composition with anti-inflammatory and anti-allergy effects comprises the following steps:
step 1, adding 30g of pseudo-ginseng roots into 400mL of 60% methanol solution, and performing reflux extraction at 70 ℃ for 1 hour to obtain pseudo-ginseng root extracting solution;
step 2, adding 20g of gardenia fruits into 400mL of 70% ethanol solution, and performing reflux extraction at 60 ℃ for 1.5h to obtain gardenia fruit extracting solution;
step 3, adding 50g of kava root into 500mL of 70% methanol solution, and performing reflux extraction at 80 ℃ for 1h to obtain kava root extracting solution;
step 4, uniformly mixing the pseudo-ginseng root extract, the gardenia fruit extract and the kava root extract according to the proportion of 3:2:5 of pseudo-ginseng root, gardenia fruit and kava root to obtain a mixed solution;
step 5, adding 2g of hydroxypropyl cyclodextrin into the mixed solution obtained in the step 4, uniformly mixing, and concentrating at 60 ℃ until no alcohol exists to obtain a concentrated solution; heating the concentrated solution at 70 deg.C for 1 hr, filtering at room temperature under-0.1 MPa, and vacuum drying at 70 deg.C for 6 hr to obtain plant extract composition DCR550 with antiinflammatory and allergy relieving effects.
Comparative example 1
The preparation method of the kava root extract comprises the following steps:
step 1, adding 100g of kava root into 1000mL of 70% ethanol solution, and carrying out reflux extraction at 70 ℃ for 2h to obtain kava root extracting solution;
and 2, drying the kava root extracting solution for 6 hours in vacuum at the temperature of 70 ℃ to obtain the kava root extract.
Comparative example 2
The preparation method of the pseudo-ginseng root extract comprises the following steps:
step 1, adding 100g of pseudo-ginseng root into 1000mL of 80% ethanol solution, and performing reflux extraction at 70 ℃ for 2 hours to obtain pseudo-ginseng root extracting solution;
and 2, drying the pseudo-ginseng root extracting solution for 6 hours in vacuum at the temperature of 70 ℃ to obtain the pseudo-ginseng root extract.
Comparative example 3
The preparation method of the gardenia fruit extract comprises the following steps:
step 1, adding 100g of gardenia fruits into 1500mL of 60% ethanol solution, and carrying out reflux extraction at 70 ℃ for 2 hours to obtain gardenia fruit extracting solution;
and 2, drying the gardenia fruit extracting solution at 70 ℃ for 6 hours in vacuum to obtain the gardenia fruit extract.
In the above examples 1-4, in step 1-3, the three substances of the kava root, the pseudo-ginseng root and the gardenia fruit can be extracted by using a methanol solution or an ethanol solution, and the three substances can be extracted by using an ethanol solution, so that the residual odor of the organic solvent after drying can be avoided; the ethanol solutions with different concentrations are adopted to respectively extract the three substances, because the solubility of the effective components of the three substances in the ethanol solutions is different, the concentration of the ethanol solution provided by the embodiment of the application can enable the effective components in the kava root, the pseudo-ginseng root and the gardenia fruit to be better dissolved in the ethanol solution with corresponding concentration, so that the finally obtained plant extract composition with anti-inflammatory and allergy-relieving functions of anti-prickling, redness relieving and burning sensation is better.
In the step 5, hydroxypropyl cyclodextrin is added into the mixed solution, and water-insoluble ingredients in the pseudo-ginseng root extract, the gardenia fruit extract and the kava root extract can be included, so that the content of some functional ingredients is improved, and the finally obtained plant extract composition with anti-inflammatory and allergy-relieving functions of anti-prickling pain, redness relieving and burning sensation are better.
The plant extract composition with anti-inflammatory and anti-allergy functions provided by the embodiments of the invention comprises the following raw materials:
notoginseng radix (Panax notoginseng) root, which is a perennial upright herb of Panax of Araliaceae, contains notoginsenoside R1, ginsenoside Rb1, Rg1, Rg2 and small amount of ginsenoside Ra, Rb2, Rb and Re. The notoginseng root can intervene NF-kB pathway by inhibiting the activity of nuclear factor kB (NF-kB), so as to reduce the generation of inflammatory factors, and further inhibit or slow down the generation of inflammation.
Gardenia fruit (Gardenia florida), dried mature fruit of Gardenia jasminoides Ellis of Rubiaceae, contains geniposide as main effective component. The geniposide is also called geniposide and is a main functional component with anti-inflammatory effect of the gardenia, and the anti-inflammatory mechanism of the geniposide is as follows: inhibiting the gene expression of partial inflammatory factors and the activation thereof. And the geniposide can play an analgesic role by controlling opioid receptor agonism and inhibiting synthesis and release of Nitric Oxide (NO).
Piper methysticum (Piper methysticum) root, Piperaceae perennial upright shrub medicinal plant of Nanpacific Islands, and its main effective components are kavalactone, kawaine, kavain, etc. The kava root can achieve the effects of relieving pain and allergy by inhibiting the expression of TRPV1 protein.
The plant extract composition having anti-inflammatory and anti-allergy effects obtained in example 1, the kava root extract obtained in comparative example 1, the notoginseng root extract obtained in comparative example 2, and the gardenia fruit extract obtained in comparative example 3 were subjected to patch test and evaluation.
1) The experimental method comprises the following steps:
the materials of example 1 and comparative examples 1-3 were dissolved and prepared into 1% solutions as experimental groups, and deionized water as blank group, and the solutions were divided into three groups. 0.5 per mill of capsicine solution is used as a patch experiment stimulating reagent, 0.5mL of capsicine solution is sucked and respectively added into three groups of 0.5mL solutions, and the three groups of solutions are placed in a patch. Screening 30 normal people with different ages and different sexes, sticking the patch on the inner side of the small arms of the two arms, and recording the feeling time and the strength of the burning sensation and the stabbing pain sensation of the arms in the test process; and simultaneously recording and comparing the red and swollen state of the skin at the spot-pasting part on the inner side of the arm before and after the test.
2) Test results
A graph showing the burning sensation of an arm as a function of time is shown in FIG. 1, and a graph showing the burning sensation of an arm as a function of time is shown in FIG. 2. In fig. 1 and 2, the ordinate represents the sensory scale, and the greater the number, the more pronounced the sensation, i.e. the stronger the tingling or burning sensation.
As can be seen from fig. 1, compared to the single kava root extract of comparative example 1, the anti-inflammatory and allergy-relieving plant extract composition obtained in the present application has a weaker pricking strength and a shorter lasting time of the pricking, under the premise of reducing the amount of kava roots, which indicates that the anti-inflammatory and allergy-relieving plant extract composition obtained in the present application has a better analgesic effect.
Compared with the single notoginseng root extract of the comparative example 2, the plant extract composition with anti-inflammatory and allergy-relieving functions has the advantages that the pricking strength is weaker and the pricking time is shorter on the premise of reducing the using amount of the notoginseng root extract, and the plant extract composition with anti-inflammatory and allergy-relieving functions has better analgesic effect.
Compared with the single gardenia fruit extract of the comparative example 3, the plant extract composition with anti-inflammatory and allergy-relieving functions obtained by the application has weaker pricking strength and shorter lasting time of the pricking on the premise of reducing the using amount of the gardenia fruit extract, and shows that the plant extract composition with anti-inflammatory and allergy-relieving functions obtained by the application has better analgesic effect.
As can be seen from fig. 2, compared to the single kava root extract of comparative example 1, the plant extract composition with anti-inflammatory and anti-allergy activities obtained in the present application has weaker burning intensity and shorter burning duration under the premise of reducing the amount of kava roots, which indicates that the plant extract composition with anti-inflammatory and anti-allergy activities obtained in the present application has better analgesic effect.
Compared with the single pseudo-ginseng root extract of the comparative example 2, the plant extract composition with anti-inflammatory and allergy-relieving functions obtained by the application has weaker burning sensation strength and shorter burning sensation duration on the premise of reducing the using amount of the pseudo-ginseng root, and shows that the plant extract composition with anti-inflammatory and allergy-relieving functions obtained by the application has better analgesic effect.
Compared with the single gardenia fruit extract of the comparative example 3, the plant extract composition with anti-inflammatory and allergy relieving functions obtained by the application has weaker burning sensation strength and shorter burning sensation duration on the premise of reducing the using amount of gardenia fruits, and shows that the plant extract composition with anti-inflammatory and allergy relieving functions obtained by the application has better analgesic effect.
The skin redness and swelling state of the inner side of the arm of 30 test people is shown in table 1, wherein the redness and swelling comparison graph of one arm is shown in fig. 3, and the upper arm is the skin redness and swelling state graph of comparative example 1; the lower arm is a red and swollen skin view of example 1.

As can be seen from Table 1, the skin redness rate of the single kava root of comparative example 1 is 93.3%, while the skin redness rate of the plant extract composition with anti-inflammatory and anti-allergy relieving effects obtained in the present application is only 6.7%, which indicates that the plant extract composition with anti-inflammatory and anti-allergy relieving effects obtained in the present application has better anti-inflammatory effects.
As can be seen from FIG. 3, the red swelling of the arms of comparative example 1 is severe as indicated by the black rectangular frame of FIG. 3, whereas the arms of example 1 of the present application have almost no red swelling, indicating that the anti-inflammatory and desensitizing plant extract composition obtained in example 1 can eliminate the red swelling caused by inflammatory factors, thus demonstrating that example 1 has a prominent anti-inflammatory effect over comparative example 1.
It should be noted that the present application is described by taking the efficacy of the plant extract composition with anti-inflammatory and anti-allergy activities of example 1 as an example, and the efficacy of the plant extract composition with anti-inflammatory and anti-allergy activities obtained in examples 2 to 4 is the same as that of example 1.
In conclusion, the experimental group of the plant extract composition DCR550 with anti-inflammatory and allergy-relieving functions has the functions of effectively resisting stabbing pain and relieving redness and burning sensation, and the efficacy and the effect of the composition are obviously better than those of a single material.
Although the present invention has been described in detail in this specification with reference to specific embodiments and illustrative embodiments, it will be apparent to those skilled in the art that modifications and improvements can be made thereto based on the present invention. Accordingly, such modifications and improvements are intended to be within the scope of the invention as claimed.
Claims (10)
1. The plant extract composition with anti-inflammatory and allergy-relieving functions is characterized by comprising the following preparation raw materials: notoginseng root, gardenia fruit and kava root.
2. The plant extract composition with anti-inflammatory and anti-allergy effects of claim 1, wherein the raw materials are used in the amounts of: 30-50 parts of pseudo-ginseng root, 20-40 parts of gardenia fruit and 30-50 parts of kava root.
3. The preparation method of the plant extract composition with anti-inflammatory and allergy relieving functions is characterized by comprising the following steps:
step 1, adding pseudo-ginseng roots into an alcohol solution, and performing reflux extraction to obtain a pseudo-ginseng root extracting solution;
step 2, adding the gardenia fruits into an alcohol solution, and performing reflux extraction to obtain a gardenia fruit extracting solution;
step 3, adding the kava root into an alcohol solution, and performing reflux extraction to obtain kava root extracting solution;
and 4, uniformly mixing the pseudo-ginseng root extracting solution, the gardenia fruit extracting solution and the kava root extracting solution to obtain a mixed solution, wherein the mixed solution is the plant extraction composition with the functions of resisting inflammation and relieving allergy.
4. The method for preparing a plant extract composition with anti-inflammatory and anti-allergy effects as claimed in claim 3, further comprising the following steps after step 4:
step 5, adding hydroxypropyl cyclodextrin into the mixed solution, uniformly mixing, and concentrating to obtain a concentrated solution; and heating the concentrated solution for reaction, filtering and drying to obtain the purified plant extract composition with anti-inflammatory and allergy-relieving effects.
5. The method for preparing a plant extract composition with anti-inflammatory and anti-allergy effects according to claim 3, wherein the alcohol solution is methanol solution or ethanol solution in steps 1-3.

The method for preparing the plant extract composition with anti-inflammatory and anti-allergy activities according to claim 5, wherein in the step 1, the mass volume ratio of the pseudo-ginseng root to the alcoholic solution is as follows: 1 (5-15); the alcoholic solution is 60-90% of ethanol solution; the temperature of the reflux extraction is 50-70 ℃, and the time of the reflux extraction is 1-3 h.

The method for preparing the plant extract composition with anti-inflammatory and anti-allergy effects as claimed in claim 5, wherein in the step 2, the mass volume ratio of the gardenia fruit to the alcoholic solution is as follows: 1, (10-25); the alcoholic solution is 40-70% of ethanol solution; the temperature of the reflux extraction is 40-70 ℃, and the time of the reflux extraction is 1-3 h.

The method for preparing a plant extract composition with anti-inflammatory and anti-allergy effects as claimed in claim 5, wherein in the step 3, the mass volume ratio of the kava root to the alcohol solution is as follows: 1, (5-15); the alcoholic solution is 60-90% of ethanol solution; the temperature of the reflux extraction is 60-80 ℃, and the time of the reflux extraction is 1-3 h.

The method for preparing a plant extract composition with anti-inflammatory and anti-allergy effects as claimed in claim 5, wherein in the step 5, the mass volume ratio of the hydroxypropyl cyclodextrin to the mixed solution is as follows: 1, (300-1000); the concentration temperature is 50-80 ℃; the heating reaction temperature is 50-80 ℃, and the heating reaction time is 1-2 h.
