Abstract
一种大豆咖啡西洋参生物活性肽的制备方法和应用,原料包括大豆、西洋参和咖啡,所述大豆、西洋参和咖啡的蛋白原料经胃蛋白酶和胰蛋白酶酶解制成,所述大豆、西洋参和咖啡肽的质量比为(1.3‑3.1):(1.3‑2.2):(1.8‑4.1),所述大豆、西洋参和咖啡复合肽的质量比为(1.3‑4.0):0.42;将大豆30‑50份,西洋参5‑15份,咖啡15‑40份比例混合,首先制备分离碱性蛋白溶液,将大豆、西洋参、咖啡分离,碱性溶液加入胃蛋白酶、胰蛋白酶、木瓜蛋白酶、木薯淀粉酶,酶解后,灭酶。本发明通过特定工艺制备获得了特定分子量区间的复合植物小分子大豆咖啡西洋参肽液,能够起到抗疲劳、抗衰老、抗休克、提高思维、改善记忆及调节内分泌、增强人体免疫力及改善心血管功能等作用。

技术领域
本发明涉及大豆咖啡西洋参生物活性肽的制备领域,尤其涉及一种大豆咖啡西洋参生物活性肽的制备方法和应用。
背景技术
生物活性肽(Bioactive Peptides,BAP)是蛋白质中20种天然氨基酸以不同组成和排列方式构成的从二肽到复杂的线性、环形结构的不同肽类的总称,是源于蛋白质的多功能化合物。生物活性肽具有多种人体代谢和生理调节功能,易消化吸收,有促进免疫、激素调节、抗菌、抗病毒、降血压、降血脂等作用,是当前国际食品界最热门的研究课题和极具发展前景的功能因子。
现有的生物活性肽种类中并没有以大豆、西洋参和咖啡为原料的,导致生物活性肽的种类不够丰富,营养成分也不够多样,无法满足消费者的需求。
发明内容
针对现有技术中的上述不足,本发明提供了一种大豆咖啡西洋参生物活性肽的制备方法和应用。
为了达到上述发明目的,本发明采用的具体方案为:
一种大豆咖啡西洋参生物活性肽的制备方法,原料包括大豆、西洋参和咖啡,所述大豆、西洋参和咖啡的蛋白原料经胃蛋白酶和胰蛋白酶酶解制成,所述大豆、西洋参和咖啡肽的质量比为(1.3-3.1):(1.3-2.2):(1.8-4.1),所述大豆、西洋参和咖啡复合肽的质量比为(1.3-4.0):0.42。
进一步,具体方法为:将大豆30-50份,西洋参5-15份,咖啡15-40份比例混合,首先制备分离碱性蛋白溶液,将大豆、西洋参、咖啡分离,碱性溶液加入胃蛋白酶、胰蛋白酶、木瓜蛋白酶、木薯淀粉酶,酶解后,灭酶。
进一步,所述复合小分子离子肽液去除表面杂质,离心后所得上清液,即获得复合大豆西洋参咖啡小分子(离子)肽。
一种大豆咖啡西洋参生物活性肽的应用,按1%-5%的比列和纯净水调合,再加入调味剂、植物防腐剂、纯净水,得到原料,制成化妆品、食品或药品及保健品。
本发明的有益效果为:
本发明通过特定工艺制备获得了特定分子量区间的复合植物小分子大豆咖啡西洋参肽液。三者或四者在合适比例下组成合物,能够以较少的使用量,起到抗疲劳、抗衰老、抗休克、提高思维、改善记忆及调节内分泌、增强人体免疫力及改善心血管功能等作用。对促进淋巴细胞的转化,诱导免疫因子生成,增强集体免疫功能的作用。具有镇静、增强学习记忆、促进神经生长、抗惊厥、阵痛、解热的作用。具有抗心率失常、抗心肌缺血和再灌损伤等作用。具有抗溶血、止血、降低血液凝固性、抑制血小板凝聚、调血脂、抗动脉粥硬化、降低血糖等作用。具有抗利尿的作用。具有抑制癌细胞增殖、抑制单纯疱疹等病毒的作用。
具体实施方式
以下通过具体实施例进一步描述本发明,但本发明不仅仅限于以下实施例。在本发明的范围内或者在不脱离本发明的内容、精神和范围内,对本发明进行的变更、组合或替换,对于本领域的技术人员来说是显而易见的,且包含在本发明的范围之内。
一种大豆咖啡西洋参生物活性肽的制备方法和应用,首先制备大豆和咖啡及西洋参按30-70份,5-15份,15-40份混合分离蛋白碱性溶液。将大豆、咖啡和西洋参分离碱性溶液加入Alcalase蛋白酶,酶解后,灭酶,离心,取得上清液,即为复合大豆咖啡西洋参肽液。在温室下对大豆西洋参咖啡肽液进行回流超滤操作,在超滤操作同时进行活性炭的吸附除臭。所述超滤操作采用9支超滤膜进行串联截留分子量为2500Da~4500Da,压力为0.2Mpa~1.2Mpa,处理5~20min,脱除相对分子质量≦2500Da的无机盐离子后,收集滤出大豆咖啡西洋参复合肽液。后在温室下加入橙子、草莓液等调味剂及低聚果糖、木糖醇、水果糖等改善成品复合肽液的口感。生产出具有保健和治疗为主的双重作用的口服液。
实施例1
原料组成(重量份):复合大豆西洋参咖啡复合肽80份、结晶果糖和食品添加剂2份。
所述食品添加剂由以下原料按重量份混合而成:甜橙香精0.5份,降苦味香精0.5份,罗汉果甜苷0.4份,甜菊糖苷0.05份。
一种植物肽的制备方法,包括以下步骤:
S1.将复合大豆西洋参咖啡肽溶于水中(物料与水的质量比为1:10),用1mol/LNaOH溶液调节溶液pH为8.5,加入碱性蛋白酶酶解,然后用1mol/L HC1溶液调节pH为7,加入中性蛋白酶酶解,添加量为2500U/kg,结束后灭酶,离心,取上清液,用截留分子量为1000u的超滤膜超滤,得到分子量小于1000u的滤过液,得到大豆西洋参咖啡复合低聚肽浓缩液。
S2.将大豆西洋参咖啡低聚肽混合,溶解于水中(物料与水的质量比为1:10),用1mol/L HC1溶液调节pH为7,加入a-糜凝乳酶和风味蛋白酶反应,所述a-糜凝乳酶的添加量为2000U/kg,所述风味蛋白酶的添加量为3000U/kg,结束后灭酶,离心,取上清液,透析,-10℃温度下预冷50min,得到复合肽浓缩液,加入结晶果糖和食品添加剂,溶解混合均匀,制得植物肽口服液。
实施例3
原料组成(重量份):大豆西洋参咖啡复合肽75份、结晶果糖15份和食品添加剂1.5份。
所述食品添加剂由以下原料按重量份混合而成:甜橙香精0.5份,降苦味香精0.5份,罗汉果甜苷0.4份,甜菊糖苷0.05份。
一种植物肽的制备方法,包括以下步骤:
S1.将大豆西洋参咖啡肽溶于水中(物料与水的质量比为1:10),用1mol/L NaOH溶液调节溶液pH为8.5,加入碱性蛋白酶酶解,然后用1mol/L HC1溶液调节pH为7,加入中性蛋白酶酶解,添加量为2500U/kg,结束后灭酶,离心,取上清液,用截留分子量为1000u的超滤膜超滤,得到分子量小于1000u的滤过液,得到大豆西洋参咖啡复合低聚液态。
S2.将大豆西洋参咖啡低聚肽混合,溶解于水中(物料与水的质量比为1:10),用1mol/L HC1溶液调节pH为6,加入a-糜凝乳酶和风味蛋白酶反应,所述a-糜凝乳酶的添加量为2000U/kg,所述风味蛋白酶的添加量为3000U/kg,结束后灭酶,离心,取上清液,得到复合肽浓缩液,加入结晶果糖和食品添加剂,溶解混合均匀,制得植物肽口服液。

将大豆西洋参咖啡肽混合,溶解于水中(物料与水的质量比为1:10),用1mol/LHC1溶液调节pH为6,加入a-糜凝乳酶和风味蛋白酶反应,所述a-糜凝乳酶的添加量为2000U/kg,所述风味蛋白酶的添加量为3000U/kg,结束后灭酶,离心,取上清液,加入结晶果糖和食品添加剂,溶解混合均匀,制得植物肽液口服液

1.一种大豆咖啡西洋参生物活性肽的制备方法,其特征在于:原料包括大豆、西洋参和咖啡,所述大豆、西洋参和咖啡的蛋白原料经胃蛋白酶和胰蛋白酶酶解制成,所述大豆、西洋参和咖啡肽的质量比为(1.3-3.1):(1.3-2.2):(1.8-4.1),所述大豆、西洋参和咖啡复合肽的质量比为(1.3-4.0):0.42。
2.根据权利要求1所述的一种大豆咖啡西洋参生物活性肽的制备方法,其特征在于,具体方法为:将大豆30-50份,西洋参5-15份,咖啡15-40份比例混合,首先制备分离碱性蛋白溶液,将大豆、西洋参、咖啡分离,碱性溶液加入胃蛋白酶、胰蛋白酶、木瓜蛋白酶、木薯淀粉酶,酶解后,灭酶。
3.根据权利要求1所述的一种大豆咖啡西洋参生物活性肽的制备方法,其特征在于:所述复合小分子离子肽液去除表面杂质,离心后所得上清液,即获得复合大豆西洋参咖啡小分子(离子)肽。
4.一种大豆咖啡西洋参生物活性肽的应用,其特征在于:按1%-5%的比列和纯净水调合,再加入调味剂、植物防腐剂、纯净水,得到原料,制成化妆品、食品或药品及保健品。
