Abstract
本发明涉及生物医药技术领域,具体涉及一种外周血造血干细胞来源致敏树突状细胞的制备方法及应用,其中促DC成熟期采用下述原料:TNF‑ɑ、IL‑6、PGE‑2、IL‑1β;本发明涉及的DC培养方法,能够大大缩短培养时间,获得高质量的DC细胞,适合与其同来源CTL细胞的培养周期,采用WT1阳性的肿瘤细胞外泌体负载DC,可满足体外扩增培养CTL细胞所需要的刺激且达到更好的抗肿瘤效果。

技术领域
本发明属于生物医药和细胞生物学领域,具体涉及一种外周血造血干细胞来源致敏树突状细胞的制备方法及应用。
背景技术
树突状细胞(Dendritic cell,DC)是目前已知的功能最强的抗原递呈细胞,是先天和过继免疫反应的桥梁,被证实与许多疑难疾病如慢性感染、肿瘤和自身免疫性疾病有关,它具有强大的T细胞激活能力,并能活化初始型T细胞、刺激淋巴细胞增殖成熟、诱导和维持细胞免疫应答,在启动和维持免疫应答的过程中起重要的作用。DC加工处理提呈抗原信息后,通过激活T淋巴细胞而发挥抗肿瘤细胞的免疫效应。CTL细胞(Cytotoxic T cell)是体内重要的免疫效应细胞,在MHC限制下直接、连续、特异性的杀伤靶细胞。CTL细胞有长期记忆功能及再次快速应答能力。对靶细胞的杀伤作用是抗原特异性的,只杀伤相应靶细胞而对其它细胞无损伤作用。肿瘤特异性CTL是由摄取加工肿瘤特异性抗原肽,协同等细胞因子的作用,体外激活淋巴细胞生成的,高效特异的CTL细胞关键在于抗原特异性提呈细胞。
在向T细胞提供抗原之前,DCs要经历两个阶段:未成熟阶段和成熟阶段。在未成熟阶段,树突状细胞具有较高的吞噬能力,但迁移和T细胞的诱导能力较低。当受到病原体或外来物的刺激时,未成熟的DCs会向成熟阶段转化,其特点是共刺激分子(CD40、CD80、CD86)的表达增加,促炎细胞因子的分泌增多,CD40、CD80、CD86的表达水平决定了DC。在生理条件下,入侵病原菌和病原菌组分(LPS等)作用于树突状细胞的Toll样受体家族,组织驻留的树突状细胞被激活,向成熟阶段转化。然而,在人工免疫中,通常只提供抗原,DCs处于半成熟状态。虽然半成熟的树突状细胞具有抗原,但同种刺激分子的低表达和有限的归巢能力极大地阻碍了T细胞的启动。因此,寻找能够促进DCs成熟和迁移的佐剂是调节免疫策略的关键。
WT1(Wilm’s tumor gene)基因在白血病及各种实体肿瘤包括肺癌和乳腺癌中高表达,且在这些恶性肿瘤中起癌基因作用,与相应肿瘤的增殖、分化、疾病的发生发展和预后有一定关系,提示WT1蛋白可以作为一个新的超表达的肿瘤抗原,用于CTL肿瘤特异性杀伤的致敏。WT1九聚肽体外树突状细胞诱导细胞毒性T淋巴细胞(CTL)产生WT1肽特异性CTL,这种CTL可以杀伤内源性表达WT1的白血病和实体瘤细胞。文献1(从外周血干细胞采集物诱导HLA-A*2402限制性WT1特异性CD8+CTL的体外实验研究,王志东等,中国科技论文在线,网络首发日期20091221)公开了:从外周血干细胞采集物黏附细胞短期培养获得成熟树突状细胞(DC),负载WT1抗原肽后,刺激非黏附细胞,采用WT1-五聚肽结合流式细胞术检测WT1特异性CTL及其免疫表型。经48小时培养获得成熟的DC2d,表达CD80、CD86、CD45RO;负载WT1肽的DC2d可诱导扩增WT1-CTL,表达CD57和CD45RO,为效应T细胞表型。短期培养的DC2d是有效的抗原递呈细胞外周血干细胞采集物可诱导扩增WT1-CTL,表型为效应记忆表型。虽然文献1培养DC的时间缩短至48h,但是其需要分离黏附细胞,仍然比较繁琐。
外泌体是特殊的膜外囊泡,其直径为30~150nm,密度为1.13~1.19g/ml,具有稳定的双分子磷脂结构,外泌体不仅可将蛋白质、RNA、DNA和脂质从一个细胞携带到另一个细胞,从而调节受体细胞的生物功能,还能保护其自身携带的DNA片段、miRNA、mRNA以及功能性蛋白等生物活性物质不被破坏。外泌体天然的材料运输特性、固有的长期循环能力和出色的生物相容性,使其具有很大的潜力成为药物递送载体,适用于递送各种化学物质、蛋白质、核酸和基因治疗剂。来自肿瘤细胞外泌体致敏的DCs能发挥抗肿瘤效应,其机制是肿瘤细胞外泌体将其携带的肿瘤抗原传递给DCs,DCs通过有效加工处理外泌体来源的肿瘤抗原激发抗肿瘤免疫应答。
传统DC培养方法时间太久,不适合与其同来源CTL细胞的培养周期,国外已有研究人员改善DC培养方法做过尝试,缩短培养时间改变培养条件,得到同样高质量的DC细胞。能更好地完善体外CTL细胞克隆体系及提升CTL细胞的肿瘤杀伤效应。专利CN105713874A(公开日期20160629)公开了一种抗肿瘤相关抗原WT1特异性CTL及其制备方法,采用具有定向靶向的纳米脂质体负载肿瘤相关性抗原WT1制备抗原特异性T淋巴细胞。采用72小时快速成熟树突状细胞,大大缩短肿瘤患者最佳治疗的时间窗口。然而本领域仍然存在进一步缩短DC培养时间的需求,如果能较好地解决DC培养这个关键问题,会为移植相关机制提供理论依据,同时为白血病的治疗开辟了一个新的方向。
发明内容
为了解决上述问题,本发明提供了一种外周血造血干细胞来源的致敏树突状细胞(DC)的体外快速制备法及其应用。
本发明涉及的DC培养方法,能够大大缩短培养时间,获得高质量的DC细胞,适合与其同来源CTL细胞的培养周期,采用WT1阳性的肿瘤细胞外泌体负载致敏DC,可满足体外扩增培养CTL细胞所需要的刺激且达到更好的抗肿瘤效果。与现有技术相比,本发明方法DC能够针对特定的高表达相应肿瘤抗原的细胞发挥更好的抗原呈递作用,发挥抗原特异性杀伤T细胞的抗肿瘤应答。
本发明通过下述技术方案实现的。
一种外周血造血干细胞来源致敏树突状细胞的制备方法,促DC成熟期采用下述原料:TNF-α、IL-6、PGE-2、IL-1β。
其中促DC成熟期采用下述浓度原料:TNF-α 5-20ng/ml、IL-6 800-1200U/ml、PGE-2 0.5-1.5μg/ml、IL-1β 5-20ng/ml。
其中促DC成熟期采用下述浓度原料:TNF-α 10ng/ml、IL-6 1000U/ml、PGE-2 1μg/ml、IL-1β 10ng/ml。
所述的一种外周血造血干细胞来源致敏树突状细胞的制备方法,其中制备方法包括:标本来源HLA-A*0201阳性的健康供者外周血造血干细胞采集物;
(1)外周血造血干细胞采集物单个核细胞(G-PBSC)分离;
(2)WT1阳性肿瘤细胞外泌体的制备;
(3)DC细胞分离与培养;
(4)促DC成熟;
即得致敏树突状细胞。
其中(1)外周血造血干细胞采集物单个核细胞(G-PBSC)分离步骤包括:
取三管离心管,分别加入Ficoll淋巴细胞分离液,每管分别上覆新鲜外周血造血干细胞采集物,离心;轻轻将白膜层转移到另外2支离心管中,PBS漂洗,离心,去除上清;重复上述步骤1次。
其中(2)WT1阳性肿瘤细胞外泌体的制备步骤包括:
a复苏WT基因高表达的慢性髓性白血病细胞系NB4细胞,置于胎牛血清的RPMI-1640培养基中,37℃,5%CO2培养至80-90%融合;
b取6只离心管,每管中用移液管加入留取的NB4细胞上清,4℃水平离心;
c取6只超速离心机专用离心管(以下简称超离管),缓慢吸取离心管中的上清中转移其中,弃去底部沉淀,每只离心管中含有液体,用PBS缓冲液配平(误差在0.01g以内),于超速离心机中4℃水平离心;
d另取6只超离管,缓慢吸取步骤b中的液体转移至新的超离管中,弃去底部沉淀,保持每只离心管中所含液体体积在,PBS缓冲液配平(误差在0.01g以内),于超速离心机中4℃水平离心;
e另取6只超离管,余下步骤与c相同,放入超速离心机后调整转速为100000g,2h;
f弃去上清,用PBS缓冲液重悬6只超离管内沉淀,配平后置于超速离心机中4℃水平离心;
g弃去上清,用移液枪取20μl外泌体悬滴加在铜网上,室温条件静置将铜网上多余液体用滤纸从侧面吸干,在铜网上滴加磷酸钨酸进行负染,将磷钨酸用滤纸吸干,室温静置在透射电镜下观察其形态;在WT1+NB4细胞的培养上清中能够提取到外泌体,观察NB4细胞外泌体在电镜下的形态,可见其呈类圆形或杯状,大小不均一,直径在30~150nm左右。
其中(3)DC细胞分离与培养包括步骤为:
RPMI-1640培养基+10%的灭活人AB血清重悬,混匀后移入在培养瓶中,5%的CO2,37℃培养箱中;吸出悬浮细胞,贴壁细胞中加入DC基础培养基含AIM-V培养液,加入人AB血清,IL-4,GM-CSF,37℃培养箱培养24小时。
其中促DC成熟步骤包括:
在步骤(3)基础培养基基础上加入TNF-α、IL-6、PGE-2、IL-1β,继续培养,采用WT1肿瘤细胞外泌体刺激,相同条件下孵育40min。即得致敏树突状细胞。
优选地,步骤1)采用G-CSF动员。
本发明一个方面提供了一种分离的DC细胞,其通过本发明上述方法制备。
本发明的有益效果:
1、本发明所涉及的DC细胞来源于经GM-CSF动员后外周血造血干细胞,细胞数量增多,细胞成分含干祖细胞。
2、大量分离纯化DC一直是制约细胞免疫的瓶颈。本发明涉及的DC培养方法,与现有传统DC培养方法相比,能够大大缩短DC的培养时间,适合与其同来源CTL细胞的培养周期,并获得高质量的DC细胞,且满足体外扩增培养CTL克隆细胞所需要的刺激。此外,在体外进行DC培养方法改进,采用HLA同型异体源DC,这在很大程度解决了CTL抗原高效提呈问题。
3、本发明采用WT1阳性的肿瘤细胞外泌体负载DC,可达到更好的抗肿瘤效果。

实施例1健康供者树突状细胞的体外培养
标本来源HLA-A*0201阳性的健康供者外周血造血干细胞采集物
(1)外周血造血干细胞采集物单个核细胞(G-PBSC)分离
取三管50ml离心管,分别加入30ml Ficoll淋巴细胞分离液,每管分别上覆20ml新鲜外周血造血干细胞采集物,1700rpm×25min离心。轻轻将白膜层转移到另外2支离心管中,10ml PBS漂洗,1500rpm×5min,去除上清。重复上述步骤1次。
(2)WT1阳性肿瘤细胞外泌体的制备:
a复苏WT基因高表达的慢性髓性白血病细胞系NB4细胞,置于含10%胎牛血清的RPMI-1640培养基中,37℃,5%CO2培养至80-90%融合。
b取6只干净的15mL离心管,每管中用移液管加入13mL留取的NB4细胞上清,4℃水平离心300g×10min。
c取6只超速离心机专用离心管(以下简称超离管),缓慢吸取15mL离心管中的上清中转移其中,弃去底部沉淀,每只离心管中含有大约10mL的液体,用PBS缓冲液配平(误差在0.01g以内),于超速离心机中4℃水平离心2000g×10min。
d另取6只超离管,缓慢吸取步骤b中的液体转移至新的超离管中,弃去底部沉淀,保持每只离心管中所含液体体积在10mL左右,PBS缓冲液配平(误差在0.01g以内),于超速离心机中4℃水平离心10000g×30min。
e另取6只超离管,余下步骤与c相同,放入超速离心机后调整转速为100000g×2h。
f弃去上清,用PBS缓冲液重悬6只超离管内沉淀,配平后置于超速离心机中4℃水平离心100000g×1h。
g弃去上清,用移液枪取20μl外泌体悬滴加在铜网上,室温条件静置1min后将铜网上多余液体用滤纸从侧面吸干,在铜网上滴加2%的磷酸钨酸30μl进行负染,1min后将磷钨酸用滤纸吸干,室温静置10min后在透射电镜下观察其形态。在WT1+NB4细胞的培养上清中能够提取到外泌体,观察NB4细胞外泌体在电镜下的形态,可见其呈类圆形或杯状,大小不均一,直径在30~150nm左右。外泌体光镜图,见图1
h采用Western blot检测NB4细胞外泌体蛋白组分,图2为采用Western blotting检测NB4细胞来源的外泌体中的WT1蛋白组分,证实外泌体中有WT1蛋白存在。
(3)DC细胞分离与培养
RPMI-1640培养基+10%的灭活人AB血清重悬,混匀后移入在75cm2培养瓶中,5%的CO2,37℃培养箱中2-4小时。吸出悬浮细胞,贴壁细胞中加入DC基础培养基含AIM-V培养液15ml,10%的人AB血清,IL-4100ng/ml,GM-CSF 80ng/ml,37℃培养箱培养24小时。此时取出部分DC细胞进行表面标记检测
(4)促DC成熟
在上述基础培养基基础上加入TNF-α 10ng/ml、IL-6 1000U/ml、PGE-2 1μg/ml、IL-1 β 10ng/ml,继续培养24小时后,采用WT1肿瘤细胞外泌体刺激,相同条件下孵育40min。
(5)DC表型鉴定
吸取所培养的DC细胞液6ml,1200rpm,离5min心后弃掉上清,PBS再次清洗后,每个检测样品设三个复管。分别加入标记荧光单抗各5ul:CD1a-PE+CD80-FITC+CD86-PECy5,CD83-FITC+HLA-DR-PECy5+CD11c-PE,对照管加入各同型对照,避光、4℃孵育30min。PBS洗2次,1300rpm,离心5min,弃上清,500μl PBS重悬细胞。调整电压及补偿,以DC细胞群圈门,检测各CD分子的表达率。
实施例2本发明方法培养体系的建立
(1)不同基础培养基浓度优化对DC细胞收获率的影响
基础培养基组1:AIM-V培养液,10%的人AB血清,IL-4 50ng/ml,GM-CSF 40ng/ml
基础培养基组2:AIM-V培养液,10%的人AB血清,IL-4 100ng/ml,GM-CSF 80ng/ml
基础培养基组3:AIM-V培养液,10%的人AB血清,IL-4 150ng/ml,GM-CSF 120ng/ml
促成熟培养基:TNF-a(10ng/ml)、IL-1β(10ng/ml)、IL-6(1000U/ml)、PEG(1μg/ml)
实验分组:①实验组1:基础培养基组1+促成熟培养基
②实验组2:基础培养基组2+促成熟培养基
③实验组3:基础培养基组3+促成熟培养基
使用15mlRPMI-1640培养基(添加10%的灭活人AB血清),将2.5×107个G-PBSC重悬,混匀后移入在75cm2培养瓶中,5%的CO2,37℃培养箱中2-4小时。吸出悬浮细胞,贴壁细胞中加入DC基础培养基含AIM-V培养液15ml,按上述实验组的不同分组加入上述不同浓度的IL-4和GM-CSF,37℃培养箱培养24小时。促DC成熟,在上述基础培养基基础上加入10ng/ml TNF-α、1000U/ml IL-6、1μg/ml PGE-2、10ng/ml IL-1β,继续培养24小时。结果见表1

根据表1结果,选择100ng/mlIL-4,80ng/mlGM-CSF,作为基础培养基里两种细胞因子的添加浓度。
(2)不同基础培养基诱导时间对DC细胞收获率的影响
实验分组:①培养时间组1:基础培养基组培养24小时+诱导培养基
②培养时间组2:基础培养基组培养48小时+诱导培养基
③培养时间组3:基础培养基组培养72小时+诱导培养基
使用15mlRPMI-1640培养基(添加10%的灭活人AB血清),将2.5×107个G-PBSC重悬,混匀后移入在75cm2培养瓶中,5%的CO2,37℃培养箱中2-4小时。吸出悬浮细胞,贴壁细胞中加入DC基础培养基含AIM-V培养液15ml,10%的人AB血清,100ng/ml IL-4,80ng/mlGM-CSF,各组分别于37℃培养箱培养24、48、72小时,72小时培养组在第48小时后半量换液,补加上述相应浓度IL-4和GM-CSF。促DC成熟,在上述基础培养基基础上加入10ng/ml TNF-α、1000U/ml IL-6、1μ g/ml PGE-2、10ng/ml IL-1β,继续培养24小时。流式细胞仪检测DC细胞得率结果见表2

(3)WT1+肿瘤细胞外泌体负载,对DC促成熟和得率的影响
实验分组:①非外泌体组:不添加外泌体刺激
②外泌体组1:WT1+外泌体5μg/ml
③外泌体组2:WT1+外泌体10μg/ml
④外泌体组3:WT1+外泌体15μg/ml
使用15mlRPMI-1640培养基(添加10%的灭活人AB血清),将2.5×107个G-PBSC重悬,混匀后移入在75cm2培养瓶中,5%的CO2,37℃培养箱中2-4小时。吸出悬浮细胞,贴壁细胞中加入DC基础培养基含AIM-V培养液15ml,10%的人AB血清,100ng/ml IL-4,80ng/mlGM-CSF,37℃培养箱培养24小时。促DC成熟,在上述基础培养基基础上加入10ng/ml TNF-α、1000U/ml IL-6、1μg/ml PGE-2、10ng/ml IL-1β,继续培养24小时。而后WT1+外泌体负载组采用不同浓度WT1肿瘤细胞外泌体刺激,相同条件下孵育40min。非外泌体组则不加任何肿瘤细胞外泌体负载。结果见表3和图3

表3结果显示,外泌体组刺激能够显著增加DC细胞的得率。
图3结果显示,外泌体刺激组与非外泌体刺激组相比,能够显著增加DC细胞CD80、CD83、CD86的表达率,且10μg/ml的外泌体刺激,即可达到明显的促进DC细胞成熟的效果,且细胞得率较好,所以选择10μg/ml的WT+细胞外泌体作为刺激浓度。
实施例3现有技术相比本发明涉及的DC制备方法的效果比较
现有技术DC细胞培养法采用试剂和仪器:1640培养基、FBS、100mm平皿、6孔板、吸管、细胞因子(GM-CSF、IL-4)。
方法具体步骤包括:(1)从液氮灌里复苏一支外周血分离单个核(PBMC)或者是新鲜分离的PBMC。(2)加10ml 1640培养基培养在100mm平皿中,培养箱中放置4小时。(3)吸走悬浮细胞(PBLs)用DMSO冻存,贴壁细胞用PBS洗涤三次。(4)再用PBS将贴壁细胞吹下来,离心后用台盼蓝进行染色计数。(5)再将细胞培养于6孔板中,同时加细胞因子40ng/mlGM-CSF和50ng/ml IL-4(6)每隔一天半量换液,同时补加细胞因子。第6天后加入细胞因子10ng/mlTNF-a(、10ng/mlIL-1β、1000U/mlIL-6、1μg/mlPEG继续培养到第7天,能明显看到细胞表面树突状增多,细胞表面积增大即为成熟的DC。
本发明DC与现有技术相比,本发明在DC基础培养步骤上增加了GM-CSF和IL-4的浓度,培养时间在原有促DC成熟步骤中增加了应用WT1+肿瘤细胞外泌体负载DC,结果显示与现有技术相比,增加了成熟DC表面CD80,CD86,CD40等标志的表达率。这种方法制备的DC成熟速度核归巢能力增加,激活T细胞的能力增强。
①本发明方法和现有技术DC细胞的形态观察
图4为倒置显微镜下观察本发明方法本发明培养DC的形态(×400),图4b为倒置显微镜下观察现有技术方法培养DC的形态(×400)。结果显示,倒置显微镜下观察两种培养方法DC细胞的形态无显著差异。
②本发明方法和现有技术培养DC细胞的表型鉴定
12人份健康供者外周血来源的DC细胞,分别采用本发明DC培养法和现有技术DC培养法,利用流式技术检测了两种方法成熟与未成熟DC表面标志的变化,
未成熟时,图5显示现有技术培养DC的CD86(11.2±2.78)及HLA-DR(7.2±1.77)要高于2天培养DC86(6.2±1.80),HLA-DR(3.30±0.8)的表达率(P<0.05)。
成熟后,图6显示和现有技术相比,本发明方法培养的DC表面标志CD40、CD80、CD86的阳性率显著升高(*P<0.01,**P<0.05)。
③两种培养法DC提呈功能的比较
比较两种培养DC对Tc细胞毒的影响,本发明方法成熟的DC与现有技术方法培养的成熟的DC,分别与的Tc效应细胞以1∶5的比例混合培养3天,激发抗原特异性Tc细胞的抗肿瘤应答。
采用CSFE标记靶细胞,调整细胞细胞浓度为1×106/ml,37℃,5%CO2条件下,终浓度为2.0μmol/L的CFSE标记10分钟。PBS清洗3次,去除残余的染料,计数,并重新悬浮于完全培养基中;效靶细胞共孵育。
Tc和DC混合细胞作为效应细胞,靶细胞选择倍增期的高表达WT1基因的人急性早幼粒细胞白血病细胞NB4细胞,效靶比为5∶1,混合效靶细胞后,放入U型96孔板中,反应体系为200μl,设置3个平行复孔,分别设对照孔(靶细胞及效应细胞)每孔加入6000个靶细胞,37℃,5%CO2孵育4小时;PI标记,收集培养的效靶细胞,PBS清洗1次,1300rpm,离心5min,500μl PBS重悬,加入25μl PI(100μg/ml)加入到U型孔中,避光、置于冰浴5分钟。
通过流式细胞技术检测靶细胞的死亡率,以此来判断成熟DC的提呈效能。6全部细胞群圈门,再以CFSE阳性的细胞群(靶细胞)第二次设门,观察PI及CFSE双阳性的细胞数(即被杀伤的靶细胞数),此细胞群位于右上象限。存活的效应细胞为左下向限(PI及CFSE双阴性细胞),死亡的效应细胞位于左上向限(被PI染色的效应细胞),右下向限为未被杀伤的靶细胞(CFSE阳性,PI阴性)。
图7结果显示本发明方法培养DC组的杀伤率85.7±6.4%,好于现有技术方法培养DC组的杀伤率为70.3±5.8%,P<0.05%。表明本发明方法培养的DC在功能上好于现有技术培养的DC。

Claims (8)
1.一种外周血造血干细胞来源致敏树突状细胞的制备方法,其特征在于:促DC成熟期采用下述原料:TNF-ɑ、IL-6、PGE-2、IL-1β。
2.根据权利要求1所述的一种外周血造血干细胞来源致敏树突状细胞的制备方法,其中促DC成熟期采用下述浓度原料:TNF-ɑ5-20ng/ml、IL-6 800-1200U/ml、PGE-2 0.5-1.5μg/ml、IL-1β5-20ng/ml。
3.根据权利要求1所述的一种外周血造血干细胞来源致敏树突状细胞的制备方法,其中促DC成熟期采用下述浓度原料:TNF-ɑ10ng/ml、IL-6 1000U/ml、PGE-2 1μg/ml、IL-1β10ng/ml。
4.根据权利要求1-4任一项所述的一种外周血造血干细胞来源致敏树突状细胞的制备方法,其中制备方法包括:标本来源HLA-A*0201阳性的健康供者外周血造血干细胞采集物;
(1)外周血造血干细胞采集物单个核细胞(G-PBSC)分离;
(2)WT1阳性肿瘤细胞外泌体的制备;
(3)DC细胞分离与培养;
(4)促DC成熟;
即得致敏树突状细胞。
5.根据权利要求4所述的一种外周血造血干细胞来源致敏树突状细胞的制备方法,其中(1)外周血造血干细胞采集物单个核细胞(G-PBSC)分离步骤包括:
取三管离心管,分别加入Ficoll淋巴细胞分离液,每管分别上覆新鲜外周血造血干细胞采集物,离心;轻轻将白膜层转移到另外离心管中,PBS漂洗,离心,去除上清;重复上述步骤1次。
6.根据权利要求4所述的一种外周血造血干细胞来源致敏树突状细胞的制备方法,其中(2)WT1阳性肿瘤细胞外泌体的制备步骤包括:
a复苏WT基因高表达的慢性髓性白血病细胞系NB4细胞,置于胎牛血清的RPMI-1640培养基中,37℃,5%CO2培养至80-90%融合;
b取离心管,每管中用移液管加入留取的NB4细胞上清,4℃水平离心;
c取超速离心机专用离心管(以下简称超离管),缓慢吸取离心管中的上清中转移其中,弃去底部沉淀,每只离心管中含有液体,用PBS缓冲液配平,于超速离心机中4℃水平离心;
d另取超离管,缓慢吸取步骤b中的液体转移至新的超离管中,弃去底部沉淀,保持每只离心管中所含液体体积在,PBS缓冲液配平,于超速离心机中4℃水平离心;
e另取超离管,余下步骤与c相同,放入超速离心机后调整转速为100000g,2h;
f弃去上清,用PBS缓冲液重悬超离管内沉淀,配平后置于超速离心机中4℃水平离心;
g弃去上清,用移液枪取外泌体悬滴加在铜网上,室温条件静置将铜网上多余液体用滤纸从侧面吸干,在铜网上滴加磷酸钨酸进行负染,将磷钨酸用滤纸吸干,室温静置在透射电镜下观察其形态;在WT1+NB4细胞的培养上清中能够提取到外泌体,观察NB4细胞外泌体在电镜下的形态,可见其呈类圆形或杯状,大小不均一,直径在30~150nm。
7.根据权利要求4所述的一种外周血造血干细胞来源致敏树突状细胞的制备方法,其中(3)DC细胞分离与培养包括步骤为:
RPMI-1640培养基+10%的灭活人AB血清重悬,混匀后移入在培养瓶中,5%的CO2,37℃培养箱中;吸出悬浮细胞,贴壁细胞中加入DC基础培养基含AIM-V培养液,加入人AB血清,IL-4,GM-CSF,37℃培养箱培养24小时。
8.根据权利要求4所述的一种外周血造血干细胞来源致敏树突状细胞的制备方法,其中促DC成熟步骤包括:
在步骤(3)基础培养基基础上加入TNF-ɑ、IL-6、PGE-2、IL-1β,继续培养,采用WT1肿瘤细胞外泌体刺激,相同条件下孵育40min;即得致敏树突状细胞。
