Abstract
本发明公开了一种预防仔猪腹泻中药精提物饲料添加剂及其制备方法与应用,该添加剂包含以下重量份原料:诃子20~25份、单宁酸5~8份、石榴皮25~32份、黄岑30~38份、党参4~8份、大蒜1~2份、菊花1~15份、菠萝皮1~1.5份、槟榔壳0.5~1.0份、益生菌2~4份、香茶菜根粉0.1~0.5份以及麸皮9~15份。本发明制备得到的饲料添加剂长期使用无毒副作用,采用中草药材的精提物作为添加成分,科学搭配营养原料,充分发挥中草药材的药理作用,可在饲养猪的全过程中进行喂养,能有效预防猪腹泻疾病的发生,药效持续时间长,而且可以提高仔猪的抵抗力,降低猪的发病率,进一步提高了仔猪的生长速率。

本发明属于生产养殖技术领域,具体涉及到一种预防仔猪腹泻中药精提物饲料添加剂及其制备方法与应用。
背景技术
猪腹泻在临床上是一种常见病,发病原因较复杂。有的是因为饲养管理不善,的是因为细菌或病毒感染引起。冬季及初春时流行的猪腹泻,症状与猪流行性腹泻和猪传染性胃肠炎非常相似,对于养殖户来说,如果猪感染了这种病,短期内整个猪场都会发病,长达1-2个月才慢慢平息,会造成很大的经济损失。造成仔猪腹泻的原因大体上可分为非病原性因素以及病原性因素两种,其中非病原性因素包括营养和饲养环境的应激,如潮湿、温度变化、断奶应激、季节温差等。
具体而言,造成猪群腹泻的原因通常为:(1)细菌性腹泻:大肠杆菌、魏氏梭菌性肠炎、猪痢疾杆菌、内劳森氏菌;(2)病毒性腹泻:传染性胃肠炎、流行性腹泻、轮状病毒、猪伪狂犬病毒;(3)寄生虫性腹泻:蛔虫、球虫、鞭虫、结肠小袋纤毛虫;(4)非感染因素:温度低血糖(饥饿性腹泻)、日粮抗原过敏性腹泻(大豆过敏)、非病原性白痢(缺铁性腹泻,酶和胃酸不足性白痢)。
猪腹泻的症状一般表现为仔猪突然发生呕吐和急剧的水样腹泻,由于失水快,来不及治疗就死亡,猪越小死亡率越高。架子猪和肥猪多为急性水泻,呈喷射状,粪便为灰白色。猪口渴,不断饮水,少数会呕吐。由于严重失水,猪体重迅速下降。
目前,预防猪腹泻的药物繁多,但是存在副作用大,预防效果不佳等缺陷。
发明内容
本发明的目的是提供一种预防仔猪腹泻中药精提物饲料添加剂及其制备方法与应用,可以在饲养猪的全过程中进行饲养,可预防仔猪腹泻疾病的发生。
为达上述目的,本发明提供了一种预防仔猪腹泻中药精提物饲料添加剂,包括以下重量份原料:诃子20~25份、单宁酸5~8份、石榴皮25~32份、黄岑30~38份、党参4~8份、大蒜1~2份、菊花1~15份、菠萝皮1~1.5份、槟榔壳0.5~1.0份、益生菌2~4份、香茶菜根粉0.1~0.5份以及麸皮9~15份。
进一步地,包括以下重量份原料:诃子25份、单宁酸6份、石榴皮30份、黄岑35份、党参6份、大蒜1份、菊花10份、菠萝皮1.5份、槟榔壳0.5份、益生菌3份、香茶菜根粉0.5份以及麸皮15份。
进一步地,益生菌为酪酸梭菌、乳酸菌、双歧杆菌以及嗜酸乳杆菌中的至少一种。

进一步地,香茶菜根粉通过以下方法制得:将香茶菜的根依次经过清洗、晾晒、烘干、研磨成粉状后制得,所述香茶菜根粉的粒径大小为6.5~13μm。
一种预防仔猪腹泻中药精提物饲料添加剂的制备方法,包括以下步骤:
(1)按照比例称取原料,将香茶菜根粉、大蒜、党参、黄岑、槟榔壳、以及麸皮粉碎至1000-2000目,将石榴皮、诃子、菊花以及菠萝皮通过冷凝回流提取法,制得混合提取液;
(2)将粉碎后的上述原料于100~180℃条件下熟化20~30min,再于水中浸泡;
(3)将浸泡后的原料煮沸后文火煮制15-20min,滤取药汁后取滤渣,于滤渣中加水煎煮,重复滤取药汁和于药渣中加水煎煮1-2次后,将滤取的药汁混合均匀;
(4)将混合提取液以及滤液合并后,加入单宁酸以及益生菌,混匀即得。
进一步地,冷凝回流提取法包括以下步骤:将石榴皮、诃子、菊花以及菠萝皮以体积分数为80%的乙醇作为提取溶剂,料液比为1:20~30g/mL的比例,提取30~60min,提取次数为1-3次。
进一步地,步骤(4)混匀后的材料为汤剂、混悬剂、粉剂或颗粒剂。

综上所述,本发明具有以下优点:
1、菊花中含有丰富的绿原酸,菠萝皮为菠萝的果皮含有丰富的果胶,两者复配可以提高肌肉拉力和改善脂肪风味;且菠萝皮含有丰富的挥发油,富有香味,可促进肥育猪进食,又可改善猪肉风味;通过益生菌的添加,可以增强猪的肠道有益菌繁殖,有助于猪充分消化吸收饲料中的养分,其中凝结芽孢杆菌可产生大量凝固素和L-乳酸等抑制有害菌的物质,对大肠杆菌、变形杆菌以及小球菌等都有抑制作用;
2、大蒜为香辛料,既可以驱寒温胃,促进肥育猪进食,还可以杀菌消毒,防止肥育猪腹泻,另外还可以起到改善猪肉风味的作用;单宁酸对多种细菌和真菌都有明显的抑制作用,与大蒜配合使用,可以起到抗病毒以及抗寄生虫的作用;同时香茶菜根粉含有多种天然药物成分,具有一定的杀菌能力,营养价值极高,与党参等连用,可以消除自身的寒性,与黄岑配合使用还可以提高配伍作用,增加杀菌效果;槟榔壳具有杀虫的作用,可以治疗绦虫、钩虫、蛔虫等寄生虫感染;
3、本发明的诃子提取物对痢疾杆菌、大肠杆菌等多种细菌都有明显的抑制作用,石榴皮对痢疾杆菌等多种细菌有明显的杀灭抑制作用,与诃子、黄岑合用,抗菌谱广,杀菌力强;
4、本发明针对仔猪阶段配料合理、营养均衡,既不充了仔猪在生长过程中所需的营养元素,提高了仔猪的生长速度,通过在添加剂中添加预防仔猪腹泻及预防仔猪肺部等感染的香茶菜根粉,可以相应提高仔猪的抵抗力以及免疫力,降低猪仔生长过程中的发病概率,提高猪肉品质。

(1)按照比例称取原料,将香茶菜根粉、大蒜、党参、黄岑、槟榔壳以及麸皮粉碎至1500目,将石榴皮、诃子、菊花以及菠萝皮通过冷凝回流提取法,制得混合提取液;其中冷凝回流包括:将石榴皮、诃子、菊花以及菠萝皮以体积分数为80%的乙醇作为提取溶剂,料液比为1:30g/mL的比例,提取60min,提取次数为2次。
(2)将粉碎后的上述原料于150℃条件下熟化30min,再于水中浸泡;
(3)将浸泡后的原料煮沸后文火煮制20min,滤取药汁后取滤渣,于滤渣中加水煎煮,重复滤取药汁和于药渣中加水煎煮2次后,将滤取的药汁混合均匀;

(1)按照比例称取原料,将香茶菜根粉、大蒜、苦参、黄岑、槟榔壳以及麸皮粉碎至2000目,将五倍子、三颗针、菊花以及菠萝皮通过冷凝回流提取法,制得混合提取液;其中冷凝回流包括:将石榴皮、诃子、菊花以及菠萝皮以体积分数为80%的乙醇作为提取溶剂,料液比为1:30g/mL的比例,提取60min,提取次数为3次。

对比例3
本对比例提供了一种预防仔猪腹泻中药精提物饲料添加剂,包括以下重量份原料:
诃子25份、单宁酸6份、石榴皮30份、黄岑35份、大蒜1份、菊花10份、菠萝皮1.5份、香茶菜根粉0.5份以及麸皮15份。
还提供了一种预防仔猪腹泻中药精提物饲料添加剂的制备方法,包括以下步骤:
(1)按照比例称取原料,将香茶菜根粉、大蒜、黄岑以及麸皮粉碎至2000目,将石榴皮、诃子、菊花以及菠萝皮通过冷凝回流提取法,制得混合提取液;其中冷凝回流包括:将石榴皮、诃子、菊花以及菠萝皮以体积分数为80%的乙醇作为提取溶剂,料液比为1:30g/mL的比例,提取60min,提取次数为3次。
(2)将粉碎后的上述原料于180℃条件下熟化30min,再于水中浸泡;
(3)将浸泡后的原料煮沸后文火煮制20min,滤取药汁后取滤渣,于滤渣中加水煎煮,重复滤取药汁和于药渣中加水煎煮2次后,将滤取的药汁混合均匀;
(4)将混合提取液以及滤液合并后,加入单宁酸,混匀制备成颗粒即得。
对比例4
本对比例提供了一种预防仔猪腹泻中药精提物饲料添加剂,包括以下重量份原料:
诃子25份、单宁酸6份、石榴皮30份、黄岑35份、党参6份、大蒜1份、菊花10份、菠萝皮1.5份、槟榔壳0.5份、凝结芽孢杆菌3份、香茶菜根粉0.5份以及麸皮15份。
还提供了一种预防仔猪腹泻中药精提物饲料添加剂的制备方法,包括以下步骤:
(1)按照比例称取原料,将除丹宁酸以及凝结芽孢杆菌以外的原料粉碎至2000目;
(2)将粉碎后的上述原料于180℃条件下熟化30min,再于水中浸泡;
(3)将浸泡后的原料煮沸后文火煮制20min,滤取药汁后取滤渣,于滤渣中加水煎煮,重复滤取药汁和于药渣中加水煎煮2次后,将滤取的药汁混合均匀;
(4)将滤液合并后,加入单宁酸以及凝结芽孢杆菌,混匀制备成颗粒即得。
试验例1
将实施例1以及对比例1-4制备得到的颗粒制备成药液,作用于大肠杆菌、沙门氏菌、金黄色葡萄球菌以及枯草芽孢杆菌(中国微生物菌种保藏中心),实验菌种的浓度为5×106~10×106CFU/mL-1,测试其抑菌效果,以抑菌圈(mm)大小为依据。
1.1细菌培养基
牛肉膏蛋白胨培养基:牛肉膏3g,蛋白胨l0g,氯化钠5g,琼脂粉20g,加入1L蒸馏水,加热煮沸,直到完全溶解,用1%氢氧化钠和50%柠檬酸钠调节其pH至7.4-7.6,121℃高压灭菌15min后备用。
1.2菌种活化
菌种保藏中心提供的菌种为冻干粉,复苏前应准备各适于该菌种生长的培养基和培养设备,所用的操作均应在无菌条件下进行。启用菌种前用75%的酒精棉消毒菌种管外壁,在酒精灯外焰上旋转灼烧,烧极热后,点凉的无菌蒸馏水,安瓶瓶裂开。用移液枪注入500μL的该菌种液体培养基并反复吹打,使安瓶瓶中冻干粉溶解,将菌液全部吸出,接种到固体培养基中,细菌放置在37℃的培养箱中,培养18-24小时。复苏后菌种传1-2代后使用。
1.3菌悬液的选择
操作皆在超净工作台中进行,从已活化的菌种培养基中用无菌接种环挑取少量菌接入相应菌种的无菌液体培养基中,使用恒温摇床培养细菌37℃下培养18-24h制成一定浓度的菌悬液。菌悬液稀释方法参照微生物学实验。
1.4抑菌圈的测定
本实验选用滤纸片法测量对供试菌种抑菌圈直径的大小,并用其来判断抑菌活性强弱。操作方法为:用打孔器在定性滤纸上打出圆形滤纸片(直径=6mm),装入干燥洁净的培养皿中于121℃高压灭菌20min,备用。按照无菌操作方法,制备好对应菌种的相应平板待用。用无菌涂布棒将无菌移液枪吸取的200μL菌悬液在培养局涂布均匀微干后,夹取滴加5μL的添加剂滤纸片贴于平板,每个平板贴三片呈正三角形放置,每组三个平行对照,并放置于恒温恒湿培养箱中细菌在37℃下培养18-24h,取三个平行对照测量抑菌圈直径大小并计算其平均值。
实验结果如表1所示:

由表1可以看出:实施例1制备得到的添加剂的抑菌圈均大于对比例1-4制备的添加剂的抑菌圈,说明本发明制备的添加剂具有较高的敏感抑菌作用。
试验例2
预防猪腹泻试验
2.1动物与分组及给药
在金瑞克动物药业下的猪养殖场,选取同种同龄的仔猪共200只,随机分为5组,每组40头,分别为空白组、实施例1组、对比例1组、对比例2组以及对比例3组,空白组常规喂养,不添加任何添加剂;
实施例1组在常规喂养的同时,添加实施例1制备得到的添加剂;
对比例1-3组在常规喂养的同时,添加对比例1-3相应的得到的添加剂,添加量与实施例1组相同;
所有组均每天给药一次,喂养的时间为3个月,考察各组仔猪腹泻发生的概率。
2.2预防猪腹泻试验的结果见表2:

由表2可知:相对于空白组,其他四个组的仔猪腹泻率显著降低;与对比例1-3组比较,本发明制备得到的添加剂对于仔猪腹泻有更好的抑制效果,还可以促消化、减少肠道疾病的发生,并且安全可靠,是绿色无污染的饲料添加配方,推广性强。
虽然对本发明的具体实施方式进行了详细地描述,但不应理解为对本专利的保护范围的限定。在权利要求书所描述的范围内,本领域技术人员不经创造性劳动即可作出的各种修改和变形仍属本专利的保护范围。
Claims (9)
1.一种预防仔猪腹泻中药精提物饲料添加剂,其特征在于,包括以下重量份原料:诃子20~25份、单宁酸5~8份、石榴皮25~32份、黄岑30~38份、党参4~8份、大蒜1~2份、菊花1~15份、菠萝皮1~1.5份、槟榔壳0.5~1.0份、益生菌2~4份、香茶菜根粉0.1~0.5份以及麸皮9~15份。
2.如权利要求1所述的预防仔猪腹泻中药精提物饲料添加剂,其特征在于,包括以下重量份原料:诃子25份、单宁酸6份、石榴皮30份、黄岑35份、党参6份、大蒜1份、菊花10份、菠萝皮1.5份、槟榔壳0.5份、益生菌3份、香茶菜根粉0.5份以及麸皮15份。
3.如权利要求1或2所述的预防仔猪腹泻中药精提物饲料添加剂,其特征在于,所述益生菌为酪酸梭菌、乳酸菌、双歧杆菌以及嗜酸乳杆菌中的至少一种。
4.如权利要求1或2所述的预防仔猪腹泻中药精提物饲料添加剂,其特征在于,所述香茶菜根粉通过以下方法制得:将香茶菜的根依次经过清洗、晾晒、烘干、研磨成粉状后制得,所述香茶菜根粉的粒径大小为6.5~13μm。
5.如权利要求1-4任一项所述的预防仔猪腹泻中药精提物饲料添加剂的制备方法,其特征在于,包括以下步骤:
(1)按照比例称取原料,将香茶菜根粉、大蒜、党参、黄岑、槟榔壳、以及麸皮粉碎至1000-2000目,将石榴皮、诃子、菊花以及菠萝皮通过冷凝回流提取法,制得混合提取液;
(2)将粉碎后的上述原料于100~180℃条件下熟化20~30min,再于水中浸泡;
(3)将浸泡后的原料煮沸后文火煮制15-20min,滤取药汁后取滤渣,于滤渣中加水煎煮,重复滤取药汁和于药渣中加水煎煮1-2次后,将滤取的药汁混合均匀;
(4)将混合提取液以及滤液合并后,加入单宁酸以及益生菌,混匀即得。
6.如权利要求5所述的预防仔猪腹泻中药精提物饲料添加剂的制备方法,其特征在于,所述冷凝回流提取法包括以下步骤:将石榴皮、诃子、菊花以及菠萝皮以体积分数为80%的乙醇作为提取溶剂,料液比为1:20~30g/mL的比例,提取30~60min,提取次数为1-3次。
7.如权利要求5所述的预防仔猪腹泻中药精提物饲料添加剂的制备方法,其特征在于,步骤(4)所述混匀后的材料为汤剂、混悬剂、粉剂或颗粒剂。
8.如权利要求1-4任一项所述的预防仔猪腹泻中药精提物饲料添加剂在制备猪饲料中的应用。
9.如权利要求8所述的应用,其特征在于,所述预防仔猪腹泻中药精提物饲料添加剂在猪饲料中的添加量为1~3kg/吨。
