Abstract
The invention relates to a traditional Chinese medicine formula of a three-rice beverage for reducing uric acid and relieving gout and a preparation process thereof, in particular to a traditional Chinese medicine formula of a three-rice beverage prepared from 8 medicinal materials of pearl barley, chicory, sophora flower bud, lophatherum gracile (also known as pheasant), kudzuvine root, hovenia dulcis thumb, heartleaf houttuynia herb and mulberry leaf according to a certain dosage proportion; according to the characteristics of the nature and taste of the traditional Chinese medicinal materials, the formula is divided into 2 groups, one group is coix seed, sophora flower bud, lophatherum gracile and hovenia dulcis thunb, the other group is houttuynia cordata, chicory, kudzu-vine root and mulberry leaf, the obtained 2 groups of medicinal material extracting solutions are respectively concentrated to proper times, then are combined, and are added with proper pH regulator, preservative and corrective, so that the three-rice drink is obtained, and has good effects of reducing uric acid and relieving gout.
Description
Technical Field
The invention belongs to the field of functional drinks prepared from medicinal and edible traditional Chinese medicines, and relates to a three-rice-drink traditional Chinese medicine formula composition for reducing uric acid and relieving gout and a preparation process thereof.
Background
Hyperuricemia is a disease in which uric acid production is increased and/or excretion is decreased due to purine metabolic disorder, and uric acid level in blood is increased. Gout is a disease that causes urate to be deposited in tissues based on hyperuricemia. The pathogenesis of hyperuricemia and gout is related to genetic factors, environmental factors, glycogen storage disease, renal insufficiency, blood diseases, medicines and the like. Drugs for regulating uric acid metabolism clinically comprise allopurinol, benzbromarone, febuxostat and the like, and drugs for treating acute gouty arthritis comprise colchicine, nonsteroidal anti-inflammatory drugs, glucocorticoid and the like, but the drugs have side effects such as headache, rash, gastrointestinal and cardiovascular adverse reactions and the like, wherein benzbromarone has definite contraindications of patients with renal calculus, and the clinical application of benzbromarone is limited. Chinese traditional medicine has definite curative effect on hyperuricemia and gout, and many scholars have carried out deep research on active ingredients of Chinese traditional medicine, gout resisting action mechanism and the like, and show good prospect. On the aspect of traditional Chinese medicine, the incidence of hyperuricemia and gout is closely related to obesity, excessive eating, fatness, greasiness, spleen deficiency for a long time, water fluid loss, dampness turbidity generation, retention in the body, generation of damp-heat by stagnation, and the traditional Chinese medicine compound with the effects of eliminating dampness, tonifying spleen, clearing heat, promoting urination, purging turbidity and treating stranguria plays a significant role in the prevention and treatment of hyperuricemia and gout. However, the process of decocting the Chinese medicinal decoction is more complicated, the taste needs to be improved, and the popularization and application of the Chinese medicaments are limited to a certain extent. The invention provides a traditional Chinese medicine formula composition of a beverage for reducing uric acid and relieving gout and a preparation process thereof, the prepared three-rice beverage has definite curative effect, and can provide a new choice for treating hyperuricemia and gout.
The Coicis semen is a Gramineae Coicis semen herbaceous plant, is rich in high-quality protein, carbohydrate, fat, mineral elements and vitamins, and also is rich in polysaccharide, fatty acid and its ester compounds, flavonoids, triterpenes, etc., and has effects of lowering blood sugar, resisting tumor, scavenging free radicals, etc. Chicory, a perennial herb of the cichorium of the compositae, is a plant used as both medicine and food, contains inulin and aromatic substances in the root, and has the effects of clearing away heat and toxic materials, inducing diuresis to reduce edema, invigorating stomach and the like. Flos Sophorae Immaturus, which is the dried bud of Sophora japonica of Leguminosae, mainly contains flavone, saponin, sterol, etc., has effects of cooling blood, stopping bleeding, clearing liver-fire, reducing capillary permeability, resisting oxidation, inflammation and virus, and inhibiting aldose reductase. Pueraria lobata (Willd.) Ohwi, a dry root of Pueraria lobata Ohwi of Leguminosae, contains flavonoids as main ingredients, has the effects of relieving muscles and fever, promoting eruption, promoting fluid production to quench thirst, invigorating yang and relieving diarrhea, and is commonly used for fever thirst, yin deficiency thirst, heat diarrhea and dysentery, spleen deficiency diarrhea, etc. Semen Hoveniae is mature seed of Hovenia dulcis Thunb and Hovenia dulcis Thunb of Rhamnaceae of Hovenia, contains flavonoids, triterpenoid saponin and phenylpropanoids as main ingredients, and has effects of relieving alcoholism, quenching thirst, relieving restlessness, relieving vomit, and facilitating urination and defecation. The herba Houttuyniae is dried aerial part of houttuynia cordata Thunb of Saururaceae, contains flavonoids, volatile oil and organic acid as main ingredients, and has antibacterial, antiviral, immunity improving, and diuretic effects. The folium Mori is dry leaf of Morus alba L of Moraceae, contains flavonoids, alkaloids, volatile oil, etc., and has effects of dispelling pathogenic wind and heat, clearing lung-heat, moistening dryness, removing liver fire and improving eyesight. Lophatherum gracile, which is also called pheasant rice and golden chicken rice according to records of national Chinese herbal medicine compilation of economic grass plants in south China, is a perennial herb plant in the family of gramineae, is sweet in nature and taste, mainly comprises flavonoid compounds, polysaccharide, amino acid and the like, and can clear heart, promote urination and eliminate dysphoria.
In conclusion, the coix seed has the effects of tonifying spleen, excreting dampness, clearing heat, promoting diuresis, and relieving swelling, and the chicory has the effects of clearing damp-turbidity, clearing damp-heat and strengthening spleen and stomach as monarch drugs; flos Sophorae Immaturus is effective in clearing liver-fire, semen Hoveniae is effective in promoting diuresis and relieving stranguria, herba Lophatheri is effective in promoting urination and herba Houttuyniae is effective in clearing heat and promoting urination, and is used as ministerial drug for removing dampness and heat, eliminating dampness and turbid urine, and promoting urination and relieving stranguria; radix Puerariae and folium Mori can be used for clearing heat, eliminating turbid pathogen and relieving alcoholism. The 8 traditional Chinese medicines in the whole formula have the effects of eliminating dampness, strengthening spleen, clearing heat, promoting urination, purging turbid pathogen and treating stranguria, and can well reduce uric acid and relieve gout.
Disclosure of Invention
The invention aims to provide a three-rice drink traditional Chinese medicine formula composition for reducing uric acid and relieving gout and a preparation process thereof.
The traditional Chinese medicine formula comprises 8 medicinal materials of coix seed, chicory, sophora flower bud, lophatherum gracile, kudzu root, hovenia dulcis thunb, houttuynia cordata and mulberry leaf, and is composed of a certain dosage proportion. According to the nature of the traditional Chinese medicinal materials, the formula is divided into 2 groups, and the 2 groups are respectively extracted, one group is coix seed, sophora flower bud, lophatherum gracile and hovenia dulcis thunb, the other group is houttuynia cordata, chicory, kudzu root and mulberry leaf, the obtained 2 groups of medicinal material extracting solutions are respectively concentrated to proper times, and then are combined, and proper pH regulator, preservative and flavoring agent are added to obtain the beverage of the three-rice beverage.
The composition of the traditional Chinese medicine formula and the preparation process thereof as claimed in claim 1, relating to the preparation of three-rice drink, which is characterized in that: the Chinese medicinal formula is divided into 2 groups, one group is coix seed, sophora flower bud, lophatherum gracile and hovenia dulcis thunb, the other group is houttuynia cordata, chicory, kudzu-vine root and mulberry leaf, the former is decocted for 3 times by 10 times of water each time and is decocted for 45 minutes each time, and the filtrate is mixed and concentrated to 10 times; the latter is decocted for 2 times with 14 times of water each time, 60 minutes each time, filtered, the filtrates are combined and concentrated to 14 times; mixing the 2 concentrated solutions, and adding appropriate pH regulator, antiseptic and correctant to obtain three-rice beverage; the extraction process can also be carried out by mixing 8 medicinal materials together or dividing into different groups other than 2 groups, and extracting for a certain time and times.
The composition and preparation process of claim 1, wherein the pH regulator is selected from sodium bicarbonate, sodium carbonate and other alkaline substances, and the above 8 herbs have synergistic effect, and can promote the reduction of uric acid and relieve gout. The preservatives and flavoring agents are selected according to the type and amount specified by the country.
In order to better understand the present invention, the effect of the present invention will be further illustrated by animal experiments to confirm that the three-rice drink has the effect of reducing uric acid and relieving gout.
1. Experiment for uric acid reducing effect of the invention
1. Experimental animals: 30 male SD rats with SPF grade of 3 months old, with the body mass (200 +/-20) g, purchased and raised by the experimental animal center of Fujian Chinese medicine university.
2. Modeling and grouping intervention: after 1 week of adaptive feeding of 30 rats, 10 rats were randomly divided into a blank group, 20 rats were modeled, and the initial blood uric acid values of the rats were measured and recorded. The model building block is injected into the abdominal cavity with 200 mg/kg potassium oxonate solution (OAPS) and 250 mg/kg ethambutol hydrochloride (EMB) suspension for intragastric administration for 1 time every day, and a HUA rat model is built; the blank group was subjected to intragastric administration with an equal volume of physiological saline. The blood uric acid level of each rat was measured on day 21 of model building for model identification. After the model identification is successful, the model-making rats are randomly divided into a model group and three-meter drinking groups, and each group contains 10 rats. According to the conversion method of the equivalent dose of the body surface areas of the human body and the rat, concentrated solution of the three-meter drinking beverage is used for the three-meter drinking group, and the stomach of the blank group and the model group is irrigated with equal dose of normal saline for 1 time every day for 21 days of continuous intervention. To maintain a stable hyperuricemic state, the rats in the remaining groups, except the blank group, were given 200 mg/kg OAPS intraperitoneal injection and 250 mg/kg EMB suspension intragastric administration simultaneously with the dosing intervention.
3. Sample collection and index detection: the abdominal aorta blood collection is carried out after the rat is anesthetized, and the uric acid value is detected by adopting a uric acid detection kit after the rat is centrifuged at 3000 r/min for 20 min.
4. The experimental results are as follows: the blood uric acid level of the rats in the three-meter drinking group is obviously reduced compared with that before the intervention (P <0.05 ); compared with the model group, the blood uric acid level of the rats in the three-rice drink group is obviously reduced, and the difference has statistical significance (P <0.05). See table 1.

2. gout relieving effect experiment of the invention

2. Modeling and grouping intervention: on the basis of the HUA model building, an ankle joint is adopted to inject MSU suspension to build a gout rat model. After the model identification is successful, the rats in the model group are randomly divided into a model group and three-meter drinking groups, and each group contains 10 rats. According to the conversion method of the equivalent dose of the body surface areas of the human body and the rat, the three-meter drinking group is filled with the concentrated solution of the three-meter drinking beverage, the blank group and the model group are filled with the equal dose of normal saline, 1 time a day is taken, and 3 days are intervened continuously.

Sample collection and index detection: measuring the ankle joint skin temperature and swelling degree of each group of rats on the next day after intervention is completed; collecting ankle joint washing liquid under anesthesia, and detecting the content of interleukin-1 beta (IL-1 beta) in the ankle joint washing liquid by ELISA.
4. The experimental results are as follows: compared with the model group, the ankle joint skin temperature, swelling degree and IL-1 beta content in joint flushing fluid of the rats in the three-meter drinking group are all obviously reduced (P <0.05). See table 2.

Detailed Description
The following examples are provided for the purpose of illustration and not limitation, and are intended to illustrate the formulation, proportion and preparation of the present invention.
Example 1:
10g of coix seeds, 5g of chicory, 5g of sophora flower buds, 5g of lophatherum gracile, 7.5g of kudzuvine roots, 5g of hovenia dulcis thunb, 7.5g of houttuynia cordata, 5g of mulberry leaves, 0.3g of sodium bicarbonate, 0.003g of stevioside and 0.09g of potassium sorbate.
Weighing the coix seed, the sophora flower bud, the lophatherum gracile and the hovenia dulcis thunb according to the formula amount, uniformly mixing, adding 10 times of water each time, decocting for 45 minutes, filtering for 3 times, combining filtrates, and concentrating to 10 times to obtain 250 ml of concentrated solution; weighing the houttuynia cordata, the chicory, the radix puerariae and the mulberry leaves according to the prescription amount, uniformly mixing, adding 14 times of water each time, decocting for 60 minutes, filtering for 2 times, combining filtrates, and concentrating to 14 times to obtain 350 ml of concentrated solution; and (3) combining the 2 groups of concentrated solutions, adding 0.3g of sodium bicarbonate, 0.003g of stevioside and 0.09g of potassium sorbate, uniformly mixing, filtering, filling and sterilizing to obtain the three-rice beverage.
Example 2:
10g of coix seeds, 5g of chicory, 5g of sophora flower buds, 5g of lophatherum gracile, 7.5g of kudzuvine roots, 5g of hovenia dulcis thunb, 7.5g of houttuynia cordata, 5g of mulberry leaves, 0.3g of sodium bicarbonate, 12g of xylitol and 0.09g of potassium sorbate.
Weighing 8 medicinal materials according to the formula amount, uniformly mixing, adding 10 times of water each time, decocting for 60 minutes, filtering for 2 times, combining filtrates, concentrating to 600 milliliters, adding 0.3g of sodium bicarbonate, 12g of xylitol and 0.09g of potassium sorbate, uniformly mixing, filtering, filling and sterilizing to obtain the three-rice beverage.
Claims (3)
1. A three-rice drinking traditional Chinese medicine formula for reducing uric acid and relieving gout is characterized in that: the Sanmi beverage is prepared from 8 medicinal materials of coix seed, chicory, sophora flower bud, lophatherum gracile, kudzu vine root, hovenia dulcis thunb, houttuynia cordata and mulberry leaf at the dose ratio of 2: 1: 1.5: 1.
2. The preparation method of the three-meter drinking traditional Chinese medicine formula for reducing uric acid and relieving gout as claimed in claim 1, characterized by comprising the following steps: the traditional Chinese medicine formula is divided into 2 groups, one group is coix seed, sophora flower bud, lophatherum gracile and hovenia dulcis thunb, the other group is houttuynia cordata, chicory, kudzu vine root and mulberry leaf, the former is decocted for 45 minutes by 10 times of water each time, filtered for 3 times, the filtrates are combined and concentrated to 10 times; the latter is decocted for 60 minutes by 14 times of water each time, filtered for 2 times, and the filtrates are combined and concentrated to 14 times of water; mixing the 2 concentrated solutions, and adding appropriate pH regulator, antiseptic and correctant to obtain three-rice beverage; or mixing the above 8 medicinal materials together, decocting with 10 times of water each time for 60 min, filtering for 2 times, mixing filtrates, concentrating to 12 times, and adding appropriate pH regulator, antiseptic and correctant to obtain the final product.
3. The preparation method of the three-meter drinking traditional Chinese medicine formula for reducing uric acid and relieving gout as claimed in claim 2, characterized in that: the pH regulator added into the three-rice beverage is sodium bicarbonate, sodium carbonate and other alkaline substances; the preservatives and flavoring agents are selected according to the type and amount specified by the country.
