Concha Margaritifera is salty in taste and cold in nature, enters liver and heart meridians, has the effects of calming liver, suppressing yang hyperactivity, tranquilizing mind, arresting convulsion, improving eyesight and removing nebula, and is commonly used for treating symptoms such as headache, dizziness, palpitation, insomnia, conjunctival congestion, nebula, blurred vision and the like.
In order to promote the medicine to penetrate through the surface layer of the skin and reach the focus, 3-6 parts by weight of mint are also added in the embodiment of the invention. The mint is pungent in flavor and cool in nature, enters lung and liver channels, and has the effects of clearing head and eyes, relieving sore throat, promoting eruption, dispelling wind and heat, soothing liver, promoting qi circulation and the like. The mint extract can not only reduce the barrier effect of the skin by increasing the fluidity of the active epidermis, but also facilitate the transdermal absorption of the medicine; can also reduce the barrier effect of skin to drugs by destroying the ordered arrangement of horny layer and horny cell, so that the drugs can penetrate into the focus of infection, thereby enhancing the curative effect. In addition, herba Menthae has antiinflammatory, antiviral, anticancer, antibacterial, antioxidant, center exciting, and radioprotective effects.
The invention also provides a preparation method of the traditional Chinese medicine composition for treating dermatitis, which comprises the following steps: adding ethanol into the raw materials, grinding for 10-15 minutes, then carrying out ultrasonic treatment at 30-45 ℃ for 1-2 hours, standing for 3-4 hours, centrifuging to obtain a supernatant, carrying out reduced pressure distillation on the supernatant until the solid content is 50-60% to obtain a concentrated solution, adding a pharmaceutical adjuvant into the concentrated solution, and mixing to obtain the traditional Chinese medicine composition for treating dermatitis, wherein the mass ratio of the concentrated solution to the pharmaceutical adjuvant is 1 (1-1.5). The method has the advantages that the cell walls of the traditional Chinese medicines are destroyed in the process of adding the raw materials into ethanol for grinding, and simultaneously, the ethanol is used as an extracting agent to be beneficial to the dissolution of medicinal components, so that the bioavailability of the raw materials is improved. Grinding for 10-15 minutes can fully grind the raw materials to increase the contact area between the raw materials and the extractant, and can fully mix the extractant and the raw materials, thereby being beneficial to the extraction of active ingredients of the medicament, saving energy and reducing production cost. In detail, in the present example, the feed-to-liquid ratio of the raw material to ethanol was 1g (20-40) mL, which is advantageous for the dissolution of the active substance, thereby improving the extraction rate of the active substance.
And then the ground slurry is subjected to ultrasonic treatment at the temperature of 30-45 ℃ for 1-2 hours, so that the activity of the medicinal components can be preserved, and the dissolution of the active components can be accelerated under the treatment condition, thereby shortening the extraction time. In the embodiment, the power of the ultrasonic wave is 40-60W, and the use of the ultrasonic wave can not only improve the dispersion degree and the extraction rate of the raw materials in the extracting agent, but also accelerate the dissolution of the active substances and shorten the extraction time. However, the power of the ultrasound should be appropriate, and excessive power may change the structure of the pharmaceutically active substance to cause its failure, and the high power may generate high temperature to impair the life of the apparatus. Standing for 3-4 hr after ultrasonic treatment to precipitate and remove large granule residue, centrifuging, and collecting supernatant to remove light impurities in the solution, thereby purifying the medicine and enhancing the drug effect of the medicine. Then the concentrated solution is stirred with the gel matrix to disperse the medicine in the gel matrix. The preparation process is simple to operate, can realize large-scale mass production, can improve the extraction rate of the medicine and the curative effect of the medicine, is convenient to carry and paint the paste-shaped medicine, and has strong practicability.

The embodiment of the invention provides a preparation method of the traditional Chinese medicine composition for treating dermatitis, which comprises the following steps: adding the raw materials into ethanol to prepare a mixture with a feed-liquid ratio of 0.04g/mL, and grinding the mixture for 13 minutes; then carrying out ultrasonic treatment on the mixture for 1.5 hours at the temperature of 40 ℃, wherein the ultrasonic power is 50W; standing for 3.5 hours after ultrasonic treatment, discarding residues, and centrifuging at the rotating speed of 750r/min for 25 minutes to obtain supernatant; distilling the supernatant under reduced pressure until the solid content is 55% to obtain concentrated solution; preparing carbomer solution with solid content of 1.5%, stirring until swelling, and adding triethanolamine to adjust pH to 7.5 to obtain medicinal adjuvant; the concentrated solution is added with the pharmaceutical excipients and mixed to obtain the traditional Chinese medicine composition for treating dermatitis, and the mass ratio of the concentrated solution to the pharmaceutical excipients is 1: 1.3.

The embodiment of the invention provides a preparation method of the traditional Chinese medicine composition for treating dermatitis, which comprises the following steps: adding ethanol into the raw materials, grinding for 14 minutes, then carrying out ultrasonic treatment at 35 ℃ for 2 hours, standing for 3.5 hours, centrifuging to obtain a supernatant, carrying out reduced pressure distillation on the supernatant until the solid content of the supernatant is 56% to obtain a concentrated solution, adding pharmaceutical excipients into the concentrated solution, and mixing to obtain the traditional Chinese medicine composition for treating dermatitis, wherein the mass ratio of the concentrated solution to the pharmaceutical excipients is 1.0: 1.1.
Examples of effects
The therapeutic effects of the Chinese medicinal compositions for treating dermatitis prepared in examples 1 to 5 of the present invention were evaluated by the following methods, and the details are shown in Table 1.
Administration: 60 SPF-grade BALB/c 6-week-old mice were randomly selected and randomly divided into 6 groups, one of which was a blank group and the other 5 groups were labeled 1-5 as model groups, and the mice of model groups 1-5 after molding were smeared with the drugs of examples 1-5 for 2 times per day for 10 days with continuous administration, and the mice of the blank group were not administered.
Molding: the mouse is firstly administrated with the medicine of the embodiment 5 for 3 days, then the mouse is radiated by ultraviolet rays with the wavelength of 308nm and at the position of 15cm for 1.5h, the corresponding medicine is smeared for 1 time after 1h and 24h, the local skin of the mouse is in a latent period of 48h, the skin is visible to be red and swollen, and a solar dermatitis model is successfully established. The evaluation standard of the appearance index of local skin tissue, "I" skin is normal and has no red and swollen; skin with reddish brown spots appears in the skin of the 'II'; "III" skin is red and swollen, no exudation; "IV" skin is red and swollen, with exudation. The practical condition evaluation standard of the experimental pathological section, "I" skin tissue is normal, the epithelium is arranged neatly, the skin attachment is distributed evenly, and no pathological change is seen; the skin tissue of II is basically normal, and a little inflammatory cells infiltrate under the mucous membrane; III, skin tissues are basically normal, a large number of neutrophils are attached to the local surface, and inflammatory cells under the mucous membrane are infiltrated; and IV, skin tissue ulcer is formed, and granulomatous inflammation is accompanied with necrosis.

10-15 parts of fructus forsythiae, 15-20 parts of houttuynia cordata, 9-12 parts of raw sanguisorba officinalis, 5-8 parts of radix sophorae flavescentis, 5-8 parts of cortex dictamni, 8-12 parts of honeysuckle, 9-12 parts of astragalus membranaceus, 9-12 parts of angelica sinensis, 6-10 parts of rhizoma alismatis, 5-8 parts of fructus cnidii and 5-10 parts of liquorice.
2. The traditional Chinese medicine composition for treating dermatitis as claimed in claim 1, which is characterized by comprising the following raw materials in parts by weight:
12 parts of fructus forsythiae, 18 parts of houttuynia cordata, 10 parts of raw sanguisorba officinalis, 6 parts of radix sophorae flavescentis, 7 parts of cortex dictamni, 10 parts of honeysuckle, 10 parts of astragalus membranaceus, 10 parts of angelica sinensis, 8 parts of rhizoma alismatis, 7 parts of fructus cnidii and 8 parts of liquorice.
3. The traditional Chinese medicine composition for treating dermatitis as claimed in claim 1, further comprising 9-12 parts by weight of cortex lycii radicis and/or 5-10 parts by weight of bitter almond oil.
4. The traditional Chinese medicine composition for treating dermatitis as claimed in claim 1, further comprising 5-10 parts of polygala tenuifolia and/or 5-10 parts of nacre by weight.
5. The traditional Chinese medicine composition for treating dermatitis as claimed in any one of claims 1 to 4, further comprising 3 to 6 parts by weight of mint.
6. A method for preparing the traditional Chinese medicine composition for treating dermatitis as claimed in any one of claims 1 to 5, comprising the following steps:
adding ethanol into the raw materials, grinding for 10-15 minutes, then carrying out ultrasonic treatment at 30-45 ℃ for 1-2 hours, standing for 3-4 hours, centrifuging, taking supernate, carrying out reduced pressure distillation on the supernate until the solid content of the supernate is 50-60% to obtain a concentrated solution, adding a pharmaceutical adjuvant into the concentrated solution, and mixing to obtain the traditional Chinese medicine composition for treating dermatitis, wherein the mass ratio of the concentrated solution to the pharmaceutical adjuvant is 1 (1-1.5).
