Description

Whitening composition and preparation method and application thereof
Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to a whitening composition and a preparation method and application thereof.
Background
The whitening cosmetics are developed for many years and have a wide variety. From simple physical covering whitening at first to exfoliating type whitening and then to synergistic whitening of various whitening components. The development trend of the whitening cosmetics at the present stage is healthy whitening, and the whitening, transparent and glossy skin state is achieved while the health of the skin is maintained. However, the whitening products in the prior art are often deficient in whitening efficacy, such as poor whitening effect or easily causing skin allergy.
Therefore, it is necessary to provide a novel whitening cosmetic which is safe and excellent in whitening effect and hardly causes skin allergy.
Disclosure of Invention
The present invention is directed to solving at least one of the problems of the prior art described above. Therefore, the invention provides a whitening composition, a preparation method and an application thereof, wherein the composition has excellent whitening effect, high whitening speed (for example, obvious whitening effect can be achieved within 28 days), safe use and certain moisturizing effect, and is not easy to cause anaphylactic reaction to sensitive skin.
The invention conception of the invention is as follows: the whitening effect of the composition is remarkably improved by utilizing the synergistic effect of the Boerhavia diffusa extract, the bletilla striata extract, the paeonol and the whitening agent (the whitening agent comprises inositol, nicotinamide and hydroxyethyl piperazine ethane sulfonic acid). The composition has high use safety, is not easy to cause anaphylactic reaction on sensitive skin, and further has a certain moisturizing effect.
In a first aspect of the present invention, a whitening composition is provided.
Specifically, the whitening composition comprises the following components: yellow core extract, bletilla striata extract, paeonol and whitening agent; the whitening agent comprises inositol, nicotinamide and hydroxyethyl piperazine ethane sulfonic acid.
Preferably, the Boerhavia diffusa extract is a Boerhavia diffusa root extract.
Preferably, the bletilla striata extract is a bletilla striata root extract.
Preferably, the composition comprises, by weight, 0.01-0.5 part of the yellow core extract, 0.02-0.6 part of the bletilla striata extract, 0.02-0.5 part of paeonol and 0.5-16 parts of a whitening agent.
More preferably, the composition comprises, by weight, 0.015 to 0.3 part of the yellow core extract, 0.025 to 0.4 part of the bletilla striata extract, 0.05 to 0.3 part of paeonol and 0.9 to 12 parts of the whitening agent.
More preferably, the composition comprises, by weight, 0.015-0.15 part of the yellow core extract, 0.025-0.25 part of the bletilla striata extract, 0.05-0.25 part of paeonol and 0.9-9.5 parts of the whitening agent.
Preferably, the whitening agent comprises 0.1-5.5 parts of inositol, 0.3-5.5 parts of nicotinamide and 0.1-5.0 parts of hydroxyethyl piperazine ethane sulfonic acid; more preferably, the whitening agent comprises 0.1-1.5 parts of inositol, 0.5-5.0 parts of nicotinamide and 0.3-3.0 parts of hydroxyethyl piperazine ethane sulfonic acid.
Preferably, the composition further comprises at least one of fruit fat or squalane of Butyrospermum PARKII (Butyrpermum PARKII). The fruit fat and squalane of the Butyrospermum PARKII (Butyrum PARKII) increase the permeability, so that effective whitening active ingredients can be better absorbed by the skin; it is useful to further improve the whitening effect of the composition.
Preferably, the composition comprises 0.01-5.5 parts of fruit fat of Butyrospermum PARKII (Butyrum PARKII); further preferably, the composition comprises 0.1-5 parts of fruit fat of Butyrospermum PARKII (Butyrspermum PARKII).

Inositol in the whitening agent can be derived from a high-purity beet extract, belongs to a natural organic penetrating agent, has no irritation, can obviously increase the expression of skin epidermal silk polyprotein, enables the skin to be tender and elastic, and has an instant brightening effect, so that the effects of quickness and long-acting whitening are achieved.
The whitening mechanism of the niacinamide in the whitening agent is to inhibit the transport of melanosomes to keratinocytes by influencing the intercellular communication between melanocytes and keratinocytes, thereby down-regulating melanin synthesis. In combination with the hydroxyethyl piperazine ethane sulfonic acid, the route of melanin from production to migration to the cell surface can be effectively controlled.
The hydroxyethyl piperazine ethanesulfonic acid also has the functions of a penetration enhancer and a cutin softener, and specifically, the hydroxyethyl piperazine ethanesulfonic acid can soften cutin, smooth and soften skin, brighten skin color and achieve the effect of whitening; promoting the absorption of functional components; the whitening active substances are promoted to better enter the skin, and the synergistic whitening effect is achieved; absorb ultraviolet rays and part of visible light, and are used for synergistically preventing sunlight.
The composition of the invention achieves the regulation of the proliferation of melanocytes and the synthesis of melanin by inhibiting MITF (microphthalmia-associated transcription factor), and the MITF directly influences the activities of various enzymes, including tyrosinase, DHICA oxidase and dopachrome isomerase, and indirectly influences the generation of melanin. The MITF mainly controls the survival and differentiation of melanocytes and the synthesis, transportation and distribution of melanin by regulating a TYR gene family, has important functions in the development processes of morphological constitution, survival, migration, proliferation and differentiation of melanocytes, and has three main action mechanisms: (1) the MITF protein can directly regulate and control Bcl2, c-Met, BIRC7, BCL2A1 and other genes required by the survival of melanocytes. (2) MITF can directly regulate melanocyte proliferation and cycle-related genes. (3) Inhibiting the expression of MITF effectively reduces the levels of TYR, TYRP1 and TYRP2 and tyrosinase, which are regulatory factors that play an important role in melanocyte proliferation and melanocyte synthesis.
The whitening pathways of the compositions of the present invention are completely different from conventional whitening pathways, which include (1) protection against ultraviolet rays; (2) inhibiting tyrosinase activity; (3) inhibiting the expression of tyrosinase; (4) inhibiting the transfer of melanin; (5) promoting keratinocyte regeneration; (6) antioxidation and anti-glycation.
In a second aspect of the present invention, there is provided a method for preparing a whitening composition.

/>
Figure BDA0003135788300000101
/>
Figure BDA0003135788300000111
The creams of comparative examples 1 to 7 were prepared in the same manner as the creams of the application examples (except for the added raw material components).
Comparative examples 8 to 11
Comparative example 8 differs from application 1 only in that 0.5 parts of phenethylresorcinol was used in place of 2 parts of niacinamide in the cream prepared in comparative example 8, and the remaining raw material components and preparation method were the same as in application example 1.
Comparative example 9 is different from application 1 only in that in comparative example 9, hydroxyethylpiperazine ethanesulfonic acid in application 1 was replaced with an equal amount of a hydroxyethylacid having a similar function to hydroxyethylpiperazine ethanesulfonic acid, and the remaining raw material components and preparation method were the same as in application example 1.
Comparative example 10 differs from application 1 only in that in comparative example 10, the hydroxyethylpiperazine ethanesulfonic acid in application 1 was replaced with an equal amount of AHA-50 having a similar function to the hydroxyethylpiperazine ethanesulfonic acid, i.e., 50 mass% of fruit-derived AHA in AHA-50 or fruit-derived AHA-50 (12-17 mass% in AHA-50 or fruit-derived AHA in fruit-50), with 28-32% lactic acid, 12-17% in glycolic acid, 2-6% in citric acid, and not more than 1.0% in tartaric acid and malic acid), and the remaining raw material components and preparation method were the same as in application 1.
Comparative example 11 is different from application 1 only in that comparative example 11 replaces hydroxyethylpiperazine ethanesulfonic acid of application 1 with the same amount of lactic acid having a similar function to hydroxyethylpiperazine ethanesulfonic acid, and the remaining raw material components and preparation method are the same as those of application 1.
Product effectiveness testing
1. Whitening effect test of cream of application examples 1 to 5 and comparative examples 1 to 7
60 female volunteers were selected, age group 25 to 45 years, healthy, skin without history of skin disease and allergy. Skin color ITA ° values were tested on a 20 ° to 41 ° portion, and all volunteers were randomly divided into 12 groups, and 5 persons of each group used the corresponding cream (creams of application examples 1-5 and comparative examples 1-7). All volunteers were able to use the cream as specified except for the ITA ° value (ITA °, i.e., individual type angle, representing individual type angle) of the test skin before the use of the cream, which is common knowledge in the art, as measured by a skin colorimeter or reflectance spectrophotometer, the calculation method or measurement method of ITA ° being the larger the ITA ° value, the lighter the skin color, and also on the 1 st, 2 nd, 4 th, and 8 th days after the continuous use of the cream, the ITA ° value of the test skin (the ITA ° value tested is the average value of ITA ° of each group) was tested, and the results are shown in table 3[ ITA ° lift (%) in table 3 (average value of ITA ° before 8 weeks-average value of ITA ° before use)/average value of ITA ° before use = 100% ].
TABLE 3
Figure BDA0003135788300000121
As can be seen from table 3, the creams prepared in application examples 1 to 5 have significantly larger average value of ITA ° than those of comparative examples 1 to 7, and the larger average value of ITA ° indicates better whitening effect. As can be seen from table 3, inositol, niacinamide, hydroxyethyl piperazine ethanesulfonic acid, huangxin extract, bletilla striata extract, and paeonol have synergistic effects in skin whitening.
2. Whitening effect test of cream in application example 1 and comparative examples 8-11
20 female volunteers, aged 25 to 45 years, were selected, healthy, skin without a history of skin disease and allergy. The skin colour ITA ° values were tested on a 20 ° to 41 ° section, and all volunteers were randomly divided into 4 groups, each of which 5 people used the corresponding cream (creams of comparative examples 8-11). All volunteers were able to use the cream as specified, and in addition to testing the skin for the ITA ° values before using the cream, the skin of the volunteers was tested for the ITA ° values on day 1, week 2, week 4, and week 8 of consecutive use of the cream (the ITA ° values tested are the average value of the ITA ° for each group), with the results shown in table 4.
TABLE 4
Figure BDA0003135788300000131
As can be seen from table 4, the cream prepared in application example 1 has a significantly larger average value of ITA ° than those of comparative examples 8 to 11, and a larger average value of ITA ° indicates a better whitening effect. As can be seen from Table 4, niacinamide in the cream of the present invention cannot be replaced with phenethyl resorcinol of the prior art. The hydroxyethylpiperazine ethanesulfonic acid in the cream of the present invention cannot be replaced with the prior art hydroxyethylacids, alpha-50 and lactic acids. The results further prove that the inositol, the nicotinamide, the hydroxyethyl piperazine ethanesulfonic acid, the yellow core extract, the bletilla striata extract and the paeonol in the invention have synergistic effect on skin whitening.
In addition, the creams prepared in application examples 1 to 5 are highly safe to use and are less likely to cause allergic reactions to sensitive skin.
