The sample carrier 34 is used to place or hold the test tube 341 to be tested.
The incubation module 35 is used for heating, controlling temperature and incubating the consumables 361 in the extraction carrier. As shown in fig. 7, incubation module 35 includes heating block 351 provided with incubation hole, attached heating film 352 of a lateral wall of heating block 351, fixed heating block 351 and incubation support 353 of heating film 352, heating block 351's incubation hole is used for inserting the consumptive material 361 that the splendid attire extracted the consumptive material, heating film 352 and industrial control module 5 electric connection, after the consumptive material 361 that extracts the consumptive material inserts heating block 351's incubation hole, industrial control module 5 control heating film 352 generates heat, heat and incubate for extracting the consumptive material through heating block 352's conduction. The magnetic attraction mode of the magnetic attraction device 37 is a translational magnetic attraction structure, as shown in fig. 8-9, the translational magnetic attraction structure includes a translational guide rail component 371, a translational magnetic attraction component 372 and a translational driving component 373, the translational guide rail component 371 includes a translational track 3711 and a translational slider 3712 in sliding fit with the translational track 3711, the translational magnetic attraction component 372 includes a magnet 3721, and the translational driving component 373 is butted with the translational slider 3712; wherein, the magnetic assembly 372 is installed on the sliding block 3712, and the driving assembly 373 is electrically connected to the industrial control module 5 for driving the magnetic assembly 372 to slide on the sliding track 3711. The magnetic attraction device 37 is a rotary magnetic attraction structure, as shown in fig. 10-12, the rotary magnetic attraction structure includes a rotary shaft 374, at least one magnet frame 375 mounted on the rotary shaft 374, and a first magnet 3722 and a second magnet 3723 mounted at two ends of the magnet frame 375, the first magnet 3722 and the second magnet 3723 respectively extend out of two sides of the rotary shaft 374, the magnet frame 375 and the rotary shaft 374 constitute a structure similar to a "seesaw", the first magnet 3722 and the second magnet 3723 are located at two ends of the "seesaw", the rotary driving component 376 drives the rotary shaft 374 to rotate and switch between a first position and a second position, when the rotary shaft 374 rotates to the first position, the first magnet 3722 is located at a first magnetic attraction station, and at this time, the first magnet 3722 is close to the bottom surface of the incubation hole (i.e. the first magnet 3722 is close to the bottom surface of the extraction consumable material 361) (as shown in fig. 12 a), when the rotating shaft 374 rotates to the second position, the second magnet 3723 is located at the second magnetic attraction station, and the second magnet 3723 is close to the side surface of the incubation hole (i.e. the second magnet 3723 is close to the side surface of the consumable part 361) (see fig. 12 b). The rotary driving motor 3761 is electrically connected to the industrial control module 5, and the industrial control module controls the rotation of the rotary driving motor. Preferably, the rotation driving motor 3761 is a stepping motor, which can control the rotation angle of the rotation shaft 374 more precisely, and further control the positions of the first magnet 3722 and the second magnet 3723.
The fluorescence detecting apparatus 38 with oscillation, as shown in fig. 13-14, includes a fluorescence detecting module 381, an oscillation mechanism 382 disposed above the fluorescence detecting module, a detecting consumable carrier 383 disposed on the oscillation mechanism, and an oscillation driving mechanism 384 for driving the detecting consumable carrier 383 to move back and forth and left and right in a horizontal direction on the oscillation mechanism 382.
The fluorescence detection module 381 includes at least one excitation light source set (not shown), at least one fluorescence light-gathering set (not shown), at least one photodiode (not shown), and a detection support frame 3811, the detection support frame 3811 is provided with an excitation light source hole and a fluorescence detection hole, the excitation light source set is disposed below the excitation light source hole, the excitation light source set includes an LED light source, an excitation light source light-gathering lens set and a filter, the fluorescence light-gathering set is disposed below the fluorescence detection hole, and the fluorescence light-gathering set includes a fluorescence light-gathering lens set and a second filter.
Preferably, the fluorescence detection module 381 is further provided with a detection module driving mechanism 3812, the detection module driving mechanism 3812 includes a pair of detection module supports 3813 arranged at intervals, a pair of detection module guide rods 3814 arranged between the pair of detection module supports 3813 in parallel, a detection module sliding seat 3815 slidably sleeved on the pair of detection module guide rods 3814, and a detection module driving device 3816 driving the detection module sliding seat 3815, the detection module wire rail 3817 and the pair of detection module guide rods 3814 are arranged on the detection module sliding seat 3815 in a horizontal direction perpendicular to the horizontal direction, and the detection support 3811 of the fluorescence detection module 381 is movably arranged on the detection module wire rail 3817 through the detection module driving device 3816. Preferably, the sensing module driver 3816 is a stepper motor, which can determine the exact position within the range of travel of the sensing module. The movement of the fluorescence detection module 381 in the horizontal plane in the horizontal and vertical directions is enabled by the detection module driving mechanism 3812, so that the detection of a plurality of consumables on the detection consumable carrier can be realized.
The oscillation mechanism 382 includes, as shown in fig. 14, a pair of oscillation support plates 3821 oppositely disposed at both sides of the fluorescence detection module 381, a pair of first oscillation guide rods 3822 fixed between both ends of the upper end portions of the pair of oscillation support plates 3821, a first oscillation slide 3823 slidably fitted over the pair of first oscillation guide rods 3822, a pair of second oscillation guide rods 3824 horizontally and vertically fixed to the first oscillation slide 3823, and a second oscillation slide 3825 slidably fitted over the pair of second oscillation guide rods 3824.
The oscillation driving mechanism 384, as shown in fig. 14, includes an oscillation driving connecting rod 3841, an oscillation driving eccentric shaft 3842 and an oscillation driving motor 3843, the oscillation driving eccentric shaft 3842 includes a main shaft portion 3844 and an eccentric shaft portion 3845 (as shown in fig. 15) provided at one end of the main shaft portion 3844 and offset with respect to the axis of the main shaft portion 3844, the main shaft portion 3844 of the oscillation driving eccentric shaft 3842 is vertically rotatably mounted on the shaft bracket through a bearing, the oscillation driving motor 3843 is vertically fixed through a motor bracket, the output end of the oscillation driving motor 3843 is connected with the lower end of the main shaft portion 3844 of the oscillation driving eccentric shaft 3842 through a timing belt, one end of the oscillation driving connecting rod 3841 is sleeved on the eccentric shaft portion 3845 at the upper end of the oscillation driving eccentric shaft 3842, and the other end of the.
The oscillation driving motor 3843 drives the oscillation driving eccentric shaft 3842 to rotate, the eccentric shaft part 3845 at the upper end of the oscillation driving eccentric shaft 382 makes a circular motion around the axis of the main shaft part 3844, a circular motion track taking the axis of the main shaft part as a circle center is formed in an overlooking manner, the oscillation driving connecting rod 3841 drives the detection consumable carrier 383 and the detection consumable to move on the oscillation mechanism 382 along the motion track of the eccentric shaft part 3845, and a reagent in the detection consumable carries out eddy current rotation along the inner wall of the detection consumable tube. The multi-channel sample adding device 4, as shown in the figure, includes an extraction head 41 for extracting a suction head 46, an injection assembly 42 connected to the extraction head 41 and communicating with each other, an extraction driving mechanism 43 for driving the extraction head 41 to move up and down, a desorption head plate 44, and a desorption head driving mechanism 45 for driving the desorption head plate to move up and down.
The head removing plate 44 is provided with a head removing hole 441, wherein the position of the head removing hole 441 corresponds to the position of the extraction head 41, the head removing hole 441 is used for allowing the extraction head 41 to pass through the extraction head 46, and the aperture of the head removing hole 441 is smaller than the aperture of the extraction head 46 extracted by the extraction head 41.
The extraction driving mechanism 43 comprises an extraction sliding rail 431 and an extraction sliding seat 432 connected with the extraction sliding rail 431 in a sliding manner, an extraction motor 433, an extraction synchronous belt 434 and an extraction screw 435, wherein the extraction motor 433 is provided with an extraction driving wheel 4331, the extraction screw 435 is provided with an extraction driven wheel 4351 linked with the extraction screw, the extraction driving wheel 4331 and the extraction driven wheel 4351 tension the extraction synchronous belt 434, the extraction sliding seat 432 is provided with an extraction screw hole, the extraction screw 435 is in threaded fit with the extraction screw hole, the extraction sliding seat 432 is provided with a base 436, an extraction head 41 is arranged on the base 436, the extraction motor 433 is electrically connected with the industrial control module 5, the industrial control module 5 drives the extraction motor 433 to rotate to drive the extraction screw 435 to rotate, the rotation of the extraction motor 433 is converted into the up-down movement of the extraction sliding seat 432 through the threaded fit of the extraction screw 435 and the extraction screw hole, and (6) pipetting. Preferably, the extraction motor 433 and the extraction screw 435 are located on the same side of the extraction timing belt 434, whereby a space above the extraction screw 435 can be saved.
The suction head removing driving mechanism 45 comprises a suction head removing slide seat 451, a suction head removing support 452 connected with the suction head removing slide seat 451, a suction head removing motor 453, a suction head removing synchronous belt 454 and a suction head removing screw 455, wherein the suction head removing motor 453 is provided with a suction head removing driving wheel 4531, the suction head removing screw 455 is provided with a suction head removing driving wheel 4551 linked with the suction head removing screw, the suction head removing driving wheel 4531 and the suction head removing driving wheel 4551 tension the suction head removing synchronous belt 454, the support 452 is provided with a suction head removing screw hole, the suction head removing screw 455 is in threaded fit with the suction head removing screw hole, a suction head removing plate 44 is installed between two wings of the suction head removing support, the suction head removing slide seat 432 is simultaneously in sliding fit with the base 436, and the suction head removing motor 453 is electrically connected with the industrial control module 5. When the industrial control module 5 drives the suction head removing motor 453 to rotate, the rotation of the suction head removing motor 453 is converted into the up-and-down movement of the suction head removing bracket 452 through the threaded matching of the suction head removing screw rod 455 and the suction head removing screw hole, and then the suction head removing plate 44 is driven to move up and down relative to the extraction head 41, so that the suction head 46 is separated from the extraction head 41 by the suction head removing plate 44. Preferably, the suction head removing motor 453 and the suction head removing screw 455 are located on the same side of the suction head timing belt 454, so that the space above the suction head removing screw 455 is saved.
The injection assembly 42, as shown in the figure, comprises a syringe 421, a piston 422 sliding in the syringe 421, and an injection driving device 423 connected with the piston 422, the extraction head 41 is connected with the syringe 421 and communicated with each other, and after the extraction head 41 extracts the suction head 46, the suction head 46 is communicated with the injection assembly through the extraction head 41. The injection drive 423 may be a motor, preferably a stepper motor. The industrial control module 5 is electrically connected with the injection driving device, when a sample or a reagent needs to be sucked, the industrial control module 5 drives the injection driving device to move, so as to drive the piston 422 to move upwards, the pressure in the suction head 46 is reduced, the sample or the reagent can be sucked by the suction head 46, when the sample or the reagent needs to be injected, the industrial control module 5 drives the injection driving device 423 to drive the piston 422 to move downwards, the pressure in the suction head 46 is increased, and the sample or the reagent is discharged from the suction head 46.
The multi-channel sample adding device 4 is slidably connected with the upper part of the casing 1 in the following way: the upper end part of the extracting slide block in the multi-channel sample adding device 4 is connected with a slide seat 47, a slide rail 16 is arranged on a top plate 13 of the machine shell 1, a driving wheel 17 driven by a motor is arranged at one end part of the slide rail 16, a driven wheel 18 and a synchronous belt 19 tensioned between the driving wheel 17 and the driven wheel 18 are arranged at the other end part of the slide rail 16, the synchronous belt 19 is matched with the slide seat 47, and the industrial control module 5 is electrically connected with the motor. When reagent and sample need to be transferred, the driving motor of the industrial control module 5 drives the synchronous belt 19 to rotate, the sliding seat 47 is driven to slide back and forth through the rotation of the synchronous belt 19, and then the multichannel sample adding device 4 is driven to move back and forth to transfer the reagent or the sample, so that the sample adding work is completed.
The industrial control module 5 comprises a touch screen industrial control computer 51 and a mounting cross beam 52, wherein the touch screen industrial control computer 51 is mounted on the rack 11 above the cabin door 2 through the mounting cross beam 52. The detection personnel operate the touch screen industrial personal computer 51 to control and operate the nucleic acid amplification detection analyzer, so that the full-automatic operation of nucleic acid extraction, amplification and detection is realized.
The working principle of the full-automatic nucleic acid amplification detection analyzer is as follows:
preparing to operate a touch screen industrial personal computer, setting corresponding parameters according to detection requirements, starting a full-automatic nucleic acid amplification detection analyzer, sending out a sliding frame of a nucleic acid extraction and amplification device, placing a test tube filled with a sample on a sample carrier, placing a suction head on the extraction carrier, and withdrawing the sliding frame;
(II) the motor of the extraction suction head rotates to drive the driving wheel to rotate, the driving synchronous belt rotates to drive the sliding seat to move, the multi-channel sampling device moves forwards to enable the extraction head to be aligned with the suction head on the extraction carrier (as shown in figure 21a), the desorption head driving mechanism drives the desorption head plate to move upwards to the uppermost end of the extraction head, the extraction driving mechanism drives the extraction head, the injection assembly and the desorption head plate to move downwards together, the extraction head is inserted into a preset position in the suction head opening, the extraction head and the suction head are connected together in an interference fit manner (as shown in figure 21b), and the extraction driving mechanism drives the extraction head and the suction head to move upwards together to separate from the extraction carrier (as;
(III) the injection driving device of the sample sucking and injecting assembly pushes the piston downwards to the lowest part of the injection tube, the multi-channel sample adding device moves to drive the suction head to be aligned with the opening of the sample test tube (as shown in figure 22a), the extraction driving mechanism drives the suction head and the desorption head plate to move downwards together, so that the suction head is inserted into the sample test tube (as shown in figure 22b), the injection driving device drives the piston to move upwards, the suction head sucks a certain amount of sample (as shown in figure 22c), and the extraction driving mechanism drives the suction head and the desorption head plate to move upwards (as shown in figure 23 a);
(IV) the sample-adding multi-channel sample-adding device moves to drive the suction head to move, so that the suction head is aligned with a consumable port on the extraction carrier (as shown in figure 23b), the extraction driving mechanism drives the suction head and the desorption head plate to move downwards, so that the suction head is inserted into the consumable, the injection driving device drives the piston to move downwards, a certain amount of sample is added into the consumable (23c), and the extraction driving mechanism drives the suction head and the desorption head plate to move upwards to finish one sample-adding work;
heating a heating membrane of the incubation module, and performing heat incubation only after extraction in consumables through conduction of a heating block; certainly, when incubating, can also make injection drive arrangement drive piston reciprocate, let the mixed liquid pass in and out the suction head many times, reach the effect of mixing mixed liquid, realize hatching simultaneously mixing.
Sixthly, moving or rotating the magnetic absorption device to approach the consumable material, and absorbing magnetism on the side surface or the bottom surface of the consumable material;
(VII) after the liquid absorption and sample addition of all the holes of the desorption head are finished, the multi-channel sample adding device moves to drive the suction head to move, so that the suction head is aligned with the suction head hole on the extraction carrier (as shown in figure 24a), the suction head and the desorption head plate move downwards and are inserted into the suction head hole (as shown in figure 24b), the desorption head driving mechanism pushes the desorption head plate to move downwards, so that the suction head is separated from the extraction head (as shown in figure 24c), the suction head falls into the suction head hole of the extraction carrier by means of gravity, and the desorption head driving mechanism drives the desorption head plate to move upwards, so that the work of the desorption head is finished;
(eighth) pipetting according to the suction head connected with the suction head, moving the multi-channel sample adding device to drive the suction head to move, aligning the suction head with the consumable on the extraction carrier (as shown in figure 22a), moving the suction head and the desorption head plate downwards by the extraction driving mechanism to be inserted into the consumable (as shown in figure 22b), moving the piston upwards by the injection driving mechanism, sucking a certain amount of liquid by the suction head (as shown in figure 22c), and moving the suction head and the desorption head plate upwards by the extraction driving mechanism (as shown in figure 23 a); the multi-channel sample adding device moves to drive the suction head and the desorption head plate to move, so that the suction head is aligned with a detection consumable hole on a detection consumable carrier (as shown in figure 23b), the suction head and the desorption head plate move downwards to be inserted into the detection consumable hole by the extraction driving mechanism, the injection driving device drives the piston to move downwards, a certain amount of liquid is added into the detection consumable (as shown in figure 23c), and the suction head and the desorption head plate are driven to move upwards by the extraction driving mechanism;
(ninth), the fluorescence detection oscillation driving mechanism drives the detection consumable carrier to oscillate on the oscillation mechanism along the horizontal direction, front, back, left and right, and drives the reagent in the detection consumable to rotate along the inner wall of the detection consumable pipe, so that uniform vortex mixing is realized; meanwhile, after the LED light source emits light, the light is condensed by the excitation light source condensing lens group, then excitation light with a specified wavelength is obtained by the filter plate, the excitation light penetrates through the excitation light source hole and irradiates on a reagent in the consumable, the reagent emits fluorescence with another wavelength to the periphery, part of the fluorescence with the other wavelength is emitted into the fluorescence detection hole, the fluorescence with the specified wavelength is obtained through the fluorescence condensing lens group and the second filter, the fluorescence with the specified wavelength is collected on the photodiode, and the intensity of analysis light can complete conventional nucleic acid amplification detection.
Preferably, the multi-channel sample adding device can also be realized as a capping gland for detecting consumables, wherein the tube cap 8 is placed on the extraction carrier. Further, the air filtering module 6 is also included, and the air filtering module 6 includes a lower cover 61 mounted on the top plate, an upper cover 62 buckled on the lower cover, filter cotton 63 disposed in the lower cover 61, an air outlet 64 disposed on the upper cover 62, and an exhaust fan 65 mounted at the lower end of the air outlet. The air containing the aerosol in the detector enters the air filtering module from the groove below the lower cover under the action of the exhaust fan, pollutants such as the aerosol are adsorbed on the filtering surface through the filtering of the filter cotton, and the filtered air meeting the emission standard is discharged from the air outlet, so that the pollution to indoor or outdoor environment can be effectively reduced.
Furthermore, a sample scanning module 7 is also provided, the sample scanning module 7 comprises a scanner 71, the scanner is used for identifying a bar code on a sample or a reagent, and the scanner 71 is fixed on a cross beam through a bracket and is arranged on the rack 11 between the cabin door and the industrial control module through the cross beam. Preferably, an ultraviolet sterilizing lamp 72 is provided on the side of the beam facing the inside of the housing to sterilize the inside of the detector with ultraviolet light. Preferably, a status indicator lamp 73 is further provided, and the status indicator lamp is mounted below the side of the cross beam and can display the status of the detector in real time.

A full-automatic nucleic acid amplification detection analyzer is characterized by comprising a machine shell with an opening on one side, a cabin door arranged at the opening of the machine shell, a nucleic acid extraction and amplification device horizontally and vertically arranged at the bottom of the machine shell towards the cabin door, a multi-channel sample adding device arranged above the nucleic acid extraction and amplification device and connected to the upper end of the machine shell in a sliding manner, and an industrial control module;
the nucleic acid extraction and amplification device comprises a sliding frame which can be sent out or retracted along the hatch door and a sending-out driving device; a sample carrier, an incubation module, an extraction carrier for loading extraction consumables, a magnetic attraction device arranged below the extraction carrier and a fluorescence detection device with oscillation are sequentially arranged on the sliding frame from the cabin door inwards;
the multichannel sample adding device comprises an extraction head for extracting a suction head, an injection assembly, an extraction driving mechanism, a suction head removing plate and a suction head removing driving mechanism, wherein the injection assembly is connected with the extraction head and is communicated with the extraction head, the extraction driving mechanism is used for driving the extraction head to move up and down, and the suction head removing driving mechanism is used for driving the suction head removing plate to move up and down.

The automatic nucleic acid amplification detection analyzer of claim 1, wherein the housing comprises a frame, a top plate and a bottom plate respectively fixed to the upper and lower ends of the frame, and side plates surrounding three sides of the frame; the lower end part of the cabin door is rotatably pivoted on the bottom plate at the opening of the shell, and a torsional spring is sleeved at the pivoting part of the cabin door and the bottom plate.
3. The automatic nucleic acid amplification detection analyzer of claim 2, wherein the sliding rack is provided with a wheel for pushing the hatch to turn outwards and open when the sliding rack is delivered.

The automatic nucleic acid amplification detection analyzer of claim 2, wherein a slide rail is disposed on a top plate of the housing, a driving wheel driven by a motor is disposed at one end of the slide rail, a driven wheel and a synchronous belt tensioned between the driving wheel and the driven wheel are disposed at the other end of the slide rail, and a slide seat is disposed at the top of the multi-channel sample adding device and driven by the synchronous belt to slide back and forth on the slide rail.

The automatic nucleic acid amplification detection analyzer of claim 2, wherein the nucleic acid extraction and amplification device further comprises a support frame, the bottom plate is provided with a stopper, the support frame is mounted on the bottom plate through the stopper, the carriage is disposed on the support frame, and the support frame cooperates with the feeding driving device to feed or retract the carriage to the chamber door.

The automatic nucleic acid amplification detection analyzer of claim 5, wherein the feeding driving device comprises a feeding motor, a feeding driving wheel, a feeding synchronous belt and two feeding driven wheels, the feeding driving wheel is disposed on an output shaft of the feeding motor, the two feeding driven wheels are disposed below the driving wheel, two ends of the feeding synchronous belt are fixed to the supporting frame, and the two feeding driven wheels press the feeding synchronous belt against the feeding driving wheel.

The automatic nucleic acid amplification detecting analyzer according to claim 2, wherein the fluorescence detecting device with oscillation comprises a fluorescence detecting module, an oscillating mechanism disposed above the fluorescence detecting module, a detecting consumable carrier disposed on the oscillating mechanism, and an oscillation driving mechanism for driving the detecting consumable carrier to move back and forth and left and right on the oscillating mechanism along a horizontal direction.

The automatic nucleic acid amplification detection analyzer of claim 2, further comprising an air filtration module, wherein the air filtration module comprises a lower cover and an upper cover, filter cotton disposed in the lower cover, an air outlet disposed on the upper cover, and an exhaust fan mounted at the lower end of the air outlet; the lower cover is installed above the top plate.
9. The automatic nucleic acid amplification detection analyzer of claim 2, wherein the industrial control module comprises a touch screen industrial personal computer and a mounting beam, the touch screen industrial personal computer is mounted on the beam, and the beam is mounted on the rack above the hatch.
10. The automated nucleic acid amplification detection analyzer of claim 9, further comprising a sample scanning module mounted on the rack between the hatch and the industrial control module.
