背景技术
氨基酸(Amino acid)是构成蛋白质的基本单位,赋予蛋白质特定的分子结构形态,使他的分子具有生化活性。蛋白质是生物体内重要的活性分子,包括催化新陈代谢的酵素和酶。氨基酸是构成生物体蛋白质并同生命活动有关的最基本的物质,是在生物体内构成蛋白质分子的基本单位,与生物的生命活动有着密切的关系。它在抗体内具有特殊的生理功能,是生物体内不可缺少的营养成分之一。
生物活性肽是蛋白质中20个天然氨基酸以不同组成和排列方式构成的从二肽到复杂的线性、环形结构的不同肽类的总称,是源于蛋白质的多功能化合物。生物活性肽具有多种人体代谢和生理调节功能,易消化吸收,有促进免疫、激素调节、抗菌、抗病毒、降血压、降血脂等作用,食用安全性极高,是当前国际食品界最热门的研究课题和极具发展前景的功能因子。
两个以上的氨基酸之间以肽键相连,形成的"氨基酸链"或"氨基酸串"就叫做肽。其中,10-15个以上氨基酸组成的肽被称为多肽,而由2至9个氨基酸组成的就叫做寡肽,由2至15个氨基酸组成的就叫做小分子肽。小分子活性肽是介于氨基酸与蛋白质之间一种生化物质,它比蛋白质分子量小,又比氨基酸分子量大,是一个蛋白质的片段。
小分子活性肽与氨基酸运输功能相比,具有吸收快速、低耗和不饱和的特点。技术人员利用其特点为一些特殊状况下的人群补充营养,如:术后伤口愈合期、康复期、身体复原期的病人;精神压力大,过度劳累,食欲不振的患者;食物中化学物质、重金属、辐射储存、残留农药,长服化学药物、中药余毒,消化系统损害引起的肠胃功能失调者;运动量大、体力劳动强度大须及时补充氮源,又不能增加肠胃功能负担者;消化系统因疾患障碍不能进食者;消化器官未发育成熟的婴幼儿,消化吸收功能开始衰退的老人。小分子活性肽对人体中的乳酸菌、双岐杆菌和酵母菌等多种有益维生物的生长有显著促进作用,这对食物营养的消化吸收具有重要意义。
现在人们常使用的氨基酸产品是人工合成的非天然复方氨基酸小分子活性肽输液。这种人工合成的非天然复方氨基酸小分子活性肽输液具有促进人体蛋白质代谢正常,纠正负氮平衡,补充蛋白质,加快伤口愈合的作用。氨基酸输液在能量供给充足的情况下,可进入组织细胞,参与蛋白质的合成代谢,获得正氮平衡,并生成酶类、激素、抗体、结构蛋白,促进组织愈合,恢复正常生理功能。但是现有人工合成的非天然复合氨基酸小分子活性肽产品都有很大的副作用,如复方氨基酸注射液可致疹样过敏反应,一旦发生需要停止用药;偶有恶心、呕吐、胸闷、心悸、发冷、发热或头痛等。人们为现有的非天然复合氨基酸小分子活性肽产品的安全性担忧。本领域技术人员渴望获得一种使用安全,从植物中提取的纯天然复合氨基酸小分子活性肽,但一直没有解决这个技术问题。

针对上述现有技术存在的问题,本发明的第一目的在于提供一种从葛仙米提取的纯天然复合氨基酸小分子活性肽;第二目的在于提供纯天然复合氨基酸小分子活性肽从植物中的提取方法;第三目的在于提供纯天然复合氨基酸小分子活性肽的制剂;第四目的在于提供纯天然复合氨基酸小分子活性肽的应用。
本发明的第一目的是这样实现的,所述的纯天然复合氨基酸小分子活性肽从葛仙米中提取,其包含有16种复合氨基酸,所述的复合氨基酸分别为门冬氨酸、丝氨酸、谷氨酸、甘氨酸、组氨酸、精氨酸、苏氨酸、丙氨酸、脯氨酸、缬氨酸、甲硫氨酸、赖氨酸、异亮氨酸、亮氨酸、苯丙氨酸、色氨酸。
葛仙米(拉丁名:Pogostemon auricularius(L.)Kassk;学名:拟球状念珠藻)俗称天仙米、天仙菜、珍珠菜,水木耳、田木耳。葛仙米蓝藻纲,念珠藻科。藻体呈胶质状、球状或其他不规则形状,蓝绿色或黄褐色。细胞球形,由多数细胞连成念珠状群体,外包胶质。异形胞位于丝体的细胞间,幼藻丝体常位于顶端。繁殖时多数产生厚壁孢子,常为串生成链状。湿润时呈绿色,干燥后呈灰黑色。附生于水中的沙石间或阴湿的泥土上。中国各地均有分布,以四川产的最著名。可以食用。具有固氮能力。含有人体必需的多种氨基酸及多糖等活性物质。具有清火、明目、抗衰老、抗感染等治疗功效。
葛仙米中含18种氨基酸,其中含人体必需的八种氨基酸,干物质葛仙米总蛋白高达56%左右,粗脂肪8.11%,碳水化合物12.69%,灰分10.88%,叶绿素30.98mg/g,抗坏血酸5.50mg/g,维生素C含量接近鲜枣,比山楂高5倍多,比柑桔高5倍;维生素B1、B2高于一般菌藻类:含矿物质15种,最丰富的有磷、硫、钙、钾、铁等,较少的有铅、硅、镁、钡、锗等,微量元素有锌、铜、锰,还有淀粉和糖类,其含钙量高于一般蔬菜,是一种极好的天然富钙营养食品。
通过对野生葛仙米的营养成分分析,得出野生葛仙米蛋白质含量达48.61%,含17种氨基酸,其中含人体7种必需氨基酸为44.619%;尤其在野生葛仙米中赖氨酸、苏氨酸的含量较高,因此将野生葛仙米与大米、玉米、小麦的加工制品配合食用,以提高蛋白质效价。
野生葛仙米脂肪酸含量为8.11%,以中碳链为主。由于中链脂肪酸组成的三酰甘油酯尚可抑制脂解作用,因而可降低血浆中必需脂肪酸的需要量,减少胆固醇的合成,另外中链脂肪酸不经过淋巴,而经过门脉血大量被肝脏截获,不会引起高脂血症。表明野生葛仙米的脂肪酸组成具有研究价值。
进一步地,所述的纯天然复合氨基酸小分子活性肽为浅黄色至棕黄色液体,其pH值为6.5-7.5。
进一步地,所述的纯天然复合氨基酸小分子活性肽在总氮含量为5.49-6.71mg/ml时,16种天然复合氨基酸总量为28.08-43.62mg/ml。
进一步地,所述的纯天然复合氨基酸小分子活性肽在总氮含量为5.49-6.71mg/ml时,16种复合氨基酸的含量分别为:门冬氨酸3.84-5.76mg/ml,丝氨酸1.82-3.38mg/ml,谷氨酸4.96-7.44mg/ml,甘氨酸1.92-2.88mg/ml,组氨酸0.72-1.08mg/ml,精氨酸0.30-1.90mg/ml,苏氨酸0.84-1.86mg/ml,丙氨酸2.00-3.00mg/ml,脯氨酸1.6-2.5mg/ml,缬氨酸1.28-1.92mg/ml,甲硫氨酸2.96-4.44mg/ml,赖氨酸2.96-4.44mg/ml,异亮氨酸0.88-1.32mg/ml,亮氨酸2.24-3.36mg/ml,苯丙氨酸0.96-1.44mg/ml,色氨酸0.28-0.52mg/ml。
本发明的第二目的是这样实现的,包括以下步骤:
S001纯化蛋白制备:取干燥的葛仙米粉碎,加入纯化水搅拌均匀,经过加热、酸解、降温、离心、过滤步骤,收集沉淀物即得葛仙米纯化蛋白。
具体为:取干葛仙米50kg,粉碎,加100-300kg纯化水,倒入提取罐中,加热至75-85±2℃,保温15-30min,加入浓盐酸250-350ml搅匀、冷却降温至10-30℃,放出提取液,以3000-5000r/min离心,倾弃上层液,收集沉淀物,即得沉淀蛋白。
S002纯化蛋白干燥:将葛仙米纯化蛋白置于干燥装置内,平铺于光滑干净的平板不锈钢容器上,平整表面,使之厚度约为1-3厘米左右,78-88±2℃条件下密闭加热干燥16-26小时,粉碎,制得葛仙米粗蛋白;
具体为,取上述沉淀蛋白平铺于光滑干净的平板不锈钢容器上,平整表面,使之厚度约为1-3厘米左右,置于鼓风干燥箱内,78-88±2℃条件下密闭加热干燥16-26小时,粉碎,得葛仙米粗蛋白。
经过研究实验,取两批纯化蛋白,在85℃下加热干燥不同时间,和一批未干燥的纯化蛋白,分别按照统一要求操作,满足酸水解条件,按照试制工艺制备,最后测定总氮及氨基酸含量生物活性小分子肽。结果如表一所示:

由此表明,纯化蛋白的干燥步骤对终产品有较大影响,相同工艺条件和处理时间内,由于干燥程度不同和未干燥,经水解后影响整个过程的中间指标控制,含水量大时,所获终产品消耗成本差别不大,但提取总氨基酸收率下降,不便于稳定控制,所以增加干燥步骤提高总氨基酸生物活性小分子肽的收率。
S003酸水解:向葛仙米粗蛋白中加入6mol/L的硫酸,最低温度110℃条件下,水解8-10小时,加水稀释,永氢氧化钙溶液调节pH7.5-8.5,过滤,取滤液即得酸水解液;
具体为,取上述干燥葛仙米粗蛋白按干燥葛仙米粗蛋白重毎1kg加入6mol/L硫酸2000-3000ml,110-125±5℃,水解8-10h,以2倍水稀释,用氢氧化钙溶液调pH值7.5-8.5,用200-300目滤布过滤,去除硫酸钙沉淀,得酸水解液。
氢氧化钙溶液为40-60g生石灰加纯化水100ml溶解制得。
S004酶解:取上述酸水解液加热升温至30-50℃,用6mol/L氢氧化钠溶液调节pH值7.0-9.0,加入按干葛仙米重0.1%-0.4%胰酶进行水解3-8小时,加热至70-100℃,保温10-30min,冷却降温至10-50℃,用300-600目滤网过滤,离心,得酶解液;
S005层析:取酶解液经阴离子交换柱后,用水冲洗至pH值6.5,用氨水洗脱,收集洗脱液,pH值在8.0-9.0,加入氢氧化钠,减压,浓缩去氨水,用6mol/L盐酸调pH至6.6-7.6,零下20℃保存24-34h,得冷冻液;
具体为:取酶解液上阴离子交换柱后,用水冲洗至pH值6.5,用1.8-2.8mol/L氨水洗脱,收集洗脱液,pH值在8.0-9.0,每公斤干葛仙米加入1.0-3.0g氢氧化钠减压浓缩去氨水,浓缩至干葛仙米重1/3的量,用6mol/L盐酸调pH至6.6-7.6,零下20℃保存24-34h,得冷冻液。
S006超滤:取冷冻液自然解冻,0-4℃,3000-6000r/min离心,分离酪氨酸及相关杂质,收集上清液,用5000分子量超滤膜超滤,收集过滤液,喷雾干燥,定溶,即得纯天然复合氨基酸小分子活性肽。
本发明的第三目的是这样实现的,在所述的纯天然复合氨基酸小分子活性肽中加入医学上接受的辅料制备成注射剂、口服剂、冻干粉针剂、片剂、颗粒剂。
本发明的第四目的是这样实现的,一种从植物中提取的纯天然复合氨基酸小分子活性肽在制备大米、玉米、小麦加工制品和富钙营养保健品中的应用。
本发明的第四目的是这样实现的,一种从植物中提取的纯天然复合氨基酸小分子活性肽在面膜、香皂、洗手液、洗面乳、护肤水制备中的应用。
与现有技术相比,本发明具有如下优点:
(1)通过工艺改进,收率提高,使用更安全。
(2)本发明适用于大批量从植物中提取纯天然复合氨基酸小分子活性肽,通过一次性处理大量植物,纯化时上样量大,可以进行天然复合氨基酸小分子活性肽规模化生产。

具体实施方式
实施例1
S001纯化蛋白制备:取干葛仙米50kg,粉碎,加100kg纯化水,倒入提取罐中,加热至75±2℃,保温15min,加入浓盐酸250ml搅匀、冷却降温至10℃,放出提取液,以3000r/min离心,倾弃上层液,收集沉淀物,即得沉淀蛋白。
S002纯化蛋白干燥:取上述沉淀蛋白平铺于光滑干净的平板不锈钢容器上,平整表面,使之厚度约为1厘米左右,置于鼓风干燥箱内,78±2℃条件下密闭加热干燥16小时,粉碎,得干燥粉葛仙米粗蛋白。
S003酸水解:取上述干燥葛仙米粗蛋白按干燥葛仙米粗蛋白重毎1kg加入6mol硫酸浓度2000ml,110±5℃,水解8h,以2倍水稀释,用氢氧化钙溶液(毎1kg猪脑加入氢氧化钙40~60g加纯化水100ml溶解)调pH值7.5~8.5,用200-300目滤布过滤,去除硫酸钙沉淀,得酸水解液。
S004酶解:取上述酸水解液加热升温至30~50℃,用6mol/L氢氧化钠溶液调节pH值7.0~9.0,加入按干葛仙米重0.1%~0.4%胰酶进行水解3~8小时,加热至70~100℃,保温10~30min,冷却降温至10~50℃,用300目滤网过滤,离心,得胰酶酶解液。
S005层析:取上述粗提液上阴离子交换柱后,用水冲洗至pH值6.5,用1.8mol/L氨水洗脱,收集洗脱液,pH值在8.0,每公斤干葛仙米加入1.0g氢氧化钠减压浓缩去氨水,浓缩至干葛仙米重1/3的量,用6mol盐酸调pH至6.6,﹣20℃保存24h,得冷冻液。
S006超滤:取上述冷冻液自然解冻,0℃,3000r/min离心,分离酪氨酸及相关杂质,收集上清液,用5000分子量超滤膜超滤,收集过滤液,喷雾干燥,定溶,按原液标准检定含量后即为目标物。
实施例2
S001纯化蛋白制备:取干葛仙米50kg,粉碎,加200kg纯化,倒入提取罐中,加热至80±2℃,保温20min,加入浓盐酸300ml搅匀、冷却降温至20℃,放出提取液,以4000r/min离心,倾弃上层液,收集沉淀物,即得沉淀蛋白。
S002纯化蛋白干燥:取上述沉淀蛋白平铺于光滑干净的平板不锈钢容器上,平整表面,使之厚度约为2厘米左右,置于鼓风干燥箱内,80±2℃条件下密闭加热干燥20小时,粉碎,得干燥粉葛仙米粗蛋白。
S003酸水解:取上述干燥葛仙米粗蛋白按干燥葛仙米粗蛋白重毎1kg加入6mol硫酸浓度2500ml,120±5℃,水解9h,以2倍水稀释,用氢氧化钙溶液(毎1kg猪脑加入氢氧化钙50g加纯化水100ml溶解)调pH值8.0,用250目滤布过滤,去除硫酸钙沉淀,得酸水解液。
S004酶解:取上述酸水解液加热升温至40℃,用6mol/L氢氧化钠溶液调节pH值8.0,加入按干葛仙米重0.3%胰酶进行水解4小时,加热至90℃,保温20min,冷却降温至40℃,用400目滤网过滤,离心,得胰酶酶解液。
S005层析:取上述粗提液上阴离子交换柱后,用水冲洗至pH值6.5,用2.0mol/L氨水洗脱,收集洗脱液,pH值在7.0,每公斤干葛仙米加入2.0g氢氧化钠减压浓缩去氨水,浓缩至干葛仙米重1/3的量,用6mol盐酸调pH至7.0,﹣20℃保存30h,得冷冻液。
S006超滤:取上述冷冻液自然解冻,2℃,5000r/min离心,分离酪氨酸及相关杂质,收集上清液,用5000分子量超滤膜超滤,收集过滤液,喷雾干燥,定溶,按原液标准检定含量后即为目标物。
实施例3
S001纯化蛋白制备:取干葛仙米50kg,粉碎,加300kg纯化,倒入提取罐中,加热至85±2℃,保温30min,加入浓盐酸350ml搅匀、冷却降温至30℃,放出提取液,以5000r/min离心,倾弃上层液,收集沉淀物,即得沉淀蛋白。
S002纯化蛋白干燥:取上述沉淀蛋白平铺于光滑干净的平板不锈钢容器上,平整表面,使之厚度约为3厘米左右,置于鼓风干燥箱内,88±2℃条件下密闭加热干燥26小时,粉碎,得干燥粉葛仙米粗蛋白。
S003酸水解:取上述干燥葛仙米粗蛋白按干燥葛仙米粗蛋白重毎1kg加入6mol硫酸浓度3000ml,125±5℃,水解10h,以2倍水稀释,用氢氧化钙溶液(毎1kg猪脑加入氢氧化钙60g加纯化水100ml溶解)调pH值8.5,用300目滤布过滤,去除硫酸钙沉淀,得酸水解液。
S004酶解:取上述酸水解液加热升温至50℃,用6mol/L氢氧化钠溶液调节pH值9.0,加入按干葛仙米重0.4%胰酶进行水解8小时,加热至100℃,保温30min,冷却降温至50℃,用600目滤网过滤,离心,得胰酶酶解液。
S005层析:取上述粗提液上阴离子交换柱后,用水冲洗至pH值6.5,用2.8mol/L氨水洗脱,收集洗脱液,pH值在9.0,每公斤干葛仙米加入3.0g氢氧化钠减压浓缩去氨水,浓缩至干葛仙米重1/3的量,用6mol盐酸调pH至7.6,﹣20℃保存24~34h,得冷冻液。
S006超滤:取上述冷冻液自然解冻,4℃,6000r/min离心,分离酪氨酸及相关杂质,收集上清液,用5000分子量超滤膜超滤,收集过滤液,喷雾干燥,定溶,按原液标准检定含量后即为目标物。
实施例4
取实施例1制备的目标物装袋成为散剂。
实施例5
取实施例1制备的目标物加入医学上接受的辅料制备成注射剂。

取实施例2制备的目标物加入医学上接受的辅料制备成口服剂。

Claims (8)
1.一种从植物中提取的纯天然复合氨基酸小分子活性肽,其特征在于,所述的纯天然复合氨基酸小分子活性肽从葛仙米中提取,其包含有16种复合氨基酸,所述的复合氨基酸分别为门冬氨酸、丝氨酸、谷氨酸、甘氨酸、组氨酸、精氨酸、苏氨酸、丙氨酸、脯氨酸、缬氨酸、甲硫氨酸、赖氨酸、异亮氨酸、亮氨酸、苯丙氨酸、色氨酸。
2.根据权利要求1所述的一种从植物中提取的纯天然复合氨基酸小分子活性肽,其特征在于,所述的纯天然复合氨基酸小分子活性肽为浅黄色至棕黄色液体,其pH值为6.5-7.5。
3.根据权利要求2所述的一种从植物中提取的纯天然复合氨基酸小分子活性肽,其特征在于,所述的纯天然复合氨基酸小分子活性肽在总氮含量为5.49-6.71mg/ml时,16种天然复合氨基酸总量为28.08-43.62mg/ml。
4.根据权利要求3所述的一种从植物中提取的纯天然复合氨基酸小分子活性肽,其特征在于,所述的纯天然复合氨基酸小分子活性肽在总氮含量为5.49-6.71mg/ml时,16种复合氨基酸的含量分别为:门冬氨酸3.84-5.76mg/ml,丝氨酸1.82-3.38mg/ml,谷氨酸4.96-7.44mg/ml,甘氨酸1.92-2.88mg/ml,组氨酸0.72-1.08mg/ml,精氨酸0.30-1.90mg/ml,苏氨酸0.84-1.86mg/ml,丙氨酸2.00-3.00mg/ml,脯氨酸1.6-2.5mg/ml,缬氨酸1.28-1.92mg/ml,甲硫氨酸2.96-4.44mg/ml,赖氨酸2.96-4.44mg/ml,异亮氨酸0.88-1.32mg/ml,亮氨酸2.24-3.36mg/ml,苯丙氨酸0.96-1.44mg/ml,色氨酸0.28-0.52mg/ml。
5.一种权利要求1-4任一所述的一种从植物中提取的纯天然复合氨基酸小分子活性肽的提取方法,其特征在于,包括以下步骤:
S001纯化蛋白制备:取干燥的葛仙米粉碎,加入纯化水搅拌均匀,经过加热、酸解、降温、离心、过滤步骤,收集沉淀物即得葛仙米纯化蛋白;
S002纯化蛋白干燥:将葛仙米纯化蛋白置于干燥装置内,平铺于光滑干净的平板不锈钢容器上,平整表面,使之厚度约为1-3厘米左右,78-88±2℃条件下密闭加热干燥16-26小时,粉碎,制得葛仙米粗蛋白;
S003酸水解:向葛仙米粗蛋白中加入6mol/L的硫酸,最低温度110℃条件下,水解8-10小时,加水稀释,永氢氧化钙溶液调节pH7.5-8.5,过滤,取滤液即得酸水解液;
S004酶解:取上述酸水解液加热升温至30-50℃,用6mol/L氢氧化钠溶液调节pH值7.0-9.0,加入按干葛仙米重0.1%-0.4%胰酶进行水解3-8小时,加热至70-100℃,保温10-30min,冷却降温至10-50℃,用300-600目滤网过滤,离心,得酶解液;
S005层析:取酶解液经阴离子交换柱后,用水冲洗至pH值6.5,用氨水洗脱,收集洗脱液,pH值在8.0-9.0,加入氢氧化钠,减压,浓缩去氨水,用6mol/L盐酸调pH至6.6-7.6,零下20℃保存24-34h,得冷冻液;
S006超滤:取冷冻液自然解冻,0-4℃,3000-6000r/min离心,分离酪氨酸及相关杂质,收集上清液,用5000分子量超滤膜超滤,收集过滤液,喷雾干燥,定溶,即得纯天然复合氨基酸小分子活性肽。
6.一种权利要求1-4任一所述的一种从植物中提取的纯天然复合氨基酸小分子活性肽的制剂,其特征在于,在所述的纯天然复合氨基酸小分子活性肽中加入医学上接受的辅料制备成注射剂、口服剂、冻干粉针剂、片剂、颗粒剂。
7.一种权利要求1-4任一所述的一种从植物中提取的纯天然复合氨基酸小分子活性肽在制备大米、玉米、小麦加工制品和富钙营养保健品中的应用。
8.一种权利要求1-4任一所述的一种从植物中提取的纯天然复合氨基酸小分子活性肽在面膜、香皂、洗手液、洗面乳、护肤水制备中的应用。
