Abstract
本发明公开了一种大建中汤半固体制剂及其制备方法。该大建中汤半固体制剂是由下述重量配比的原料药制备而成:花椒4.2‑7.8份、干姜8.4‑15.6份、人参4.2‑7.8份、饴糖21‑39份。本发明大建中汤半固体制剂具有温中补虚、降逆止痛的功效,同时兼有滋补、缓和的治疗作用;该大建中汤半固体制剂能够改善脾阳虚证大鼠的一般状态,减轻消瘦、神疲乏力、形寒肢冷、泄泻等症状,增加进食量和体重,升高肛温,使游泳耐力显著提高(p<0.01),使脏器指数明显升高(p<0.01),能够有效治疗脾阳虚证。此外,本发明大建中汤半固体制剂具有优异的色泽、杂质、风味和组织形态,其体积小、稳定性好、易保存、口感好、服用方便,具有很好临床推广应用价值。

技术领域
本发明具体涉及一种大建中汤半固体制剂及其制备方法。
背景技术
大建中汤出自《金匮要略》,由人参(处方多用党参)、干姜、蜀椒、饴糖组成,为温里剂,具有温中补虚,降逆止痛之功效。主治中阳衰弱,阴寒内盛之脘腹剧痛证,现在多用于改善术后肠梗阻。
中药水煎液剂量大,易变质,不稳定,煎膏剂是中药传统剂型之一,它的优点是浓缩制成,体积小,便于服用,另外,膏剂含有炼蜜或糖,有滋补强身作用,所以中医多用来治疗慢性病,而且是滋补类成药中较好的剂型,而目前,并没有关于大建中汤半固体制剂的报道。
发明内容
为解决上述问题,本发明提供了一种大建中汤半固体制剂,它是由下述重量配比的原料药制备而成:
花椒4.2-7.8份、干姜8.4-15.6份、人参4.2-7.8份、饴糖21-39份。
进一步地,它是由下述重量配比的原料药制备而成:
花椒6份、干姜12份、人参6份、饴糖30份。
进一步地,它是以所述原料药的药粉、或水或有机溶剂提取物为活性成分,加上药学或保健品中常用的辅料制备而成的制剂。
进一步地,所述辅料由0.7-1.3重量份混合胶和0.7-1.3重量份矫味剂组成,优选为由1.0重量份混合胶和1.0重量份矫味剂组成。
进一步地,所述半固体制剂为煎膏剂或中药果冻。
本发明还提供了上述煎膏剂的制备方法,它包括以下步骤:
(1)按照权利要求1或2所述配比称取原料药;
(2)取花椒、干姜、人参,混匀,加6~10倍重量的水煎煮30~80min,过滤,加饴糖浓缩至相对密度为1.30~1.50,即得。
进一步地,步骤(2)中,所述水的加入量为10倍重量;所述煎煮时间为30min;所述相对密度为1.40。
本发明还提供了上述中药果冻的制备方法,它包括以下步骤:
(1)按照权利要求3或4所述配比称取原辅料;
(2)按照权利要求6或7所述方法制备煎膏剂;
(3)取混合胶,搅拌状态下依次加入步骤(2)所得煎膏剂、矫味剂,加热熬煮,趁热倒入模具,冷却,灭菌即得。
本发明还提供了上述的半固体制剂在制备温中补虚的药物、食品或保健食品中的用途。

(2)取花椒、干姜、人参,混匀,加10倍量(v/w;ml/g)水煎煮30min,过滤,加饴糖浓缩至相对密度为1.30~1.40,即得煎膏剂;

(3)取混合胶,搅拌状态下依次加入煎膏剂、矫味剂,浓缩至固定体积,趁热将胶体倒入模具中,静置冷却,至凝胶成型,灭菌即得大建中汤果冻。
实施例4、本发明大建中汤半固体制剂的制备
配方:花椒6g、干姜12g、人参6g、饴糖30g、混合胶1g、矫味剂0.7g。
制备方法:同实施例1。
实施例5、本发明大建中汤半固体制剂的制备
配方:花椒6g、干姜12g、人参6g、饴糖30g、混合胶1g、矫味剂1.3g。
制备方法:同实施例1。
以下通过实验例证明本发明大建中汤半固体制剂的有益效果。
实验例1:本发明药物制剂的口感测试
(1)实验方法
评价方法:选取30名有食品专业背景的实验人员,分别从色泽、杂质、风味、组织形态四个方面对实施例1~5制得的大建中汤果冻的口感进行评价。每个方面5分,口感总分20分。
口感评价标准如下:
色泽:制剂应具有其应有的色泽,且均匀一致,无分层无斑点。5分:制剂呈棕黄色,且均匀一致,无分层无斑点;4分:制剂呈棕褐色或浅棕黄色,且均匀一致,无分层无斑点;3分:制剂呈棕黄色、棕褐色或浅棕黄色,且基本均匀一致,无明显分层和斑点;2分:制剂呈棕黄色、棕褐色或浅棕黄色,颜色稍有分层,上浅下深;1分:制剂呈棕黄色、棕褐色或浅棕黄色,颜色有明显分层,上浅下深。
杂质:制剂应不含正常视力可见外来杂质。5分:无正常视力可见外来杂质;4分:正常视力偶见外来杂质;3分:正常视力可见少量外来杂质;2分:正常视力可见明显外来杂质;1分:正常视力可见较多外来杂质。
风味:制剂应具有品种应有的风味,无异味。5分:酸甜适宜,口感柔和,回味麻味不刺激,略带苦味;4分:酸甜味不足或过重,回味麻味不刺激,略带苦味;3分:偏酸或偏甜,回味麻味不刺激,略带苦味;2分:酸甜适宜,回味麻味刺激,略带苦味和涩味;1分:酸甜味不足或过重、偏酸或偏甜,回味麻味刺激,略带苦味和涩味。
组织形态:制剂呈凝胶状,脱离包装容器后能基本保持原有形态,组织柔软适中,按压富有弹性。5分:脱离包装容器后能基本保持原有形态,组织柔软适中,按压富有弹性;4分:脱离包装容器后能基本保持原有形态,组织略偏硬或偏软,按压弹性不足;3分:脱离包装容器后能基本保持原有形态,组织过硬,按压变形后较难复原;2分:脱离包装容器后能基本保持原有形态,组织过软,按压易破碎;1分:脱离包装容器后能较难保持原有形态,粘连容器壁或者破碎。
(2)实验结果

口感评价结果如表1所示,可以看出,本发明实施例1制得的大建中汤半固体制剂在色泽、杂质、风味、组织形态四个方面均取得了满分,综合效果最佳。
所以,本发明实施例1特定原辅料配比下制得的大建中汤半固体制剂呈棕黄色,且均匀一致,无分层无斑点,无正常视力可见外来杂质,酸甜适宜,口感柔和,回味麻味不刺激,略带苦味,脱离包装容器后能基本保持原有形态,组织柔软适中,按压富有弹性,口感评分最高。
实验例2:本发明药物制剂的治疗效果
(1)实验药物
大建中汤:配方:花椒6g、干姜12g、人参6g、饴糖30g。制备方法:(1)按照配方称取花椒、干姜、人参,混匀,加10倍量(v/w;ml/g)水煎煮30min,过滤,加饴糖浓缩至浓度为0.563g/ml,即得大建中汤。
附子理中丸:购买市售产品。
(2)实验方法
取体质量200g左右的SPF级SD大鼠,将大鼠随机分为4组(模型组、中药组、阳性组、正常组),每组12只。采用“饮食失节+劳倦+苦寒泻下”法复制脾阳虚证动物模型:模型组、中药组、阳性组大鼠于实验第1天开始饱食1日,禁食2日,不禁水,同时每日游泳至力竭,连续14d;正常组大鼠正常饮水进食,在其他组大鼠游泳同时置于水深5cm的同温水域。模型组、中药组、阳性组大鼠于实验第15d起灌服100%番泻叶水煎液,1mL/100g,每日1次,连续14d;正常组大鼠灌服37℃等剂量纯水。
于动物模型复制第28d,通过对体质量、肛温、进食量、游泳时间的组间差异性比较,客观化评价动物模型复制是否成功,对不符合评价标准的实验动物予以剔除。脾阳虚证动物模型复制成功后,于实验第29d晚上给药,中药组大鼠灌服0.563g/ml大建中汤,阳性组大鼠灌服0.15g/ml附子理中丸溶液;1mL/100g,一日一次,连续14d,自由进食饮水;空白组和模型组灌服37℃等剂量纯水,自由进食饮水。给药同时,每天早上模型组、中药组、阳性组按照脾阳虚模型复制(即灌服番泻叶)所述相应干预因素同期处理。末次给药后称量大鼠大鼠体重,观察皮毛状态,测量进食量、肛温、游泳时间,处死大鼠后取出脾脏、胸腺称重,然后根据大鼠一般状态、体质量、进食量、游泳时间、肛温、脏器指数结合SPSS 21.0进行数据分析,评价药物的作用。
(3)实验结果

与正常组比*p<0.05,**p<0.01;与模型组比#p<0.05,##p<0.01。
实验结果如表2、表3所示。从实验结果可以看出,本发明的大建中汤制剂能够改善脾阳虚证大鼠的一般状态,减轻消瘦、神疲乏力、形寒肢冷、泄泻等症状,增加进食量和体重,升高肛温,使游泳耐力显著提高(p<0.01),使脏器指数明显升高(p<0.01),有效治疗脾阳虚证。
实验结果表明,本发明提供的大建中汤能够明显改善脾阳虚证大鼠消瘦、泄泻、神疲乏力等各项症状,说明本发明提供的大建中汤能够有效治疗脾阳虚证。
综上,本发明提供的大建中汤半固体制剂具有滋补、缓和的作用,同时在治疗中阳衰弱,阴寒内盛之脘腹剧痛证方面具有显著效果;该大建中汤半固体制剂能够有效治疗脾阳虚证。而且该大建中汤半固体制剂本身具有优异的色泽、杂质、风味和组织形态,其稳定性好、易保存、口感好、服用方便,具有很好的临床推广应用价值。
Claims (10)
1.一种大建中汤半固体制剂,其特征在于:它是由下述重量配比的原料药制备而成:
花椒4.2-7.8份、干姜8.4-15.6份、人参4.2-7.8份、饴糖21-39份。
2.根据权利要求1所述的半固体制剂,其特征在于:它是由下述重量配比的原料药制备而成:
花椒6份、干姜12份、人参6份、饴糖30份。
3.根据权利要求1或2所述的半固体制剂,其特征在于:它是以所述原料药的药粉、或水或有机溶剂提取物为活性成分,加上药学或保健品中常用的辅料制备而成的制剂。
4.根据权利要求3所述的半固体制剂,其特征在于:所述辅料由0.7-1.3重量份混合胶和0.7-1.3重量份矫味剂组成,优选为由1.0重量份混合胶和1.0重量份矫味剂组成。
5.根据权利要求1~4任一项所述的半固体制剂,其特征在于:所述半固体制剂为煎膏剂或中药果冻。
6.权利要求5所述煎膏剂的制备方法,其特征在于:它包括以下步骤:
(1)按照权利要求1或2所述配比称取原料药;
(2)取花椒、干姜、人参,混匀,加6~10倍重量的水煎煮30~80min,过滤,加饴糖浓缩至相对密度为1.30~1.50,即得。
7.权利要求6所述煎膏剂的制备方法,其特征在于:步骤(2)中,所述水的加入量为10倍重量;所述煎煮时间为30min;所述相对密度为1.40。
8.权利要求5所述中药果冻的制备方法,其特征在于:它包括以下步骤:
(1)按照权利要求3或4所述配比称取原辅料;
(2)按照权利要求6或7所述方法制备煎膏剂;
(3)取混合胶,搅拌状态下依次加入步骤(2)所得煎膏剂、矫味剂,加热熬煮,趁热倒入模具,冷却,灭菌即得。
9.权利要求1~5任一项所述的半固体制剂在制备温中补虚的药物、食品或保健食品中的用途。
10.根据权利要求9所述的用途,其特征在于:所述药物或保健食品能够治疗脾阳虚证。
