Abstract
本发明提供了一种能补气、养颜的复方人参阿胶片的制备方法，其特征在于制备过程中将人参粉碎、过筛、干燥得到人参粉，通过水热浸提法提取人参皂甙Rg1；再以提取人参皂甙Rg1的剩余物为原料超声提取得到人参多糖；将生姜粉碎机粉碎、加入95%乙醇浸提得到姜辣素；黄芪粉、白芍粉、黄连粉一同放入中药提取设备中，回流提取得清膏，减压干燥、粉碎得细粉，将其与阿胶粉混合；将上述所得物料与微晶纤维素、糊精、麦芽糊精、食用葡萄糖混合均匀，再缓慢加入粘合剂食用酒精，制成软硬适中的材料；将混合制成的软材紧握成团，制备软材，制粒，干燥，过筛，整粒，烘干，加入微粉硅胶、润滑剂硬脂酸镁，混合均匀，用单冲压片机压片，按照生产规格包装。
Description
技术领域
本发明属于中药制剂领域，涉及了一种含有阿胶和人参、生姜、黄芪、白芍、黄连几种中药的有效活性成分配伍制备成一种片剂的制备方法，这种片剂具有补气、养颜的功效。
背景技术
人参能加强动物高级神经活动的兴奋和抑制过程。能增强机体对一切非特异性刺激的适应能力，能减少疲劳感。研究表明人参皂甙Rg1的抗疲劳作用显著，中性皂甙（Rb1、Rb2、Rc等）无抗疲劳作用。人参皂甙中具有收敛作用的成分较多，能调节真皮水份平衡，可抑制黑色素的形成，使皮肤变得光滑、柔嫩。而人参多糖是一种免疫增强剂，对免疫力低、抵抗力差等问题具有调理作用。
生姜可解毒、发汗解表、温中止呕、润肺止咳；用于脾胃虚寒、胃气不和、食欲减退、恶心呕吐、风寒或咳嗽，以及风寒感冒、恶风发热、鼻塞头痛等症。尤其是生姜中的活性成分姜辣素能促消化姜所含的姜辣素能刺激舌头上的味觉神经，也能促进胃肠蠕动，增加消化液的分泌，增强消化功能，可以增进食欲。生姜中的姜辣素还有抗衰老作用，可以清除人体由于新陈代谢作用产生的氧自由基，这种能力比维生素E还要强。
阿胶具有补血滋阴，润燥，止血。用于血虚萎黄，眩晕心悸，心烦不眠，肺燥咳嗽等作用。阿胶通过补血而滋润皮肤，利于皮肤保健，长期服用可使脸色红润，肌肤细嫩有光泽，是滋养皮肤、美容养颜之佳品。
黄芪有增强免疫功能、增强机体耐缺氧及应激能力、促进机体代谢、改善心功能、降压作用、保肝作用、调节血糖、抗菌及抑制病毒作用。预防传染病现代医学研究表明，黄芪内含而多种抗菌有效成分，而且能增强机体的免疫功能，因此还能用于预防某些传染病的发生。黄芪对治疗慢性阑尾炎也有效果，因为黄芪具有脱毒、生肌的功效。
白芍能降低转氨酶含量，让病变的肝脏逐渐恢复，还能分解各种有毒性的物质。对于肠胃疼痛可以很好的缓解。此外白芍可以有效防止色素的沉着，让皮肤变得细腻有光泽，因此具有美容护肤的功效，对于月经不调也有很好的调理作用。
黄连味苦，寒。归心、脾、胃、肝、胆、大肠经。清热燥湿，泻火解毒。用于湿热痞满，呕吐吞酸，泻痢，黄疸，高热神昏，心火亢盛，心烦不寐，心悸不宁等。黄连外用研末调敷或摊纱布上贴敷，有收敛止痒之效，也可以用于炎症性、化脓性皮肤病，如脓疱疮、湿疹、黄水疮等。黄连内服，对于急性皮肤炎症或皮肤化脓性感染既清热泻火解毒，又能从深层次洁肤护肤。
发明内容
本发明提供了一种能补气、美容养颜的复方人参阿胶片的制备方法，其特征在于，其中包含了人参、生姜、黄连、黄芩、白芍和阿胶几种中药的合理配伍，并通过了不同提取工艺，将其中几种药材特定的有效成分提取出来，通过现代压片技术将其制备成易于携带、方便服用的片剂。
本发明采用的技术方案如下：
一种能补气、养颜的复方人参阿胶片的制备方法，其特征在于，由以下步骤共同制成：
（1）水热浸提法提取人参皂甙Rg1：
将15-25重量份人参用中药粉碎机粉碎，过80-100目筛，在40-50℃的烘箱中烘干，得到人参粉，待用；水热浸提法提取人参皂甙Rg1：向粉碎的人参粉末中按1:15-20加入蒸馏水至中药提取罐中，充分搅拌均匀，60-70℃下浸提2-3次，每次6h，离心，取上层清液，得到人参皂甙Rg1的滤液、浓缩、减压干燥得人参皂甙Rg1粗粉，粉碎或者研磨待用；
（2）超声辅助提取人参多糖：
向（1）提取人参皂甙Rg1后的剩余物中，根据原人参粉加入量，按1:30-40加入蒸馏水，混匀浸泡，在50-55℃下超声提取40-50min，离心，将上清液浓缩、冷却，边搅拌边加入乙醇，于4-6℃冰箱中静置过夜，离心，沉淀依次用乙醚、丙酮洗涤3-5次后，放入60-70℃干燥箱中烘干至恒重，得到人参多糖；
（3）乙醇浸提法提取生姜姜辣素：
将4-8重量份生姜粉碎，按1:40-50加入90-95%乙醇混匀浸泡，在60-65℃下浸提60-70min，离心，将上清液浓缩、冷却，放入45-60℃干燥箱中烘干至恒重，得到姜辣素；
（4）阿胶粉与黄芪粉、白芍粉、黄连粉水提物的混合：
将12-20重量份阿胶用粉碎机粉碎，过80-100目筛，得到可直接入药的阿胶细粉；将8-10重量份黄芪粉、6-7重量份白芍粉、3-4重量份黄连粉混合，按1:20-25加入蒸馏水，加入到中药提取设备中，密封放置，浸泡12h，加热至60-80℃回流提取1-2h，过滤，将过滤浓缩至相对密度为1.2-1.3的清膏，在60-75℃下减压干燥至恒重，冷却，粉碎成细粉，与阿胶细粉混合，待用；
（5）复方人参阿胶片的制备：
将5-10重量份微晶纤维素、12-18重量份糊精、混合，研磨粉碎，过40目筛后，与（1）、（2）、（3）（4）中所得物料混合，加入1-2重量份麦芽糊精、2-4重量份食用葡萄糖，混合均匀，缓慢加入粘合剂食用酒精，制成软硬适中的材料；将混合制成的软材紧握成团，制备软材，制粒，于65-75℃干燥30-40min，过16-18目筛，整粒，再置于65-70℃烘箱内烘干20-30min，加入1-2重量份微粉硅胶、0.5-1重量份润滑剂硬脂酸镁，混合均匀，用单冲压片及压片，按生产规格包装。
其中步骤（2）中边搅拌边加入乙醇使乙醇的量达到80%-85%（V/V），且超声功率为200W，离心转速为6000-8000rpm处理5-8min。步骤（5）中所述干燥后物料含水量为4%-6%，每片规格重0.4g-0.5g。
与现有技术相比本发明的有益效果：
将人参经过粉碎、过筛、烘干后得到人参细粉，用水热浸提法提取得到具有抗氧化、防治衰老功效的人参皂甙；以热水浸提人参皂甙后的剩余物为原料，超声提取得到具有清除自由基、抗脂质过氧化能力的人参多糖；运用乙醇浸提法对生姜粉浸提得到具有抗氧化活性的姜辣素，对DPPH自由基和H2O2的清除能力较强。
将阿胶粉碎成细粉与黄芩、白芍、黄连用水煎煮后的提取物混合，再与人参皂甙、人参多糖、姜辣素等活性成分协同抗氧化，加入辅料混合制粒、压片，包装。
具体实施方式
下面通过具体的实施例对本发明做进一步的详细描述，不是对本发明的限制。
实施例1
（1）人参皂甙Rg1提取物的制备：
将20重量份人参用中药粉碎机粉碎，过80目筛，在50℃的烘箱中烘干，得到人参粉，待用；水热浸提法提取人参皂甙：向人参粉末中加入1:15的蒸馏水，加入到中药提取罐中，充分搅拌均匀，在65℃温度下浸提3次，每次6h，离心，取上层清液，上清液浓缩、减压干燥得到人参皂甙Rg1提取物，粉碎或者研磨待用；
（2）人参多糖的提取：
向（1）提取人参皂甙Rg1后的剩余物中，根据原人参粉加入量，按1:40加入蒸馏水，混匀浸泡，在50-55℃下超声提取40min，离心，将上清液浓缩、冷却，边搅拌边加入乙醇，于5℃冰箱中静置过夜，离心、沉淀依次用乙醚、丙酮洗涤3-5次后，放入70℃干燥箱中烘干至恒重，得到人参多糖；
（3）姜辣素的提取：
将4重量份的生姜粉碎，按1:45加入95%乙醇混匀浸泡，在65℃下浸提60min，离心，将上清液浓缩、冷却，放入45-60℃干燥箱中烘干至恒重，得到姜辣素提取物；
（4）阿胶粉与黄芪粉、白芍粉、黄连粉水提物的混合：
将15重量份阿胶用粉碎机粉碎，过80目筛，得到可直接入药的阿胶细粉；将8重量份黄芪粉、6重量份白芍粉、3重量份黄连粉混合，按1:25加入蒸馏水，加入到中药提取设备中，密封放置，浸泡12h，加热至70℃回流提取2h，过滤，将过滤浓缩至相对密度为1.2-1.3的清膏，在60℃下减压干燥至恒重，冷却，粉碎或研磨成细粉，与阿胶细粉混合，待用；
（5）复方人参阿胶片的制备：
将8重量份微晶纤维素、15重量份糊精、混合，研磨粉碎，过40目筛后，与（1）、（2）、（3）（4）中所得物料混合，加入2重量份麦芽糊精、3重量份食用葡萄糖，混合均匀，缓慢加入粘合剂食用酒精，制成软硬适中的材料；将混合制成的软材紧握成团，制备软材，制粒，于65℃干燥40min，过16-18目筛，整粒，再置于65℃烘箱内烘干30min，加入2重量份微粉硅胶、0.5重量份润滑剂硬脂酸镁，混合均匀，用单冲压片机压片，按生产规格包装。
其中步骤（2）中边搅拌边加入乙醇使乙醇的量达到80%（V/V），且超声功率为200W，离心转速为8000rpm处理5min。步骤（5）中所述干燥后物料含水量为5%，每片规格重0.5g。
Claims (4)
1.一种能补气、养颜的复方人参阿胶片的制备方法，其特征在于，将人参粉碎、过筛、干燥得到人参粉，通过水热浸提法提取人参皂甙Rg1；再以人参皂甙Rg1提取后的剩余物为原料超声提取得到人参多糖；将生姜粉碎机粉碎、乙醇浸提得到姜辣素；黄芪粉、白芍粉、黄连粉一同放入中药提取设备中回流提取得清膏，减压干燥、粉碎得细粉，将其与阿胶粉混合；将上述所得物料与微晶纤维素、糊精、麦芽糊精、食用葡萄糖混合均匀，再缓慢加入粘合剂食用酒精，制成软硬适中的材料；将混合制成的软材紧握成团，制备软材，制粒，干燥，过筛，整粒，烘干，加入微粉硅胶、润滑剂硬脂酸镁，混合均匀，用单冲压片机压片，按照生产规格包装。
2.根据权利要求1中所述的一种补气、养颜的复方人参阿胶片的制备方法，其特征在于：
（1）水热浸提法提取人参皂甙Rg1：
将15-25重量份人参用中药粉碎机粉碎，过80-100目筛，在40-50℃的烘箱中烘干，得到人参粉，待用；水热浸提法提取人参皂甙Rg1：向粉碎的人参粉末中按料液比1:15-20加入蒸馏水投至中药提取罐，充分搅拌均匀，60-70℃下浸提2-3次，每次6h，离心，取上层清液，得到人参皂甙Rg1的滤液、浓缩、减压干燥得人参皂甙Rg1粗粉，粉碎待用；
（2）超声提取人参多糖：
向（1）提取人参皂甙Rg1的剩余物中以原始人参粉的量按1:30-40加入蒸馏水，混匀浸泡，在50-55℃下超声提取40-50min，离心，将上清液浓缩、冷却，边搅拌边加入乙醇，于4-6℃冰箱中静置过夜，离心、沉淀用乙醚、丙酮洗涤3-5次后，放入60-70℃干燥箱中烘干至恒重，得到人参多糖；
（3）乙醇浸提法提取生姜姜辣素：
将4-8重量份生姜粉碎、按1:40-50加入90-95%乙醇混匀浸泡，在60-65℃下浸提60-70min，离心，将上清液浓缩、冷却，放入45-60℃干燥箱中烘干至恒重，得到姜辣素；
（4）阿胶粉与黄芪粉、白芍粉、黄连粉水提物的混合：
将12-20重量份阿胶用粉碎机粉碎，过80-100目筛，得到可直接入药的阿胶细粉；将8-10重量份黄芪粉、6-7重量份白芍粉、3-4重量份黄连粉混合，按1:20-25加入蒸馏水，加入到中药提取设备中，密封放置，浸泡12h，加热至60-80℃回流提取1-2h，过滤，将过滤浓缩至相对密度为1.2-1.3的清膏，在60-75℃下减压干燥至恒重，冷却，粉碎成细粉，与阿胶细粉混合，待用；
（5）复方人参阿胶片的制备：
将5-10重量份微晶纤维素、12-18重量份糊精、混合，研磨粉碎，过40目筛后，与（1）、（2）、（3）（4）中所得物料混合，加入1-2重量份麦芽糊精、2-4重量份食用葡萄糖，混合均匀，缓慢加入粘合剂食用酒精，制成软硬适中的材料；将混合制成的软材紧握成团，制备软材，制粒，于65-75℃干燥30-40min，过16-18目筛，整粒，再置于65-70℃烘箱内烘干20-30min，加入1-2重量份微粉硅胶、0.5-1重量份润滑剂硬脂酸镁，混合均匀，用单冲压片及压片，按生产规格包装。
3.权利要求2中所述的一种补气、养颜的复方人参阿胶片的制备方法，其特征在于，步骤（2）中边搅拌边加入乙醇使乙醇的量达到80%-85%（V/V），且超声功率为200W，离心转速为6000-8000rpm处理5-8min。
4.权利要求2中所述的一种补气、养颜的复方人参阿胶片的制备方法，其特征在于，步骤（5）中所述干燥物料含水量为4%-6%，每片重0.4g-0.5g。
