(3) sending the alcohol extraction material in the step (2) into a liquid preparation tank, adding purified water, boiling, cooling, sequentially adding sodium dodecyl sulfate, propylene glycol and glycerol, uniformly stirring, and performing heat preservation treatment to obtain a mixed solution;
(4) and (3) adding chlorhexidine acetate into the mixed solution in the step (3), uniformly stirring, adding the auxiliary material solution in the step (1), uniformly stirring, cooling, adding an aromatic dissolved by ethanol, uniformly stirring, metering volume by using purified water, continuously stirring uniformly, and subpackaging to obtain the antibacterial washing solution.
Further, in the step (2), the concentration is carried out until the relative density of the materials is 1.05-1.08 at the temperature of 60-70 ℃; adding ethanol to adjust the ethanol content of the material to 70%.

Many Chinese herbs and herbs have the functions of clearing away heat and toxic material, purging fire and drying dampness, and killing parasites and relieving itching. The human vulva and perineum are easy to generate damp-heat due to the special physiological and anatomical characteristics of the human vulva and perineum, and the medicines are usually decocted with water for fumigation and washing in clinical traditional Chinese medicine, so that the curative effect is better. In order to discuss the action mechanism and verify the clinical curative effect, pharmacodynamic experimental researches such as anti-inflammation, bacteriostasis, disinsection and the like are carried out on the externally applied foam type lavender antibacterial lotion.
1. Experimental Material
Foam type lavender antibacterial lotion (manufactured by Jinji pharmaceutical Co., Ltd., lot No. 181010), fluocinonide cream (manufactured by Guangdong Henjian pharmaceutical Co., Ltd., lot No. 181028);
escherichia coli (8099), Staphylococcus aureus (ATCC6538), and Candida albicans (ATCC10231), all provided by Kyork, Guangdong, and Trichomonas vaginalis isolated by Hunan mountain Water testing, Inc.;
a common-grade healthy adult New Zealand rabbit, provided by Taiping Biotechnology Limited, Hunan;
kunming mouse, provided by Tianqin biotechnology limited of Changsha, Hunan;
guinea pigs, supplied by the company Taiping, Hunan.
2. Method and results
2.1 anti-inflammatory assay
In a mouse concha swelling test, 80 mice are divided into a lavender foam type bacteriostatic lotion high dose group, a lavender foam type bacteriostatic lotion medium dose group, a lavender foam type bacteriostatic lotion low dose group, a fluocinolone acetonide group and a blank control group randomly, wherein 16 mice are used in each group, and the weights of the mice are 18-22 g and half of the mice are female and male. 0.03mL of dimethylbenzene was dropped on the right ear shell of each mouse, and after 30min, the right ear shell was rubbed with the corresponding drug 1 time. 1.5h after wiping, the ear shell is cut off, the left ear shell is taken as a normal control, the ear piece is taken by an 8mm puncher and weighed, and the weight difference of the left ear piece and the right ear piece is taken as the swelling degree of the ear shell. Compared with a blank control group, the result shows that the high-dose lavender foam type bacteriostatic lotion has an obvious inhibiting effect on mouse concha swelling caused by xylene (P is less than 0.05). The experimental results are shown in table 1 below.

2.2 bacteriostatic test
2.2.1 sterility test
Diluting the original foam type lavender antibacterial lotion with sterilized normal saline, and mixing with nutrient agar uniformly to make the concentrations of the foam type lavender antibacterial lotion contained in the culture medium respectively equal to original medicines, 1/2 and 1/5. After incubation at 37 ℃ for 72h, the plate was counted for bacterial and fungal colonies, respectively. And bacteria and mould do not grow in the foam lavender bacteriostatic lotion original medicine and the culture media of the dilution solutions.
2.2.2 bacteriostatic test
Coating Staphylococcus aureus and Escherichia coli on the surface of bacterial nutrition agar plate, coating Streptococcus type B on the surface of blood agar plate, coating gonococcus on the surface of gonococcus isolation culture plate, and coating Candida albicans on the surface of fungal nutrition agar plate. And (3) respectively punching holes on the flat plates by using a puncher with the aperture of 5mm, and quantitatively injecting foam type lavender bacteriostatic lotion raw medicine into the holes. After incubation at 37 ℃ for 24h, the zone diameter of each plate was measured. The results of the experiment are shown in table 2 below.

As can be seen from the experimental results in Table 2, the foam type lavender bacteriostatic lotion stock solution has obvious inhibiting and killing effects on staphylococcus aureus, escherichia coli, type B streptococcus, gonococcus and candida albicans within different action time, and the average bacteriostatic rate is higher than 93.52%; the growth of the 5 bacteria is completely inhibited after the bacteriostatic washing liquid is diluted by 1 time, and the growth of staphylococcus aureus, escherichia coli and streptococcus b can still be completely inhibited after the bacteriostatic washing liquid is diluted by 5 times. The foam type lavender antibacterial lotion has strong antibacterial effects on staphylococcus aureus, escherichia coli, streptococcus B, gonococcus and candida albicans.
2.2.3 determination of minimum inhibitory concentration
Diluting the foam type lavender antibacterial lotion with sterilized normal saline, mixing with nutrient agar uniformly to make the final concentration of the antibacterial lotion in the culture medium 1/1, 1/2 and 1/5, and pouring into a flat plate. Diluting each strain with sterilized normal saline to 3 hundred million mL-1One loop was inoculated on each plate and blank control plate, 3 groups of each strain were prepared. After culturing at 37 ℃ for 24h, observing, and taking the highest dilution of the foam type lavender bacteriostatic lotion which can completely inhibit the bacterial growth as the MIC of the lavender bacteriostatic lotion. The result shows that the bacteriostatic lotion diluted by 1 time has inhibitory effect on staphylococcus aureus, escherichia coli, streptococcus B, gonococcus and candida albicans. After the original liquid medicine is diluted by 5 times, the growth of the 5 bacteria is completely inhibited.
2.3 insecticidal test
Subculturing vaginal infusorian collected from clinical patients at 1640, subculturing at 37 deg.C, adding foam type lavender antibacterial lotion crude drug into each tube when the vaginal infusorian grows to reach concentration of more than 20 infusorian bodies in each visual field under microscope to make the concentrations in the tubes respectively 1/2, 1/4, and 1/5, making 3 groups in parallel, shaking, and culturing. And performing microscopic examination on the death condition of the trichomonas in 1min, 2min, 5min, 10min, 20min, 40min, 60min, 80min, 120min, 12h and 24h after adding the lavender foam type antibacterial lotion. The results are shown in Table 3.

As can be seen from the experimental results in Table 3, the foam type lavender bacteriostatic lotion stock solution and the foam type lavender bacteriostatic lotion diluent have an obvious killing effect on trichomonas vaginalis.
2.4 multiple skin irritation tests
3 white New Zealand rabbits (2 males and 1 female) with the weight of 2.0-2.5 kg, wherein hairs on two sides of the spine of the back of the New Zealand rabbits are cut off before the test, and the hair removing ranges are about 3cm multiplied by 3cm on the left side and the right side respectively. In the test, 0.5mL of foam type lavender antibacterial lotion is uniformly smeared on the left unhaired skin, the range is 2.5cm multiplied by 2.5cm, and the right skin is used as a blank control. After 4h of application, the skin was gently rinsed with warm water to remove residue. Applied once a day for 14 days with hair removal necessary before each application. The results were observed 24h after each application. Experimental results show that the foam type lavender antibacterial lotion has no irritation to the rabbit skin irritation test result for many times.
2.5 vaginal mucosal irritation test
6 female New Zealand rabbits with the weight of 2.0-2.5 kg are divided into an experimental group and a control group, and each group comprises 3 rabbits. Inserting a blunt hose into the vagina of a New Zealand rabbit for 4-5 cm, sucking foam type lavender antibacterial lotion by using an injector, and slowly injecting 2mL of antibacterial lotion into the vagina of the New Zealand rabbit of an experimental group through a catheter; the control group was injected with an equal amount of sterilized normal saline. Killing the New Zealand rabbits 24h after the infection, dissecting, observing the vaginal tissue of the New Zealand rabbits, and taking the vaginal tissue for pathological examination. The vaginal mucosa of all tested animals in the experimental group is basically intact through visual observation, and congestion, edema and other abnormal manifestations are not seen; all the tested animals in the control group have intact vaginal mucosa and no obvious congestion, edema and other manifestations are seen. Experimental results show that the foam type lavender antibacterial lotion has no irritation to vaginal mucosa of New Zealand rabbits.
2.6 skin allergy test
White guinea pigs 32, 16 each female and male, weighing 261.0 + -16.0 g, were divided into test and control groups of 16 animals (half female and male) before testing. Hairs within 3cm by 3cm of the left and right sides of the spinal column of the back of the guinea pig were removed. During the test, 0.5mL of foam type lavender antibacterial lotion is dropped on two layers of gauze with the size of 2cm multiplied by 2cm, then the foam type lavender antibacterial lotion is pasted on the unhaired skin on the left side of the back of a guinea pig, the unhaired skin is covered by a layer of cellophane, the unhaired skin is sealed and fixed by a non-irritant adhesive tape for 6 hours, and the 7 th day and the 14 th day are respectively repeated once by the same method. Control guinea pigs were not exposed to sensitization. At 14d after the last sensitization, the test spots were applied to the depilated skin on the right side of the back of the test guinea pig as described above. The skin reactions tested were observed after 24h and 48h after drug removal. The control group was challenged with the test substance in the same manner. No irritation reaction such as erythema, edema and the like is found on the skin of the tested guinea pigs 24 hours and 48 hours after the test object is stimulated and contacted. The experimental result shows that the result of the test of the foam type lavender antibacterial lotion on the skin allergy of the guinea pig shows that no skin allergy is found.
The research proves that the externally-used foam type antibacterial lotion has the functions of clearing heat, removing dampness, killing parasites and relieving itching, and provides a theoretical basis for clinical application of the antibacterial lotion.
Due to the adoption of the technical scheme, the invention has the following beneficial effects:
(1) the invention aims to provide a pudendum washing lotion which is used for cleaning and inhibiting bacteria in private parts of men and women, gynecological inflammation and bacterial infection symptoms, has lubrication, no stimulation and good cleaning and inhibiting effects, has the characteristics of strong pertinence, remarkable curative effect, uncomplicated formula and moderate price, and has the effects of inhibiting bacteria and eliminating peculiar smell.
(2) In long-term medical practice, the inventor of the invention creatively develops a private part external-use foam type bacteriostatic lotion which has good curative effect, wide medicine source, moderate price, advanced and feasible production process and more convenient use according to the traditional Chinese medicine theory and the modern preparation technology and through years of research and verification of the effect through research experiments of pharmacodynamics, pharmacology, clinic and the like.
(3) According to the foam type lavender bacteriostatic lotion, lavender, golden cypress, radix sophorae flavescentis, fructus cnidii, borneol and menthol which are traditional Chinese medicine raw materials, chlorhexidine acetate which is a chemical medicine, glycerin, propylene glycol, polysorbate, sodium dodecyl sulfate and lavender essence which is an aromatic are adopted, and the foam type lavender bacteriostatic lotion with the effects of cleaning, bacteriostasis and peculiar smell elimination is prepared by further combining the creatively obtained preparation method. The technical problem that the acid-base imbalance of the pudendum is caused and more pathogenic bacteria are invaded due to the fact that most of female pudendum washing liquid improved in the background technology is alkaline components and the pudendum washing liquid containing the alkaline components is frequently used for washing is solved.
(4) The extrusion of the bacteriostatic lotion is in a foam state, and rich foam can be generated by light kneading, so that compared with liquid lotion, the action of kneading and foaming is omitted, and the washing is more labor-saving; the weak acid formula can keep the resistance of a weak acid environment in private parts, can effectively inhibit pudendum infection and skin infection caused by candida albicans, staphylococcus aureus and other pathogenic bacteria, and has higher effective rate and no toxic or side effect on various gynecological inflammations and bacterial infections, female puerperium, induced abortion, cleaning and bacteriostasis of menstrual periods and male and female pudendum, cleaning before and after sexual life, daily health care and the like.

Further, the aromatic is lavender essence.
The preparation method of the foam type lavender bacteriostatic lotion comprises the following steps:

(4) adding chlorhexidine acetate into the mixed solution obtained in the step (3), stirring uniformly, adding the auxiliary material solution obtained in the step (1), stirring uniformly, cooling, adding an aromatic dissolved in ethanol, stirring uniformly, fixing the volume with purified water, continuing stirring uniformly, and subpackaging to obtain the antibacterial washing solution; the temperature reduction is to reduce the temperature of the material to 46 ℃; before subpackaging, the method also comprises adjusting the pH value, namely adjusting the pH value to 6.5 by using citric acid.

A foam type lavender antibacterial lotion is prepared from the following raw materials in parts by weight: 0.6 part of lavender, 0.6 part of phellodendron bark, 0.3 part of lightyellow sophora root, 0.3 part of common cnidium fruit, 0.06 part of borneol, 0.06 part of menthol, 0.6 part of chlorhexidine acetate, 15 parts of propylene glycol, 60 parts of glycerol, 3 parts of sodium dodecyl sulfate, 1.2 parts of polysorbate and 1.25 parts of aromatic.

the concentration is to concentrate the materials to have a relative density of 1.06 at a temperature of 65 ℃; adding ethanol to adjust the ethanol content of the material to 70%; the amount of water added for the first time of decoction is 8.0 times of the weight of the materials, and the amount of water added for the second time is 6.0 times of the weight of the materials;
(3) sending the alcohol extraction material in the step (2) into a liquid preparation tank, adding purified water, boiling, cooling, sequentially adding sodium dodecyl sulfate, propylene glycol and glycerol, uniformly stirring, and performing heat preservation treatment to obtain a mixed solution; the boiling time is 18min, the cooling is to cool the materials to 70 ℃, and the heat preservation treatment is to keep the temperature at 70 ℃ for 25-30 min;
(4) adding chlorhexidine acetate into the mixed solution obtained in the step (3), stirring uniformly, adding the auxiliary material solution obtained in the step (1), stirring uniformly, cooling, adding an aromatic dissolved in ethanol, stirring uniformly, fixing the volume with purified water, continuing stirring uniformly, and subpackaging to obtain the antibacterial washing solution; the temperature reduction is to reduce the temperature of the materials to 45 ℃; before subpackaging, the method also comprises adjusting the pH value, namely adjusting the pH value to 6 by using citric acid.

the method of examples 1-5 and comparative examples 1-4 is adopted to prepare the washing liquid, the prepared washing liquid is subjected to insecticidal and bacteriostatic tests according to the method, the bacteriostatic effects of 5min, 10min and 20min are observed, and the insecticidal effects of 1min, 60min, 120min and 12h are simultaneously observed. The results of the experiment are shown in table 4 below.

As can be seen from the experimental data in Table 4, the bacteriostatic lotion prepared by the method has good bacteriostatic and insecticidal effects.

The extrusion is in a foam state, and rich foam can be generated by light kneading, so that the action of kneading and foaming is saved compared with liquid washing liquid, and the washing is more labor-saving; the weak acid formula can keep the resistance of a weak acid environment in private parts, can effectively inhibit pudendum infection and skin infection caused by candida albicans, staphylococcus aureus and other pathogenic bacteria, and has higher effective rate and no toxic or side effect on various gynecological inflammations and bacterial infections, female puerperium, induced abortion, cleaning and bacteriostasis of menstrual periods and male and female pudendum, cleaning before and after sexual life, daily health care and the like.

A foam type lavender antibacterial lotion is characterized by being prepared from the following raw materials in parts by weight: 0.2-1 part of lavender, 0.2-1 part of phellodendron bark, 0.1-0.5 part of sophora flavescens, 0.1-0.5 part of fructus cnidii, 0.02-0.1 part of borneol, 0.02-0.1 part of menthol, 0.2-1 part of chlorhexidine acetate, 5-25 parts of propylene glycol, 20-100 parts of glycerol, 1-5 parts of sodium dodecyl sulfonate, 0.4-2 parts of polysorbate and 0.25-2 parts of aromatic.

The foam type lavender bacteriostatic lotion according to claim 1, which is characterized by being prepared from the following raw materials in parts by weight: 0.3-0.9 part of lavender, 0.3-0.9 part of phellodendron bark, 0.2-0.4 part of sophora flavescens, 0.2-0.4 part of fructus cnidii, 0.03-0.09 part of borneol, 0.03-0.09 part of menthol, 0.3-0.9 part of chlorhexidine acetate, 10-20 parts of propylene glycol, 30-90 parts of glycerol, 2-4 parts of sodium dodecyl sulfonate, 0.5-1.9 parts of polysorbate and 0.3-1.9 parts of aromatic.

The foam type lavender bacteriostatic lotion according to claim 1, which is characterized by being prepared from the following raw materials in parts by weight: 0.6 part of lavender, 0.6 part of phellodendron bark, 0.3 part of lightyellow sophora root, 0.3 part of common cnidium fruit, 0.06 part of borneol, 0.06 part of menthol, 0.6 part of chlorhexidine acetate, 15 parts of propylene glycol, 60 parts of glycerol, 3 parts of sodium dodecyl sulfate, 1.2 parts of polysorbate and 1.25 parts of aromatic.
4. The foam type lavender bacteriostatic lotion according to any one of claims 1-3, wherein the foam type lavender bacteriostatic lotion is characterized in that: the aromatic is lavender essence.
5. A preparation method of the foam type lavender bacteriostatic lotion according to any one of claims 1-4, which is characterized by comprising the following steps:
(1) weighing borneol and menthol according to the parts by weight, dissolving the borneol and the menthol in ethanol respectively, adding the mixture into polysorbate, and stirring the mixture fully and uniformly to obtain an auxiliary material liquid;
(2) weighing lavender, phellodendron amurense, radix sophorae flavescentis and fructus cnidii according to parts by weight, adding water for decocting, filtering, continuously adding water for decocting filter residues, filtering, combining the two filtrates, concentrating, cooling, adding ethanol, uniformly mixing, standing, taking supernate, and recovering ethanol to obtain ethanol extraction materials;
(3) sending the alcohol extraction material in the step (2) into a liquid preparation tank, adding purified water, boiling, cooling, sequentially adding sodium dodecyl sulfate, propylene glycol and glycerol, uniformly stirring, and performing heat preservation treatment to obtain a mixed solution;
(4) and (3) adding chlorhexidine acetate into the mixed solution in the step (3), uniformly stirring, adding the auxiliary material solution in the step (1), uniformly stirring, cooling, adding an aromatic dissolved by ethanol, uniformly stirring, metering volume by using purified water, continuously stirring uniformly, and subpackaging to obtain the antibacterial washing solution.
