The inventor also finds that the complex use of the rosa canina fruit and the tremella polysaccharide can promote the transdermal absorption of active ingredients of the rosa canina fruit extract, has the capability of removing superoxide radicals and hydroxyl radicals, prevents the skin from aging, increases the skin cell activity, promotes the skin regeneration, delays the skin aging, repairs the photodamaged skin, improves the skin nutrient metabolism, makes the skin tender and smooth, removes wrinkles, increases the elasticity and prevents the aging by inhibiting the peroxidation of cell lipid.
In the invention, the preparation method of the active composition with the function of delaying skin aging comprises the following steps: mixing Rosa canina fruit extract, Tremella polysaccharide and sodium hyaluronate uniformly.
The second aspect of the invention provides application of the active composition with the function of delaying skin aging, which is applied to the fields of skin care products and cosmetics.
In a preferred embodiment, the active composition is present in the skin care product, cosmetic, from 10 to 30% by weight.

Examples
Example 1
Example 1 provides an active composition with a function of delaying skin aging, and the active composition is prepared from the following raw materials in percentage by weight: 10% of rosa canina fruit extract, 10% of tremella polysaccharide, 6% of sodium hyaluronate and 100% of water; the weight ratio of the high molecular sodium hyaluronate to the medium molecular sodium hyaluronate to the low molecular sodium hyaluronate is 1: 3: 0.4; the tremella polysaccharide and the hyaluronic acid are purchased from Shandong focus biotechnology limited company;
the preparation method of the rosa canina fruit extract comprises the following steps:
(1) freeze-drying Rosa canina fruit, crushing into powder, and sieving with 800 mesh sieve to obtain Rosa canina fruit jelly dry powder;
(2) ultrasonically washing the Rosa canina fruit jelly dry powder with aqueous solution of 80% methanol and 0.1% hydrochloric acid, collecting the washing solution, placing in a refrigerator at 4 deg.C, and standing for 24 hr; taking out, filtering to obtain filtrate I, and drying under reduced pressure at 50 deg.C to obtain dried substance;
(3) adding water into the dried product to prepare a solution, wherein the ratio of the material to the liquid is 1: 30(g/mL), placing in a refrigerator at 4 ℃, and standing for 24 hours; taking out and filtering to obtain a filtrate II, extracting for 3 times by using ethyl acetate, collecting an extract liquor II, and then carrying out reduced pressure distillation to obtain the product.
The preparation method of the active composition with the function of delaying skin aging comprises the following steps: mixing Rosa canina fruit extract, Tremella polysaccharide and sodium hyaluronate uniformly.
Example 2
Example 2 differs from example 1 in that the raw materials for preparing the active composition comprise the following components in percentage by weight: 0.5 percent of rosa canina fruit extract, 0.5 percent of tremella polysaccharide, 1 percent of sodium hyaluronate and water for 100 percent.
Example 3
Example 3 differs from example 1 in that the raw materials for preparing the active composition comprise the following components in percentage by weight: 0.5% of rosa canina fruit extract, 0.5% of tremella polysaccharide, 1% of sodium hyaluronate and water to make up to 100%; the Rosa canina fruit extract is commercially available.
Examples 4 to 6
Examples 4 to 6 each provide a cleansing liquid containing the active composition having the function of delaying skin aging, and the specific components of the essence are shown in table 1.
Comparative examples 1 to 3
Comparative examples 1-3 provide a serum having the specific composition shown in table 1.

The preparation method of the essence comprises the following steps: mixing deionized water, vitamin E acetate, glycerol, 1, 3-propylene glycol, PEG-40 hydrogenated castor oil, dipotassium glycyrrhizinate and octyl glycol, and stirring at 75 deg.C; cooling to 50 deg.C, adding the active composition and PEG-100, and mixing; adding triethanolamine to adjust pH to 5.5-7.0; cooling to below 40 deg.C, adding phenoxyethanol, ethylhexyl glycerol and essence, and mixing.
Performance testing
Detection of radical scavenging Activity by DPPH method
(1) The test principle is as follows: 1, 1-Diphenyl-2-trinitrophenylhydrazine (1, 1-Diphenyl-2-piperidinylhydrazyl, DPPH) is a stable nitrogen center organic free radical, and an ethanol solution of the stable nitrogen center organic free radical is dark purple and has strong absorption at about 517 nm; in the presence of free radical scavengers, DPPH absorption is diminished by pairing with its single electron, and gradually disappears. The degree of color fading of the DPPH ethanol solution is in a quantitative relation with the number of electrons received by the DPPH ethanol solution, so that a spectrophotometer can be used for carrying out rapid quantitative analysis, detecting the condition of scavenging free radicals and evaluating the capacity of the antioxidant for scavenging the free radicals.
(2) The test method comprises the following steps:
preparing a DPPH ethanol solution: 6mg of DPPH was weighed out accurately, dissolved in 95% ethanol and placed in a 50mL volumetric flask, and dissolved with the aid of ultrasound. The application method comprises diluting with 95% ethanol for four times to obtain DPPH ethanol solution with concentration of 3mg/100mL (76 μ M), and storing in dark (0-4 deg.C).
Pretreating a sample to be detected: the ascorbic acid standards of examples 1-3 were dissolved in deionized water and diluted to the appropriate concentration with deionized water for use.
The experimental steps are as follows: adding 2mL of sample solution to be detected into a test tube, then adding 2mL of ethanol solution of LDPPH, and uniformly mixing. Reacting in dark place at room temperature for 30min in the dark place in a dark place, and measuring absorbance Abs at 525 nm; except for the sample group, a sample solvent control group, a blank control group and a solvent control group (solvent of DPPH) are respectively arranged, and the positive control group is an ascorbic acid group.

In the formula:
T0: adding the absorbance of the sample solution to be detected with equal volume of 95% ethanol (sample solvent control group);
t: adding the absorbance of the sample solution to be detected and an isopyknic DPPH solution (sample group);
C0: absorbance of distilled water plus equal volume of 95% ethanol (solvent control);
c: absorbance of distilled water plus an equal volume of DPPH solution (blank control).
The test results are shown in Table 2.

As can be seen from Table 2, the active composition has a DPPH radical scavenging effect. Compared with a blank control group, the active composition has stronger capacity of scavenging free radicals, and the higher the content of the active ingredients, the higher the DPPH clearance rate.
2. Detection of Elastase Activity inhibition
The experimental steps are as follows: mu.L of a sample solution and 130. mu.L of a 0.1M Tris-HCl buffer solution (pH8.0) containing 1.015mM of the reaction substrate Succ-Ala-Ala-p-nitroanilide were added to a 96-well plate, incubated at 25 ℃ for 5 minutes, 15. mu.L of an elastase solution (0.5U/mL) was added, and incubation at 25 ℃ was continued for 30 minutes, and then absorbance A410 at a wavelength of 410nm was measured by a microplate reader, the sample solution including the solution prepared in examples 1-3 and an Elastatin solution (both diluted with deionized water to an appropriate concentration), wherein Elastatin solution (Elastatin enzyme inhibitor, purchased) was used as a positive control group. The absorbance was also measured as a reference solution by replacing the sample aqueous solution with deionized water.
The calculation formula of the elastase activity inhibition rate is as follows: inhibition ratio (%) ═ a0-(A410-B))/A0×100%;
Wherein:
A0: absorbance of a reference solution;
A410: is the absorbance of the sample solution;
b: absorbance of sample blank solution.
The test results are shown in Table 3.

As can be seen from table 3, the active composition has an elastase inhibitory effect, and compared to the blank control group, the active composition has a stronger elastase inhibitory effect, and the higher the content of the active ingredient is, the higher the inhibitory effect is.
3. Skin texture test
Selecting the left and right cheekbones of the face of a subject, the left and right cheeks at the intersection of the nose tip and the pupil, and the left and right sides at the intersection of the temple and the mouth corner as test areas, firstly, measuring the skin texture of the test part before applying the cosmetics by a technician by using a visia facial image analyzer, measuring five times at each point, and taking an average value. The subjects tested skin texture again after four weeks of continuous use of the cosmetics in the examples and comparative examples. The number of each measurement at the test site of the subject was counted, and the change law of the skin texture was analyzed, thereby determining the efficacy of the example and comparative example cosmetics for the improvement of the skin texture.

4. Anti-wrinkle experiment on human skin
The anti-wrinkle test of human skin is the most direct method for testing the anti-wrinkle effect. The test population is female wrinkled people with the average age of 35-45 years. The left and right cheekbones of the face, the left and right cheeks at the intersection of the nose tip and the pupil, and the left and right sides at the intersection of the temple and the mouth corner of the subject were selected as the test areas. The elasticity value of the applied part of the subject was measured using a skin elasticity tester (MPA580) before the test. The gel is applied twice a day in the morning and at night, and the photo before application is used as a control. During the experiment, the subject was unable to apply any other cosmetic. After the subjects continued to use the anti-wrinkle cosmetic for four weeks, the skin elasticity values before and after the use of the example and comparative cosmetics were compared. As shown in tables 4 and 5, the skin elasticity and texture improvement effect of the subjects of comparative examples 1 to 3 is general after 4 weeks of use, while the skin elasticity and texture improvement effect of the subjects of examples 4 to 6 is obvious after 4 weeks of use, which indicates that the rosa canina fruit extract of the present invention can fully exert its antioxidant activity, and has synergistic effect with tremella polysaccharide and hyaluronic acid, and the anti-aging effect of the product is obviously improved.

Claims (10)

1. An active composition with the function of delaying skin aging, which is characterized by comprising the following components: rosa canina fruit extract, Tremella polysaccharide and sodium hyaluronate.
2. The active composition with the function of delaying skin aging according to claim 1, wherein the weight ratio of the rosa canina fruit extract, the tremella polysaccharides and the sodium hyaluronate is (0.5-10): (0.5-10): (1-6).
3. The active composition according to claim 1, wherein the weight ratio of the rosa canina fruit extract, the tremella polysaccharide and the sodium hyaluronate is 1: 1: (0.5-0.6).
4. The active composition with the function of delaying skin aging as claimed in claim 1, wherein the preparation method of the rosa canina fruit extract comprises the following steps:
(1) freeze-drying Rosa canina fruit, pulverizing into powder, and sieving with 400-1000 mesh sieve to obtain Rosa canina fruit jelly dry powder;
(2) washing the Rosa canina fruit jelly dry powder with an alcohol aqueous solution for several times, collecting the washing liquid, placing in a refrigerator at-5-5 deg.C, and standing for 10-30 hr; taking out, filtering to obtain filtrate I, and drying at 40-60 deg.C to obtain dried substance;
(3) adding water into the dried product to prepare a solution, placing the solution in a refrigerator with the temperature of between 5 ℃ below zero and 5 ℃, and standing the solution for 10 to 30 hours; taking out and filtering to obtain a filtrate II, extracting for a plurality of times by using an ester organic solvent, collecting an organic phase extract, and then removing the solvent to obtain the organic phase-free water-based paint.
5. The active composition with skin aging delaying function according to claim 4, wherein the volume concentration of the alcohol in the alcohol aqueous solution is not less than 70%.
6. The active composition with the function of delaying skin aging according to claim 4, wherein the drying treatment is reduced pressure drying.
7. The active composition with the function of delaying skin aging according to claim 4, wherein the feed-liquid ratio of the dry matter to the water is 1: (10-100); the unit of the feed-liquid ratio is g/mL.
8. The active composition with the function of delaying skin aging according to claim 1, wherein the sodium hyaluronate is a mixture of high molecular sodium hyaluronate, medium molecular sodium hyaluronate and low molecular sodium hyaluronate; the molecular weight of the high molecular sodium hyaluronate is not less than 160 ten thousand daltons; the molecular weight of the middle molecule sodium hyaluronate is 20-160 ten thousand daltons; the molecular weight of the low molecular weight sodium hyaluronate is 1 ten thousand to 20 ten thousand daltons.
9. Use of the active composition with skin aging delaying function according to any one of claims 1 to 8, characterized by being applied to the fields of skin care products and cosmetics.
