3) inactivating enzymes in the leaching liquor obtained in the step 2), performing centrifugal separation at a high speed, and collecting upper-layer liquid;
4) adding ether into the collected upper layer liquid, extracting for 2-3 times, extracting to collect oil layer, and evaporating to remove ether.
Example 4
The invention provides a ceramide compound muscle-soothing and skin-activating factor which comprises the following components in parts by weight: 123 parts of ceramide, 11 parts of plant compound oil, 9 parts of lupin seed extract, 10 parts of platycladi seed extract, 7 parts of cocklebur fruit extract, 5 parts of crocus flower extract, 4 parts of scutellaria root extract, 3 parts of actinidia arguta extract and 2.5 parts of snail secretion filtrate.
The plant composite oil comprises 2 parts of calendula oil, 1 part of rice bran oil and 8 parts of composite oil prepared by the following steps in sequence:
1) cleaning tea seeds, cowberry fruit seeds, caraway seeds and grape seeds, drying, uniformly mixing according to the mass ratio of 2:2:2:2, and crushing to obtain mixed powder;
2) adding 1.2% (w/w) of hydrolytic protease, 0.2% (w/w) of cellulase, 0.4% (w/w) of xylanase and deionized water (the solid-to-liquid ratio is controlled to be 1g:6mL) into the mixed powder, uniformly mixing, adjusting to be alkaline, placing in an ultrasonic instrument, and carrying out enzymolysis at 42 ℃ for 35min to obtain a leaching solution;
3) inactivating enzymes in the leaching liquor obtained in the step 2), performing centrifugal separation at a high speed, and collecting upper-layer liquid;
4) adding ether into the collected upper layer liquid, extracting for 2 times, extracting the collected oil layer, and evaporating to remove ether.
Example 5
The invention provides a ceramide compound muscle-soothing and skin-activating factor which comprises the following components in parts by weight: 12 parts of ceramide, 12 parts of plant compound oil, 10 parts of lupin seed extract, 10 parts of platycladi seed extract, 7 parts of cocklebur fruit extract, 5 parts of crocus flower extract, 5 parts of scutellaria baicalensis root extract, 3 parts of actinidia arguta extract, 3 parts of snail secretion filtrate and 2 parts of yeast/rice fermentation product filtrate.
The plant composite oil comprises 2 parts of calendula oil, 2 parts of rice bran oil and 8 parts of composite oil prepared by the following steps in sequence:
1) cleaning tea seeds, cowberry fruit seeds, caraway seeds and grape seeds, drying, uniformly mixing according to the mass ratio of 3:3:1.5:1, and crushing to obtain mixed powder;
2) adding 1% (w/w) of hydrolytic protease, 0.25% (w/w) of cellulase, 0.5% (w/w) of xylanase and deionized water (the solid-to-liquid ratio is controlled to be 1g:6mL) into the mixed powder, uniformly mixing, adjusting to be alkaline, placing in an ultrasonic instrument, and carrying out enzymolysis at 38 ℃ for 38min to obtain a leaching solution;
3) inactivating enzymes in the leaching liquor obtained in the step 2), performing centrifugal separation at a high speed, and collecting upper-layer liquid;
4) adding ether into the collected upper layer liquid, extracting for 2-3 times, extracting to collect oil layer, and evaporating to remove ether.
Example 6
The invention provides a ceramide compound muscle-soothing and skin-activating factor which comprises the following components in parts by weight: 12 parts of ceramide, 12 parts of plant compound oil, 10 parts of lupin seed extract, 10 parts of platycladi seed extract, 7 parts of cocklebur fruit extract, 5 parts of crocus flower extract, 5 parts of scutellaria baicalensis root extract, 3 parts of actinidia arguta extract, 3 parts of snail secretion filtrate and 3 parts of yeast/rice fermentation product filtrate.
The plant composite oil comprises 1 part of calendula oil, 2 parts of rice bran oil and 9 parts of composite oil prepared by the following steps in sequence:
1) cleaning and drying the tea seeds, the cowberry seeds, the caraway seeds and the grape seeds according to a mass ratio of 2: 3: 2.5:1, uniformly mixing, and crushing to obtain mixed powder;
2) adding 1.2% (w/w) of hydrolytic protease, 0.2% (w/w) of cellulase, 0.5% (w/w) of xylanase and deionized water (the solid-to-liquid ratio is controlled to be 1g:6mL) into the mixed powder, uniformly mixing, adjusting to be alkaline, placing in an ultrasonic instrument, and carrying out enzymolysis at 45 ℃ for 32min to obtain a leaching solution;
3) inactivating enzymes in the leaching liquor obtained in the step 2), performing centrifugal separation at a high speed, and collecting upper-layer liquid;
4) adding ether into the collected upper layer liquid, extracting for 2-3 times, extracting to collect oil layer, and evaporating to remove ether.
Example 7
The invention provides a cream containing ceramide compound anmei activating factor, which is prepared from 1% of example 5 ceramide compound anmei activating factor and 99% of matrix, wherein the total mass of the cream is 100%.
The matrix comprises the following components in percentage by mass based on 100% of the total mass of the matrix: 3% of squalane, 3.5% of potassium cetyl phosphate, 2% of cetearyl isononanoate, 3% of tetrahydro-methyl-pyrimidine carboxylic acid, 0.1% of sorbitan stearate and 1.2% of hydrogenated lecithin; 0.1% of cyclotetrasiloxane, 1% of cyclohexasiloxane, 0.8% of phytosterol oleate, 0.1% of EDTA disodium, 3% of dipropylene glycol, 1% of Arabic gum and 0.5% of sodium hyaluronate; 1% of poly gamma-sodium glutamate, 0.25% of carbomer, 0.6% of tocopheryl acetate, 0.5% of plant preservative and the balance of deionized water.
The plant preservative is prepared by mixing chamomile essential oil, a sage extract, a magnolia flower extract and a zanthoxylum fruit extract according to a mass ratio of 5:5:1: 2.
Example 8
The invention provides a cream containing ceramide compound anmei activating factor, which is prepared from 5% of example 5 ceramide compound anmei activating factor and 95% of matrix, wherein the total mass of the cream is 100%.
The matrix comprises the following components in percentage by mass based on 100% of the total mass of the matrix: squalane 6%, potassium cetyl phosphate 1%, cetearyl isononanoate 4%, tetrahydro-methyl-pyrimidinecarboxylic acid 1%, sorbitan stearate 1%, hydrogenated lecithin 0.3%; 2% of cyclotetrasiloxane, 0.5% of cyclohexasiloxane, 2% of phytosterol oleate, 0.02% of disodium EDTA, 8% of dipropylene glycol, 0.5% of Arabic gum and 0.8% of sodium hyaluronate; 0.5% of poly-gamma-sodium glutamate, 0.5% of carbomer, 0.2% of tocopheryl acetate, 2% of plant preservative and the balance of deionized water.

Example 9
The invention provides a cream containing ceramide compound anmei activating factor, which is prepared from 3% of ceramide compound anmei activating factor in example 1 and 97% of matrix, wherein the total mass of the cream is 100%.
The matrix comprises the following components in percentage by mass based on 100% of the total mass of the matrix: 4% of squalane, 3% of potassium cetyl phosphate, 3% of cetearyl isononanoate, 2% of tetrahydro-methyl-pyrimidinecarboxylic acid, 0.5% of sorbitan stearate and 0.8% of hydrogenated lecithin; 0.8% of cyclotetrasiloxane, 0.6% of cyclohexasiloxane, 1.5% of phytosterol oleate, 0.05% of disodium EDTA (ethylene diamine tetraacetic acid), 5% of dipropylene glycol, 0.8% of Arabic gum and 0.6% of sodium hyaluronate; 0.8% of poly-gamma-sodium glutamate, 0.35% of carbomer, 0.4% of tocopheryl acetate, 1% of plant preservative and the balance of deionized water.
The plant preservative is prepared by mixing chamomile essential oil, a sage extract, a magnolia flower extract and a zanthoxylum fruit extract according to a mass ratio of 2:5:1: 2.
Example 10
The invention provides a cream containing ceramide compound anmei activating factor, which is prepared from 3% of example 3 ceramide compound anmei activating factor and 97% of matrix, wherein the total mass of the cream is 100%; the matrix was identical in composition and content to the matrix in example 9.
Example 11
The invention provides a cream containing ceramide compound anmei activating factor, which is prepared from 3% of example 4 ceramide compound anmei activating factor and 97% of matrix by the total mass of the cream being 100%, wherein the matrix is consistent with the components and the content of the matrix in example 9.
Example 12
The invention provides a cream containing ceramide compound anmei activating factor, which is prepared from 3% of example 5 ceramide compound anmei activating factor and 97% of matrix by the total mass of the cream being 100%, wherein the matrix is consistent with the components and content of the matrix in example 9.
The ceramide polypeptide compound Anmyo skin-activating cream described in examples 7 to 12 is prepared according to the conventional process in the field; and evaluating the safety of the prepared ceramide polypeptide compound skin-activating cream for soothing the muscles.

To better demonstrate the efficacy of the present invention, the efficacy experiments of the present application, as well as the efficacy experiments of the comparative examples, are given below:

The other components were in the same amounts as in example 12, except that the same amount of the platycladi seed extract was used instead of the scutellaria root extract in example 12.

The other components were in the same amounts as in example 12, except that the same amount of crocus sativus flower extract was used instead of the actinidia arguta extract in example 12.
Comparative example 4
The other components and contents are the same as those in example 12, except that the vegetable composite oil is prepared by separately purchasing catechu seed oil, cowberry seed oil, caraway seed oil and grape seed oil in a mass ratio of 3:3:1.5:1.
Comparative example 5
The other ingredients were the same as in example 12 except that phenoxyethanol/ethylhexylglycerin (0.5%) was used in place of the plant preservative in example 1 and the remainder was supplemented with deionized water.
1. Moisture retention test:
the specific test method is as follows:
reference standard: refer to QB/T4256-.
The testing personnel: 30 healthy volunteers were selected, and the ages were 30-40 years old, respectively.
And (3) testing conditions are as follows: the capacitance method skin moisture tester Corneometer CM825 is used for testing the temperature of 20-22 ℃ and the humidity of 40-60%.
The test principle is as follows: the capacitance method is used for measuring the moisture content of the human skin stratum corneum, and is based on the obvious difference of dielectric constants of water and other substances, the capacitance values of the measured skin are different according to the moisture content of the skin stratum corneum, and the parameters can represent the moisture content (MMV) of the skin.
Test samples: the experimental groups were the products provided in examples 9-12 and the control groups were the products provided in comparative examples 1-5.
The test method comprises the following steps: each area is fixedly measured at three points according to a certain sequence, and an average value of the three points is obtained. 2. Skin elasticity test
Test samples: the experimental groups were the products provided in examples 9-12 and the control groups were the products provided in comparative examples 1-5.
The experimental method comprises the following steps: the testing personnel: 45 healthy volunteers aged 30-40 years respectively were selected and randomly grouped into 5 groups. The samples were applied twice daily in the morning and evening. During the experiment, the subject was not able to apply any other cosmetic product at the experimental site.
Skin elasticity test: the skin condition at week 0 and week 4 was measured using the skin elastometer MPA580, the test site was the cheekbone, the probe repeated 3 times in the same test area during the measurement, and the average was taken, and the test results are shown in Table 3:

3. DPPH free radical scavenging test
0.2mmol/L DPPH solution is prepared by 50% ethanol and stored away from light. In the experimental group, 0.2mg/mL of solution to be tested is prepared from the products provided in examples 9-12 and the comparison group 1-5, the absorbance A of the solution to be tested is measured at 517nm, the absorbance A is measured in parallel for 3 times, the average value is taken, and the experimental result is shown in Table 3.

4: skin roughness test
1) Test samples: the experimental groups were the products provided in examples 9-12 and the control groups were the products provided in comparative examples 1-5.
2) The test method comprises the following steps: 90 wrinkled female volunteers with the average age of 30-40 years old were taken, randomly divided into 9 groups of 10, and the above test samples were used respectively. The subject takes a proper amount of cream to apply on the face, and the cream is applied once every morning and evening for 28 days continuously. During the experiment, the subject was not able to apply any other cosmetic product at the experimental site.
Data were collected on day 0 and day 28 using a skin wrinkle tester vision meter SV600, and the test site was the zygomatic region. The test parameter is skin roughness R (the difference between the highest peak and lowest valley of the skin lines), the greater the value of R, the rougher the skin and the more severe the wrinkles. The test results are shown in table 4.

As is apparent from tables 1 to 4, the technical scheme provided by the invention can deeply penetrate into the bottom layer of the skin to supplement moisture, keep moisture for a long time, effectively remove free radicals, recover the elasticity of the skin, promote the skin to recover a good barrier function, effectively lock moisture, carefully nourish the skin, and have good skin-care and anti-aging effects.
Human body trial
1. The test population: 100 people, age 30-40 years.

the matrix comprises the following components in percentage by mass based on 100% of the total mass of the matrix: 3-6% of squalane, 1-3.5% of potassium cetyl phosphate, 2-4% of cetearyl isononanoate, 1-3% of tetrahydro-methyl pyrimidine carboxylic acid, 0.1-1% of sorbitan stearate and 0.3-1.2% of hydrogenated lecithin; 0.1-2% of cyclotetrasiloxane, 0.5-1% of cyclohexasiloxane, 0.8-2% of phytosterol oleate, 0.02-0.1% of disodium EDTA (ethylene diamine tetraacetic acid), 3-8% of dipropylene glycol, 0.5-1% of Arabic gum and 0.5-0.8% of sodium hyaluronate; 0.5-1% of poly gamma-sodium glutamate, 0.25-0.5% of carbomer, 0.2-0.6% of tocopheryl acetate, 0.5-2% of plant preservative and the balance of deionized water; the plant preservative is prepared by mixing chamomile essential oil, a sage extract, a magnolia flower extract and a zanthoxylum fruit extract according to the mass ratio of 2-5:2-5:1-3: 1-2;
the ceramide compound muscle-soothing and skin-activating factor comprises the following components in parts by weight: 120-27 parts of ceramide, 10-13 parts of plant compound oil, 8-11 parts of lupin seed extract, 8-11 parts of platycladi seed extract, 6-9 parts of cocklebur fruit extract, 3-6 parts of crocus sativus flower extract, 3-6 parts of scutellaria baicalensis root extract, 2-4 parts of actinidia arguta extract and 2-4 parts of snail secretion filtrate;
the plant composite oil is mainly prepared by extracting camellia seed, cranberry seed, caraway seed and grape seed sequentially through the following steps:
1) cleaning and drying the tea seeds, the cowberry seeds, the caraway seeds and the grape seeds according to a mass ratio of 2-3: 1.5-2.5: 1-2, uniformly mixing, and crushing to obtain mixed powder;
2) adding hydrolytic protease, xylanase, cellulase and deionized water into the mixed powder, mixing uniformly, adjusting to be alkaline, placing in an ultrasonic instrument, and carrying out enzymolysis for 30-45min at 35-45 ℃ to obtain a leaching solution;
3) inactivating enzymes in the leaching liquor obtained in the step 2), performing centrifugal separation at a high speed, and collecting upper-layer liquid;
4) adding extractant into the collected upper layer liquid, extracting for 2-3 times, and collecting the grease layer.
2. The cream containing the ceramide compound anmei activating factor is characterized in that the ceramide compound anmei activating factor also comprises 2-4 parts of yeast/rice fermentation product filtrate.
3. The cream containing the ceramide compound anmei skin-activating factor as claimed in claim 1, wherein the plant compound oil in the ceramide compound anmei skin-activating factor further comprises 1-2 parts of calendula oil.
4. The cream containing the ceramide compound anmei skin-activating factor as claimed in claim 3, wherein the plant compound oil in the ceramide compound anmei skin-activating factor further comprises 1-2 parts of rice bran oil.
5. The cream containing ceramide compound anmei activating factor is characterized in that the addition amount of the hydrolytic protease is 0.5-1.5%, the addition amount of the cellulase is 0.1-0.25%, and the addition amount of the xylanase is 0.3-0.5%.
