The ceramide polypeptide compound anmei emulsion described in example 8 to example 12, and comparative emulsion sample 1 to comparative emulsion sample 8, product safety was evaluated as follows:
(1) microbiological test item experiments: according to the determination method of the total number of bacterial colonies, faecal coliform, staphylococcus aureus, pseudomonas, mould and saccharomycetes in the cosmetic hygiene standard, the microorganism detection of the ceramide polypeptide compound anmei emulsion provided by the invention is all qualified.
(2) And (3) testing sanitary chemical inspection projects: according to the method for measuring total mercury, total arsenic and total lead in the cosmetic hygiene standard, the metal elements of the ceramide polypeptide compound anmei emulsion provided by the invention are all qualified.
Efficacy experiments:
1. anti-allergy and anti-irritation
1) According to the basic principle of animal skin irritation or corrosiveness tests specified in the cosmetic hygiene code-2007, requirements and methods for the skin irritation integral mean values of the ceramide polypeptide compound anmei emulsion provided by the example 8 to the example 12 and the comparative emulsion samples 1 to 7 are less than 0.2, which indicates no irritation, and the skin irritation integral mean value of the comparative emulsion sample 8 is 1.0, which has slight irritation.
2) 20 sensitive muscle volunteers were selected and the ceramide polypeptide compound Anmyo emulsion in example 12 was continuously used for 7 days, resulting in 0 person with discomfort or allergy symptoms and 0% allergy rate. One week after the end of the trial, the volunteers of this group used a control emulsion sample of 8 for 7 days, giving rise to discomfort or allergic symptoms in 2 persons with an allergy rate of 10%. Namely, the ceramide polypeptide compound anmeiye of the invention is mild, has no stimulation and has strong anti-allergy property.
2. Moisture retention test:
the specific test method is as follows:
reference standard: refer to QB/T4256-.
The testing personnel: 55 healthy volunteers were selected, aged 30-40 years each.
And (3) testing conditions are as follows: the capacitance method skin moisture tester Corneometer CM825 is used for testing the temperature of 20-22 ℃ and the humidity of 40-60%.
The test principle is as follows: the capacitance method is used for measuring the moisture content of the human skin stratum corneum, and is based on the obvious difference of dielectric constants of water and other substances, the capacitance values of the measured skin are different according to the moisture content of the skin stratum corneum, and the parameters can represent the moisture content (MMV) of the skin.
Test samples: the experimental groups were the products provided for examples 8, 9, 12 and the control groups were comparative emulsion sample 1 to comparative emulsion sample 8.
The test method comprises the following steps: each area is fixedly measured at three points according to a certain sequence, and an average value of the three points is obtained. Then, the ceramide polypeptide compound anmei emulsion is smeared on a tested part (according to (2.0 +/-0.1) mg/cm)2The amount of (c) was applied in a single application), the skin moisture content of the test site was measured at 0h, 1h, 4h and 8h using a probe, and the average value of each group was calculated for the corresponding time point, and the results are shown in table 1.
TABLE 1
The skin moisture content increase rate calculation formula is as follows:
skin moisture content increase rate [ (% MMVt-MMV) ]0)/MMV0×100%
MMV0MMV on the skin before application
MMVtMMV of the skin t periods after application
The results are shown in Table 2.
TABLE 2
3. Skin moisture loss test.
The skin moisture loss (TEWL) tester TM300 of the German CK company is adopted for detection, and the testing method is basically consistent with the skin moisture content (MMV). The test area is a left forearm, each test area is tested for 3 times to obtain an average value, and the percutaneous water loss TEWL value is smaller, the water loss is smaller, and the water locking capacity is stronger; conversely, the weaker the water-holding capacity. The rate of change, and the rate of change of TEWL values, the lower the value, the better the water-locking effect, the results are shown in Table 3.
The lower the rate of change (TEWLt-TEWL0)/TEWL0 x 100%, the lower the water loss, the stronger the skin barrier function of the test sample, and the better the moisturizing effect. During the experiment, the subject was not able to apply any other cosmetic product at the experimental site.
TABLE 3 skin TEWL assay results
4, skin elasticity test
Test samples: the experimental groups were the products provided for examples 8, 9, 12 and the control groups were comparative emulsion sample 1 to comparative emulsion sample 8.
The experimental method comprises the following steps: the testing personnel: 55 healthy volunteers aged 30-40 years were selected, and randomly grouped into 5 groups. The samples were applied twice daily in the morning and evening. During the experiment, the subject was not able to apply any other cosmetic product at the experimental site.
Skin elasticity test: the skin condition of week 0 and week 4 was measured using the skin elastometer MPA580, the test site was the cheekbone, the probe repeated 3 times in the same test area during the measurement, and the average was taken.
The skin elasticity increase rate W ═ NT-N0)/N0×100%
N0-skin elasticity test value before application.
NT-elasticity test value T period after application.
The results of the specific skin elasticity tests are shown in table 4:
