Disclosure of Invention

The invention aims to provide a composition with anti-allergy and relieving effects, which has the functions of reducing stimulation of a allergen, inhibiting histamine release and hyaluronidase activity and repairing damaged barriers of sensitive skin. The invention also provides the essence milk containing the composition, which can relieve and repair sensitive skin, and meanwhile, the essence milk has excellent stability, excellent moisturizing and skin moistening performance, mild and low irritation and excellent skin repairing effect.

In a first aspect, the present invention provides a composition with anti-allergy and soothing effects, comprising, based on 100 parts by weight of the composition:

20-30 parts by weight of centella asiatica (CENTELLA ASIATICA) extract;

20-30 parts by weight of rhizoma Polygoni Cuspidati (Polygonum CUSPIDATUM) root extract;

10-20 parts by weight of radix Scutellariae (Scutellaria BAICALENSIS) extract;

10-20 parts by weight of tea (CAMELLIA SINENSIS) leaf extract;

5-10 parts by weight of GLYCYRRHIZA GLABRA (GLYCYRRHIZA GLABRA) root extract;

5-10 parts by weight of a CHAMOMILLA (CHAMOMILLA RECUTITA) flower extract;

5-10 weight parts of herba Rosmarini OFFICINALIS (Rosmarinus OFFICINALIS) leaf extract.

The centella asiatica (CENTELLA ASIATICA) extract is numbered 03169 in the catalogue of used cosmetic raw materials (2021 edition) in China, the POLYGONUM CUSPIDATUM (POLYGONUM CUSPIDATUM) root extract is numbered 02961 in the catalogue of used cosmetic raw materials (2021 edition) in China, and the centella asiatica (CENTELLA ASIATICA) extract and the POLYGONUM CUSPIDATUM (POLYGONUM CUSPIDATUM) root extract can generate a synergistic effect, so that the permeability and the skin thickness of capillary vessels can be improved, sensitive skin is relieved, sensitive symptoms such as irritation, redness and erythema are reduced, and an excellent relieving and anti-allergy effect is achieved.

Specific component parameters for the anti-allergy soothing compositions provided in examples 1-3 are shown in table 1.

The specific component parameters of the compositions with anti-allergy soothing effect provided by comparative examples 1-7 are shown in table 2.

It should be noted that the hydrolyzed corn starch used in comparative examples 1 to 7, which was numbered 06387 in the catalog of names of used cosmetic raw materials (2021 edition) in china, had no anti-allergy, soothing, antibacterial and anti-inflammatory effects.

The compositions provided in examples 1-3 and comparative examples 1-7 were tested as follows:

-preparing an acetic acid buffer solution with a pH of 5.0 with a glacial acetic acid diluted solution and an aqueous sodium acetate crystal solution; then, preparing a hyaluronidase solution (with the final working concentration of 1250U/mL) and a sodium hyaluronate solution (with the concentration of 0.5g/L) by using the prepared acetic acid buffer solution;

-preparing calcium chloride into an aqueous solution with a concentration of 0.25 mmol/L;

-sodium hydroxide is prepared into an aqueous solution with a concentration of 0.4 mol/L;

after sodium carbonate is prepared into an aqueous solution with the concentration of 1mol/L, 50ml of the aqueous solution of sodium carbonate is uniformly mixed with 3.5ml of acetylacetone to prepare an acetylacetone solution for later use;

weighing 1.6g of p-dimethylaminobenzaldehyde, and dissolving the p-dimethylaminobenzaldehyde in 30mL of concentrated hydrochloric acid and 30mL of absolute ethyl alcohol to prepare an Ellisib reagent;

-preparation of red blood cell suspension: phosphate Buffer (PBS) at pH 7.4 (1.8 g/L glucose was added) and citric acid (C) as anticoagulant were prepared in advance ₆ H ₅ Na ₃ O ₇ ·2H ₂ O0.02 g/ml, citric acid C ₆ H ₈ O ₇ 0.008g/ml) to treat fresh blood; subpackaging the collected blood sample by using a centrifugal tube, centrifuging for 15min at 5000r/min, removing supernatant, adding PBS, gently shaking, mixing, centrifuging again, repeatedly washing until the supernatant is colorless and transparent, finally diluting the precipitate with PBS to 2% of red blood cell suspension by mass concentration, and storing in a refrigerator at 4 ℃ for later use;

determination of suitable experimental concentrations of Sodium Dodecyl Sulfate (SDS): 0.5ml of a solution of samples to be detected (SDS) with different concentrations, which is diluted by PBS in advance, and 0.5ml of a suspension of red blood cells with the mass concentration of 2 percent are respectively added into a centrifugal tube and fully and uniformly mixed. The haemolysis rate of erythrocytes after the test composition has interacted with SDS: diluting the composition with PBS to 1% by mass, mixing 0.5ml of the solution with 0.5ml of 2% by mass of red blood cell suspension in advance, adding 0.5ml of SDS solution with experimental concentration, fully mixing uniformly, performing subsequent incubation and centrifugation, and calculating the hemolysis rate.

According to the test results in table 3, the composition of the present invention has a better anti-allergy effect through the synergistic effect between the components: the composition has obvious inhibition rate on hyaluronidase, and the composition has the effects of resisting inflammation, relieving and synergistically moisturizing; has certain inhibition effect on RBC hemolysis induced by SDS, and proves that the composition of the invention has the effects of reducing stimulation, relieving and resisting allergy; so that the red swelling of the 3 percent SLS-stimulated left forearm area is quickly eliminated, and the composition has obvious anti-stimulation and anti-red swelling effects.

Examples 4 to 6 all provide a serum, the serum is prepared by the same conventional process, the anti-allergy and soothing composition provided in example 3 is used for the serum, and other components of the serum are commercially available products.

Specific composition parameters of the essence milks of examples 4 to 6 are shown in table 4.

The essences of examples 4 to 6 used thickeners consisting of acrylic acid (ester)/C10-30 alkanol acrylate crosspolymer, xanthan gum and acacia gum (ACACIA SENEGAL) gum, which comprises, based on 100 parts by weight of the thickener: 40 parts by weight of acrylic acid (acrylate)/C10-30 alkanol acrylate crosspolymer, 30 parts by weight of xanthan gum and 30 parts by weight of acacia gum (ACACIA SENEGAL) gum.

The essential milk of examples 4 to 6 uses surfactants consisting of sodium polyacrylate, C18-21 alkane, and trideceth-6, the surfactants comprising, based on 100 parts by weight of the surfactants: 10 parts by weight of sodium polyacrylate, 40 parts by weight of C18-21 alkane and 50 parts by weight of tridecyl polyether-6.

The essential milks of examples 4 to 6 used the same stabilizers, which comprise, based on 100 parts by weight of the stabilizers:

0.8 parts by weight of ceramide NP;

5 parts by weight of caprylic/capric triglyceride;

3 parts by weight of hydrogenated lecithin;

3 parts by weight of polysorbate-80;

1.5 parts by weight of 1, 2-hexanediol;

2 parts by weight of butanediol;

0.5 parts by weight of cetyl ethylhexanoate;

0.08 parts by weight of disodium ethylenediaminetetraacetate (disodium EDTA);

the balance being water.

The essential milk provided in example 6 was tested as follows:

The essence of example 6 was applied to the left leg of 20 subjects with sensitive and itching leg skin, and a commercially available anti-sensitivity soothing cream was applied to the other leg, and TEWL values of the tested sites were measured at 0h, 6h and 12h using a Tewameter, which are important parameters for evaluating skin barrier function and skin repair after using the product, and the smaller the value, the better the skin barrier and the better the repair.

The experimental results are as follows: at 6h and 12h, the TEWL values of the area of the left leg to which the serum of example 6 was applied were decreased by 65.2% and 73.6%, respectively, while the TEWL values of the area of the right leg to which a commercial anti-allergy soothing cream was applied were decreased by 40.6% and 55.2%, respectively. The essential milk of example 6 of the present invention is capable of improving the moisture content of sensitive skin and has an excellent barrier repair function.

[ clinical experiments ]

Selecting volunteers with skin sensitivity and pruritus symptoms in the age range of 25-45 years, 40 male volunteers and 40 female volunteers, randomly dividing into two groups, namely a test group and a control group, and 40 persons in each group. Test groups: after cleaning the face every day, the essence cream of example 6 was applied before sleeping. Control group: after face cleaning every day, a certain commercially available anti-allergy soothing cream is smeared before sleep. Both groups did not use other skin care products. The service life is one month.

Evaluation criteria: the pruritus frequency is obviously reduced, the red swelling disappears, and the desquamation phenomenon of the skin is reduced to present a moist state.

The experimental results are as follows: by using the essence milk of the embodiment 6, the redness and swelling of the skin can be obviously improved after one week, the skin pruritus frequency is obviously reduced, 40 effective cases are obtained in a test group, and the effective rate reaches 100%; the anti-allergy and relieving cream sold in the market has 20 effective cases, and the effective rate is only about 50 percent. After the anti-allergic health care product is used for one month, the effective rate of a test group reaches 100 percent, and the effective rate of a commercially available anti-allergic product is only about 70 percent.

The essence milk disclosed by the invention can repair the skin barrier, and the barrier repair has an effect on relieving the skin itch. The essence lotion disclosed by the invention can be used for relieving and repairing sensitive skin. The essence milk provided by the invention has good stability, is mild and non-irritating, and has a remarkable anti-allergy repair effect.

The technical features of the embodiments described above may be arbitrarily combined, and for the sake of brevity, all possible combinations of the technical features in the embodiments described above are not described, but should be considered as being within the scope of the present specification as long as there is no contradiction between the combinations of the technical features.

1. A composition having anti-allergy soothing efficacy, comprising, based on 100 parts by weight of the composition:

20-30 parts by weight of centella asiatica (CENTELLA ASIATICA) extract;

10-20 parts by weight of radix Scutellariae (Scutellaria BAICALENSIS) extract;

10-20 parts by weight of tea (CAMELLIA SINENSIS) leaf extract;

5-10 parts by weight of GLYCYRRHIZA GLABRA (GLYCYRRHIZA GLABRA) root extract;

5-10 parts by weight of a chamomile (CHAMOMILLA RECUTITA) flower extract;

2. Use of the composition of claim 1 as an additive in skin care and cosmetics.

3. An essence milk, comprising, based on 100 parts by weight of the essence milk:

2-5 parts by weight of caprylic/capric triglyceride;

0.5-2 parts by weight of white chinlon (LIMNANTHES ALBA) seed oil;

0.1-1 parts by weight of tocopheryl acetate;

0.2-1 parts by weight of glyceryl stearate citrate;

0.1-1 parts by weight of polyglyceryl-3-methylglucose distearate;

0.2 to 0.6 part by weight of a thickener;

0.1 to 1 part by weight of a surfactant;

0.1 to 3 parts by weight of the composition of claim 1;

0.5-1 parts by weight of a stabilizer;

2-5 parts by weight of 1, 3-propanediol;

2-5 parts by weight of glycerol;

0.1 to 1 part by weight of 1, 2-hexanediol;

1-5 parts by weight of butanediol;

1-5 parts by weight of propylene glycol;

0.5-2 parts by weight of trehalose;

2-5 parts by weight of betaine;

0.01-0.1 parts by weight of oat (AVENA SATIVA) beta-glucan;

0.1-0.2 parts by weight of allantoin;

0.1 to 1 part by weight of panthenol;

0.01-0.1 parts by weight of sodium hyaluronate;

0.01-0.1 weight parts of dendrobium officinale extract;

0.1-1 weight parts of dipotassium glycyrrhizinate;

0.1-1 weight parts of p-hydroxyacetophenone;

0.01-0.05 parts by weight of essence;

the balance being water.

4. The serum according to claim 3, characterized in that the thickener consists of acrylates/C10-30 alkanol acrylate crosspolymer, xanthan gum and acacia gum (ACACIA SENEGAL).

5. The essence of claim 3, wherein the surfactant is composed of sodium polyacrylate, C18-21 alkane and trideceth-6.

6. The essence of claim 3, wherein the stabilizer comprises, based on 100 parts by weight of the stabilizer:

0.1-1 parts by weight of ceramide NP;

3-8 parts by weight of caprylic/capric triglyceride;

2-4 parts by weight of hydrogenated lecithin;

2-4 parts by weight of polysorbate-80;

1-2 parts by weight of 1, 2-hexanediol;

1-3 parts by weight of butanediol;

0.1-0.5 parts by weight of cetyl ethylhexanoate;

0.05-0.1 weight part of disodium ethylene diamine tetraacetate;

the balance being water.
