In addition, the tremella polysaccharide and the high-molecular sodium hyaluronate are compounded for use, so that a synergistic effect can be generated, the hydration capacity of skin cells is obviously enhanced, the water content of skin cuticle is increased, a moisturizing and water-locking protective film with a certain thickness can be formed on the skin surface, the percutaneous loss of active ingredients and water molecules can be reduced, the double effects of water retention and water locking are also realized, and skin is moist, does not dry, is not tight and is smooth after the tremella polysaccharide and the high-molecular sodium hyaluronate are used.
The inventor also finds that the compound use of the dog rose-hip and tremella polysaccharide can promote the transdermal absorption of active ingredients of the dog rose-hip extract, has the capability of removing superoxide radicals and hydroxyl radicals, prevents skin aging by inhibiting peroxidation of cell lipid, increases skin cell activity, promotes skin regeneration, delays skin aging, repairs light damaged skin, and simultaneously can improve skin nutrition metabolism, tender, smooth, wrinkle-removing, elasticity-increasing and aging-preventing skin.
In the invention, the preparation method of the active composition with the function of delaying skin aging comprises the following steps: mixing fructus Rosae Davuricae extract, tremella polysaccharide and sodium hyaluronate.
The second aspect of the invention provides application of the active composition with the function of delaying skin aging, and the active composition is applied to the fields of skin care products and cosmetics.
In a preferred embodiment, the active composition is present in skin care products, cosmetics in an amount of 10 to 30wt%.
The present invention will be specifically described below by way of examples. It is noted herein that the following examples are given solely for the purpose of further illustration and are not to be construed as limitations on the scope of the invention, as will be apparent to those skilled in the art in light of the foregoing disclosure.
In addition, the raw materials used are commercially available unless otherwise indicated.
Examples
Example 1
Example 1 provides an active composition with skin aging delaying function, which is prepared from the following raw materials in percentage by weight: 10% of dog tooth rose hip extract, 10% of tremella polysaccharide, 6% of sodium hyaluronate and 100% of water; the weight ratio of the high molecular sodium hyaluronate, the medium molecular sodium hyaluronate and the low molecular sodium hyaluronate is 1:3:0.4; the tremella polysaccharide and the hyaluronic acid are purchased from Shandong focus biotechnology limited company;
the preparation method of the Rosa canina fruit extract comprises the following steps:
(1) Freeze-drying fructus Rosae Davuricae, pulverizing into powder, and sieving with 800 mesh sieve to obtain fructus Rosae Davuricae powder;
(2) Ultrasonic washing fructus Rosae Davuricae lyophilized powder with 80% methanol and 0.1% hydrochloric acid water solution for 3 times, collecting the washing liquid, placing in a refrigerator at 4deg.C, and standing for 24 hr; taking out and filtering to obtain filtrate I, and drying under reduced pressure at 50deg.C to obtain dried product;
(3) Adding water into the dried product to prepare a solution, wherein the feed liquid ratio is 1:30 (g/mL) and placed in a refrigerator at 4℃for 24 hours; taking out and filtering to obtain filtrate II, extracting with ethyl acetate for 3 times, collecting extract II, and distilling under reduced pressure.
The preparation method of the active composition with the function of delaying skin aging comprises the following steps: mixing fructus Rosae Davuricae extract, tremella polysaccharide and sodium hyaluronate.
Example 2
Example 2 differs from example 1 in that the active composition is prepared from the following raw materials in percentage by weight: 0.5% of Rosa canina fruit extract, 0.5% of tremella polysaccharide, 1% of sodium hyaluronate and 100% of water.
Example 3
Example 3 differs from example 1 in that the active composition is prepared from the following ingredients in weight percent: 0.5% of dog tooth rose hip extract, 0.5% of tremella polysaccharide, 1% of sodium hyaluronate and 100% of water; the Rosa canina fruit extract is commercially available.
Examples 4 to 6
Examples 4 to 6 each provide a cleansing liquid comprising the active composition having a skin aging delaying function, and specific components of the essence are shown in table 1.
Comparative examples 1 to 3
Comparative examples 1-3 provide a serum with specific compositions as shown in table 1.

The preparation method of the essence comprises the following steps: mixing deionized water, vitamin E acetate, glycerol, 1, 3-propylene glycol, PEG-40 hydrogenated castor oil, dipotassium glycyrrhizinate and octanediol, and stirring at 75deg.C; cooling to 50 ℃, adding the active composition and PEG-100, and uniformly mixing; adding triethanolamine to adjust pH to 5.5-7.0; cooling to below 40deg.C, adding phenoxyethanol, ethylhexyl glycerol and essence, and mixing.
Performance testing
DPPH method scavenging free radical Activity detection
(1) The test principle is as follows: 1, 1-Diphenyl-2-trinitrophenylhydrazine (DPPH) is a stable nitrogen-centered organic radical, and the ethanol solution is dark purple and has strong absorption about 517 nm; when a radical scavenger exists, DPPH absorption is weakened and gradually disappears due to single electron pairing with the radical scavenger. The fading degree of the DPPH ethanol solution is quantitatively related to the number of electrons accepted by the DPPH ethanol solution, so that the rapid quantitative analysis can be carried out by using a spectrophotometer, the free radical removal condition is detected, and the free radical removal capability of the antioxidant is evaluated.
(2) The testing method comprises the following steps:
preparing a DPPH ethanol solution: accurately weighing 6mg DPPH, dissolving with 95% ethanol, and dissolving with ultrasonic assistance in a 50mL volumetric flask. When in use, the solution is diluted four times by 95% ethanol to obtain DPPH ethanol solution with the concentration of 3mg/100mL (76 mu M), and the solution is preserved in a dark place (0-4 ℃).
Pretreatment of a sample to be tested: examples 1-3, the ascorbic acid standard, were dissolved in deionized water and diluted with deionized water to the appropriate concentration for use.
The experimental steps are as follows: and adding 2mL of the sample solution to be detected into a test tube, then adding 2mL of the PPH ethanol solution, and uniformly mixing. The reaction is carried out for 30min at room temperature in dark place and the absorbance Abs is measured at 525 nm; in addition to the sample group, a sample solvent control group, a blank control group and a solvent control group (DPPH solvent) are respectively arranged, and the positive control group is an ascorbic acid group.
Clearance I (%) = [1- (T-T) 0 )/(C-C 0 )]×100%
Wherein:
T 0 : absorbance of the sample solution to be measured with 95% ethanol (sample solvent control group) of equal volume;
t: absorbance (sample group) of the sample solution to be measured added with the equal volume of DPPH solution;
C 0 : absorbance of distilled water plus equal volume of 95% ethanol (solvent control);
c: absorbance of distilled water plus an equal volume of DPPH solution (blank).
The test results are shown in Table 2.

As can be seen from Table 2, the active composition has the effect of scavenging DPPH free radicals. Compared with the blank control group, the active composition has stronger capability of scavenging free radicals, and the higher the content of the active ingredient, the higher the DPPH scavenging rate.
2. Detection of inhibition of elastase Activity
The experimental steps are as follows: mu.L of a sample solution comprising the solutions prepared in examples 1-3 and elastinal solution (both diluted to a suitable concentration with deionization) and 130. Mu.L of 0.1M Tris-HCl buffer (pH 8.0) containing 1.015mM reaction substrate Succ-Ala-Ala-p-nitroanilide were added to a 96-well plate, incubated at 25℃for 5 minutes, 15. Mu.L of elastase solution (0.5U/mL) was added, incubation was continued at 25℃for 30 minutes, and absorbance A410 at a wavelength of 410nm was measured with a microplate reader, wherein the elastinal solution (elastase inhibitor, outsourced) was used as a positive control group. The absorbance was also measured by replacing the aqueous sample solution with deionized water as a reference solution.
The elastase activity inhibition rate is calculated as: inhibition (%) = (a) 0 -(A 410 -B))/A 0 ×100%;
Wherein:
A 0 : absorbance of the reference solution;
A 410 : is the absorbance of the sample solution;
b: absorbance of sample blank solution.
The test results are shown in Table 3.

As can be seen from table 3, the active composition has an elastase inhibitory effect, and the active composition has a stronger elastase inhibitory effect than the blank, and the higher the content of the active ingredient, the higher the inhibitory effect.
3. Skin texture testing
Selecting left and right cheeks at the intersection line of the facial cheeks, the nasal tip and the pupil of a subject, and left and right sides at the intersection line of the temple and the mouth corner of the subject as test areas, firstly, measuring the skin texture degree of the test area by a technician by using a visual facial image analyzer before applying cosmetics, measuring five times per point, and taking an average value. The subjects were again tested for skin texture after four weeks of continuous use of the cosmetics in examples and comparative examples. And counting the numerical value measured by each time of the experimental part of the subject, and analyzing the change rule of the skin texture of the experimental part of the subject, so as to determine the effect of the cosmetics of the examples and the comparative examples on improving the skin texture.

4. Anti-wrinkle test of human skin
The anti-wrinkle test of human skin is the most direct method for testing anti-wrinkle effect. The subject population was wrinkled females with an average age of 35-45 years. The left and right cheeks at the intersection of the facial cheeks, nasal tip and pupil, and the left and right sides at the intersection of the temple and the corner of the mouth of the subject are selected as test areas. The elasticity value of the site of application of the subject was determined prior to testing using a skin elasticity tester (MPA 580). The composition was applied twice a day in the morning and evening, and the photo before application was used as a control. During the experiment, the subjects were unable to apply any other cosmetics. After four weeks of continuous use of the anti-wrinkle cosmetics, the subjects were compared with the skin elasticity values before and after use of the cosmetics of examples and comparative examples. And counting the numerical value measured by the experimental part of the subject each time, and analyzing the skin elasticity change rule to evaluate the skin elasticity change condition.
The test results are shown in Table 5.

As shown in tables 4 and 5, the subjects of comparative examples 1 to 3 have general skin elasticity and texture improvement effects after using for 4 weeks, while the subjects of examples 4 to 6 have remarkable skin elasticity and texture improvement effects after using for 4 weeks, which indicates that the Rosa canina fruit extract of the present invention can fully exert its antioxidant activity, and has synergistic effects with tremella polysaccharide and hyaluronic acid, and the anti-aging effect of the product is remarkably improved.

Claims (6)

1. An active composition with skin aging delaying function is characterized in that,
the active composition comprises the following components: fructus Rosae Davuricae extract, tremella polysaccharide and sodium hyaluronate; the weight ratio of the dog tooth rose hip extract to tremella polysaccharide to sodium hyaluronate is 1:1: (0.5-0.6);
the preparation method of the Rosa canina fruit extract comprises the following steps:
(1) Freeze-drying fructus Rosae Davuricae, pulverizing into powder, and sieving with 400-1000 mesh sieve to obtain fructus Rosae Davuricae powder;
(2) Washing the dog rose jelly dry powder with an alcohol aqueous solution for a plurality of times, collecting washing liquid, and placing in a refrigerator at the temperature of-5 to 5 ℃ for standing for 10 to 30 hours; taking out and filtering to obtain filtrate I, and drying at 40-60deg.C to obtain dried product;
(3) Adding water into the dried product to prepare a solution, and placing the solution in a refrigerator at the temperature of-5 to 5 ℃ for standing for 10 to 30 hours; taking out and filtering to obtain filtrate II, extracting with ester organic solvent for several times, collecting organic phase extractive solution, and removing solvent to obtain the final product;
the sodium hyaluronate is a mixture of high molecular sodium hyaluronate, medium molecular sodium hyaluronate and low molecular sodium hyaluronate; the molecular weight of the high molecular sodium hyaluronate is not less than 160 ten thousand daltons; the molecular weight of the medium molecular sodium hyaluronate is 20 ten thousand-160 ten thousand daltons; the molecular weight of the low molecular weight sodium hyaluronate is 1 ten thousand to 20 ten thousand daltons;
the weight ratio of the high molecular sodium hyaluronate, the medium molecular sodium hyaluronate and the low molecular sodium hyaluronate is 1: (1-5): (0.1-1).
2. The active composition having a function of delaying skin aging according to claim 1, wherein the volume concentration of the alcohol in the aqueous solution of the alcohol is not less than 70%.
3. The active composition having a function of delaying skin aging according to claim 1, wherein the drying treatment is reduced pressure drying.
4. The active composition having a skin aging delaying function according to claim 1, wherein the ratio of the dry matter to water is 1: (10-100); the unit of the feed liquid ratio is g/mL.
5. Use of an active composition with a function of delaying skin aging according to any one of claims 1 to 4, characterized in that it is applied in the field of skin care products, cosmetics.
6. Use of an active composition having a skin aging delaying function according to claim 5, wherein the active composition is contained in an amount of 10 to 30wt% in skin care products, cosmetics.
