Abstract
The invention provides a patch type mask containing an astaxanthin inclusion compound and a preparation method thereof, relating to the technical field of masks. The patch type mask is prepared from essence containing an astaxanthin inclusion compound and mask cloth, the formula of the mask essence is scientific and reasonable, the astaxanthin is included by adopting cyclic oligosaccharide hydroxypropyl-beta-cyclodextrin with hydrophilicity outside and lipophilic inner cavity inside through compounding of various effective components, so that the astaxanthin forms a water-soluble inclusion compound, the astaxanthin can be promoted to effectively permeate the skin without changing the skin structure, the skin permeability is increased, the bioavailability of the astaxanthin is improved, and a good anti-aging effect is exerted; the astaxanthin inclusion compound is compounded with a plurality of effective components by the hydroxypropyl-beta-cyclodextrin, so that the whitening effect is achieved while multiple effects of anti-aging are achieved; the mask cloth improves the fitting property, air permeability and moisture permeability of the mask, so that the mask essence is fully absorbed by the skin, deeply moisturizes the skin and has multiple effects of skin rejuvenation.
Description
Technical Field
The invention relates to the technical field of facial masks, in particular to a patch type facial mask containing an astaxanthin inclusion compound and a preparation method thereof.
Background
With the rapid increase of the global aging population and the increased pressure of people in modern society, people continuously deepen the cognition of skin injury caused by the sharp increase of oxidation pressure in the increasingly severe environment, pursue the health and beauty of the skin, discuss the anti-aging problem and increasingly increase the demand of anti-aging products. The facial mask is beneficial to the skin to remove products of epidermal cell metabolism and accumulated grease substances, so that water and other active ingredients in the facial mask penetrate into the horny layer of the epidermis, the skin becomes soft, and the skin is naturally bright and elastic.
Astaxanthin (Astaxanthin) is used as a fat-soluble ketone carotenoid, also called Astaxanthin or Astaxanthin, and is classified as a carotenoid group, is a strong scavenger of free radicals, active oxygen and active nitrogen and can obviously reduce damage of ROS and NMP to dermal collagen and elastin. Meanwhile, the self repair mechanism of the skin can reconstruct the damaged collagen, and the normal metabolism of the skin is ensured. The astaxanthin has antioxidant and anti-wrinkle effects. Research shows that compared with carotene, vitamin E, etc., astaxanthin has stronger bioactivity and is a substance safe for human body. Astaxanthin, which has been considered to be "super vitamin E", has an antioxidant activity about 10 times higher than that of beta-carotene and about 500 times higher than that of vitamin E.
Haematococcus pluvialis (Haematococcus pluvias) is a unicellular green freshwater alga, and is divided into two stages of swimming vegetative growth and motionless transformation to produce astaxanthin throughout the life cycle of Haematococcus pluvialis: under the conditions of weak illumination intensity and enough nitrogen and phosphorus salts, the swimming green vegetative cells grow and develop, and when the environment is not suitable, such as high temperature, high salt, high illumination or nutrient salt starvation, a large amount of astaxanthin can be accumulated. Astaxanthin is widely present in organisms, particularly feathers of shrimps, crabs, fish and birds, and Haematococcus pluvialis is the highest accumulated species in all known astaxanthin-synthesizing organisms, can accumulate more than 1-3% of the dry weight of the astaxanthin, and is recognized as the most ideal source for producing natural astaxanthin in the natural world.
Hydroxypropyl-beta-cyclodextrin (HP-beta-CD) is a cyclic oligosaccharide, has hydrophilic outside and lipophilic inside, can form a water-soluble inclusion compound with a fat-soluble drug, promotes the drug to permeate the skin without changing the skin structure, is a high-quality inclusion material, and can remarkably improve the solubility and the stability of the drug. The astaxanthin has poor water solubility and is sensitive to external factors such as illumination, oxygen, strong acid and the like, so that the astaxanthin has low bioavailability and a plurality of limitations in practical application.
Therefore, the technical scheme provided by the invention obtains the stable inclusion compound by preparing the astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound, and solves the problems that the astaxanthin is poor in water solubility, sensitive to external factors such as illumination, oxygen, strong acid and the like, low in bioavailability and incapable of being fully absorbed by skin.
Disclosure of Invention
The invention provides a patch type mask containing an astaxanthin inclusion compound and a preparation method thereof aiming at overcoming the defects of the prior art, and aims to solve the problems that astaxanthin is poor in water solubility, sensitive to external factors such as illumination, oxygen, strong acid and the like, low in bioavailability and incapable of being fully absorbed by skin.
The invention solves the technical problems through the following technical means:
the invention provides a patch type facial mask containing an astaxanthin inclusion compound, which is prepared from facial mask essence containing the astaxanthin inclusion compound and facial mask cloth;
the facial mask essence comprises the following components in percentage by weight: 0.05-10% of astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound, 0.02-5% of tea polyphenol, 0.2-1% of bisabolol, 0.5-3% of tranexamic acid, 0.1-5% of damask rose hydrosol, 1-4% of 1, 2-pentanediol, 2-5% of glycerol, 4-10% of 1, 3-butanediol, 0.02-5% of sodium hyaluronate, 0.02-0.2% of EDTA-2Na, 0.05-2% of xanthan gum, 0.2-0.5% of sodium alginate, 0.3-1% of jojoba wax PEG-1200.05, 0.3-1% of PEG-40 hydrogenated castor oil, 0.1-1% of preservative and the balance of deionized water.
As a further improvement of the invention, the preparation method of the astaxanthin clathrate compound comprises the following steps:
s1, mixing the components in a mass ratio of 1: 5-20, weighing astaxanthin and hydroxypropyl-beta-cyclodextrin, respectively dissolving the astaxanthin and the hydroxypropyl-beta-cyclodextrin in ethanol and ethanol with the concentration of 95%, introducing nitrogen for protection, keeping the temperature, magnetically stirring, and carrying out an embedding reaction to balance to obtain an inclusion compound solution;
s2, placing the clathrate compound solution in a round-bottom flask, performing rotary evaporation under reduced pressure in a water bath condition to remove ethanol, and placing the concentrate at room temperature for drying;
s3, adding deionized water into the dried concentrate, and fully dissolving the inclusion compound to obtain an inclusion compound solution;
s4, carrying out suction filtration on the inclusion compound solution to remove insoluble substances to obtain an orange transparent solution;
s5, freeze-drying the orange transparent solution to obtain the astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound of the orange solid compound.
As a further improvement scheme of the invention, the water bath temperature in the step S2 is 37-40 ℃, and the freeze drying time in the step S5 is 24-48 hours.
In a further improvement of the invention, the astaxanthin is derived from haematococcus pluvialis, and the astaxanthin clathrate compound is prepared from astaxanthin and hydroxypropyl-beta-cyclodextrin according to a mass ratio of 1: 5-20.
As a further improvement of the invention, the preservative is selected from one or more of phenoxyethanol, methyl paraben and moutan bark extract.
As a further improvement of the invention, the mask cloth is selected from 384 silk film cloths.
The invention also provides a preparation method of the patch type mask containing the astaxanthin inclusion compound, the patch type mask is prepared from mask essence containing the astaxanthin inclusion compound and mask cloth, and the preparation method of the essence comprises the following steps:
s11, adding xanthan gum and sodium alginate into deionized water, heating, stirring and dissolving uniformly to obtain a mixture a, and standing at room temperature for later use;
s12, adding glycerol, 1, 3-butanediol, 1, 2-pentanediol, sodium hyaluronate, EDTA-2Na, jojoba wax PEG-120 and PEG-40 hydrogenated castor oil into deionized water, heating and stirring uniformly, adding into the mixture a to obtain a mixture b, and standing at room temperature for later use;
and S13, adding the astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound, the tea polyphenol, the tranexamic acid, the bisabolol and the rosa damascena pure dew into the mixture b, uniformly stirring, adding water to full dose, and uniformly stirring to obtain the essence.
The invention has the beneficial effects that:
1. the patch type mask is prepared from essence containing an astaxanthin inclusion compound and mask cloth, the formula of the mask essence is scientific and reasonable, the astaxanthin is included by adopting cyclic oligosaccharide hydroxypropyl-beta-cyclodextrin with hydrophilicity outside and lipophilic inner cavity inside through compounding of various effective components, so that the astaxanthin forms a water-soluble inclusion compound, the astaxanthin can be promoted to effectively permeate the skin without changing the skin structure, the skin permeability is increased, the bioavailability of the astaxanthin is improved, and a good anti-aging effect is exerted.
2. The astaxanthin clathrate compound is compounded with a plurality of effective components such as tea polyphenol, bisabolol, tranexamic acid, damascus rose hydrosol and the like by using hydroxypropyl-beta-cyclodextrin, so that the whitening effect is exerted while the multiple effects of resisting aging are achieved; the mask cloth improves the fitting property, air permeability and moisture permeability of the mask, so that the mask essence is fully absorbed by the skin, deeply moisturizes the skin and has multiple effects of skin rejuvenation.

The preparation method of the astaxanthin clathrate compound comprises the following steps:
s1, mixing the components in a mass ratio of 1: 5-20, weighing astaxanthin and hydroxypropyl-beta-cyclodextrin, respectively dissolving the astaxanthin and the hydroxypropyl-beta-cyclodextrin in ethanol and ethanol with the concentration of 95%, introducing nitrogen for protection, keeping the temperature, magnetically stirring, and carrying out an embedding reaction to balance to obtain an inclusion compound solution;
s2, placing the clathrate compound solution in a round-bottom flask, performing rotary evaporation under reduced pressure in a water bath condition to remove ethanol, and placing the concentrate at room temperature for drying; wherein the temperature of the water bath is 37-40 ℃;
s3, adding deionized water into the dried concentrate, and fully dissolving the inclusion compound to obtain an inclusion compound solution;
s4, carrying out suction filtration on the inclusion compound solution to remove insoluble substances to obtain an orange transparent solution;
s5, freeze-drying the orange transparent solution to obtain an astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound of an orange solid compound; wherein the freeze drying time is 24-48 hours.

In the following examples, various starting materials were commercially available. Wherein, the sodium hyaluronate is purchased from Dongying Foster bioengineering limited company, the xanthan gum and the glycerol are purchased from Shanghai Youso chemical technology limited company, the tea polyphenol is purchased from Zhejiang Yuelaiyi pharmaceutical industry limited company, the jojoba wax PEG-120 is purchased from Shanghai Alloco biotechnological limited company, the Damascus rose hydrolat is purchased from Beijing Ainomama subfamily technology limited company, the bisabolol is purchased from Guangzhou Luzi biotechnical limited company, the hydroxypropyl-beta-cyclodextrin, the biogenic acid, the 1, 2-pentanediol, the 1, 3-butanediol, the EDTA-2Na, the sodium alginate and the PEG-40 hydrogenated castor oil are purchased from Shanghai Merlin Biotech limited company.
Example 1
The patch type facial mask containing the astaxanthin inclusion compound is prepared from facial mask essence containing the astaxanthin inclusion compound and facial mask cloth. Wherein the facial mask cloth is selected from 384 silk film cloth.
Specifically, the facial mask essence consists of the following components in percentage by weight: 2% of astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound, 0.02% of tea polyphenol, 0.2% of bisabolol, 0.5% of tranexamic acid, 1% of damask rose hydrosol, 1% of 1, 2-pentanediol, 2% of glycerol, 4% of 1, 3-butanediol, 0.02% of sodium hyaluronate, 0.02% of EDTA-2Na, 0.05% of xanthan gum, 0.2% of sodium alginate, 0.1% of jojoba wax PEG-1200.05%, 0.3% of PEG-40 hydrogenated castor oil, 0.1% of preservative and the balance of deionized water. Wherein the preservative is selected from phenoxyethanol.
Wherein the astaxanthin is derived from haematococcus pluvialis, and the astaxanthin inclusion compound is prepared from astaxanthin and hydroxypropyl-beta-cyclodextrin according to a mass ratio of 1: 5, and preparing the product.
The preparation method of the astaxanthin clathrate compound comprises the following steps:
s1, mixing the components in a mass ratio of 1: weighing astaxanthin and hydroxypropyl-beta-cyclodextrin, respectively dissolving in ethanol and 95% ethanol, introducing nitrogen gas for protection, keeping the temperature, magnetically stirring, and allowing the embedding reaction to reach balance to obtain an inclusion compound solution;
s2, placing the clathrate compound solution in a round-bottom flask, performing rotary evaporation under reduced pressure in a water bath condition to remove ethanol, and placing the concentrate at room temperature for drying; wherein the water bath temperature is 37 ℃;
s3, adding deionized water into the dried concentrate, and fully dissolving the inclusion compound to obtain an inclusion compound solution;
s4, carrying out suction filtration on the inclusion compound solution to remove insoluble substances to obtain an orange transparent solution;
s5, freeze-drying the orange transparent solution to obtain an astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound of an orange solid compound; wherein the freeze-drying time is 24 hours.

Specifically, the facial mask essence consists of the following components in percentage by weight: 3% of astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound, 0.05% of tea polyphenol, 0.5% of bisabolol, 1% of tranexamic acid, 2% of rose hydrosol of Damascus, 1.5% of 1, 2-pentanediol, 2.5% of glycerol, 4.5% of 1, 3-butanediol, 0.05% of sodium hyaluronate, 0.05% of EDTA-2Na, 0.1% of xanthan gum, 0.25% of sodium alginate, 0.2% of jojoba wax PEG-1200.1%, 0.4% of PEG-40 hydrogenated castor oil, 0.2% of preservative and the balance of deionized water. Wherein the preservative is selected from phenoxyethanol.
Wherein the astaxanthin is derived from haematococcus pluvialis, and the astaxanthin inclusion compound is prepared from astaxanthin and hydroxypropyl-beta-cyclodextrin according to a mass ratio of 1: 10 is prepared.
The preparation method of the astaxanthin clathrate compound comprises the following steps:
s1, mixing the components in a mass ratio of 1: 10 weighing astaxanthin and hydroxypropyl-beta-cyclodextrin, respectively dissolving in ethanol and 95% ethanol, introducing nitrogen for protection, keeping the temperature, magnetically stirring, and allowing the embedding reaction to reach balance to obtain an inclusion compound solution;
s2, placing the clathrate compound solution in a round-bottom flask, performing rotary evaporation under reduced pressure in a water bath condition to remove ethanol, and placing the concentrate at room temperature for drying; wherein the water bath temperature is 37 ℃;
s3, adding deionized water into the dried concentrate, and fully dissolving the inclusion compound to obtain an inclusion compound solution;
s4, carrying out suction filtration on the inclusion compound solution to remove insoluble substances to obtain an orange transparent solution;
s5, freeze-drying the orange transparent solution to obtain an astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound of an orange solid compound; wherein the freeze-drying time is 24 hours.
The preparation method of the patch type facial mask containing the astaxanthin clathrate compound comprises the following steps of preparing a facial mask essence containing the astaxanthin clathrate compound and facial mask cloth. The preparation method of the essence is the same as that of example 1.
Example 3
The patch type facial mask containing the astaxanthin inclusion compound is prepared from facial mask essence containing the astaxanthin inclusion compound and facial mask cloth. Wherein the facial mask cloth is selected from 384 silk film cloth.
Specifically, the facial mask essence consists of the following components in percentage by weight: astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound 4%, tea polyphenol 0.08%, bisabolol 0.6%, tranexamic acid 1.2%, rose hydrosol 3%, 1, 2-pentanediol 2%, glycerin 2%, 1, 3-butanediol 5%, sodium hyaluronate 0.1%, EDTA-2Na 0.1%, xanthan gum 0.15%, sodium alginate 0.3%, jojoba wax PEG-1200.2%, PEG-40 hydrogenated castor oil 0.45%, preservative 0.5%, and the balance of deionized water. Wherein the preservative is selected from phenoxyethanol.
Wherein the astaxanthin is derived from haematococcus pluvialis, and the astaxanthin inclusion compound is prepared from astaxanthin and hydroxypropyl-beta-cyclodextrin according to a mass ratio of 1: 20 is prepared.
The preparation method of the astaxanthin clathrate compound comprises the following steps:
s1, mixing the components in a mass ratio of 1: 20 weighing astaxanthin and hydroxypropyl-beta-cyclodextrin, respectively dissolving in ethanol and 95% ethanol, introducing nitrogen gas for protection, keeping the temperature, magnetically stirring, and allowing the embedding reaction to reach balance to obtain an inclusion compound solution;
s2, placing the clathrate compound solution in a round-bottom flask, performing rotary evaporation under reduced pressure in a water bath condition to remove ethanol, and placing the concentrate at room temperature for drying; wherein the water bath temperature is 37 ℃;
s3, adding deionized water into the dried concentrate, and fully dissolving the inclusion compound to obtain an inclusion compound solution;
s4, carrying out suction filtration on the inclusion compound solution to remove insoluble substances to obtain an orange transparent solution;
s5, freeze-drying the orange transparent solution to obtain an astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound of an orange solid compound; wherein the freeze-drying time is 24 hours.
The preparation method of the patch type facial mask containing the astaxanthin clathrate compound comprises the following steps of preparing a facial mask essence containing the astaxanthin clathrate compound and facial mask cloth. The preparation method of the essence is the same as that of example 1.
Test examples
The astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound in the patch type mask essence prepared by the embodiment of the invention is subjected to a relevant test.
Test example 1: observation by electron microscope
Test samples: examples 1,2, 3;
the test method comprises the following steps: sampling a proper amount of astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound according to the requirements of a transmission electron microscope, and scanning a sample;
and (3) testing results: the results are shown in FIGS. 1,2 and 3.

the facial mask essence comprises the following components in percentage by weight: 0.05-10% of astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound, 0.02-5% of tea polyphenol, 0.2-1% of bisabolol, 0.5-3% of tranexamic acid, 0.1-5% of damask rose hydrosol, 1-4% of 1, 2-pentanediol, 2-5% of glycerol, 4-10% of 1, 3-butanediol, 0.02-5% of sodium hyaluronate, 0.02-0.2% of EDTA-2Na, 0.05-2% of xanthan gum, 0.2-0.5% of sodium alginate, 0.3-1% of jojoba wax PEG-1200.05, 0.3-1% of PEG-40 hydrogenated castor oil, 0.1-1% of preservative and the balance of deionized water.
2. The patch type mask containing the astaxanthin clathrate compound according to claim 1, wherein the preparation method of the astaxanthin clathrate compound comprises the following steps:
s1, mixing the components in a mass ratio of 1: 5-20, weighing astaxanthin and hydroxypropyl-beta-cyclodextrin, respectively dissolving the astaxanthin and the hydroxypropyl-beta-cyclodextrin in ethanol and ethanol with the concentration of 95%, introducing nitrogen for protection, keeping the temperature, magnetically stirring, and carrying out an embedding reaction to balance to obtain an inclusion compound solution;
s2, placing the clathrate compound solution in a round-bottom flask, performing rotary evaporation under reduced pressure in a water bath condition to remove ethanol, and placing the concentrate at room temperature for drying;
s3, adding deionized water into the dried concentrate, and fully dissolving the inclusion compound to obtain an inclusion compound solution;
s4, carrying out suction filtration on the inclusion compound solution to remove insoluble substances to obtain an orange transparent solution;
s5, freeze-drying the orange transparent solution to obtain the astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound of the orange solid compound.
3. The patch mask containing the astaxanthin clathrate compound according to claim 2, wherein the water bath temperature in the step S2 is 37-40 ℃, and the freeze-drying time in the step S5 is 24-48 hours.
4. The patch mask according to claim 1, wherein the astaxanthin is derived from Haematococcus pluvialis, and the astaxanthin inclusion compound is prepared from astaxanthin and hydroxypropyl-beta-cyclodextrin in a mass ratio of 1: 5-20.
5. The patch mask containing the astaxanthin clathrate according to claim 1, wherein the preservative is selected from one or more of phenoxyethanol, methylparaben and moutan bark extract.
6. The patch type mask containing the astaxanthin clathrate compound according to claim 1, wherein the mask cloth is selected from 384 silk mask cloths.

The method for preparing the astaxanthin-containing inclusion compound patch mask according to any one of claims 1 to 6, wherein the patch mask is prepared from a mask essence containing the astaxanthin inclusion compound and a mask cloth, and the preparation method of the essence comprises the following steps:
s11, adding xanthan gum and sodium alginate into deionized water, heating, stirring and dissolving uniformly to obtain a mixture a, and standing at room temperature for later use;
s12, adding glycerol, 1, 3-butanediol, 1, 2-pentanediol, sodium hyaluronate, EDTA-2Na, jojoba wax PEG-120 and PEG-40 hydrogenated castor oil into deionized water, heating and stirring uniformly, adding into the mixture a to obtain a mixture b, and standing at room temperature for later use;
and S13, adding the astaxanthin/hydroxypropyl-beta-cyclodextrin inclusion compound, the tea polyphenol, the tranexamic acid, the bisabolol and the rosa damascena pure dew into the mixture b, uniformly stirring, adding water to full dose, and uniformly stirring to obtain the essence.
