Abstract
本发明涉及在不使抗体功能降低的情况下,施予至患者后在肝脏内被容易地代谢、减少放射性核素向肝脏等器官的集聚的标记技术,并提供包含IgG抗体和与其结合的IgG结合肽的修饰抗体。IgG结合肽包含GPDCAYH(Xaa1)GELVWCTFH(式中,Xaa1为赖氨酸残基、半胱氨酸残基、天冬氨酸残基、谷氨酸残基、2‑氨基辛二酸、或二氨基丙酸)等由13~17个氨基酸残基组成的氨基酸序列,在IgG结合肽的N末端,介由修饰接头而与下式(II‑1)表示的化合物于其赖氨酸残基的位置处连接。
Description
技术领域
本发明涉及对IgG抗体的修饰及放射性金属标记。
背景技术
抗体由于其针对靶分子的特异性而在各种研究·开发中被用于靶分子的检测,医疗领域中,通过将可与癌症等的患部特异性结合的抗体用放射性核素标记并施予至体内来实施利用单光子发射计算机断层成像(Single Photon Emission Computed Tomography)(SPECT)、正电子发射断层成像(Positron Emission Tomography)(PET)的诊断,另外,用放射性核素标记的抗体作为用于治疗疾病的医药品也受到瞩目。
作为用放射性核素标记抗体的方法,将螯合化合物与抗体结合、并使该螯合化合物担载放射性金属元素的方法正在被使用(非专利文献1)。上述螯合化合物的结合迄今为止主要介由抗体所含的赖氨酸的氨基、半胱氨酸的巯基、及被活化的羧基等来进行,但即使其针对官能团具有特异性,也没有位点特异性,因此存在因对于抗体的抗原结合位点的修饰等而导致抗体活性降低的问题、难以控制结合化合物的数量等问题。
为了克服上述问题,已在使用以位点特异性方式引入了特定官能团的抗体,从而对抗体进行修饰。例如,通过利用基因工程修饰将非天然氨基酸(非专利文献2~4)、游离的半胱氨酸(非专利文献5~6)引入特定位点,从而能够实现特定位点处的修饰。虽然正在不断开发这样的位点特异性的抗体修饰技术,但多数情况下,需要以抗体工程的方式来修饰抗体自身,考虑到与该修饰相伴而使得抗体功能降低、开发成本高,不能必然认为是有利的方法。
因此,作为能够特异性且简便地修饰抗体的方法,提出了下述方法:通过将配体与针对IgG抗体的Fc结构域的特定位点具有结合性的IgG结合肽连接,使该IgG结合肽与抗体结合,从而无需改变抗体分子的序列,因此能够在不会导致与抗体分子的基因修饰相伴的功能降低的情况下利用放射性金属核素来标记IgG(专利文献1及2)。
另一方面,经放射性金属核素标记的低分子多肽在施予至生物体时,自施予早期起在肾脏中长期观察到放射活性,因此,将放射性金属标记抗体施予至生物体内时,存在肾脏的辐射暴露及肾损伤的可能性,但提出了通过使用特定的低分子多肽来使非特异性肾集聚减少的放射性标记化合物(专利文献3及4、非专利文献7及8)。

发明内容
但是,一直以来,将放射性金属标记抗体施予至生物体内时,确认到向肝脏、脾脏等网状内皮系统发达的器官的高度集聚,存在对于患者的正常器官造成不必要的辐射暴露的担忧。虽然专利文献3及4的技术中减少了肾集聚,但存在由于与Fab片段结合而导致抗体功能降低的担忧。
本发明是鉴于上述状况而完成的,其课题在于提供在不使抗体功能降低的情况下施予至患者后在肝脏内被容易地代谢、能够减少放射性核素向肝脏等器官的集聚的标记技术。
本发明的一个方式提供修饰抗体,其包含IgG抗体、和与所述IgG抗体结合的IgG结合肽,所述IgG结合肽包含由下式(I)表示的、由13~17个氨基酸残基组成的氨基酸序列,在所述IgG结合肽的N末端,介由修饰接头而与下式(II-1)表示的化合物于其赖氨酸残基的位置处连接。

上述式(II)的化合物能够介由赖氨酸侧链末端的NR1R2表示的基团而与IgG抗体、IgG结合肽等蛋白质的官能团结合,因此,能够用于包括IgG抗体在内的各种靶分子的放射性金属标记。因此,本发明的其他方式提供使可与靶分子结合的多肽与上述式(II)的化合物结合而成的化合物或其盐。
根据本发明,由于在抗体的Fc区具备具有苯丙氨酸残基及赖氨酸残基的排泄促进接头,因此,能够得到通过肝脏代谢而促进放射性金属从体内排泄、体内动力学优异的放射性金属标记抗体。

具体实施方式
<修饰IgG结合肽>
本发明的一个方式中,修饰IgG结合肽包含由上述式(I)表示的、由13~17个氨基酸残基组成的IgG结合肽,且该IgG结合肽的N末端被上述式(III)表示的DBCO接头修饰。
式(I)的IgG结合肽只要是可与IgG结合的肽即可,没有特别限定,优选WO2017/217347中记载的IgG结合肽。
以下,对本发明中使用的IgG结合肽进行详细说明。
本说明书中使用的“IgG”是指哺乳动物、例如人及黑猩猩等灵长类、大鼠、小鼠、及兔等实验动物、猪、牛、马、绵羊、及山羊等家畜动物、以及狗及猫等宠物的IgG,优选为人的IgG(IgG1、IgG2、IgG3或IgG4)。本说明书中的IgG进一步优选为人IgG1、IgG2、或IgG4、或者兔IgG,特别优选为人IgG1、IgG2、或IgG4。
本发明中使用的IgG结合肽包含由上述式(I)表示的、由13~17个氨基酸残基组成的氨基酸序列,且能与人IgG结合。
上述式(I)中,N末端或C末端的X1-3这一符号表示半胱氨酸(C或Cys)以外的独立且任意的氨基酸残基X以1~3个连续,构成其的氨基酸残基是相同或不同的残基,但优选由3个并非全部相同的残基的序列组成。同样地,X2表示半胱氨酸(C或Cys)以外的独立且任意的氨基酸残基X以2个连续,构成其的氨基酸残基是相同或不同的残基,但优选由该2个连续的氨基酸残基为不同残基的序列组成。
式(I)的2个半胱氨酸残基能够以二硫键键合而形成环肽。通常,式(I)的IgG结合肽中,(Xaa1为半胱氨酸残基的情况下,排除Xaa1)外侧的2个半胱氨酸残基以二硫键键合。或者,式(I)的肽中,外侧的2个半胱氨酸残基中的硫醚基经由下式表示的接头而连接,

上式中的虚线部分表示与硫醚基键合的部分。该接头与通常的二硫键相比,对于还原反应等更稳定。因此,该接头在例如使用锆等能使二硫键不稳定化的放射性金属核素时优选使用。
上述的式(I)的IgG结合肽的氨基酸序列中,为17个氨基酸残基的情况下的、自N末端起第1位及第2位以及第16位及第17位的氨基酸残基X可以缺失,这样的肽由13个氨基酸组成。
本说明书中使用的“为17个氨基酸残基的情况下的”是在将肽的氨基酸残基以氨基酸编号称呼时,用于针对式(I)的IgG结合肽,自作为最长氨基酸长度的17个残基的N末端起依次从第1位到第17位加以编号的、方便表达的术语。
此外,上述的式(I)的肽的氨基酸序列的半胱氨酸(C)以外的氨基酸残基、即为17个氨基酸残基的情况下的自N末端起第1~3、5、6、15~17位的各氨基酸残基,优选选自以下氨基酸残基。此处,各大写字母为氨基酸的单字母记载:
第1位的氨基酸残基=S、G、F、R或无;
第2位的氨基酸残基=D、G、A、S、P、高半胱氨酸或无;
第3位的氨基酸残基=S、D、T、N、E或R;

如前所述,本发明涉及的上述式(I)的IgG结合肽的特征在于,在各氨基酸序列之中具有分隔的至少2个半胱氨酸(C)残基,并以能在这些半胱氨酸残基之间形成二硫键的方式配置有半胱氨酸残基,就优选的肽而言,2个半胱氨酸残基以二硫键键合而形成环肽,在各半胱氨酸残基的N末端侧及C末端侧可以具有1~3个半胱氨酸以外的任意的氨基酸残基。在各半胱氨酸残基的N末端侧及C末端侧具有1~3个氨基酸残基的情况下,为17个氨基酸残基的情况下的自N末端起第1~2、16~17位的各氨基酸残基为上述例示的氨基酸残基。
如上所述,本发明中使用的IgG结合肽中,Xaa1为赖氨酸残基、半胱氨酸残基、天冬氨酸残基、及谷氨酸残基等蛋白质构成氨基酸、以及二氨基丙酸及2-氨基辛二酸等非蛋白质构成氨基酸、优选为赖氨酸残基。Xaa1优选能利用后述的交联剂进行修饰。本说明书中“非蛋白质构成氨基酸”是指无法用于在生物体中构成蛋白质的氨基酸。为了提高将本发明中使用的IgG结合肽利用交联剂修饰时的位点特异性,本发明中使用的IgG结合肽优选在其序列中完全不具有、或几乎不具有(例如,仅具有1个或2个)与Xaa1相同的残基。例如,Xaa1为赖氨酸残基时,本发明中使用的IgG结合肽优选在其序列中于Xaa1以外的位置完全不具有、或几乎不具有赖氨酸残基。
就本发明中使用的IgG结合肽而言,与人IgG的结合亲和性相较于其他人免疫球蛋白(IgA、IgE、IgM)而言高约10倍以上、优选高约50倍以上、更优选高约200倍以上。与本发明中使用的IgG结合肽与人IgG的结合相关的解离常数(Kd)可以通过表面等离子共振光谱分析(使用例如BIACORE系统)来确定,例如小于1×10-1M~1×10-3M,优选小于1×10-4M,更优选小于1×10-5M。
本发明中使用的IgG结合肽与IgG抗体的Fc结构域结合。本发明中使用的IgG结合肽如WO2017/217347的实施例中所示,在上述Xaa1中,与IgG Fc的特定区域、即人IgG Fc中的基于Eu编号的Lys248残基(以下,在本说明书中缩略记为“Lys248”,相当于人IgG CH2(序列号5)的第18位的残基)或Lys246残基(以下,在本说明书中缩略记为“Lys246”,相当于人IgG CH2(序列号5)的第16位的残基)、优选Lys248接近。
本发明中使用的IgG结合肽可以利用例如WO2017/217347中记载的方法来得到。具体而言,可以利用惯用的液相合成法、固相合成法等肽合成方法、基于自动肽合成仪的肽合成等(Kelley等,Genetics Engineering Principles and Methods,Setlow,J.K.eds.,Plenum Press NY.(1990)Vol.12,p.1-19;Stewart等,Solid-Phase Peptide Synthesis(1989)W.H.Freeman Co.;Houghten,Proc.Natl.Acad.Sci.USA(1985)82:p.5132;“新生化学实验讲座1蛋白质IV”(1992)日本生化学会编,东京化学同人)来制造。或者,可以利用使用了编码本发明中使用的IgG结合肽的核酸的基因重组法、噬菌体展示法等来制造肽。通过将例如编码本发明中使用的IgG结合肽的氨基酸序列的DNA组入表达载体中,导入宿主细胞中并进行培养,可以制造目标肽。制得的肽可以通过常规方法、例如凝胶过滤色谱、离子交换柱色谱、亲和色谱、反相柱色谱、HPLC等色谱、硫酸铵分级、超滤、及免疫吸附法等来进行回收或纯化。
肽合成中,例如,准备对各氨基酸(是天然或非天然均可)的、除了要键合的α-氨基和α-羧基以外的官能团进行了保护的氨基酸类,在各氨基酸的α-氨基与α-羧基之间进行肽键形成反应。通常,预先将位于肽的C末端的氨基酸残基的羧基介由合适的间隔基团或接头而与固相结合。选择性地除去如此操作得到的二肽的氨基末端的保护基团,在与下一氨基酸的α-羧基之间形成肽键。连续实施这样的操作从而制造侧链基团被保护的肽,最后,除去全部的保护基团,从固相分离。保护基团的种类、保护方法、肽结合方法的细节详细记载于上述文献中。
利用基因重组法的制造例如可以通过包括下述步骤的方法来进行:将编码本发明的肽的DNA插入合适的表达载体中,将载体导入至合适的宿主细胞,培养细胞,从细胞内或细胞外液中回收目标肽。载体没有限定,有例如质粒、噬菌体、粘粒、噬菌粒、及病毒等载体。作为质粒载体,没有限定,可举出来源于大肠杆菌的质粒(例如pET22b(+)、pBR322、pBR325、pUC118、pUC119、pUC18、pUC19、pBluescript等)、来源于枯草芽孢杆菌的质粒(例如pUB110、pTP5等)、及来源于酵母的质粒(例如YEp13、YCp50等)等。作为噬菌体载体,没有限定,可举出T7噬菌体展示载体(T7Select10-3b、T7Select1-1b、T7Select1-2a、T7Select1-2b、T7Select1-2c等(Novagen))、及λ噬菌体载体(Charon4A、Charon21A、EMBL3、EMBL4、λgt10、λgt11、λZAP、λZAPII等)。作为病毒载体,没有限定,可举出例如逆转录病毒、腺病毒、腺相关病毒、牛痘病毒、及仙台病毒等动物病毒、以及杆状病毒等昆虫病毒等。作为粘粒载体,没有限定,可举出Lorist 6、Charomid9-20、及Charomid9-42等。作为噬菌粒载体,没有限定,例如pSKAN、pBluescript、pBK、及pComb3H等是已知的。载体中可以以能表达目标DNA的方式含有调控序列、用于筛选含有目标DNA的载体的筛选标记、用于插入目标DNA的多克隆位点等。这样的调控序列中包括启动子、增强子、终止子、S-D序列或核糖体结合位点、复制起点、及多聚A位点等。另外,筛选标记可以使用例如氨苄青霉素抗性基因、新霉素抗性基因、卡那霉素抗性基因、及二氢叶酸还原酶基因等。用于导入载体的宿主细胞为大肠杆菌、枯草芽孢杆菌等细菌、酵母细胞、昆虫细胞、动物细胞(例如,哺乳动物细胞)、及植物细胞等,向这些细胞的转化或转染包括例如磷酸钙法、电穿孔法、脂质体转染法、粒子轰击(particle gun)法、及PEG法等。转化细胞的培养按照用于培养宿主生物的常规方法进行。例如,大肠杆菌、酵母细胞等微生物的培养液含有宿主微生物能同化的碳源、氮源、及无机盐类等。为了使本发明中使用的IgG结合肽的回收变得容易,优选使通过表达而生成的肽分泌至细胞外。这可以通过将编码能实现肽从其细胞中分泌的肽序列的DNA与编码目标肽的DNA的5'末端侧结合来进行。利用信号肽酶切割转移至细胞膜的融合肽,将目标肽分泌释放至培养基中。或者,也可以回收集聚至细胞内的目标肽。这种情况下,将细胞物理性或化学性地破坏,使用蛋白质纯化技术回收目标肽。
因此,本发明还涉及编码本发明中使用的肽的核酸。此处,核酸包括DNA或RNA(例如mRNA)。
使本发明中使用的IgG结合肽与其他蛋白质融合的情况下,可以分别制备IgG结合肽和其他蛋白质,然后根据需要使用接头使IgG结合肽与蛋白质融合,也可以利用基因重组法,根据需要加入合适的接头而作为融合蛋白质进行制作。这种情况下,优选以不损害本发明的IgG结合肽与IgG的结合性的方式制作融合蛋白质。
本发明中使用的上述IgG结合肽优选利用交联剂进行了修饰。如上所述,本发明中使用的IgG结合肽如后述实施例中所示,在上述Xaa1中,与IgG Fc的特定区域、即人IgG Fc中的基于Eu编号的Lys248或Lys246、优选Lys248接近。因此,通过将本发明中使用的IgG结合肽的Xaa1用交联剂修饰,使其与IgG进行交联反应,从而能够在IgG结合肽的Xaa1与IgGFc的Lys248或Lys246、优选Lys248之间位点特异性地形成交联结构。如上述那样,将本发明中使用的IgG结合肽的Xaa1用交联剂及各种化合物修饰,使其与IgG进行交联反应,能够将各种化合物特异性且简便地引入IgG。另外,根据本发明,能够介由IgG结合肽而引入上述式(I)的化合物、其他的化合物,因此,能够将各种结构的化合物引入IgG。此外,本发明由于得到的产物的收率高、并且未伴随抗体自身的改变,因此也具有使抗体功能降低的可能性低的优点。
本发明中使用的IgG结合肽也可以针对人以外的动物、优选哺乳动物的IgG使用。这种情况下,本发明中使用的IgG结合肽所结合的IgG中的位点可以由阅读了本说明书的本领域技术人员通过例如将人IgG的序列与其他动物的IgG的序列进行比对来容易地确定。
本发明中,“交联剂”是用于使本发明中使用的IgG结合肽与IgG Fc介由共价键而连接的化学物质。本发明的交联剂可以由本领域技术人员适当选择,可以是至少具有2处能与所期望的氨基酸(例如,赖氨酸残基、半胱氨酸残基、天冬氨酸残基、谷氨酸残基、2-氨基辛二酸、或二氨基丙酸、及精氨酸等)结合的位点的化合物。作为其例子,没有限定,可举出DSG(戊二酸二琥珀酰亚胺基酯,disuccinimidyl glutarate)、DSS(辛二酸二琥珀酰亚胺基酯,disuccinimidyl suberate)等优选含有2个以上琥珀酰亚胺基的交联剂、DMA(己二酰亚胺酸二甲酯二盐酸盐,dimethyl adipimidate·2HCl)、DMP(庚二酰亚胺酸二甲酯二盐酸盐,dimethyl pimelimidate·2HCl)、及DMS(辛二酰亚胺酸二甲酯二盐酸盐,dimethylsuberimidate·2HCl)等优选含有2个以上酰亚胺酸部分的交联剂、以及DTBP(3,3’-二硫代双丙酰亚胺酸二甲酯二盐酸盐,dimethyl 3,3’-dithiobispropionimidate·2HCl)及DSP(二硫代双(丙酸琥珀酰亚胺基酯),ithiobis(succinimidyl propionate))等具有SS键的交联剂。
本发明的利用交联剂进行了修饰的IgG结合肽可以利用WO2017/217437中记载的方法来制造。
另外,本发明中使用的IgG结合肽可以基于提高其稳定性等目的而通过C末端的酰胺化等进行修饰。
本发明中使用的IgG结合肽在N末端导入了修饰接头。作为修饰接头,可举出例如上述式(III)表示的修饰接头。
L1的聚乙二醇(PEG)接头的PEG的分子数量没有特别限定,可以是例如1~50个分子、1~20个分子、2~10个分子、2~6个分子。
L2为由0~5个以下氨基酸残基组成的氨基酸序列,优选至少含有半胱氨酸。
L3只要是至少含有DBCO(二苯并环辛炔)基的基团即可,可以选择市售的各种DBCO试剂(例如,DBCO-C6-酸、二苄基环辛炔胺、二苄基环辛炔-马来酰亚胺、DBCO-PEG酸、DBCO-PEG-NHS酯、DBCO-PEG-醇、DBCO-PEG-胺、DBCO-PEG-NH-Boc、羧基罗丹明-PEG-DBCO、磺酰罗丹明-PEG-DBCO、TAMRA-PEG-DBCO、DBCO-PEG-生物素、DBCO-PEG-DBCO、DBCO-PEG-马来酰亚胺、TCO-PEG-DBCO、DBCO-mPEG等)与L1或L2键合而形成的基团,作为一例,可举出上述式(IV)表示的DBCO-马来酰亚胺与L2的半胱氨酸残基反应而形成的基团。
式(III)的修饰接头具有成为点击反应的底物的DBCO,因此,能够与具有叠氮基的配体通过点击反应而结合。点击反应可以使用已知的条件,但只要在反应溶剂中于25~50℃的温度维持20~40分钟的时间即可。作为反应溶剂,除了磷酸缓冲液、Tris缓冲液、乙酸缓冲液、柠檬酸缓冲液等缓冲液以外,还可以使用二甲基亚砜等有机溶剂与少量缓冲液混合而成的混合溶剂。
<IgG抗体>
一个方式中,本发明涉及结合有本发明的修饰IgG结合肽的IgG抗体。该IgG抗体可以通过将修饰IgG结合肽与IgG抗体混合、使修饰IgG结合肽与IgG之间发生交联反应来形成。优选上述式(I)表示的IgG结合肽的上述Xaa1的氨基酸残基与IgG Fc的Lys248或Lys246、优选Lys248能够以位点特异性的方式发生交联反应。该交联反应可以利用WO2017/217437中记载的方法来实施。
适用本发明的IgG抗体只要包含至少1条重链即可,没有特别限定。重链的分子量为约50kDa,至少包含抗原结合区及Fc区。重链可以介由二硫键而结合有分子量为约25kDa的轻链。本发明的IgG抗体的分子量满足上述构成即可,没有特别限定,优选为约100~170kDa左右,更优选为约120~170kDa。
本发明中,“约”表示±10%的数值范围。
本发明的IgG抗体是通过于Fc区进行位点特异性的交联反应而形成的,因此,该交联反应对IgG的活性造成负面影响的可能性较小。另外,通过将修饰IgG结合肽与IgG连接,能够向IgG附加新的功能性。也可以以IgG的形式使用各种抗体医药。
<配体>
本发明的其他方式中,提供上述式(II)表示的肽系配体。
上述式(II)的化合物可以介由赖氨酸的侧链末端的NR1R2表示的基团而与IgG结合肽、修饰IgG结合肽、IgG抗体、抗体片段等多肽、其他可与靶分子结合的配体等靶分子元件结合,因此,能够用于各种靶分子元件的放射性金属标记。另外,上述式(II)的化合物具备具有苯丙氨酸残基及赖氨酸残基的肽链作为主链,该主链在肝脏中被代谢,因此,放射性金属向肝脏的集聚得到抑制,向尿中的排泄得到促进,体内动力学优异。由此,可以应用于不仅使用短半衰期放射性金属核素、也使用释放细胞损伤性的α射线、β射线的放射性金属核素的放射线治疗药,另外,由于放射性图像诊断中腹部的成为背景的放射性较少,因此,腹部的病灶部位的检测·诊断变得容易。
上述式(II)中R1及R2键合而与氮原子一同形成马来酰亚胺基的情况下,能够与半胱氨酸的SH基键合,R1及R2中的一方为氢原子且另一方为CO(CH2)4N3表示的基团的情况下,能够与DBCO等炔进行点击反应而结合。因此,通过使用上述式(III)表示的修饰接头、且使用下式(II-2)表示的化合物作为配体,能够使上述修饰IgG结合肽、或结合有所述修饰IgG结合肽的IgG抗体与下式(II-2)表示的配体结合。

<修饰抗体>
本发明的其他方式中,提供修饰抗体,其包含IgG抗体、和与所述IgG抗体结合的IgG结合肽,该IgG结合肽包含由上述的式(I)表示的、由13~17个氨基酸残基组成的氨基酸序列,在该IgG结合肽的N末端介由修饰接头而与上述式(II-1)表示的化合物于其赖氨酸残基的位置处连接。
本发明的修饰抗体中,优选上述式(II-1)表示的化合物的赖氨酸残基与修饰接头的结合位点通过DBCO基与叠氮基进行点击反应而形成。更优选的是,可以使用向上述式(II-1)表示的化合物的赖氨酸残基引入碳原子数为1~10的烷基叠氮基而得到的化合物,进一步优选使用上述式(II-2)表示的化合物。另外,此时,修饰接头优选为上述式(III)表示的修饰接头。点击反应可以使用已知的条件,但只要在反应溶剂中于25~50℃的温度维持20~40分钟的时间即可。作为反应溶剂,除了磷酸缓冲液、Tris缓冲液、乙酸缓冲液、柠檬酸缓冲液等缓冲液以外,还可以使用二甲基亚砜等有机溶剂与少量缓冲液混合而成的混合溶剂。
<放射性金属标记抗体>
本发明的其他方式中,提供放射性金属与上述修饰抗体配位而成的放射性金属标记抗体。
作为放射性金属核素的例子,可举出111In(铟)、89Zr(锆)、67/68Ga(镓)、64Cu(铜)、90Y(钇)、213Bi(铋)、225Ac(锕)、177Lu(镥)。结合至IgG结合肽的放射性金属核素可以根据本发明的放射性金属标记抗体的用途来选择。例如,癌症的检测·诊断可以使用111In、89Zr、64Cu及67/68Ga,例如PET(正电子发射断层成像,Positron Emission Tomography)可以使用89Zr及64Cu,SPECT(单光子发射计算机断层成像,Single Photon Emission ComputedTomography)可以使用111In。作为治疗用途,可以使用225Ac及177Lu。
为了使放射性金属与本发明的修饰抗体配位,例如,在反应溶剂中将两者于25~120℃的温度维持30~180分钟的时间即可。作为反应溶剂,可以使用磷酸缓冲液、Tris缓冲液、乙酸缓冲液、柠檬酸缓冲液、HEPES缓冲液等。
<放射性医药>
本发明的其他方式中,提供含有上述放射性金属标记抗体的放射性医药。
本发明的放射性医药可以作为例如核医学图像诊断剂或癌症的诊断剂使用。这种情况下,IgG抗体可以根据癌症种类而适当选择,例如,为乳腺癌时可以使用曲妥珠单抗(Trastuzumab)、为大肠癌时可以使用西妥昔单抗(Cetuximab)。核医学图像诊断剂或癌症的诊断剂可以按照常规方法制剂化(请参见例如Remington's Pharmaceutical Science,最新版,Mark Publishing Company,Easton,美国)。
实施例
以下,记载实施例而进一步详细地说明本发明,但本发明不受这些内容的限制。
实施例1:含有DBCO的IgG抗体的制作
制作了以下所示的结合有DBCO修饰IgG结合肽的IgG抗体。
[化学式5]
1)DBCO修饰IgG结合肽的制作
利用Fmoc固相合成法按照常规方法合成了将N末端的氨基用乙酰基修饰而成的氨基PEG5化IgG结合肽GPDCAYHKGELVWCTFH(序列号2,分子内的2个Cys介由二氯丙酮而交联,C末端经酰胺化)。去保护后,将纯化的IgG结合肽9.1mg溶解于含有0.5%吡啶的DMSO 100μL(33.3mM)中,向其中添加含有100mM DBCO-马来酰亚胺且含有0.5%吡啶的DMSO 50μL(以肽的摩尔比计为1.5倍的量),于室温使其反应30分钟。然后,向该肽溶液加入溶解于乙腈中的DSG(500mM)200μL(最终浓度为286mM,相对于肽而言为30倍摩尔当量),使其于50℃反应3小时。将总量(350μL)分成2份,稀释于含有0.1%TFA的10%乙腈5mL中,将离心后的上清液注入Inert Sustain(注册商标)C18色谱柱(7.6mm 1×250mm,GLScience),以从含有0.1%TFA的10%乙腈到含有0.1%TFA的60%乙腈的梯度洗脱。对洗脱物进行质谱分析,回收作为目标物质的DBCO修饰IgG结合肽后,除去有机溶剂,然后冷冻干燥。
2)含有DBCO的IgG抗体的制作
将上述1)中制备的修饰IgG结合肽以4mM的浓度溶解于DMSO中,将所得溶液25.2μL与溶解于10mM乙酸缓冲液(pH5.5)中的20μM的抗HER2人抗体(曲妥珠单抗)(中外制药)16.8mL混合,使其于37℃反应3小时(肽与抗体的摩尔比=3:1)。将如此制备的含有DBCO的抗体使用阳离子交换色谱柱CIM multus SO3-8(8mL,BIA separation)在10mM乙酸缓冲液(pH 5.5)中以从0到0.35M NaCl的梯度洗脱进行纯化。确认了除了未反应的抗体以外,生成了结合有1个肽的一价抗体和结合有2个肽的二价抗体。在此分取一价抗体的峰,然后在Vivaspin(截留分子量为10000Da,GE Healthcare)上以3000g离心,由此实施脱盐浓缩。
实施例2:DO3ABn-Phe-Lys((CH2)4-N3)-OH的合成
通过以下的路线(其中,步骤(a)的产物为化合物(4b),步骤(b)的产物为化合物(5b))合成了DO3ABn-Phe-Lys((CH2)4-N3)-OH(式(II-2)中,n为0的化合物)。需要说明的是,利用1H-NMR的分析使用了JEOLECS-400光谱仪(日本电子株式会社),利用ESI-MS的分析使用了HPLC1200系列6130四级杆LC/MS质谱仪(Agilent Technologies Japan,Ltd.)。

5-叠氮戊酸乙酯(Ethyl5-azidovalerate)(化合物(1))的合成
将5-溴代戊酸乙酯(Ethyl 5-bromovalerate)(1g,4.78mmol)溶解于N,N-二甲基甲酰胺(N,N-dimethylformamide)(DMF,3mL)中,加入NaN3(0.5g,7.7mmol),于85℃搅拌一夜。将混悬液用水(40mL)稀释,使用乙醚(diethyl ether)(40mL×3)萃取。将溶液减压浓缩,将所得溶液直接用于下一反应。
5-叠氮戊酸(5-Azidovaleric acid)(化合物(2))的合成
向化合物(1)的溶液中加入1N NaOH水溶液(8.2mL)后,加入甲醇(methanol)直到混合液呈均匀为止。于室温搅拌4小时后,将反应液减压浓缩,将得到的水溶液用乙醚(diethyl ether)(5mL)洗涤。然后,加入5%柠檬酸水溶液,使溶液成为酸性后,用乙醚(diethyl ether)(5mL×3)萃取。向有机层加入Na2SO4进行干燥后,减压蒸馏除去溶剂,由此得到化合物(2)(360mg,52.6%)。

5-叠氮戊酸2,3,5,6-四氟苯基酯(2,3,5,6-tetrafluorophenyl 5-
azidovalerate)(化合物(3))的合成
将化合物(2)(17.9mg,0.125μmol)、2,3,5,6-四氟苯酚(2,3,5,6-tetrafluorophenol)(44.0mg,0.250μmol)、N,N-二异丙基乙胺(N,N-diisopropylethylamine)(DIEA,53.2μL,0.312μmol)溶解于CH2Cl2(2.0mL)中后,在冰冷下加入1-乙基-3-(3-二甲基氨基丙基)碳二亚胺·盐酸盐(1-Ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride)(EDC,48.0mg,0.250μmol)。使溶液恢复至室温,搅拌1.5小时后,加入乙酸乙酯(ethyl acetate)(10mL),用饱和氯化铵溶液(3mL×3)洗涤。向有机层加入Na2SO4进行干燥后,减压蒸馏除去溶剂,由此得到化合物(3)(39.9mg)。得到的粗产物含有2,3,5,6-四氟苯酚(2,3,5,6-tetrafluorophenol),但直接用于下一反应。
DO3ABn-Phe-Lys-OH(化合物(4b))的合成
使Phe-Lys通过肽延伸反应与Cl-Trt(2-Cl)-Reisn(渡边化学工业,制品编号:A00187)结合,针对(tBu)3DO3ABn-COOH(16-20mg),加入1.3等量的结合有Phe-Lys的树脂、O-(7-氮杂苯并三唑-1-基)-N,N,N’,N’-四甲基脲六氟磷酸盐(HATU,2.5等量)、1-羟基-7-氮杂苯并三唑(HOAt,2.5等量)、DIEA(5.0等量),在DMF(200μL)中于室温搅拌一夜。使用DMF洗涤8次,然后使用CH2Cl2洗涤4次,然后将树脂减压干燥。向干燥后的树脂加入三氟乙酸(TFA)/三异丙基硅烷(triisopropylsilane)/水=95/2.5/2.5的混合液,于室温搅拌3小时。减压蒸馏除去溶剂后,加入乙醚(diethyl ether)(5mL),由此使其结晶化。滤取晶体,用乙醚(diethyl ether)洗涤后,减压干燥,由此以白色晶体的形式得到了化合物(4b)的TFA盐7.7mg(25.7%)。
DO3ABn-Phe-Lys((CH2)4-N3)-OH(化合物(5b))的合成
将化合物(4b)溶解于DMF(0.5mL)中,加入化合物(3)(7.5等量)、DIEA(4.0等量),于室温搅拌3小时。减压蒸馏除去溶剂后,再次溶解于水(5mL)中。将得到的水溶液用CHCl3(3mL×3)洗涤后,减压浓缩,通过分离提取用HPLC进行纯化,由此以白色晶体的形式得到了化合物(5b)的TFA盐1.7mg(20.0%)。ESI-MS(M+H)+:m/z 881,found:881
需要说明的是,在利用分离提取用HPLC的纯化中,使用Imtakt Cadenza C18(20mm×150mm)作为色谱柱,A相使用0.1%TFA/水,B相使用0.1%TFA/甲醇(methanol),利用B相于0~45分钟从10%上升至50%、于45~55分钟从50%上升至100%的梯度系统,以流速5mL/分钟分取保留时间为38.0分钟的峰。
另外,质谱分析通过以下方式实施:使用Imtakt US-C18(4.6mm×150mm)作为色谱柱,A相使用0.1%TFA/水,B相使用0.1%TFA/乙腈(acetonitrile)、利用B相于0~30分钟内从0上升至45%、于30~40分钟内从45上升至100%的梯度系统,以流速1mL/分钟对保留时间为25.0分钟的峰进行分析。
实施例3:DO3ABn-Phe-Gly-Lys((CH2)4-N3)-OH的合成通过实施例2所示的路线(其中,步骤(a)的产物为化合物(4c),步骤(b)的产物为化合物(5c))合成了DO3ABn-Phe-Lys((CH2)4-N3)-OH(式(II-2)中,n为1的化合物)。
DO3ABn-Phe-Gly-Lys-OH(化合物(4c))的合成
除了使用结合有Phe-Gly-Lys代替Phe-Lys的树脂以外,实施与实施例2的化合物(4b)同样的操作,以白色晶体的形式得到了化合物(4c)的TFA盐30.0mg(76.8%)。
DO3ABn-Phe-Gly-Lys((CH2)4-N3)-OH(化合物(5c))的合成除了使用化合物(4c)代替化合物(4b)以外,实施与实施例2的化合物(5b)同样的操作,以白色晶体的形式得到了化合物(5c)的TFA盐4.6mg(14.0%)。

需要说明的是,利用HPLC的纯化中,与实施例2不同,使用Imtakt Cadenza C18(20mm×150mm)作为色谱柱,A相使用0.1%TFA/水,B相使用0.1%TFA/乙腈(acetonitrile),利用B相于0~45分钟从10%上升至50%、于45~55分钟从50%上升至100%的梯度系统,以流速5mL/分钟分取保留时间为33.9分钟的峰。
另外,质谱分析的条件与实施例2相同,但保留时间为24.7分钟。
比较例1:DO3ABn-Gly-Lys((CH2)4-N3)-OH的合成
通过实施例2所示的路线(其中,步骤(a)的产物为化合物(4a),步骤(b)的产物为化合物(5a))合成了DO3ABn-Gly-Lys((CH2)4-N3)-OH(式(II-2)中,n为1、且不具有苯基残基)。
DO3ABn-Gly-Lys-OH(化合物(4a))的合成
除了使用结合有Gly-Lys代替Phe-Lys的树脂以外,实施与实施例2的化合物(4b)同样的操作,以白色晶体的形式得到了化合物(4a)的TFA盐8.3mg(25.3%)。
DO3ABn-Gly-Lys((CH2)4-N3)-OH(化合物(5a))的合成
除了使用化合物(4a)代替化合物(4b)以外,实施与实施例2的化合物(5b)同样的操作,以白色晶体的形式得到了化合物(5a)的TFA盐1.5mg(16.3%)。

其中,HPLC纯化中的保留时间为22.6分钟,质谱分析中的保留时间为19.3分钟。
制造例1:DBCO-NGA的制备
使用与Journal of Medicinal Chemistry 1997,40,2643-2652同样的方法,制备了利用2-亚氨基硫杂环戊烷(2-iminothiolane)引入巯基而得到的NGA溶液10mg/mL。向该溶液100μL中添加溶解有相对于半乳糖结合白蛋白(NGA)而言为50等量的DBCO-马来酰亚胺(maleimide)(购自Aldrich公司)的DMSO溶液2μL,于37℃反应1.5小时。反应结束后,将溶液使用以用D-PBS(-)平衡化后的sephadex G-50作为担载体的离心柱(spin column)进行纯化,由此制作了以平均结合数2.1结合有DBCO的DBCO-NGA。
制造例2:111In-DO3ABn-Phe-Lys-NGA的制作
DO3ABn-Phe-Lys((CH2)4-N3)-OH的111In标记及与DBCO-NGA的点击反应按照以下的路线(其中,步骤(a)的产物为化合物(6b),步骤(b)的产物为化合物(7b))进行合成。

111In-DO3ABn-Phe-Lys((CH2)4-N3)-OH(化合物(6b))的制作向111InCl3溶液(1.48MBq/60μL)添加1M乙酸缓冲液(pH5.5,20μL),于室温静置5分钟,向所得溶液添加以5×10-4M的浓度溶解于0.1M乙酸缓冲液(pH 5.5,20μL)的化合物(5b)。使溶液于95℃反应5分钟后,恢复至室温,加入0.2M DTPA溶液10μL,于室温静置5分钟。将反应液利用分析用HPLC纯化。将含有目标物质的组分利用Sep-pak将溶剂置换为甲醇,向得到的溶液中加入D-PBS(-)30μL,将溶剂减压浓缩。A相使用0.05M乙酸缓冲液(pH 5.5),B相使用甲醇(methanol),利用B相于0~30分钟从20%上升至60%、于30~35分钟从60%上升至100%的梯度系统,以流速1mL/分钟,分取保留时间为20.2分钟的峰,从而进行纯化,以放射化学收率>99%得到了化合物(6b)。
111In-DO3ABn-Phe-Lys-NGA(7b)的制作
将化合物(7b)的D-PBS(-)溶液30μL添加至DBCO-NGA溶液10μL,于室温反应14小时。将反应后的溶液使用以用D-PBS(-)平衡化后的sephadex G-50作为担载体的离心柱进行纯化,由此以放射化学收率43.0%得到了作为目标的111In标记NGA(化合物(7b))。
制造例3:111In-DO3ABn-Phe-Gly-Lys-NGA的制作
DO3ABn-Phe-Gly-Lys((CH2)4-N3)-OH的111In标记及与DBCO-NGA的点击反应按照制造例2所示的路线(其中,步骤(a)的产物为化合物(6c),步骤(b)的产物为化合物(7c))进行合成。
111In-DO3ABn-Phe-Gly-Lys((CH2)4-N3)-OH(化合物(6c))的制作
除了使用化合物(5c)代替化合物(5b)以外,实施与制造例2的化合物(6b)同样的操作,以放射化学收率98.7%得到了化合物(6c)。
其中,HPLC纯化中的保留时间为18.7分钟。
111In-DO3ABn-Phe-Gly-Lys-NGA(7c)的制作
除了使用化合物(6c)代替化合物(6b)以外,实施与制造例2的化合物(6b)同样的操作,以放射化学收率50.0%得到了化合物(7c)。
制造例4:111In-DO3ABn-Gly-Lys-NGA的制作
DO3ABn-Gly-Lys((CH2)4-N3)-OH的111In标记及与DBCO-NGA的点击反应按照制造例2所示的路线(其中,步骤(a)的产物为化合物(6a),步骤(b)的产物为化合物(7a))进行合成。
111In-DO3ABn-Gly-Lys((CH2)4-N3)-OH(化合物(6c))的制作
除了使用化合物(5a)代替化合物(5b)以外,实施与制造例2的化合物(6b)同样的操作,以放射化学收率93.2%得到了化合物(6a)。
其中,利用分析用HPLC的纯化中,使用Imtakt US-C18(4.6mm×150mm)作为色谱柱,A相使用0.05M乙酸缓冲液(pH 5.5)、B相使用甲醇(methanol),利用B相于0~30分钟从0上升至40%、于30~35分钟从40%上升至100%的梯度系统,以流速1mL/分钟分取保留时间为19.1分钟的峰,由此进行纯化。
除了使用化合物(6a)代替化合物(6b)以外,实施与制造例2的化合物(6b)同样的操作,以放射化学收率38.0%得到了化合物(7a)。
制造例5:CHX-A”-DTPA-NGA的制作
将NGA溶解于0.1M硼酸缓冲液(pH 8.5)中,制备NGA溶液(250μg/50μL)。另外,将p-SCN-CHX-A”-DTPA((R)-2-氨基-3-(4-异硫氰酸酯基苯基)丙基]-反-(S,S)-环己烷-1,2-二胺-五乙酸,(R)-2-Amino-3-(4-isothiocyanatophenyl)propyl]-trans-(S,S)-cyclohexane-1,2-diamine-pentaacetic acid,购自Aldrich公司)溶解于0.1M硼酸缓冲液(pH 8.5)中(6mg/mL),加入0.1N NaOH,将pH调节为8~9后,使用0.1M硼酸缓冲液(pH 8.5)稀释,由此制备p-SCN-CHX-A”-DTPA溶液(5mg/mL)。将20等量的p-SCN-CHX-A”-DTPA溶液加入NGA溶液中,于室温反应一夜。
反应结束后,将溶液使用以用0.25M乙酸缓冲液(pH 5.5)平衡化后的sephadex G-50作为担载体的离心柱进行纯化,由此得到作为目标的CHX-A”-DTPA-NGA溶液。
制造例6:111In-CHX-A”-DTPA-NGA的制作
将111InCl3溶液(1.48MBq/20μL)添加至1M乙酸缓冲液(pH5.5,5μL)中,于室温静置5分钟,向所得溶液中添加CHX-A”-DTPA-NGA溶液(30μg/15μL)。使溶液于37℃反应1.5小时后,恢复至室温,加入0.2M DTPA溶液10μL,于室温静置5分钟。将反应后的溶液使用以用D-PBS(-)平衡化后的sephadex G-50作为担载体的离心柱进行纯化,由此得到作为目标的111In-CHX-A”-DTPA-NGA。
实施例4:放射性金属标记IgG抗体[111In]H-FGK-DOTA的制作
使实施例1中得到的含有DBCO的IgG抗体1.4mg(1.5nmol)及实施例3中得到的DO3ABn-Phe-Gly-Lys((CH2)4-N3)-OH 0.1mg(75nmol)在10mM乙酸缓冲液167.5μL中于室温下进行点击反应。反应结束后,通过超滤进行纯化,由此制作了介由DBCO修饰IgG结合肽使曲妥珠单抗(Trastuzumab)与DOTA-FGK结合而成的修饰抗体(以下为“H-FGK-DOTA”)。将得到的H-FGK-DOTA在111InCl3溶液200μL(19.7MBq,pH6.5)和100mM HEPES缓冲液50μL中混合。需要说明的是,以相对于[111In]而言的H-FGK-DOTA的摩尔数成为20~100倍的方式进行了制备。通过使其于45℃反应2小时来进行络合物形成反应,络合物形成后,通过超滤进行纯化。使用放射性同位素活度计测定放射性活度,结果,得到的[111In]H-FGK-DOTA的放射性活度为2.33MBq,校正衰减而算出的放射化学收率为14.0%。
另外,将[111In]H-FGK-DOTA滴落至滤纸,然后利用100mM柠檬酸钠缓冲液展开。展开结束后,使用TLC扫描仪(Gita Star,raytest公司)测定,结果,放射化学纯度为97.1%。
实施例5:放射性金属标记IgG抗体[111In]H-DOTA的制作
使实施例1中得到的含有DBCO的IgG抗体及1,4,7,10-四氮杂环十二烷-1,4,7-三(乙酸)-10-(叠氮基丙基乙基乙酰胺)(Azide-mono-amide-DOTA,Macrocyclics公司)0.04mg(75nmol)在乙酸缓冲液167.5μL于室温下进行点击反应。反应结束后,通过超滤进行纯化,由此制作了介由DBCO修饰IgG结合肽使曲妥珠单抗与DOTA结合的IgG抗体(以下为“H-DOTA”)。将得到的H-DOTA在111InCl3溶液200μL(21.3MBq,pH6.5)和100mM HEPES缓冲液50μL中混合,由此进行络合物形成反应,络合物形成后,通过超滤进行纯化,由此得到[111In]H-DOTA 2.99MBq。与实施例3同样地算出放射化学收率及放射化学的纯度,结果,放射化学收率为19.5%,放射化学纯度为99.6%。
评价1:动物实验(1)
将制造例2~4及6中得到的各111In标记NGA溶液以成为(11.1kBq/8μg蛋白/100μL/只小鼠)的浓度的方式使用D-PBS(-)稀释·调节,经由ddY、6周龄的雄性小鼠的尾静脉施予。在施予5分钟、或1小时后宰杀1组5只小鼠,回收血液及所关注的器官,测定器官重量后,利用伽马射线自动计数仪(Auto-well Gamma Counter)测定放射性。

表1示出施予5分钟后的各器官放射性(%ID)。表2示出施予1小时后的各器官放射性(%ID)。Gly-Lys表示施予了制造例4中制作的111In-DO3ABn-Gly-Lys-NGA的结果,Phe-Lys表示施予了制造例2中制作的111In-DO3ABn-Phe-Lys-NGA的结果,Phe-Gly-Lys表示施予了制造例3中制作的111In-DO3ABn-Phe-Gly-Lys-NGA的结果,SCN表示制造例6中制作的111In-CHX-A”-DTPA-NGA的结果。
如表1所示,施予5分钟的情况下,在肝脏中确认了放射性集聚,由于包含Phe-Lys及Phe-Gly-Lys的氨基酸序列,因此确认到从肝脏的排泄。
评价2:动物实验(2)
出于使用实施例4中得到的[111In]H-FGK-DOTA及实施例5中得到的[111In]H-DOTA来调查FGK序列的有效性的目的,实施了体内(in vivo)实验。
将溶剂置换为磷酸缓冲生理盐水(PBS)的[111In]H-FGK-DOTA及[111In]H-DOTA以成为每100μL为200kBq的方式调节后,在异氟烷麻醉下,向3只6周龄的雄性小鼠(ddY)各自尾静脉施予100μL,施予后,在第1小时、6小时、24小时通过放血致死进行宰杀。摘出器官(心脏、肺、肝脏、脾脏、肾脏、胃、小肠、大肠、下肢骨、肌肉、膀胱),与血液、尿液、粪便、剩余全身一同测量重量后,测定放射性活度。各小时时间点的放射性分布(注射量%(ID))的平均值及标准偏差示于图1。另外,图2中示出所关注的器官的体内动力学(%ID)。统计分析使用学生t检验进行。另外,显著性差异的判定为P<0.05。
图1A为示出施予后1小时的结果的图,图1B为示出施予后6小时的结果的图,图1C为示出施予后24小时的结果的图。
图2A为示出肝脏中的放射性分布(%ID)的经时变化的图,图2B为示出肾脏中的放射性分布(%ID)的经时变化的图。图2C中,“肾脏及泌尿系统”表示肾脏、膀胱及尿液中的放射性分布(%ID)的合计的经时变化,“肝脏及胆道系统”表示肝脏、小肠、大肠及粪便中的放射性分布(%ID)的合计的经时变化。
根据在施予后6小时时间点[111In]H-DOTA与[111In]H-FGK-DOTA的肝脏摄入存在差异的情况,以及在施予后24小时时间点[111In]H-FGK-DOTA向尿中的排泄显著上升的情况,表明利用FGK序列成功地促进了放射性核素的排泄,使向肝脏的集聚减少。
本申请以2018年4月16日提出申请的日本申请特愿2018-78487号为基础主张优先权,其全部公开内容并入本说明书中。
Claims (12)
1.修饰抗体,其是包含IgG抗体、和与所述IgG抗体结合的IgG结合肽的修饰抗体,其中,
所述IgG结合肽包含由下式(I)表示的、由13~17个氨基酸残基组成的氨基酸序列,
(X1-3)-C-(X2)-H-(Xaa1)-G-(Xaa2)-L-V-W-C-(X1-3) (I)
式(I)中,
X各自独立地为半胱氨酸以外的任意的氨基酸残基,
C为半胱氨酸残基,
H为组氨酸残基,
Xaa1为赖氨酸残基、半胱氨酸残基、天冬氨酸残基、谷氨酸残基、2-氨基辛二酸、或二氨基丙酸,
G为甘氨酸残基,
Xaa2为谷氨酸残基、谷氨酰胺残基或天冬酰胺残基,
L为亮氨酸残基,
V为缬氨酸残基,且
W为色氨酸残基,
2个半胱氨酸残基可以彼此介由二硫键或接头而结合,C末端可以经过酰胺化,
在所述IgG结合肽的N末端,介由修饰接头而与下式(II-1)表示的化合物于其赖氨酸残基的位置处连接,
式(II-1)中,n为0或1。
2.如权利要求1所述的修饰抗体,其中,所述式(II-1)表示的化合物的赖氨酸残基与所述修饰接头的结合位点通过DBCO(二苯并环辛)基与叠氮基进行点击反应而形成。
3.如权利要求2所述的修饰抗体,其中,所述修饰接头为下式(III)表示的修饰接头,
L1-L2-L3 (III)
L1为与所述IgG结合肽的N末端结合的聚乙二醇接头,
L2为由0个以上且5个以下氨基酸残基组成的氨基酸序列,
L3为在末端具有DBCO(二苯并环辛)基的基团,
通过向所述式(II)表示的化合物的赖氨酸残基引入碳原子数为1~10的烷基叠氮基,从而经由点击反应而形成所述赖氨酸残基与所述修饰接头的结合位点。
4.如权利要求1~3中任一项所述的修饰抗体,其中,所述式(I)表示的IgG结合肽包含下述的氨基酸序列,
GPDCAYHKGELVWCTFH(序列号2)
所述氨基酸序列中,2个半胱氨酸残基可以彼此以二硫键结合,C末端可以经过酰胺化。
6.放射性金属标记抗体,其包含配位有放射性金属的权利要求1~5中任一项所述的修饰抗体。
7.放射性医药,其含有权利要求6所述的放射性金属标记抗体。
8.修饰IgG结合肽,其包含:
IgG结合肽,其包含由下式(I)表示的、由13~17个氨基酸残基组成的氨基酸序列;和
与所述IgG结合肽的N末端结合的、下式(III)表示的修饰接头,
(X1-3)-C-(X2)-H-(Xaa1)-G-(Xaa2)-L-V-W-C-(X1-3) (I)
式(I)中,

9.IgG抗体,其结合有权利要求8所述的修饰IgG结合肽。
11.使可与靶分子结合的多肽与权利要求10所述的化合物结合而成的化合物或其盐。
12.如权利要求11所述的化合物或其盐,其中,所述多肽为权利要求8所述的修饰IgG结合肽、或权利要求9所述的IgG抗体。
