Description

Fullerene maintenance antibacterial gel and preparation method thereof
Technical Field
The invention belongs to the technical field of gynecological product nursing, and particularly relates to a fullerene maintenance antibacterial gel and a preparation method thereof.
Background
Along with the improvement of living standard of people, the health protection consciousness of people is gradually strengthened, and more people begin to pay attention to personal health and protection, especially the private maintenance of women. Women begin to age more than 20 years, and with the age, the body of the women ages, the fat in private parts begins to decrease, and gradually becomes loose and aged with the age. When women approach the menopause, their intravaginal membranes begin to thin, blood flow decreases, and pelvic floor muscles weaken. The loose pudendum can affect the female sexual confidence, and the skin of the pudendum can be excessively stretched by unreasonable movement, which causes injury.
With age, parts of the body are constantly exposed to oxygen, particularly organs affected by oxygen including reactive oxygen species, lipid peroxides, and the like. The theory of free radicals considers that free radicals are an important factor affecting human aging. Endogenous oxygen free radicals are produced during the metabolism of the body, and the free radicals produced by the metabolism are the main sources of free radicals in the human body. The excessive free radical aggregation of human body can cause body damage, the damage accumulation exceeds the repair capability of the body, various macromolecular damages of skin can be caused, the cell differentiation state is changed and even lost, therefore, the aging of the body part is accelerated, and the free radical is an important factor for causing skin damage and aging in the aging process of the body part.
The existing private products generally have the bacteriostatic function, do not have the function of scavenging free radicals, and have single efficacy and poor bacteriostatic effect.

2. Bacteriostasis test
The bacteriostatic test mainly adopts a suspension method to test the bacteriostatic effect of the fullerene curing bacteriostatic gel on candida albicans, staphylococcus aureus and escherichia coli, and the specific operation method is as follows:
1. preparing a bacterial suspension: before experiment, the working strain is streaked and inoculated on an agar slant, the agar slant is cultured for 24h at 36.0 ℃, and the slant of the cultured test strain is washed by 0.03mol/LPBS to prepare bacterial suspension.
2. The detection method comprises the following steps: 5mL of the test sample solution and 4 tubes of the control sample solution were each sampled.
And (3) taking the bacterial suspension, respectively dripping 100 mu L of the bacterial suspension on each sample to be tested and each control sample, uniformly mixing, starting timing, acting for 2min, 5min, 10min and 20min, sucking 0.5mL of sample liquid, adding the sample liquid into a test tube containing 5.0mL of PBS, fully and uniformly mixing, diluting, then taking 2-3 dilutions, sucking 0.5mL of the dilutions respectively, placing the dilutions in 2 plates, rotating the plates by using a culture medium cooled to 45 ℃, fully and uniformly stirring, turning the plates after agar is solidified, culturing for 48 hours at 36.0 ℃, and counting viable bacteria colonies.
The bacteriostatic rate was calculated as follows:
X4=(A-B)/A×100%
in the formula: x4Percent is the bacteriostatic rate;
a is the average colony number of the control sample;
b is the average colony number of the test sample.
The bacteriostatic rate is more than or equal to 50-90%, the product has bacteriostatic action, the bacteriostatic rate is more than or equal to 90%, the product has strong bacteriostatic action, examples 1-6 and comparative examples 1-3, and the bacteriostatic test results of the existing gels (DoctorYang bacteriostatic gel and Sunday whisper bacteriostatic gel) in the market are shown in Table 2:
TABLE 2 results of bacteriostasis tests on examples 1-6, comparative examples 1-3 and commercial gels
Figure BDA0002589198110000111
Figure BDA0002589198110000121
The test results in table 2 show that the fullerene maintenance antibacterial gel of the embodiments 1 to 6 of the invention can not only increase the skin elasticity, but also has excellent antibacterial effects on candida albicans, staphylococcus aureus and escherichia coli; the gels of comparative examples 1-3 have relatively poor effect on candida albicans, and the carrier Yang bacteriostatic gel and summer whisper bacteriostatic gel on the market have no bacteriostatic effect as the fullerene curing bacteriostatic gel.
The gel disclosed by the invention adopts antibacterial components in a reasonable proportion, the components are safe and non-irritant, and the gel disclosed by the invention has an obvious effect on resisting free radicals and preventing organism aging through synergistic interaction among the components, and has excellent antibacterial performance.
The antibacterial gel is prepared by compounding fullerene C60, gene recombinant human collagen, a humectant, a plant extract, a thickening agent, a pH regulator, an anti-inflammatory agent, a bacteriostatic agent and purified water according to the mass ratio of 1:1, wherein the fullerene C60 and the gene recombinant human collagen can resist free radicals of organisms, activate cells and play an external protection effect, and the gene recombinant human collagen has excellent biocompatibility, can adjust the synthesis of human collagen and plays an internal maintenance effect; the Hamamelis virginiana extract selected by the invention has the effects of anti-inflammation and convergence, the phellodendron extract and the cacumen biotae extract have excellent antibacterial and antiviral effects, and the purslane extract has the effects of anti-allergy and anti-inflammation.
Because the plant extracts have anti-inflammatory or antibacterial and antiviral effects, when the fullerene curing and bacteriostasis gel is prepared, the antibacterial effect and the effect of removing free radicals can be achieved by selecting different plant extracts, and the anti-inflammatory or antibacterial and antiviral emphasis points are different.
The fullerene C60 used in the invention is the minimum molecule reaching a stable structure in a fullerene spherical structure, so that the fullerene C60 has relatively small molecular weight and is easy to be absorbed by skin while having stable property, can be compatible with free radicals generated by organisms, has high-efficiency oxidation resistance, can play a role in activating cells and preventing skin from fading, and can remarkably enhance the skin elasticity and tighten the skin after being used for a long time by adding the fullerene C60 into a system;
the gene recombinant human collagen selected by the invention is type III collagen, is collagen with very rich content in fibrous collagen, has complete affinity and excellent biocompatibility with human source fragments, is more beneficial to skin absorption, can increase fibroblasts after long-term use, regulates the collagen synthesis of a human body, and promotes the skin elasticity to increase.
