The invention provides a composition containing a compound extract of kochia scoparia and cnidium fruit, which comprises the following components in parts by weight: 7g of compound extract of kochia scoparia and cnidium fruit, 10g of extract of folium Rubi Bungeanae, 4g of extract of folium orthosiphoni, 5g of extract of folium Viticis negundo, 25g of butanediol, 9g of 1, 2-pentanediol and 40g of water.
Example 6
The invention provides a composition containing a compound extract of kochia scoparia and cnidium fruit, which comprises the following components in parts by weight: 7g of compound extract of kochia scoparia and cnidium fruit, 8g of extract of folium Rubi Bungeanae, 6g of extract of folium orthosiphoni, 5g of extract of folium Viticis negundo, 25g of butanediol, 9g of 1, 2-pentanediol and 40g of water.
The kochia scoparia and cnidium fruit composite extracts described in examples 5-6 are prepared by the following steps in sequence:
(1) pretreatment of raw materials: respectively crushing the dried fructus kochiae and the dried fructus cnidii, sieving the crushed fructus kochiae and the dried fructus cnidii with a 60-mesh sieve, and mixing the ground fructus kochiae and the dried fructus cnidii in a mass ratio of 2:5 for later use;
(2) enzymatic hydrolysis: adding water into the mixed powder in the step (1) according to the material-liquid ratio of 1:10(g: mL), adding 0.2 wt% of mannose polymerase (enzyme activity of 30000u/g) and xylanase (enzyme activity of 50000u/g) in a mass ratio of 6:1 into the mixed powder, stirring for 70min at the pH of 4.0 and the temperature of 55 ℃, and heating to boil to inactivate the enzyme activity;
(3) pressure extraction: adding absolute ethanol into the enzymolysis liquid obtained in the step (2) to obtain a 60% ethanol solution, and placing the ethanol solution in a high-temperature high-pressure reaction container, wherein the pressure is 0.4MPa, the temperature is 70 ℃, and the extraction time is 35 min;
(4) and (3) purification: filtering the extracting solution obtained in the step (3), concentrating under reduced pressure to 1/6 of the original volume, passing the concentrated solution through an AB-8 macroporous adsorption resin column, eluting with 3 times of column volume of water, eluting with 4 times of column volume of 10% ethanol solution, discarding the eluent, eluting with 3 times of column volume of 70% ethanol solution at an elution rate of 1.5BV/h, recovering the eluent, and concentrating under reduced pressure to dryness to obtain the compound extract of kochia fruit and cnidium fruit.
Example 7
The invention provides a composition containing a compound extract of kochia scoparia and cnidium fruit, which comprises the following components in parts by weight: 7g of compound extract of kochia scoparia and cnidium fruit, 8g of extract of folium Rubi Bungeanae, 6g of extract of folium orthosiphoni, 5g of extract of folium Viticis negundo, 25g of butanediol, 9g of 1, 2-pentanediol and 40g of water.
The compound extract of kochia scoparia and cnidium fruit in example 7 is prepared by the following steps in sequence:
(1) pretreatment of raw materials: respectively crushing the dried fructus kochiae and the dried fructus cnidii, sieving the crushed fructus kochiae and the dried fructus cnidii with a 60-mesh sieve, and mixing the ground fructus kochiae and the dried fructus cnidii in a mass ratio of 1:3 for later use;
(2) enzymatic hydrolysis: adding water into the mixed powder in the step (1) according to the material-liquid ratio of 1:10(g: mL), adding 0.2 wt% of the mixed powder, namely, mannose polymerase (enzyme activity of 30000u/g) and xylanase (enzyme activity of 50000u/g) in the mass ratio of 2:1, stirring for 70min at the temperature of 55 ℃ at the pH value of 4.0, and heating to boil to inactivate the enzyme;
(3) pressure extraction: adding absolute ethanol into the enzymolysis liquid obtained in the step (2) to obtain a 60% ethanol solution, and placing the ethanol solution in a high-temperature high-pressure reaction container, wherein the pressure is 0.4MPa, the temperature is 70 ℃, and the extraction time is 35 min;
(4) and (3) purification: filtering the extracting solution obtained in the step (3), concentrating under reduced pressure to 1/6 of the original volume, passing the concentrated solution through an AB-8 macroporous adsorption resin column, eluting with 3 times of column volume of water, eluting with 4 times of column volume of 10% ethanol solution, discarding the eluent, eluting with 3 times of column volume of 70% ethanol solution at an elution rate of 1.5BV/h, recovering the eluent, and concentrating under reduced pressure to dryness to obtain the compound extract of kochia fruit and cnidium fruit.
The preparation method of the composition containing the compound extracts of kochia scoparia and cnidium fruit in any one of the above examples 1 to 7 is as follows:
(1) mixing phenoxyethanol with small amount of water uniformly in advance for use (when phenoxyethanol is not present, the step is omitted);
(2) heating the residual water to 40-45 ℃, adding the polyhydric alcohol, uniformly stirring, adding the mixture obtained in the step (1) which is treated in advance, uniformly stirring, adding other materials, and uniformly stirring to obtain the water-based paint.
To demonstrate the effect of the present invention, comparative experimental examples of the present application are given below:
comparative example 1
A composition comprising the following components in parts by weight: 5g of compound extract of kochia scoparia and cnidium fruit, 21g of extract of folium orostachydis scholaris, 36g of butanediol and 38g of water.
The preparation method of the compound extract of kochia scoparia and cnidium fruit is the same as that of example 1.
Compared with example 1, only the clerodendranthus spicatus extract was replaced with the same amount of the extract of the leaf of the Rubus sibirica, and the other ingredients were the same.
Comparative example 2
A composition comprising the following components in parts by weight: 6g of the folium oroxyli extract, 2g of the clerodendranthus spicatus extract, 18g of the vitex negundo extract, 30g of butanediol, 10g of 1, 2-hexanediol and 34g of water.
In comparison with example 3, only the combined extract of kochia scoparia and cnidium fruit was replaced with the same amount of the extract of vitex negundo, and the other ingredients were the same.
Comparative example 3
A composition comprising the following components in parts by weight: 7g of compound extract of kochia scoparia and cnidium fruit, 8g of extract of folium Rubi Bungeanae, 6g of extract of folium orthosiphoni, 5g of extract of folium Viticis negundo, 25g of butanediol, 9g of 1, 2-pentanediol and 40g of water.
The compound extract of the belvedere fruit and the cnidium fruit is prepared by the following steps in sequence:
(1) pretreatment of raw materials: respectively crushing the dried fructus kochiae and the dried fructus cnidii, sieving the crushed fructus kochiae and the dried fructus cnidii with a 60-mesh sieve, and mixing the ground fructus kochiae and the dried fructus cnidii in a mass ratio of 2:5 for later use;
(2) enzymatic hydrolysis: adding water into the mixed powder in the step (1) according to the feed-liquid ratio of 1:10(g: mL), adding 0.2% of xylanase (enzyme activity of 50000u/g) by weight of the mixed powder, stirring for 70min at the pH of 4.0 and the temperature of 55 ℃, and heating to boil to inactivate the enzyme activity;
(3) pressure extraction: adding absolute ethanol into the enzymolysis liquid obtained in the step (2) to obtain a 60% ethanol solution, and placing the ethanol solution in a high-temperature high-pressure reaction container, wherein the pressure is 0.4MPa, the temperature is 70 ℃, and the extraction time is 35 min;
(4) and (3) purification: filtering the extracting solution obtained in the step (3), concentrating under reduced pressure to 1/6 of the original volume, passing the concentrated solution through an AB-8 macroporous adsorption resin column, eluting with 3 times of column volume of water, eluting with 4 times of column volume of 10% ethanol solution, discarding the eluent, eluting with 3 times of column volume of 70% ethanol solution at an elution rate of 1.5BV/h, recovering the eluent, and concentrating under reduced pressure to dryness to obtain the compound extract of kochia fruit and cnidium fruit.
In comparison with example 6, the composition had the same raw material composition. But the compound extract of the kochia scoparia and the cnidium fruit is only added with xylanase for enzymolysis in the step (2) during preparation.
Comparative example 4
A composition comprising the following components in parts by weight: 2g of kochia scoparia extract, 5g of cnidium fruit extract, 8g of folium rambutae, 6g of clerodendranthus spicatus extract, 5g of vitex negundo leaf extract, 25g of butanediol, 9g of 1, 2-pentanediol and 40g of water.
The above Kochia scoparia extract and cnidium officinale extract are commercially available. Kochia scoparia extract and cnidium officinale extract were obtained from Lebo cosmeceutical technology (Shanghai) Co., Ltd.
Comparative examples 1-4 the compositions of any one of the groups were prepared in the same manner as in the examples.
The composition containing the compound extracts of the kochia scoparia and the cnidium fruit can be used as an active ingredient for preparing daily necessities for killing and inhibiting mites. When the composition is used specifically, the composition prepared by the invention is directly added into a daily necessity matrix, and the addition amount is 0.2-10 wt%.
To better illustrate the advantages of the present invention, the efficacy test of the composition containing the compound extract of kochia scoparia and cnidium fruit provided by the present invention is given below:
test 1: skin irritation test
(1) Test samples: compositions prepared in examples 1-7.
(2) The test method comprises the following steps: 30 healthy volunteers are selected, the age is 18-45 years, the test subjects meet the volunteer selection standard, no history of allergic dermatitis exists, and no irritant product is used in the previous month.
Taking about 0.020-0.025 ml of a test sample, coating the test sample in a small chamber of a spot tester (Beijing science popularization instrument factory), applying a special adhesive tape for external application on the back of a testee, removing the spot tester after 24 hours, slightly wiping off the residual product by using a wet paper towel after the residual product exists, and recording the result according to the skin reaction grading standard in technical Specification for cosmetic safety (2015) at 0.5 hour, 24 hours and 48 hours respectively. The identification criteria are shown in Table 1.

As can be seen from the results of the closed patch test, 30 subjects showed no adverse reaction and no erythema, burning sensation, itching, etc. at 0.5 hour, 24 hours, and 48 hours on the compositions containing the compound extracts of Kochia scoparia and cnidium fruit prepared in examples 1 to 7. The composition of the invention is safe and mild to use and has no stimulation to skin.
And (3) testing 2: efficacy test for inhibition of sebum secretion
(1) Test samples: the compositions of examples 1 to 7 and comparative examples 1 to 4 were added to the following cleanser bases, respectively, in an amount of 2 wt%, to obtain cosmetics containing the compositions of examples 1 to 7 and comparative examples 1 to 4, which were designated as cleansers 1 to 7, comparative cleansers 1 to 4, respectively, and the cleanser bases as controls.
Phase A: 4g of lauric acid, 6g of stearic acid, 5g of palmitic acid, 3g of myristic acid, 2g of ethylene glycol distearate, 2.5g of white beeswax, 62g of ceteareth-62, 1.0g of PEG-150 distearate and a proper amount of preservative;
phase B: 10g of glycerol, 5g of polyethylene glycol-4005 g of butanediol, 3g of sodium laureth sulfate, 0.5g of hydroxyethyl cellulose// sodium acetate// water// isopropanol// cellulose, 0.3g of disodium EDTA and adding water to 100 g;
and C phase: 2.5g of potassium hydroxide and 6g of water;
phase D: 5g of cocoyl sodium glycinate, 3g of sodium lactate and 10g of water;
phase E: 2g of the compositions of examples 1-7 or comparative examples 1-4, 0.3g of polyethylene and a proper amount of essence.
The preparation methods of the facial cleanser 1-7 and the comparative facial cleanser 1-4 are as follows:
firstly, adding the phase A raw material into a stirring pot, stirring and heating to 85 ℃, and uniformly stirring and dispersing for later use;
secondly, adding the phase B raw material into a vacuum emulsifying pot, stirring and heating to 85 ℃, adding the phase C raw material, homogenizing for 3 minutes, and keeping the temperature for 30 minutes until all the materials are dissolved;
slowly filtering the raw materials in the step I, pumping the raw materials into the vacuum emulsifying pot in the step II, keeping the temperature in vacuum for stirring for 1.5 hours after pumping, and cooling after defoaming is completed;
cooling to 60 ℃, adding the D-phase raw material which is mixed and dispersed uniformly in advance, and stirring uniformly in vacuum at a low speed;
and fifthly, cooling to 40 ℃, adding the E-phase raw material, and uniformly stirring.
(2) The test method comprises the following steps: selecting 60 subjects with the ages of 18-45 years and the basic value of the oil content in the forehead test area which is larger than 180 (namely oily or mixed oily skin subjects), randomly dividing the subjects into 12 groups, measuring the oil content of the forehead by using a skin oil test probe sebumeterSM815 before using the product and on the 28 th day of using the product respectively by 5 persons in each group, measuring the oil content of the forehead twice in each area, and averaging; during the test period, the test subjects used the test sample face-cleaning once a day, each group of subjects was prescribed to use 0.6g of sample each time, and the test environment temperature was: 20-22 ℃, humidity is as follows: 40-60%. The results are shown in Table 2.

The experimental results in table 2 show that the reduction of skin oil is significantly higher than that of the comparative facial cleanser after the facial cleanser prepared by the composition of the invention is continuously used for 28 days, which shows that the components of the composition of the invention act synergistically to inhibit the secretion of skin oil more effectively, clear pores and achieve a good oil control effect. The oil control effect of the facial cleanser 6 is superior to that of the facial cleanser 5, and the result shows that when the clerodendranthus spicatus extract and the folium orostachydis sieboldii extract are compounded in a specific ratio, the effect of inhibiting sebum secretion is better, so that the food supply of mites can be greatly reduced, and the propagation speed of the mites is reduced.
And (3) testing: test for bacteriostatic efficacy
Staphylococcus aureus is the most common pathogenic bacterium in human pyogenic infection, and is gram-positive as Staphylococcus epidermidis growing on the surface of organisms. As can be seen from the mechanism of demodex disease development, these two bacteria are present in the skin of skin lesions, with 94.3% staphylococcus epidermidis and 1.1% staphylococcus aureus. The bacteriostatic efficacy of the compositions of the present invention was evaluated in this experiment by determining the inhibitory rate of the compositions against staphylococcus epidermidis and staphylococcus aureus.
(1) Test samples: the face washes 1, 3, 5, 6, 7 in test 2 were used, and the face washes 1, 3, 4 were compared.
(2) The test method comprises the following steps: in order to verify the antibacterial effect of the composition, the antibacterial performance of the composition is determined according to GB15979-2002 hygienic Standard for Disposable sanitary articles appendix C4 'test method for antibacterial performance of dissolution antibacterial (bacteriostatic) product'.
Preparation of bacterial suspension: washing 24h culture of Staphylococcus epidermidis and Staphylococcus aureus with 0.03mol/L phosphate buffer solution to obtain 104~106CFU/mL bacterial suspension;
respectively taking 1g of test sample, adding 0.1mL of the bacterial suspension, uniformly coating and mixing, starting timing, adding 9mL0.03mol/L of phosphate buffer solution after 20min, fully and uniformly mixing, taking 3 dilutions, respectively taking 1mL of the dilutions, placing the dilutions in two parallel plates, adding corresponding culture medium, and culturing for 24 hours at 36 +/-1 ℃ under an anaerobic condition to count bacterial colonies. The above experiment was repeated 3 times, and the average was taken and a blank control group was set.
The calculation formula of the bacteriostatic rate is as follows: and X is (A-B)/A X100%, wherein X is the bacteriostasis rate, A is the average recovered colony of the control sample, and B is the average recovered colony of the test sample.

The experimental results in table 3 show that the facial cleanser containing the composition has strong bacteriostatic effect on staphylococcus epidermidis and staphylococcus aureus, while the bacteriostatic rate of the comparative facial cleanser is lower than that of the facial cleansers in the examples, which shows that the components of the composition cooperate with each other to exert the best bacteriostatic action and prevent the combined infection of mites and bacteria on the surface of the skin.
And (4) testing: mite eradication efficacy testing
(1) Test samples: the sample from test 3 was still used.
(2) The test method comprises the following steps: and performing mite expelling and killing tests according to the industry standard NY/T1151.2-2006. Wherein, the test mite species: dust mites (dermatophagoidesfarina), hermaphrodite and nymphs. The evaluation indexes of the drug effect are shown in a table 4, and the test results of the mite expelling and killing are shown in a table 5.

As can be seen from the experimental results in Table 5, both the mite-expelling rate and the mite-killing rate of the facial cleanser containing the composition can reach B level, and the mite-expelling and mite-killing effects of the facial cleanser containing the composition are obviously superior to those of the facial cleanser in the comparative example. Particularly, when the mixture of the cnidium fruit and the kochia scoparia fruit is compounded in a specific proportion, and under the combined enzymolysis action of the mannose polymerase and the xylanase, the compound extract of the cnidium fruit and the kochia scoparia fruit is cooperated with other components, and the mite expelling and killing performance of the composition is obviously improved.

A composition containing a compound extract of Kochia scoparia and cnidium fruit is characterized by mainly comprising the following components in parts by weight: 5-10 parts of compound extracts of kochia scoparia and cnidium fruit, 6-15 parts of folium ramosiae, 2-9 parts of clerodendranthus spicatus, 3-8 parts of vitex negundo, 30-40 parts of polyhydric alcohol and 34-44 parts of water;
the preparation method of the compound extract of the belvedere fruit and the cnidium fruit comprises the following steps:
(1) pretreatment of raw materials: respectively pulverizing dried Kochia scoparia and cnidium fruit, sieving with 40-70 mesh sieve, and mixing to obtain mixed powder;
(2) enzymatic hydrolysis: adding water into the mixed powder in the step (1) according to the material-liquid ratio of 1:10-1:15/g: mL, adding complex enzyme accounting for 0.2-0.4% of the weight of the mixed powder, stirring for 60-80min at the pH of 4.0-5.0 and the temperature of 45-55 ℃, and heating to boil to inactivate enzyme;
(3) pressure extraction: adding absolute ethanol into the enzymolysis liquid in the step (2) to obtain 50-70% ethanol solution, placing the ethanol solution in a high-temperature high-pressure reaction container, wherein the pressure is 0.4-0.7MPa, the temperature is 70-90 ℃, and the extraction time is 25-35 min;
(4) and (3) purification: filtering the extracting solution obtained in the step (3), concentrating under reduced pressure to 1/6 of the original volume, passing the concentrated solution through an AB-8 macroporous adsorption resin column, eluting with 3 times of column volume of water, eluting with 4 times of column volume of 10% ethanol solution, discarding the eluent, eluting with 3-6 times of column volume of 40-70% ethanol solution at an elution rate of 1.0-2.0BV/h, recovering the eluent, and concentrating under reduced pressure to dryness to obtain the compound extract of kochia fruit and cnidium fruit.
2. The composition containing the compound extract of kochia scoparia and cnidium fruit as claimed in claim 1, wherein the mass ratio of the extract of the leaves of the Amur buster fruit to the extract of the clerodendranthus spicatus is 4: 3.
3. The composition containing the compound extract of kochia scoparia and cnidium fruit as claimed in claim 1, wherein the polyhydric alcohol is one or more of propylene glycol, butylene glycol, 1, 2-pentanediol, glycerol and 1, 2-hexanediol.
4. The composition containing the compound extract of kochia scoparia and cnidium fruit as claimed in claim 1, wherein the mass ratio of kochia scoparia to cnidium fruit in step (1) is 1: 3.

The composition containing the compound extract of kochia scoparia and cnidium fruit as claimed in claim 1, wherein the compound enzyme in step (2) comprises mannosidase with enzyme activity of 30000IU/g and xylanase with enzyme activity of 50000IU/g, and the mass ratio of the mannosidase to the xylanase is 2: 1.
6. Use of the composition comprising the complex extract of kochia scoparia and cnidium fruit as set forth in any one of claims 1 to 5 for the preparation of a daily necessity.
7. The use of the composition containing the compound extract of kochia scoparia and cnidium monnieri fruit as claimed in claim 6 in the preparation of daily necessities, wherein the daily necessities are one of water aqua, lotion, cream, jelly and powder.
